In this study, we investigated the in vitro effect of tomentosin on cell proliferation by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay, reactive oxygen species by 2',7'-dichlorofluorescein diacetate staining assay, apoptosis (AO/EtBr, propidium iodide, and 4',6-diamidino-2-phenylindole staining, mitochondrial membrane potential), cell adherent, cell migration, inflammation, apoptosis, and oxidative stress from gastric cancer cells (GCCs) AGS. Upon their relative cell proliferative, inflammatory, and apoptotic molecular markers were analyzed by using the enzyme-linked immunosorbent assay and Western blot analysis method. Treatment with tomentosin (IC50 = 20 µM) significantly inhibited cell proliferation and oxidative stress-induced anti-cell proliferative (proliferating cell nuclear antigen and cyclin-D1) also regulated expression, drastically diminished tumor necrosis factor-α, nuclear factor-κB, interleukin-6, and interleukin-1ß expression levels, significantly upregulated Bcl-2 and Bax expression. Thus, this tomentosin can significantly reduce GCC proliferation via cytotoxicity which is stimulated apoptosis markers via morphology staining changes and inhibitory inflammatory markers. The tomentosin-induced oxidative stress may be involved to stimulate apoptotic mechanisms via mitochondria-mediated signaling by the inhibition of inflammation. Taken together, our findings suggest a possible future use of chemotherapeutic agents for pharmacological benefits and as an anti-cancer treatment option.

In present study, we discovered unusual solvent-mediated aggregation-enhanced emission (AEE) character of 11-mercaptoundecanoic acid capped gold nanoclusters (MUA-Au NCs). When aggregated in aqueous media, the MUA-Au NCs showed strong emission, which was weakened by adding ethanol. Interestingly, the suppressed emission was selectively enhanced in the presence of hydrogen sulfide (H2S) because H2S was absorbed onto Au NCs through the strong sulfur-gold bonding affinity. The hydrolyzed H2S, namely, HS-, made the Au NCs negatively charged, which aggregated again due to decreased solubility. The H2S-mediated fluorescence enhancement can be further amplified by introducing a hydrophilic thiolate (glutathione, GSH) onto the surface of Au NCs (GSH/MUA-Au NCs), which enabled sensitive determination of H2S. Under the optimized condition, a detection limit of 35 nM was achieved. The determination was not interfered by other anions such as F-, Cl-, Br-, I-, OAc-, N3-, NO3-, HCO3-, SCN-, SO32-, and SO42-. This excellent sensing performance allowed practical application of the GSH/MUA-Au NC-based sensing platform to accurate determination of H2S in human serum samples. RESUMO

A series of tetracyclic furazan-triazole compounds have been synthesized and fully characterized. The predicted detonation performance and tested mechanical sensitivities showed their high-energy performance and insensitive features. Quantum chemical calculations and crystal structure analysis were applied to study the intrinsic structure-property relationship among these compounds. In addition, the detonation test shows their promising potential as secondary explosives.

Cyprinid herpesvirus 3 (CyHV-3) is the etiological agent of koi herpersvirus disease (KHVD), which causes serious economic losses in global common carp and ornamental koi carp production of larvae as well as adult type fish. To control KHVD, vaccines against CyHV-3 utilizing different immunization routes have been developed, among them, oral vaccination is the most desirable method to prevent fish diseases occurring at the early larval stage. Here, we developed an oral subunit vaccine through the Saccharomyces cerevisiae cell surface display of CyHV-3 envelope protein pORF65, then, the recombinant yeast fed to Artemia which served as bio-encapsulation vector by subsequently feeding the common carp (Cyprinus carpio var. Jian) larvae. The fluorescent observation showed that the Artemia and S. cerevisiae could deliver intact antigen to the hindgut of carp larvae suggesting the possibility of the vector for oral immunization. On this basis, after three immunizations at a week interval, the oral vaccine induced high level of specific anti-pORF65 antibody. Meanwhile, a significant difference of immune-related genes expression occurred including cxca, IL-1ß, IFN-a1, lysozyme, IgM and CD8α between vaccined group and blank control group. In addition, 30% of relative percent survival of carp larvae after immunization was obtained post the animal infection assay, offered an certain immune protection. Our results indicated that the oral pORF65 subunit vaccine bioencapsulated in Artemia induced the activation of immune response and high level of antibodies, which could be served as an oral vaccine candidate for the prevention of CyHV-3 infection.

Colon carcinoma is a recurring type of cancer that affects the intestine epithelial with a poor survival rate. It was already proven the anticancer property of hesperidin in various cancers but the bioavailability hesperidin is poor, which hinders the hesperidin usage. In this investigation we synthesized hesperidin loaded Zn2+@ SA/PCT nanocomposites and assessed its anticancer potential against colon cancer (HCT116) cells. Hesperidin loaded Zn2+@ SA/PCT nanocomposites were characterized using Fourier transform infrared (FTIR), X-ray diffraction (XRD), scanning electron microscope (SEM) and transmission electron microscope (TEM) analysis. The drug releasing capacity and cytotoxic property was assessed via drug releasing assay, MTT assay with HCT116 cells. The anticancer potency of hesperidin nanocomposites were evaluated with TUNEL, DAPI staining, reactive oxygen species (ROS) generation assay and it is confirmed with flow cytometry analysis of MMP disruption in colon cancer (HCT116) cell line. Further the immunoblotting analysis of cysteine proteases Caspases 3, 9, PARP, proapoptotic protein Bax and antiapoptotic protein Bcl2 were performed. The results of FTIR, XRD and electroscopic analyses confirmed the synthesized hesperidin nanocomposites accomplish the properties of potent nanodrug and the MTT assay authentically confirmed that the synthesized hesperidin nanocomposite inhibited the HCT116 cell growth, and the results of fluorescent staining proved that the hesperidin nanocomposite induced the apoptotic mediated cell necrosis via promoting the expression of apoptotic proteins thereby induced the apoptosis in colon cancer (HCT116) cells. Hence, it was concluded that the, hesperidin loaded nanocomposites persuasively inhibited proliferation of colon carcinoma cell and induced apoptosis in in vitro condition.

In this work, a series of pyrazole-derived energetic compounds were successfully synthesized. These energetic compounds were fully characterized by NMR spectroscopy, IR spectroscopy, and elemental analysis. The structures of compounds 5, 6, 7 and 7a were determined by single crystal X-ray diffraction. The physicochemical and energetic properties of all synthesized energetic compounds, including density, thermal stability and energetic performance, were investigated. The structure-property relationship was illustrated using two-dimensional fingerprint plots based on Hirshfeld surfaces, NCI plots and ESP of 7 and 7a. Among these energetic compounds, the hydroxylammonium salt 7b exhibited satisfactory calculated detonation performance (8700 m s-1), which was comparable to the commonly used highly explosive RDX (8748 m s-1). The potassium salt 5 was tested for its detonation ability by detonating RDX. The result indicates that compound 5 could be used as a potential green primary explosive.

AIMS: Diabetic retinopathy (DR) is the main cause of blindness in adults and investigating new therapeutic targets for DR is necessary. This study aimed to investigate the effect of high-mobility group box 1 (HMGB1) protein and its mechanism in diabetic retinopathy (DR) were investigated. MAIN METHODS: Human retinal endothelial cells (HREC) were uesd for chip-seq. Sprague Dawley (SD) rats were randomly divided into control group, HMGB1 group, diabetes mellitus (DM) combined with HMGB1 siRNA group, and DM group. Next, eyeballs were removed and retinas were detached for western blot. The DM model of cell was built by increasing the glucose concentration in cell culture medium. The regulation of HMGB1 was achieved by short hairpin (sh)-HMGB1 transfection, then, the transfected cells were harvested for luciferase assay, western blot and qRT-PCR analyses as well as proliferation and apoptosis detection. KEY FINDINGS: Chip-seq and luciferase assay showed the possible transcription factor functions of HMGB1 and IKB-α was one of the HMGB1 binding sites. In vivo and in vitro results indicated high expression of HMGB1 and NF-kB and low expression of IKB-α in DR and the expression of IKB-α and NF-kB was regulated by HMGB1. Moreover, cell assays showed that HMGB1 inhibited cell proliferation and promoted apoptosis. SIGNIFICANCE: The results from the present study showed that HMGB1 may be involved in the pathogenesis of DR as a transcription factor through NF-kB pathway. Therefore, blockade of HMGB1 may be a new method for the treatment of DR.

Advanced treatment is of great significance to water reclamation and reuse, which can improve water quality, control microbial risks and guarantee the safety of water reuse. This study evaluates the microbial quantity and bacterial community dynamics during advanced wastewater treatment and reuse processes (i.e. denitrification biofilter (DNBF), ultrafiltration (UF), ozonation, ultraviolet (UV) disinfection) at a large-scale water reclamation plant. It is found that different treatment processes had significant influence on the cultivability of total bacteria and the log reduction values of fecal coliform at DNBF, UF, ozonation and UV are calculated as 0.38, 2.46, 0.38 and 1.63 respectively. Moreover, the bacterial diversity in the treatment process showed apparent spatial differences, among which the effluent from ozonation process had the lowest bacterial diversity. Sequencing analysis indicated the existence of pathogenic bacterium such as Arcobacter, Bacteroides and Pseudomonas in the secondary effluent. Notably, Pseudomonas remained the most dominant species (relative abundance 41.9% in UV effluent) in reclaimed water after advanced treatment processes, which calls for high attention to sustainable water reuse. In order to inhibit bacterial regrowth in the storage tank, chlorine disinfection is recommended to improve the continuous disinfection capability of the system.

The rapid and sensitive visualization of 2,6-dipicolinic acid (DPA, a unique anthrax biomarker) is essential to prevent anthrax disease or biological terrorist attack. In this study, a Eu3+-labeled ethylenediaminetetraacetic acid loaded hyperbranched polyethyleneimine carbon nanodot (hPEI-CD-EDTA-Eu3+) nanoprobe has been proposed for the ratiometric DPA detection. The sensing mechanism is based on the rapid DPA-Eu3+ chelation within 30 s and subsequent enhanced fluorescence emission through the antenna effect. With the introduction of EDTA chelating unit, the resulted fluorescence of Eu3+-complex is greatly enhanced, which endows sensitive DPA perception. By employing hPEI-CD as the internal reference, ratiometric DPA sensing is realized with a good linearity in the concentration range from 1.0 to 100 nM, with a limit of detection of 190 pM (S/N = 3). The specific chelation affinity between Eu3+ and DPA provides satisfying selectivity over other amino acids and ions. Using nanoprobe-loaded polyvinylidene fluoride paper as the analytical device, point-of-care DPA visualization is achieved. Furthermore, the practical application of designed paper device is validated by the visual detection of metabolic DPA-release from Bacillus subtilis spores.

Zika virus (ZIKV) is a mosquito-borne flavivirus that has been associated with neuropathology in fetuses and adults, imposing a serious health concern. Therefore, the development of a vaccine is a global health priority. Notably, neutralization tests have a significant value for vaccine development and virus diagnosis. The cytopathic effect (CPE)-based neutralization test (Nt-CPE) is a common neutralization method for ZIKV. However, this method has some drawbacks, such as being time-consuming and labor-intensive and having low-throughput, which precludes its application in the detection of large numbers of specimens. To improve this problem, we developed a neutralization test based on an enzyme-linked immunospot assay (Nt-ELISPOT) for ZIKV and performed the assay in a 96-well format. A monoclonal antibody (mAb), 11C11, with high affinity and reactivity to ZIKV was used to detect ZIKV-infected cells. To optimize this method, the infectious dose of ZIKV was set at a multiplicity of infection (MOI) of 0.0625, and a detection experiment was performed after incubating for 24 h. As a result, under these conditions, the Nt-ELISPOT had good consistency with the traditional Nt-CPE to measure neutralizing titers of sera and neutralizing antibodies. Additionally, three neutralizing antibodies against ZIKV were screened by this method. Overall, we successfully developed an efficient neutralization test for ZIKV that is high-throughput and rapid. This Nt-ELISPOT can potentially be applied to detecting neutralizing titers of large numbers of specimens in vaccine evaluation and neutralizing antibody screening for ZIKV.

Gold nanoparticles with special surface plasmon resonance have been widely used in sensing and therapy because of their easy preparation, unique optical properties, excellent biocompatibility, etc. The applications of gold nanoparticles in chemo/biosensing, imaging, and therapy reported in 2016-2019, are summarized in this review. Regarding the gold nanoparticle-based sensing or imaging, sensing mechanisms and strategies are provided to illustrate the concepts for designing sensitive and selective detection platforms. Gold nanoparticlemediated therapy is introduced by surface plasmon resonance-based therapy and delivery-based therapy. Beyond the sole therapeutic system, platforms through synergistic therapy are also discussed. In the end, discussion of the challenges and future trends of gold nanoparticle-based sensing and therapy systems is described.

BACKGROUND: Long non-coding RNAs (lncRNAs) are increasingly investigated in numerous carcinomas containing gastric cancer (GC). The aim of our research is to inquire about the expression profile and role of LBX2-AS1 in GC. METHODS: The expressions of LBX2-AS1, miR-219a-2-3p, FUS and LBX2 were measured by qRT-PCR. Western blot evaluated FUS and LBX2 protein levels. Cell proliferation and apoptosis were, respectively, evaluated by CCK-8, colony formation, EdU, flow cytometry and TUNEL assays. FISH and subcellular fractionation assays examined the position of LBX2-AS1. The binding between genes were certified by RIP, RNA pull-down, ChIP and luciferase reporter assays. Pearson correlation analysis analyzed the association of genes. Kaplan-Meier method detected the relationship of LBX2-AS1 expression with overall survival. RESULTS: The up-regulation of LBX2-AS1 in GC tissues and cells was verified. Function assays proved that LBX2-AS1 down-regulation restricted the proliferation ability. Then, we unveiled the LBX2-AS1/miR-219a-2-3p/FUS axis. Additionally, LBX2-AS1 positively regulated LBX2 mRNA stability via FUS. LBX2 transcriptionally modulated LBX2-AS1. In the end, rescue and in vivo experiments validated the whole regulatory mechanism. CONCLUSION: LBX2-AS1/miR-219a-2-3p/FUS/LBX2 positive feedback loop mainly affected the proliferation and apoptosis abilities of GC cells, offering novel therapeutic targets for the treatment of patients with GC.

This paper presents a droplet-based immunoassay chip allowing each droplet to be positioned in a passive droplet-positioning cavern under continuous flow. In addition, the chip surface can immobilize any kind of histidine-tagged capture agents for performing simultaneous multiplex immunoassays. Distinct families of monodispersed droplets were generated since a diaphragm, which is a thin elastomeric flap film suspended from the top of the main channel, forms a double T junction for shearing the aqueous liquids by the carrier flow. These two types of monodispersed droplets traverse the main channel to the downstream detection area and enter the passive positioning caverns for further immunoassay. A layer of Ni-Co film was coated on the substrate by electrodeposition in order to immobilize the multiplex histidine-tagged capture molecules. In this study, the tumor suppressor protein p53 and the extracellular signal-related kinase 1 (ERK1) were used as the capture agents. Then, both histidine-tagged proteins p53 and ERK1 were immobilized by the Ni-Co layer in a microarray format for subsequent immunoassay and fluorescence detection. The experimental results show that the detected fluorescence intensity is proportioned to the concentration of the encapsulated content in a small droplet. This proposed droplet-based immunoassay chip can immobilize multiplex histidine-tagged proteins, irrelevant to the species of proteins, to carry out simultaneous immunoassays and allow the operation sequence to be conducted automatically through the manipulation of droplets.

OBJECTIVE: To observe the effect of cinobufagin on transient outward potassium current (IA) in rat dorsal root ganglion cells of cancer-induced bone pain (CIBP) and explore the possible analgesic mechanism of cinobufagin. METHODS: Whole cell patch clamp technique was used to examine the effect of cionbufagin on IA in acutely isolated dorsal root ganglion (DRG) cells from normal SD rats and rats with bone cancer pain. RESULTS: The DRG cells from rats with CIBP showed obviously decreased IA current density, an activation curve shift to the right, and an inactivation curve shift to the left. Cinobufagin treatment significantly increased the IA current density and reversed the changes in the activation and inactivation curves in the DRG cells. CONCLUSIONS: IA current is decreased in DRG neurons from rats with CIBP. Cinobufagin can regulate the activation and inactivation of IA current in the DRG cells, which may be related to its analgesic mechanism.

Polyamines (putrescine, spermidine, and spermine) are essential polycations that play important roles in various physiological and pathophysiological processes in mammalian cells. The study was to investigate their role in cardioprotection against ischemia/reperfusion (I/R) injury and the underlying mechanism. Isolated hearts from male Sprague-Dawley rats were Langendorff-perfused and cardiac I/R was achieved by 30 min of global ischemia followed by 120 min of reperfusion. Different concentrations of polyamines (0.1, 1, 10, and 15 µmol/L of putrescine, spermidine, and spermine), cyclosporin A (0.2 µmol/L), or atractyloside (20 µmol/L) were given 10 min before the onset of reperfusion. The hemodynamics were monitored; the lactate dehydrogenase (LDH) levels in the coronary effluent were measured spectrophotometrically; infarct size was determined by the 2,3,5-triphenyltetrazolium chloride staining method; and mitochondrial permeability transition pore (MPTP) opening was determined spectrophotometrically by the Ca2+-induced swelling of isolated cardiac mitochondria. The results showed that compared to I/R alone, 0.1 and 1 µmol/L polyamines treatment improved heart function, reduced LDH release, decreased infarct size, and these effects were inhibited by atractyloside (MPTP activator). In isolated mitochondria from normal rats, 0.1 and 1 µmol/L polyamines treatment inhibited MPTP opening. However, 10 and 15 µmol/L polyamines treatment had the opposite effects, and these effects were inhibited by cyclosporin A (MPTP inhibitor). Our findings showed that polyamines may have either protective or damaging effects on hearts suffering from I/R by inhibiting or activating MPTP opening.

Several energetic materials, which are composed of furoxan and 1,2,4-oxadiazole backbones, were synthesized by nitrating 3,3'-bis(5-amino-1,2,4-oxadiazol-3-yl)-4,4'-azofuroxan (2) under 100 wt% HNO3 or 100 wt% HNO3/Ac2O followed by a cation metathesis. All synthesized compounds were fully characterized by multinuclear NMR spectroscopy, IR spectroscopy, and elemental analysis, while 3,3'-bis(1,2,4-oxadiazol-5(4H)-one-3-yl)-4,4'-azofuroxan (3) and diammonium 3,3'-bis(5-nitramino-1,2,4-oxadiazole-3-yl)-4,4'-azofuroxan (4a) were confirmed by single crystal X-ray diffraction. The physicochemical and energetic properties of these compounds including density, thermal stability and sensitivity were investigated. Compounds 3 and 4 have high densities (3: 1.90 g cm-3, 4: 1.92 g cm-3), which are comparable to that of HMX (1.91 g cm-3). All energetic compounds show relatively high calculated heat of formation in the range from 504.79 kJ mol-1 to 1405.62 kJ mol-1. Their detonation properties were evaluated by EXPLO5 code using the measured density and calculated heat of formation. Among them, compounds 3 and 4 have good detonation performance (3: D = 8891 m s-1, P = 34.7 GPa, 4: D = 9505 m s-1, P = 41.3 GPa) and acceptable sensitivities (3: IS = 10 J, 4: IS = 4 J), which indicate their potential applications as high-performance energetic materials.

Silver nanowires (AgNWs) with the diameter down to 16 nm have been synthesized by using tetrabutyl ammonium dibromochloride (TBADBC) as the organic auxiliary. Moderate dosage of TBADBC was proven to be favorable for the in situ growth of Ag pentahedral seeds on the surface of small AgClBr2 particles and have a passivation effect on the Ag{100} crystal facet during the growth of AgNWs, eventually resulting in the formation of thin AgNWs. Furthermore, the transparent conductive film fabricated by using the obtained AgNWs exhibited excellent photoelectric properties, which has a transmittance of 90.60%, a haze of 0.95%, and a sheet resistance of 110.8 Ω/sq, indicating large potential for application in flexible transparent conductors.

In order to further explore the relationship between water body eutrophication of some reaches of Yellow River and phosphorus species in sediments, and evaluate phosphorus release potential from sediments to overlying water, we investigated the distribution of P species and their release risk in the surface sediments from different reaches along Yellow River, as well as the influence of artificial dam on phosphorus cycle in Yellow River. The results show that the content of calcium phosphorus (PCa) is higher than the content of bioavailable phosphorus (BP) (BP = Pex + PFe; in the formula, Pex means exchangeable phosphorus and PFe means iron phosphorus) in the surface sediments from Yellow River. Among all the surface sediment samples from 21 stations, only Dayudu section (H15) has a ratio of W (BP)/W (PCa) higher than 0.5; the results show that the intensity of phosphorus release from H15 is high, and there is a potential risk of eutrophication in water, while the phosphorus release level of water sediment in other reaches of Yellow River is low. The content of BP and total phosphorus (∑P) in surface sediments along the Yellow River is in descending order: middle reaches of Yellow River > upper reaches of Yellow River > Yellow River downstream, while the content order of total phosphorus (TP) in the overlying water is as follows: Yellow river downstream > middle reaches of Yellow River > upper reaches of Yellow River (except the H7 station), indicating that extensive artificial corresponding damming in Yellow River basin makes the concentration of TP increasing significantly in overlying from upstream to downstream. The establishment of Haibowan water conservancy project in Yellow River makes the total particulate matter (TPM) concentration in the water to reduce to very low lever in Wuhai H7 section, and the phosphorus concentration in the overlying water reaches 0.136 mg L-1.

RESUMO

BACKGROUND: This study aims to systematically assess the efficacy and safety of fasudil for the treatment of aneurysmal subarachnoid hemorrhage (ASH). METHODS: This study will include all of randomized controlled trials on the efficacy and safety of fasudil for the treatment of ASH. Ten electronic databases of PubMed, Embase, Cochrane Library, Google Scholar, Web of Science, Ovid, Cumulative Index to Nursing and Allied Health Literature, Allied and Complementary Medicine Database, Chinese Biomedical Literature Database, and China National Knowledge Infrastructure will be searched from inception to the May 1, 2019 without language restrictions. We will also search gray literatures to avoid missing any other potential studies. Two authors will independently perform study selection, data extraction and management, and methodologic quality assessment. The primary outcome is limbs function. The secondary outcomes comprise of muscle strength, muscle tone, quality of life, and adverse events. RESULTS: This study will provide a comprehensive literature search on the current evidence of fasudil for the treatment of ASH from primary and secondary outcomes. CONCLUSION: The results of this study will present evidence to determine whether fasudil is an effective and safety treatment for patients with ASH.
