RESUMO

Two new norlignans ( 1 - 2 ), together with two known phenylpropanoids ( 3 - 4 ) were isolated from whole herb of Anemone vitifolia. All compounds were reported from this plant for the first time. The structures of these compounds were identified by comprehensive HR-ESI-MS, 1D and 2D NMR spectroscopic data analysis and comparison with literature data. Additionally, bioactivity study results showed two new compounds have potential anti-inflammatory activity. The plausible biosynthetic pathway for these compounds were also speculated in the paper.

Activation of inflammatory cells and upregulations of a number of cytokines in the central nervous system (CNS) of patients with prion diseases are frequently observed. To evaluate the potential changes of some brain cytokines that were rarely addressed during prion infection, the levels of 17 different cytokines in the brain homogenates of mice infected with different scrapie mouse-adapted agents were firstly screened with Luminex assay. Significant upregulations of interferon gamma-induced protein 10 (IP10), keratinocyte chemoattractant (KC) and macrophage colony stimulating factor (M-CSF) were frequently detected in the brain lysates of many strains of scrapie infected mice. The upregulations of those three cytokines in the brains of scrapie infected mice were further validated by the individual specific ELISA and immunohistochemical assay. Increased specific mRNAs of IP10, M-CSF and KC in the brains of scrapie infected mice were also detected by the individual specific qRT-PCRs and IP10-specific digital PCR. Dynamic analyses of the brain samples collected at different time points post infection revealed the time-dependent increases of those three cytokines, particularly IP10 during the incubation period of scrapie infection. In addition, we also found that the levels of IP10 in cerebral spinal fluid (CSF) of 45 sporadic Creutzfeldt-Jakob disease (sCJD) patients were slightly but significantly higher than those of the cases who were excluded the diagnosis of prion diseases. These data give us a better understanding of inflammatory reaction during prion infection and progression of prion disease.

Although perovskite light-emitting diodes (PeLEDs) are promising for next-generation displays and lighting, their efficiency is still considerably below that of conventional inorganic and organic counterparts. Significant efforts in various aspects of the electroluminescence process are required to achieve high-performance PeLEDs. Here, we present an improved flexible PeLED structure based on the rational interface engineering for energy-efficient photon generation and enhanced light outcoupling. The interface-stimulated crystallization and defect passivation of the perovskite emitter are synergistically realized by tuning the underlying interlayer, leading to the suppression of trap-mediated non-radiative recombination losses. Besides approaching highly emissive perovskite layer, the outcoupling of trapped light is also enhanced by combining the silver nanowires-based electrode with quasi-random nanopatterns on flexible plastic substrate. Upon the collective optimization of the device structure, a record external quantum efficiency of 24.5% is achieved for flexible PeLEDs based on green-emitting CsPbBr3 perovskite.

BACKGROUND: Insulinoma is a subtype of pancreatic neuroendocrine tumors. Many patients with insulinoma are obese due to frequent food intake. Ghrelin is associated with obesity and blood levels of insulin. It is not clear if plasma levels of ghrelin in insulinoma patients correlate with hyperinsulinemia and obesity. Expression of ghrelin and its receptor has not been well demonstrated in insulinoma. OBJECTIVE: To study if plasma levels of ghrelin is associated with obesity and hyperinsulinemia or hyperproinsulinemia in patients with insulinoma, and to detect the expression of ghrelin and its receptor in insulinoma. METHODS: Plasma levels of acylated ghrelin, insulin, and proinsulin were measured in 37 patients with insulinoma and 25 controls by ELISA. Expression of ghrelin and its receptor GHS-R1A was examined in 20 insulinoma and paired pancreatic specimens by immunostaining. P ≤ 0.05 was considered significant. RESULTS: The plasma levels of acylated ghrelin in patients with insulinoma were significantly lower than that in the controls (median 15 pg/ml vs. 19 pg/ml, respectively, P = 0.016). The reduced plasma levels of acylated ghrelin in patients were significantly correlated with obesity, hyperinsulinemia, and hyperproinsulinemia (P = 0.029 and P = 0.028, respectively). Expression of ghrelin and its receptor GHS-R1A was shown in the majority of insulinoma specimens. The expression of GHS-R1A was positively correlated with ghrelin expression in insulinoma (P = 0.014). CONCLUSIONS: Plasma levels of acylated ghrelin decreased in patients with insulinoma, probably due to the hyperinsulinemia and obesity in the patients. Expression of both ghrelin and its receptor is common in insulinoma.

Antibiotic residues in the environment pose a great risk to global public health. They increase antibiotic resistance by enhancing plasmid conjugation among bacteria or mutations within bacterial genomes. However, little is known about whether the putative environmental levels of antibiotics are sufficient to influence plasmid-mediated transformability. In this study, we explored the effect of eight kinds of representative antibiotics and several other compounds on the plasmid transformability of competent Escherichia coli. Only levofloxacin (LEV) at the putative environmental levels was found to facilitate the frequency of PBR322-or RP4-plasmid-mediated transformation by up to 5.3-fold. Additionally, PBR322 transformation frequency could be further enhanced by copper ion or ammonia nitrogen but inhibited by humic acid. However, when competent E. coli was exposed to the minimal inhibitory concentrations (MIC) of the antibiotics, an enhanced plasmid-assimilation ability was observed and plasmid transformation frequency was increased by up to 98.6-fold for all the tested antibiotics. Furthermore, E. coli exhibited a preference for the uptake of plasmids harbouring the resistance genes to the antibiotics it had been exposed to. Among these antibiotics, cephalexin, tetracycline, and kanamycin induced the highest uptake of RP4. The putative environmental levels of LEV enhanced plasmid transformability regardless of the presence of corresponding antibiotic resistance gene (ARG) on the genetic elements, suggesting environmental LEV residues may facilitate dissemination of antibiotic resistance by any plasmid-mediated transformability, thereby posing a great risk to health.

With data for distributions and diversities of macroinvertebrates from 12 main subtidal zones in the bays of Fujian on August 2010, we investigated species composition, biodiversity and community structure, as well as their relationships with environmental factors. The results showed that 382 macroinvertebrates were recorded, which included 170 annelids, 75 crustaceans, 78 mollusks, 19 echinodermata, and 40 other species. The species richness, individual abundance, biomass and biodiversity indices showed significantly spatial variability. The average of species richness was (55±21), with the highest (92) in Luoyuan Bay and the lowest species (25) in Jiuzhen Bay. Polychaetes was dominant species in each bay, with an average percentage of (51.8±5.5)%. Zhaoan Bay had the highest average individual abundance with (1330±1094) ind·m-2, followed by Fuqing Bay, Xiamen Bay and Meizhou Bay, while Quanzhou bay had the lowest. Xinghua Bay had the highest average biomass with (821.2±2387.7) g·m-2, followed by Fuqing Bay and Xiamen Bay. Both richness index and Shannon diversity were much higher in Luoyuan Bay, Meizhou Bay, Shenhu Bay, Xiamen Bay and Dongshan Bay, and their values were low in Fuqing Bay and Jiuzhen Bay. The mean value for d and H across all bays was (0.80±0.09) and (2.73±0.64), respectively. The macroinvertebrates of the study area were divided into 14 groups using a criterion of 20% similarity. The stations in Luoyuan Bay had the highest similarity and the lowest spatial variability, and followed by Sansha Bay, Fuqing Bay, Shenhu Bay and Zhaoan Bay. The higher variability of community composition was found among the stations for the rest bays. On the basis of Spearman correlations among species richness, biodiversity indices and environmental factors, as well as PCA analy-sis, Cu in the sediment, water depth, and dissolved inorganic phosphorus in bottom water were the main factors driving the spatial variability of composition and distribution of macroinvertebrates in the study area.

As a folk medicine, Phlomis likiangensis is traditionally used in China to activate collaterals and protect cardiovascular system. We hypothesized that the beneficial effects of Phlomis likiangensis may be related to vasodilatation. In the present study, twelve known iridoid glucosides (1-12) were isolated from Phlomis likiangensis. The vasodilatory effects and the underlying mechanisms of the main components (iridoid glucosides) of Phlomis likiangensis on rat aortic rings were investigated. The result showed that iridoid glucosides significantly increased the vasodilatation in rat aortic rings, which was abolished by removing the endothelium of the vessels or by eliminating the generation of nitric oxide. Finally, the structure-activity relationship of compounds 1-12 was also speculated. Our findings provide the first evidence that the iridoid glucosides of Phlomis likiangensis may be the pharmacodynamic basis for its traditional efficacy.

Paraquat (PQ) promotes cell senescence in brain tissue, which contributes to Parkinson's disease. Furthermore, PQ induces heart failure and oxidative damage, but it remains unknown whether and how PQ induces cardiac aging. Here, we demonstrate that PQ induces phenotypes associated with senescence of cardiomyocyte cell lines and results in cardiac aging-associated phenotypes including cardiac remodeling and dysfunction in vivo. Moreover, PQ inhibits the activation of Forkhead box O3 (FoxO3), an important longevity factor, both in vitro and in vivo. We found that PQ-induced senescence phenotypes, including proliferation inhibition, apoptosis, senescence-associated ß-galactosidase activity, and p16INK4a expression, were significantly enhanced by FoxO3 deficiency in cardiomyocytes. Notably, PQ-induced cardiac remolding, apoptosis, oxidative damage, and p16INK4a expression in hearts were exacerbated by FoxO3 deficiency. In addition, both in vitro deficiency and in vivo deficiency of FoxO3 greatly suppressed the activation of antioxidant enzymes including catalase (CAT) and superoxide dismutase 2 (SOD2) in the presence of PQ, which was accompanied by attenuation in cardiac function. The direct in vivo binding of FoxO3 to the promoters of the Cat and Sod2 genes in the heart was verified by chromatin immunoprecipitation (ChIP). Functionally, overexpression of Cat or Sod2 alleviated the PQ-induced senescence phenotypes in FoxO3-deficient cardiomyocyte cell lines. Overexpression of FoxO3 and CAT in hearts greatly suppressed the PQ-induced heart injury and phenotypes associated with aging. Collectively, these results suggest that FoxO3 protects the heart against an aging-associated decline in cardiac function in mice exposed to PQ, at least in part by upregulating the expression of antioxidant enzymes and suppressing oxidative stress.

Nematicidal activity at different concentrations of fosthiazate against soybean cyst nematode (Heterodera glycines) was evaluated in this paper. The mortality rates of second-stage juvenile (J2) reached 13.43, 48.39, 66.82, 79.77, and 86.35% at 12 hr after exposure to 2.18, 3.44, 5.45, 8.61, and 13.62 mg/l of fosthiazate, respectively, whereas cumulative hatching rates totaled 58.24, 53.88, 42.54, 24.11, and 13.69% at 18 days after exposure to concentrations. J2s dead by exposure to fosthiazate exhibited shrunk and twisted body shape, whose length of nematode body, stylet, and esophageal glands to head were significantly shorter than that of the control (p < 0.05). A pot test was also performed to count the numbers of cysts on soybean roots, showing reduction of 43.64-97.94% due to application of fosthiazate at 5.45, 13.62, 34.04, and 85.10 mg/l concentrations. Fos-like antigens (Fosl) including Fosl1 and Fosl2 exclusively heterodimerize with Jun members to form AP-1 complex, thereby participating in various cellular progresses including cell cycle regulation. However, expression patterns of these two genes during embryonic development remains largely unknown. In the present study, both temporal and spatial expression patterns of fosl1 and fosl2 were examined during embryonic development of Xenopus tropicalis. Real-time quantitative PCR results showed that the expression of the two genes was increased from stage 2 to stage 42. However, expression level of fosl1 is much higher than that of fosl2 at stage 42. Whole-mount in situ hybridization showed that fosl1 was expressed in eyes, branchial arch, notochord, otic vesicle, and liver. However, fosl2 was expressed in lung primordium from stage 34 to stage 38, in addition to the moderate expression in eyes and branchial arch at stage 42. Thus, the developmental expression patterns of these two fosl genes is different in Xenopus embryos. These results provide a basis for further functional study of these two genes.

Three Legionella-like strains, designed km488T, km489 and km521, were isolated from freshwater samples in China. Cells were Gram-stain-negative, rod-shaped and non-spore-forming. Growth was observed on BCYEα agar, but not on BCYEα agar without l-cysteine, chocolate agar with PolyViteX or Columbia blood agar. The major fatty acids (>5 %) of strains km488T, km489 and km521 were C16 : 0, anteiso-C15 : 0, iso-C16 : 0 and anteiso-C17 : 0. The mip gene sequences (574 nt) showed the isolates were almost identical with more than 99.7 % sequence similarities, and closely matched to L. gormanii ATCC 33297T with 95.4-95.6 % sequence similarities. Phylogenetic analyses based on concatenated gene (16S rRNA, mip, rpoB and rnpB) sequences indicated that the isolates formed a distinct cluster along with L. gormanii within the genus Legionella. Matrix-assisted laser desorption ionization time-of-flight analyses also demonstrated a clear separation between the isolates and other closely and distantly related Legionella species. DNA-DNA hybridization studies demonstrated that the isolates were closely related (92.0 -95.0 % DNA-DNA relatedness) but differentiated from their phylogenetic neighbours (<70 % DNA-DNA relatedness). The whole genome of km488T was sequenced, and showed a G+C content of 37.8 mol%. Based on the findings from this polyphasic taxonomic study, the isolates are considered to represent a single novel species, for which the name Legionella qingyii sp. nov. is proposed. The type strain is km488T (KCTC 15636T=CCTCC AB 2018025T=NRBC 113223T).

Paraffin wax (PW) is widely used as a phase change material (PCM) in the thermal energy storage field, whereas the leakage and strong rigidity of PW have hindered its practical applications. In this work, binary melamine foam (MF)/PW blends with simultaneous thermal energy storage and shape memory properties were prepared through vacuum impregnation. Herein, PW performs as a latent heat storage material and as a switching phase for shape fixation and MF serves as a supporting material to prevent the leakage and as a permanent phase for shape recovery. Due to the light weight and super-elasticity of MF, the MF/PW PCMs possess not only good encapsulation ability and a high latent heat, but also excellent shape-fixing and recovery properties (shape-fixing and recovery ratios are about 100%). Besides, the MF/PW PCMs can be fabricated into arbitrary shapes using MF as a template, and they exhibit excellent shape memory cyclic performance and thermal reliability. A temperature-sensitive and temperature-controlled deployable panel is further established, which can be installed in the electronic device and used for temperature protection. With high thermal energy storage capability, excellent shape memory properties, shape designability, and stable cycling reliability, this multifunctional MF/PW PCM shows a promising application in thermal energy management systems.

RESUMO

As a high-level cognitive function of actively regulating human behaviors, cognitive control plays essential roles in conflict processing, working memory, decision making and so on. However, it is still under debate whether a universal cognitive control mechanism underlies the processing of various conflicts. Many existing theories tend to hold that cognitive control is domain-general; however, this view has been challenged by recent empirical studies. The logics of studying generality/specificity mainly include transferability, parallel comparison, correlation and resources competition, etc. Current empirical findings support that cognitive control is domain-general, domain-specific or both, respectively. To tackle this controversy, future studies about cognitive control can be performed from the perspectives of life-span development, the dynamic brain network, combination of multiple logics, causal relationship from brain injury, computational modeling, cognitive flexibility and functional connectivity.

Three new phenylacetamide glycosides (1-3) together with one known phenylacetamide glycoside (4) and two known flavonoid glycosides (5-6) were isolated from whole plants of Dracocephalum tanguticum. The structure of all compounds were elucidated based on spectroscopic data analysis and comparison with data reported in related literature. Compounds (1-3) were evaluated for their anti-hyperglycemic and anti-fungal (Candida albicans) activities, the results revealed that all of them showed moderate activity with 3T3-L1 adipocytes glucose consumption rate of 20.80 ± 1.47%, 21.48 ± 2.44%, and 21.57 ± 1.35%, respectively at the final concentration of 25 µM. However, none of them showed obvious Candida albicans inhibitory activity.

Two new xanthone glycosides (1-2), together with seven known analogues (3-9), were isolated from whole herb of Swertia punicea. The structures of these metabolites were established on the basis of detailed spectroscopic analysis and comparison with data reported in the literature. In an in vitro test, all isolates were evaluated for their anti-inflammatory activity. The results revealed that all of them showed significant anti-inflammatory activity with IC50 values ranging from 1.237 to 3.319 mM. Compounds 3, 4, and 5 (IC50 values in the range 1.237-1.987 mM) displayed more potent anti-inflammatory activity than the positive control, indomethacin (IC50 value of 2.004 mM).

OBJECTIVE: This study aimed to determine the effects of diet-induced paternal obesity on cognitive function in mice offspring. METHODS: Male mice (F0) were randomized to receive either a control diet (10 kcal% fat) or a high-fat diet (HFD; 60 kcal% fat) for 10 weeks before being mated with normal females to generate F1 offspring. Male F1 offspring were mated with normal females to generate F2 offspring. Behavioral tests were used to assess cognitive functions in F1 and F2 offspring. Reduced representation bisulfite sequencing was used to the explore mechanisms of epigenetic inheritance. RESULTS: HFD-induced paternal obesity resulted in cognitive impairments in F1 offspring, potentially due, at least in part, to increased methylation of the BDNF gene promoter, which was inherited from F0 spermatozoa. BDNF/tyrosine receptor kinase B signaling was associated with cognitive impairments in HFD-fed F1 offspring. However, there were no significant changes in F2 offspring. CONCLUSIONS: The findings provide evidence of intergenerational effects of paternal obesity on cognitive function in offspring occurring via epigenetic spermatozoan modifications.

BACKGROUND: In Traditional Chinese Medicine (TCM) theories, the typical clinical manifestations of gout are attributed to the "dampness-heat pouring downward." Therefore, TCM practitioners always consider prescribing the formulae which are believed to clear heat and drain dampness for the management of gout. This clinical trial aims: (1) to determine the hypouricemic effect of "Yellow-dragon Wonderful-seed Formula" (YWF) decoction in gout patients with dampness-heat pouring downward pattern and (2) to determine if gypsum could provide additional significant benefits to YWF. METHODS: A total of 72 hyperuricemic individuals with gout and dampness-heat pouring downward pattern were included with 62 of them completing the trial. Participants were randomly assigned to the YWF group, the YWF + gypsum group, or the allopurinol group. YWF and YWF + gypsum decoctions were orally administered for four weeks. Allopurinol was also orally administered for four weeks as the active control. Serum uric acid (sUA) level was the primary outcome measure. Urine urate level, scores on the SF-36 scale, erythrocyte sedimentation rate (ESR), X ray film, and C reactive protein (CRP) level were the secondary outcome measures. RESULTS: Compared with the values at week 0, YWF and YWF + gypsum did not significantly decrease the sUA level at each weekend reading. YWF, YWF + gypsum, and allopurinol decreased the urine urate levels and there were significant differences between the YWF group and the YWF + gypsum group. All the changes in the eight structures of SF-36 during the intervention period were not significantly different among the three groups and there was no significant difference in the CRP level among the three groups at each weekend reading. CONCLUSIONS: YWM, which modified on the basis of Two Wonderful Herbs Powder (2WHP), does not show significant hypouricemic effect. There is a possibility that Gypsum Fibrosum may provide additional effects to YWF in decreasing the urine urate levels but cannot add benefits to YWF in other outcome measures. The North Yellow Sea is a major aquaculture production area for the scallop Patinopecten yessoensis. In this study, the temporal and spatial variation of phycotoxins in scallops, phytoplankton, and their cysts were analyzed during a survey conducted from June 2011 to April 2012 around Zhangzi Island. The study area is a semi-enclosed epicontinental sea surrounded by the Shandong Peninsula, the Liaodong Peninsula and the Korean Peninsula. The three main results of the study were as follows: (1) The saxitoxin-group toxins, okadaic acid and analogues, and pectenotoxins were the major phycotoxin residues found in scallops; (2) Six kinds of toxic microalgae were identified, Protoperidinium spp., Gonyaulax spp., and Alexandrium spp. were the dominant taxa; Seven types of potential marine toxin-producing dinoflagellates, A. tamarense, A. catenella, Dinophysis fortii, G. catenatum, Gambierdiscus toxicus, Azadinium poporum, and Pseudo-nitzschia pungen were identified as the primary source of phycotoxins and were present at relatively high density from June to October; and (3) azaspiracids and domoic acid might be new potential sources of toxin pollution. This study represents the first assessment to phycotoxins around Zhangzi Island in the North Yellow Sea.

Three benthic biotic indices, AZTI marine biotic index (AMBI), benthic opportunistic polychaetes amphipods (BOPA), and benthic polychaetes amphipods (BPA), combined with Shannon diversity index (H) were applied in ecological quality status (EQS) assessment, to investigate their suitability at four bays and an estuary in Fujian Province. The results showed that there were substantial differences in the performance of these indices. There were only four sites with the same assessment grades using different indices, accounting for 8.7% of the applied sites. AMBI classified 76.1% of the sites as "Good", while 89.1% of the sites were classified as "High" using BOPA. The assessment results of BPA and H showed obvious gradient changes. For the bay areas, all the BOPA, BPA and AMBI values had no significant correlation with dissolved inorganic phosphate (DIP) and dissolved inorganic nitrogen (DIN) concentrations, suggesting that these indices did not respond to eutrophication pressure. The H value was significantly and negatively correlated with DIN concentration. For the estuarine area, the three indices, i.e . BOPA, BPA and AMBI, had significantly positive correlations with DIN and DIP, and there was no clear spatial variation in the assessment grades of these indices with the distance toward sea. The results suggested that BOPA, BPA and AMBI would overestimate the EQS and would show no response to the eutrophication pressure in estuarine area. Similar to the result from the bay areas,H value in the estuary area was significantly and negatively correlated to DIN. Meanwhile, the assessment grades of the seven sections in the estuary tended to increase with the distance toward the sea. In summary, our results suggested that BOPA, BPA and AMBI would not be suitable for the EQS assessment for Fujian coastal area, while H would be more suitable as it could respond to the main anthropogenic pressures.

An asymmetric trapping field was generated from an asymmetric half-round rod electrode linear ion trap (A-HreLIT), and its performance of unidirectional ion ejection was studied. Two different asymmetric structures of A-HreLITs were constructed, one rotating y electrode pairs toward an x electrode with an angle θ, and the other stretching one x electrode with a distance α. The center of trapping field was displaced away from the geometrical center of the ion trap, defined to be the midpoint along the axis of y between x electrodes, which leads to unidirectional ion ejection through one x electrode. Computer simulations were used to investigate the relationship between asymmetric geometric parameter of θ (or α) and analytical performance. Both structures could result in similar asymmetric trapping fields, which mainly composed of dipole, quadrupole, and hexapole fields. The dipole and hexapole fields were approximately proportional to the asymmetric geometric parameter of rotation angle θ (or stretch distance α). In simulation, ion trajectories and ion kinetic energy were calculated. For ions with m/z 609 Th, the simulation results showed that mass resolution of over 2400 (FWHM) and ion unidirectional ejection efficiency of nearly 90% were achieved in an optimized A-HreLIT. Ion detection efficiency of A-HreLIT could be improved significantly with only one ion detector, while maintaining a considerable mass resolution. Furthermore, the A-HreLIT could be driven by a traditional balanced RF power supply. These advantages make A-HreLIT suitable for developing miniaturized mass spectrometer with high performance.
