RESUMO

Deoxynivalenol (DON), as a widespread Fusarium mycotoxin in cereals, food products, and animal feed, is detrimental to both human and animal health. The liver is not only the primary organ responsible for DON metabolism but also the principal organ affected by DON toxicity. Taurine is well known to display various physiological and pharmacological functions due to its antioxidant and anti-inflammatory properties. However, the information regarding taurine supplementation counteracting DON-induced liver injury in piglets is still unclear. In our work, twenty-four weaned piglets were subjected to four groups for a 24-day period, including the BD group (a basal diet), the DON group (3 mg/kg DON-contaminated diet), the DON+LT group (3 mg/kg DON-contaminated diet + 0.3% taurine), and the DON+HT group (3 mg/kg DON-contaminated diet + 0.6% taurine). Our findings indicated that taurine supplementation improved growth performance and alleviated DON-induced liver injury, as evidenced by the reduced pathological and serum biochemical changes (ALT, AST, ALP, and LDH), especially in the group with the 0.3% taurine. Taurine could counteract hepatic oxidative stress in piglets exposed to DON, as it reduced ROS, 8-OHdG, and MDA concentrations and improved the activity of antioxidant enzymes. Concurrently, taurine was observed to upregulate the expression of key factors involved in mitochondrial function and the Nrf2 signaling pathway. Furthermore, taurine treatment effectively attenuated DON-induced hepatocyte apoptosis, as verified through the decreased proportion of TUNEL-positive cells and regulation of the mitochondria-mediated apoptosis pathway. Finally, the administration of taurine was able to reduce liver inflammation due to DON, by inactivating the NF-κB signaling pathway and declining the production of pro-inflammatory cytokines. In summary, our results implied that taurine effectively improved DON-induced liver injury. The underlying mechanism should be that taurine restored mitochondrial normal function and antagonized oxidative stress, thereby reducing apoptosis and inflammatory responses in the liver of weaned piglets.

Sarcopenia is a geriatric disease characterized by reduced muscle function and mass. The capacity to self-renew and myogenesis of satellite cells (SCs) declines with age, resulting in age-related sarcopenia. MicroRNAs (miRNAs) can regulate the proliferation and myogenesis of SCs. In this study, we identified that miR-467a-3p and miR-874-5p could respectively mediate the stemness and myogenesis of SCs by performing bioinformatics analysis. AntagomiR-467a-3p (ant-467a) and antagomiR-874-5p (ant-874) improved the stemness and myogenesis of SCs, respectively. SC-targeting extracellular vesicles (EVs) were constructed by overexpressing TSG101 on the surface of EVs isolated from bone marrow mesenchymal stem cells (BMSCs). Ant-467a loaded EVs (EVs-467a) and ant-874 loaded EVs (EVs-874) were prepared by transferring ant-467a and ant-874 into SC-targeting EVs. EVs-467a and EVs-874 were more effective than ant-467a and ant-874 in promoting the stemness and myogenesis of SCs. Sequentially intermuscular injection of EVs-467a and EVs-874 significantly improved sarcopenia in ovariectomy (OVX) mice. The effects of multiple injections of EVs-467a and EVs-874 in the treatment of sarcopenia could be achieved by using a hierarchically injectable hydrogel to sustainedly release EVs-467a and EVs-874 in vivo. Our findings provide an EV-based SC-targeting antagomiRNAs controlled release strategy as a novel therapy against sarcopenia. This article is protected by copyright. All rights reserved.

Osteoporotic fracture is a major health problem plaguing the ageing society, and improving its treatment is an urgent challenge. How to ameliorate bone loss determines the recovery of such fractures. Extracellular vesicle (EV)-loaded hydrogel has the capacity to treat osteoporotic fractures due to its pro-osteogenic property. And balancing proliferation and maturation of osteoblast precursors (OBPs) is of great significance to avoid OBP depletion, which is lacking in current treatment. Based on osteoblastogenic miRNAs, this study aimed to explore the efficacies of the combination of hierarchical hydrogel and EVs altering functional miRNAs level in bone loss. Through bioinformatics analyses, we screened out proliferative gene-targeting miR-200b-3p and osteogenic gene-targeting miR-130b-3p. And antagomiR-200b-3p (ant-200b) enhanced OBP proliferation, and antagomiR-130b-3p (ant-130b) promoted OBP differentiation. After confirming the directional effect of Fibronectin (Fn1) on OBPs, we prepared OBP-targeting EVs. Furthermore, encapsulation of two antagomiRNAs in EVs enhanced the respective effect of ant-200b and ant-130b. Notably, hierarchically injectable hydrogel exerted an effective function in promoting the sequential delivery of EVs-200b and EVs-130b. Importantly, hierarchical hydrogel containing dual EVs effectively ameliorated bone loss. Overall, hierarchical hydrogel based on two antagomiRNAs effectively improves bone loss in vivo due to its role in promoting OBP proliferation and maturation sequentially.

A hollow microtubular covalent organic framework (denoted as TatDha-COF) was synthesized by solvothermal method for the enrichment and determination of quinones. The TatDha-COF showed large specific surface area (2057 m2 g-1), good crystal structure, ordered pore size distribution (2.3 nm), stable chemical properties and good reusability. Accordingly, a simple and efficient method based on dispersive solid-phase extraction (d-SPE) and atmospheric pressure gas chromatography tandem mass spectrometry (APGC-MS/MS) was developed for the determination of quinones in complex samples. The established method demonstrated a wide liner range, good linearity (r>0.9990), high enrichment factors (EFs, 24-69-folds) and low detection limits (LODs, 0.200-30.0 pg L-1, S/N≥3). On this basis, the suggested method was successfully applied to sensitively detect the eight ultratrace quinones in mice plasma. Overall, the established method has provided a powerful tool for the enrichment and detection of ultratrace quinones in complex samples, presenting the promising application of TatDha-COF in sample pretreatment.

Preservative is of importance to retard fruit deterioration and prolong the shelf-life. The suitability of using water-soluble polysaccharide extracted from waste macroalgae Enteromorpha prolifera (EPP) for cherry tomato preservation was evaluated. As compared with the control, the EPP-treated cherry tomatoes exhibited better fruit appearance, lower disease index and rot index during storage. Around 47 % EPP-treated cherry tomatoes were commercially acceptable after 36 days, which was however only 15.6 % for untreated cherry tomatoes, indicating the satisfactory preservation effectiveness of EPP-rich solution for cherry tomatoes. The post-extraction residue was commonly underutilized, we herein attempted to employ an emerging thermochemical conversion technique, hydrothermal liquefaction, to produce crude bio-oil (biocrude) from post-extraction E. prolifera. A biocrude yield of ∼23 wt% (dry-ash-free, daf) was obtained, and fatty acids and phenolics were identified to be the two main components in biocrude. The biocrude contained ∼70 % carbon and the higher heating value was ∼30 MJ/kg.

RATIONALE: Homocysteine (hcy) is a metabolite in the human body and an important determinant of cardiovascular health. To assist in the assessment of human cardiovascular safety, a rapid and accurate quantitative analysis method must be developed. Laser desorption/ionization mass spectrometry (LDI-MS) has been widely used in biological, chemical, and medical analyses due to its excellent characteristics of high throughput, low sample consumption, and high speed. Here, an LDI-MS method based on covalent organic framework (COF) film was developed for the rapid and sensitive determination of hcy in human serum using an isotope-labeled internal standard. METHODS: We tried to cultivate COFs on indium-tin oxide substrate at room temperature to form a thin film, which was used in LDI-MS. Compared with the traditional organic matrices, the COF film had a clean background and a high signal response for the detection of hcy. In addition, using COF film as the substrate, with a high signal intensity and a clean background, can be helpful to analyze a series of small molecules such as amino acids, bisphenols (Bps), estrogens, and drugs. We evaluated the limit of detection (LOD) and the reproducibility of this method. Finally, the calibration curve was constructed for the quantification of serum hcy using an isotope-labeled internal standard and was applied for the rapid determination of hcy in clinical human serum. RESULTS: COF film-assisted LDI-MS had a higher response signal and a cleaner background compared to four conventional organic matrices. The LOD of the method for hcy was 0.5 µmol/L, equivalent to 500 fmol. This method exhibited excellent performance for small-molecule compounds, including amino acids, Bps, drugs, and estrogens. The reproducibilities of this method for shot-to-shot and dot-to-dot were determined to be good. This method was applied for the rapid analysis of hcy in clinical human serum, showing good correlation with those obtained by hospital testing. CONCLUSION: The COF film-based LDI-MS method had simple sample preparation, short analysis time, clean background, and good reproducibility for hcy analysis. As an important indicator of human health, the detection of serum hcy is of great significance to human health.

The development of novel stationary phases with specific functionality is of great importance in chromatographic separation. Herein, we fabricated fluoro-functionalized spherical covalent organic frameworks (SF-COFs) via a bottom-up strategy as stationary phases for high-performance liquid chromatography (HPLC). Benefiting from the significant monodispersity, narrow size distribution, and high fluorine content, the SF-COFs packed column showed high column efficiency and excellent resolution for the separation of the organic fluorides involving polyfluorobenzenes, polychlorobenzenes, polybromobenzenes, perfluoroalkyl methacrylates, and halogenated trifluorotoluenes, which cannot be separated on the fluorine-free spherical covalent organic frameworks packed column. Especially, the column efficiency of 20 100-38 500 plates/m was obtained for polyfluorobenzenes, and the relative standard deviations of the retention time for continuous 10 separations of polychlorobenzenes and polybromobenzenes were less than 0.98%. Furthermore, the prepared SF-COFs packed column showed overwhelming superiority in the separation of organic halides compared with commercial C18 and pentafluorophenyl (PFP) packed columns. In addition, the compounds with different hydrophobicity or aromatic ring structure were also successfully separated on the SF-COFs packed column. This work extended the application of spherical COFs and provided a new way to introduce specific functional groups into the COF-based stationary phase for HPLC.

The Matching Familiar Figures Test (MFFT) is a well-known and extensively used behavioral measure of reflection-impulsivity. However, the instrument has several deficiencies, including images designed for school-age children in the United States during the 1960s. Most importantly, an adult version of the instrument is currently unavailable and the lack of a single repository for the images raises questions regarding the MFFT's validity and reliability. We developed a 21st century version of the MFFT using images that are familiar to adults and reside in a freely accessible repository. We conducted two studies examining validity and reliability issues. In Study 1, participants interacting with the MFFT-2021, versus those interacting with the original MFFT20, spent more time on the task, took more time in making their first response, and were more likely to complete the task without errors, even though the average number of errors was higher than the comparison group. The coherence of these results is evidence of convergent validity. Regarding predictive validity, the MFFT-2021 remained a reliable predictor of rational thinking, such that participants who demonstrated more reflection (less impulsivity) tended to avoid rational thinking errors. Also, performance on the MFFT-2021 predicted higher quality judgments in processing job characteristic cues with embedded interactions, a form of configural information processing. We also found evidence of concurrent validity: performance on the MFFT-2021 differed in a predictable manner for participants grouped by their performance on the Cognitive Reflection Test. In Study 2, we tested discriminant validity by comparing participant performance on the MFFT-2021 to their performance on the Information Sampling Task (IST), another behavioral measure of reflection-impulsivity used in studies of psychopharmacological and addiction behaviors. For our participants (undergraduate business students), we found that the MFFT was a stronger predictor of performance on rational thinking tasks, and, contrary to prior studies, our exploratory factor analysis identified separate factors for the MFFT-2021 and the IST, supporting discriminant validity, indicating that these two instruments measure different subtypes of reflection-impulsivity.

Complex transformations of heavy metals in the mega-river-estuary continuum limit our understanding of their pollution history. This study investigated sedimentary compositions of heavy metals, major elements, total organic carbon, grain size, and radionuclides to study spatiotemporal variations in heavy metal accumulation patterns and their controlling mechanisms in four sediment cores (E1-E4) from the Yangtze River Estuary (YRE). Results show that only E3 in the distal YRE front mirrors well the heavy metal pollution history due to its continuous deposition in a stable sedimentary environment, while E1 and E2 record the influence of riverine and estuarine projects and processes apparently. E1 in the proximal YRE front registers intense human disturbance through sediment dredging and dumping activities to produce a thick layer of abnormal low 210Pbex and minor heavy metal concentrations. E2 in the intermediate YRE front demonstrates the recently increasing influence of reduced sediment discharge by its upcore coarsening trend with decreased heavy metal concentrations. Flood and storm events left different imprints in core sediments of E2 and E3 by their coarse stratal units with fewer heavy metal concentrations. The source analysis indicates that heavy metals in estuarine sediments mainly come from natural processes but are significantly affected by human activities. A direct linkage of the heavy metal accumulation history with the socioeconomic development in recent decades is found by a comparison study of core data from the tidal river to the estuary, albeit with a remarkable spatiotemporal difference, which is jointly determined by grain size, offshore distance, hydrodynamic condition, depositional status, and sedimentation rate besides estuarine processes. This warns us to carefully interpret the heavy metal history from single or sparse core data in a mega estuary system with intense natural forces and human disturbances analogous to the YRE.

To explore the effect of MYOZ1 in the muscle growth and development of meat ducks, MYOZ1 single-nucleotide polymorphism loci were screened at the DNA level in the meat duck population with highest and lowest feed conversion rates. The expression of MYOZ1 was detected using reverse-transcription quantitative polymerase chain reaction. The protein expression of MYOZ1 was detected using Western blotting at the protein level. The results showed that there was a base mutation site at 30 bp and 158 bp in the fourth exon of MYOZ1, which was mutated from C to T (exon4 C30T) and from G to A (exon4 G158A), respectively. The allele frequency of the locus was significantly different between the high and low feed conversion rate groups (p < 0.01). The relative expression of MYOZ1 mRNA in breast muscle tissue of HF ducks was significantly higher than that of LF ducks (p < 0.01). The MYOZ1 protein expression of HF ducks was significantly higher than that of LF ducks (p < 0.01). In general, MYOZ1 has a positive regulatory effect on the muscle growth and development of meat ducks. The results of this study lay a certain theoretical basis for the muscle growth and development of meat ducks.

Objective: To evaluate the effects of antiretrovirals on cardiovascular function and some biochemical indexes in gestational female rats. Methods: Nineteen 9-week-old female and six 10-week-old male SD rats were divided into normal control group (CON) and highly active antiretroviral therapy group (HARRT), 9/10 female rats and 3 male rats were combined into one cage, totally 2 cages. Female rats in CON group were intragastrically given with normal saline (NS, 10 ml/kg) every morning and evening, while female rats in HARRT group were treated with equal volume antiretrovirals (AZT 31.25 mg/kg + 3TC 15.63 mg/kg + LPV/r (41.67/10.42) mg/kg) for 3 months. The body weight and survival rate of female rats were recorded. Echocardiography and multichannel physiological recorder were used to detect arterial blood pressure and cardiac hemodynamic parameters. The levels of blood glucose, blood lipids, myocardial enzymes and liver enzymes were detected by corresponding kits. Myocardial collagen fibers were observed by Masson staining and the ultrastructure of myocardial cells were observed by transmission electron microscopy. Results: All female rats in CON group survived (9/9), while only 6 rats in HARRT group survived (6/10). Compared with CON group, the body weight of female rats in HAART group was decreased significantly(P<0.01); the levels of left ventricular end diastolic diameter (LVDd), interventricular septal thickness (IVST), thickness of left ventricular posterior wall (LVPWT) , left atrial diameter (LAD) and arterial diastolic pressure were increased significantly (P<0.05); the level of LVP+dP/dtmax was decreased (P<0.01). The levels of triglyceride, creatine kinase, and glutamic oxaloacetic transaminase were decreased (P<0.05 or P<0.01), while the level of glucose was increased (P<0.05). The collagen fibers were increased in myocardial tissue, and ultrastructure of myocardial cells was abnormal. Conclusion: Antiretrovirals during gestation can cause cardiovascular diseases in female rats.

Highly efficient enrichment of N-glycopeptides from complicated biosamples based on mass spectrometry is essential for biomedical applications, especially in disease biomarker research. In this work, glutathione (GSH)-modified hierarchical flower-like hollow covalent organic frameworks loaded with Au nanoparticles (HFH-COFs@Au@GSH) were synthesized for N-glycopeptide enrichment. Due to the abundant accessibility sites, high specific surface area, and inherent high stability of the hierarchical flower-like hollow structure, a large number of Au NPs and hydrophilic GSH can be modified on the HFH-COFs. The HFH-COFs@Au@GSH displayed excellent hydrophilicity and remarkable enrichment performance for N-glycopeptides: low detection limit (0.1 fmol µL-1), large adsorption capacity (200 µg mg-1), great selectivity (1 : 1000, HRP to BSA), and good reusability (at least 5 times). Furthermore, the HFH-COFs@Au@GSH were successfully applied to capture N-linked glycopeptides in human serum, and 308 N-glycosylation peptides corresponding to 84 N-glycosylation proteins with 123 N-glycosylation sites were detected. Gene ontology analyses were used to elucidate the cellular component, biological process and molecular function of detected glycoproteins in human serum, demonstrating the great potential of the HFH-COFs@Au@GSH in N-glycopeptide enrichment for glycoproteomic analysis of complex biological samples.

Objective: Osteoporosis is associated with senescence of bone marrow mesenchymal stem cells (BMSCs). Extracellular vesicles derived from senescent BMSCs (BMSC-EVs) could be uptaken by muscle satellite cells (SCs). We hypothesized that inhibiting the uptake of harmful BMSC-EVs by SCs could prevent patients with osteoporosis complicated with sarcopenia. Methods: Bioinformatics analysis was used to analyze senescent SCs. Myogenic potential of SCs was measured using myogenesis assay and immunofluorescence while muscle atrophy was measured using histological evaluation. And the interaction of cluster of differentiation (CD) 81 and the membrane proteins of SCs was verified using biotin pulldown assay.. CD81-specific siRNA (si-CD81) was used to knockdown CD81 and anti-CD81 antibody (anti-CD81 Ab) was used to block CD81. Results: Differentially expressed genes in senescent SCs were enriched in muscle cell differentiation. The myogenic potential of senescent SCs was significantly decreased. Senescent BMSC-EVs impaired myogenesis of SCs. CD81 on the surface of BMSC-EVs could bind to membrane proteins of SCs. Both knockdown of CD81 and blocking CD81 prevented the uptake of senescent BMSC-EVs by SCs, thus relieving harmful effects of senescent BMSC-EVs on muscle atrophy. Conclusion: Blocking CD81 on the surface of senescent BMSC-EVs attenuates sarcopenia in aged mice, which could be useful for prevention of sarcopenia in patients with osteoporosis in clinical practice. Translational potential of this article: Inhibiting uptake of extracellular vesicles derived from senescent bone marrow mesenchymal stem cells by muscle satellite cells can prevent muscle atrophy in aged mice and has potential for application in treating sarcopenia.

BACKGROUND: To determine the maturity of cantaloupe, measuring the soluble solid content (SSC) as the indicator of sugar content based on the refractometric index is commonly practised. This method, however, is destructive and limited to a small number of samples. In this study, the coupling of a convolutional neural network (CNN) with machine vision was proposed in detecting the SSC of cantaloupe. The cantaloupe images were first acquired under controlled and uncontrolled conditions and subsequently fed to the CNN to predict the class to which each cantaloupe belonged. Four hand-crafted classical machine-learning classifiers were used to compare against the performance of the CNN. RESULTS: Experimental results showed that the CNN method significantly outperformed others, with an improvement of >100% being achieved in terms of classification accuracy, considering the data acquired under the uncontrolled environment. CONCLUSION: The results demonstrated the potential benefit to operationalize CNNs in practice for SSC determination of cantaloupe before harvesting. © 2022 Society of Chemical Industry.

BACKGROUND: In this study, the duration of high-pressure processing (HPP) required to achieve a 5 log reduction of Escherichia coli O157:H7 in fruit purees was evaluated. Banana, cantaloupe, and dragon fruit purees were subjected to HPP at 600 MPa for 300, 270, and 270 s, respectively, and their physicochemical properties and enzyme activities were then analysed. Diabetic mice were fed fresh and HPP-treated purees to observe their effects on the glycemic index (GI) and postprandial blood glucose response. RESULTS: Compared with their fresh counterparts, the HPP-treated banana and dragon fruit purees exhibited significantly higher viscosities, lower glucose concentrations, and higher glucose dialysis retardation indices and showed disrupted sucrose invertase and polygalacturonase activities. The GI and postprandial blood glucose response were not significantly different between the fresh and HPP-treated cantaloupe purees. By contrast, the peak time of glucose response (Tmax ) was delayed from 30 min to 60 min, and the area under the receiver operating characteristic curve was reduced by 40% in the mice fed HPP-treated banana and dragon fruit purees. The GIs of the HPP-treated banana and dragon fruit purees (were 50.3 and 44.8, respectively) were significantly lower than those of their fresh counterparts (85.1 and 75.2, respectively). CONCLUSION: HPP can change the physicochemical properties of fruit purees, resulting in stabilized blood glucose levels and lower GIs after consumption. Therefore, purees processed in this manner would benefit consumers and patients with diabetes/pre-diabetes who need to maintain stable blood glucose levels (Fig. S1). © 2022 Society of Chemical Industry.

Subcutaneous fat and intramuscular fat (IMF) deposition are closely related to meat production and pork quality. Dingyuan pig, as a native pig breed in China, low selection leads to obvious genetic and phenotypic differences in the population. Individuals with extreme fat content in the population are ideal models for studying the mechanism of fat deposition. In this study, we used RNA-Seq and tandem mass tags-based (TMT) proteomics to analyze the key pathways and genes that specifically regulate subcutaneous fat and IMF deposition in Dingyuan pigs. We identified 191 differentially expressed genes (DEGs) and 61 differentially abundant proteins (DAPs) in the high and low back fat thickness (HBF, LBF) groups, 85 DEGs and 12 DAPs were obtained in the high and low intramuscular fat (HIMF, LIMF) groups. The functional analysis showed that the DEGs and DAPs in the backfat groups were mainly involved in carbohydrates, amino acids, and fatty acids metabolism, whereas the IMF groups were involved in the insulin pathway, longevity, and some disease-related pathways. We found 40 candidate genes that might tissue-specifically lipids deposition for subcutaneous and intramuscular fat. Our research provides theoretical reference materials for the improvement of fat deposition traits of local pig breeds in my country.

Phthalate esters (PAEs) are a typical group of endocrine-disrupting chemicals (EDCs) and widely found in the environment, which have attracted great concern due to the potential risks to environment and human health. It is urgent to develop a high-efficiency and high-sensitivity analytical approach for enrichment and determination of ultra-trace PAEs. Herein, magnetic covalent organic frameworks (Fe3O4@TAPB-DVA) were fabricated via a one-pot approach at room temperature and explored as a magnetic enrichment probe with high surface area (275.50 m2/g), good crystallinity, regular pore size distribution (3.1 nm) and outstanding saturation magnetization (26.5 emu/g). Accordingly, a simple and sensitive method based on magnetic solid-phase extraction (MSPE) combined with HPLC-MS/MS was established to detect ultra-trace PAEs from fine particulate matter (PM2.5) samples. The proposed method showed wide linear ranges (1-400 ng/L), good linearity (r > 0.9978) and high enrichment factors (EFs, 6.99-95.01 folds) with low detection limits (LODs) ranged from 0.01 to 0.5 ng/L (S/N ≥ 3). On this basis, the proposed method was substantially employed for sensitive detection of low- abundance PAEs from PM2.5 samples, suggesting the promising application of the Fe3O4@TAPB-DVA in sample enrichment.

In this work, boron dipyrromethene (BODIPY) is for the first time employed as electron-deficient core (A') to construct an A-D-A'-D-A type nonfused-ring electron acceptor (NFREA) for polymer solar cells (PSCs). Among, cyclopentadithiophene (CPDT) and fluorinated dicyanoindanone (DFIC) are involved as electron-donating (D) bridges and terminal A groups, respectively. Bearing with the steric BODIPY core, tMBCIC exhibits twisted configuration with dihedral angles >45°  between BODIPY and CPDT bridges. Thus, compared with the BODIPY-free planar A-D-D-A structured bCIC, reduced aggregation, weakened intramolecular D-A interactions with up-shifted lowest unoccupied molecular orbital by 0.4 eV as well as blueshifted absorption by up to 150 nm is observed in tMBCIC. Moreover, owing to the intrinsic large molar extinction coefficient from BODIPY, promoted light-harvest ability is achieved for tMBCIC, particularly in its blend films. Therefore, PSCs by using PBDB-T as donor, tMBCIC as NFREA afford superior power conversion efficiency (PCE) of 9.22% and higher open-circuit voltage (Voc ) of 0.954 V compared to 4.47% and 0.739 V from bCIC-devices. Moreover, compared to other BODIPY-flanked electron acceptors (<5%) reported so far, BODIPY-cored tMBCIC realizes a remarkable progress in PCE.

Based on previous reports,toll-like receptors (TLRs) are recognition molecules common in various aquatic animals and play a vital role in innate immunity. In this study, a novel TLR CgToll-3 with leucine-rich repeats (LRRs) and a TIR (Toll-interleukin 1-resistance) domain was cloned in Crassostrea gigas. CgToll-3 with sixteen potential extracellular N-linked glycosylation sites and shares the closest phylogenic relationship with molluscan TLRs. Alignment of LRRs and TIR domains indicated that CgToll-3 was highly conserved compared to other LRRs of mollusks which could respond against Vibrio or other bacterial molecules, and contained three conserved functionally important motifs (Box 1, Box 2, and Box 3). The Hex Molecular Docking result showed that CgToll-3 could interact with CgMyd88 via the TIR domain. Subcellular Co-localization and BiFC Assay confirmed this interaction, and they could induce NF-κB activation. CgToll-3 was moderately expressed in the digestive gland, and its expression level was significantly up-regulated after Vibrio alginolyticus challenge. Taken together, CgToll-3 might be involved in the innate immune response to V. alginolyticus for C. gigas through a MyD88-dependent TLR mediated signaling pathway.

Hepatic ischemia-reperfusion injury (HIRI) is an additional injury to ischemic tissue after hepatic revascularization, and its pathological mechanism is complex. HIRI is not only involved in the molecular targets that mediate cell death, such as ion channel activation, abnormal protease activation and mitochondrial dysfunction, but also related to the down-regulation of endogenous protective signals. As a by-product of normal aerobic metabolism, reactive oxygen species (ROS) act as a multi effect physiological signal factor at low concentration. However, liver ischemia-reperfusion will lead to excessive ROS accumulation, destroy redox homeostasis, lead to oxidative stress, cause cell death through a variety of mechanisms, and drive the further damage of ischemic liver. Recent studies have found that the antioxidant treatment of nano selenium can reduce the excessive production of ROS and play a potential protective role in reducing HIRI. This paper reviews the molecular mechanism of the antioxidant effect of nano selenium for the prevention and treatment of HIRI, in order to provide further experimental basis for the clinical prevention and treatment of HIRI.
