RESUMO

Bone remodeling is dynamic and is tightly regulated through bone resorption dominated by osteoclasts and bone formation dominated by osteoblasts. Imbalances in this process can cause various pathological conditions, such as osteoporosis. Bone morphogenetic protein 9 (BMP9), a biomolecule produced and secreted by the liver, has many pharmacological effects, including anti-liver fibrosis, antitumor, anti-heart failure, and antidiabetic activities. However, the effects of BMP9 on the regulation of osteoblast and osteoclast functions and the underlying molecular mechanism(s) have not yet been investigated. In this study, BMP9 increased the expression of osteoblastogenic gene markers, such as ALP, Cola1, OCN, RUNX2, and OSX, and ALP activity in MC3T3-E1 cells by upregulating LGR6 and activating the Wnt/ß-catenin pathway. BMP9 also suppressed receptor activator of nuclear factor-κB (NF-κB) ligand (RANKL)-induced osteoclast differentiation of bone marrow macrophages (BMMs) by inhibiting the Akt-NF-κB-NFATc1 pathway. More importantly, in an ovariectomy (OVX) mouse model, BMP9 attenuated bone loss and improved bone biomechanical properties in vivo by increasing bone-forming activity and suppressing bone resorption activity. Accordingly, our current work highlights the dual regulatory effects that BMP9 exerts on bone remodeling by promoting bone anabolic activity and inhibiting osteoclast differentiation in OVX mice. © 2020 American Society for Bone and Mineral Research.

Inorganic phosphate (Pi) starvation severely affects the normal growth and development of plants. Here, a Pi-responsive gene, named MdMYB2 (MDP0000823458), was cloned and functionally identified in apple. Overexpression of MdMYB2 regulated the expression of Pi starvation-induced (PSI) genes and then promoted phosphate assimilation and utilization. The ectopic expression of MdMYB2 in Arabidopsis influenced plant growth and flowering, which was partially rescued by application of exogenous gibberellin (GA). These results indicated that MdMYB2 may be an essential regulator in phosphate utilization and GA-regulated plant growth and development.

Idiopathic hypogonadotropic hypogonadism (IHH) patients are characterized by the absence of puberty and varying degrees of deteriorated metabolic conditions. Osteocalcin (OC) could regulate testosterone secretion and energy metabolism, but it remains unknown whether such an effect exists in IHH patients. Our study is aimed to examine the relationship between serum OC levels with testosterone and its responsiveness to gonadotropin stimulation and metabolic profiles in male IHH patients. A total of 99 male patients aged 18-37 years and diagnosed with IHH were enrolled in the current study, and the relationships between OC and testicular volume, baseline total testosterone (TT), free testosterone (FT), and peak TT (Tmax) levels after human chorionic gonadotropin (hCG) stimulation, gonadotropin responsiveness index (GRI), which is calculated by dividing Tmax by testicular volume, as well as metabolic profiles, such as 2-h post-challenge glucose (2hPG) and fat percentage (fat%), were analyzed. The results showed that OC had an independent negative relationship with testicular volume (r = -0.253, P = 0.012) and a positive association with Tmax (r = 0.262, P = 0.014) after adjusting for confounders. In addition, OC was a major determinant of GRI (adjusted R 2 for the model = 0.164, P = 0.012), fat% (adjusted R 2 for the model = 0.100, P = 0.004), and 2hPG (adjusted R 2 for the model = 0.054, P = 0.013) in IHH patients. In conclusion, OC is associated with testosterone secretion upon gonadotropin stimulation, glucose metabolism, and fat mass variations in IHH. To study the correlation between ultra high performance liquid chromatography( UPLC) fingerprint of Smilax china and its anti-pelvic inflammatory effect,and to explore the pharmacodynamic material basis of S. china against pelvic inflammatory disease.UPLC fingerprints of 10 batches of S. china from different habitats were established,and the values of SOD,MDA,TNF-α,and IL-6 in rats with pelvic inflammation were measured. The weight of each single pharmacodynamics index to the total efficacy was determined by analytic hierarchy process,and the contribution of each peak in fingerprints to the each single pharmacodynamics index and total efficacy was analyzed by the grey relational analysis. Then the structures of chemical constituents at the identified peaks were confirmed by comparing with the reference substance. The 27 common characteristic peaks of UPLC fingerprints were all related to the anti-pelvic inflammation effect of S. china,of which 13 peaks were identified as peak 2( 3,5-dihydroxy-2-methylbenzoic acid-3-O-glucoside),peak 3( chlorogenic acid),peak 5( 2,7,4-trihydroxydihydroflavone-5-O-glucoside),peak 6( 7,4-dihydroxydihydroflavonol-5-O-glucoside),peak 7( taxifolin-7-O-glucoside),peak 9( taxifolin),peak 10( polydatin),peak 11( oxyresveratrol),peak 12( astilbin),peak15( resveratrol),peak 16( quercitrin),peak 18( engeletin) and peak 24( kaempferol). The correlation degree of 21 peaks and the total efficacy was greater than 0. 8,and the top 10 ranked by correlation degree were as follows: peak 1,3,7,19,18,17,4,11,16,and 21. The results showed that the anti-pelvic inflammation effect of S. china was achieved by the combined action of pharmacodynamic substances. In order to control the quality of S. china and its prepared slices more effectively,the index components of content detection should be selected reasonably.

The 14-3-3 proteins are a family of highly conserved phosphoserine-binding proteins that participate in the regulation of diverse physiological and developmental processes. In this research, twenty 14-3-3 genes in apples, which contained a highly conserved 14-3-3 domain, were identified and divided into two subgroups. Among them, MdGRF11 was further cloned and investigated. qRT-PCR analyses and GUS staining show that MdGRF11 is expressed in various organs and tissues with the highest expression levels found in the fruit. MdGRF11 was upregulated by polyethylene glycol 6000 (PEG 6000), NaCl, abscisic acid (ABA) and low temperature (4 °C) treatments. MdGRF11-overexpressing transgenic Arabidopsis and apple calli exhibited reduced sensitivity to salt and PEG 6000 treatments. Moreover, the ectopic expression of MdGRF11 improved the tolerance of transgenic tobacco to salt and drought stresses, which grew longer roots, underwent more growth, and presented higher chlorophyll levels than the wild-type control under salt and drought stress conditions. Furthermore, MdGRF11 expression remarkably reduced electrolyte leakage, malondialdehyde content levels, H2O2 and O2- accumulation under salt and drought stress conditions, which relied on the regulation of ROS-scavenging signaling to reduce oxidative damage of cells after salt and drought stress treatment. MdGRF11 also enhanced tolerance to stress by upregulating expression levels of ROS-scavenging and stress-related genes, especially improving responses to drought stress by modifying the water loss rates and stomatal aperture. Moreover, MdGRF11 could interact with MdAREB/ABF transcription factors through yeast two hybrid analyses. In conclusion, our results indicate that MdGRF11 acts as a positive regulator of salt and drought stress responses through regulating ROS scavenging and other signaling systems.

An unusual tetra-nuclear linear cyanido-bridged complex [Ru2 (µ-ap)4 -CN-Ru2 (µ-ap)4 ](BPh4 ) (1) (ap=2-anilinopyridinate) has been synthesized and well characterized. The crystallographic data, magnetic measurement, IR, EPR and theoretical calculation results demonstrate that complex 1 is the first example of mixed spin Ru2 5+ -based complex with uncommon electronic configurations of S=1/2 for the cyanido-C bound Ru2 5+ and S=3/2 for the cyanido-N bound Ru2 5+ . This phenomenon can be understood by the theoretical calculation results that from the precursor Ru2 (µ-ap)4 (CN) (S=3/2) to complex 1 the energy gap between π* and δ* orbitals of the cyanido-C bound Ru2 5+ core increases from 0.57 to 1.61 eV due to the enhancement of asymmetrical π back-bonding effect, but that of the cyanido-N bound Ru2 5+ core is essential identical (0.56 eV). Besides, the analysis of UV/Vis-NIR spectra suggests that there exists metal to metal charge transfer (MMCT) from the cyanido-N bound Ru2 5+ (S=3/2) to the cyanido-C bound Ru2 5+ (S=1/2), supported by the TDDFT calculations.

To investigate MMCT excited states of MV complexes, two symmetrical tetranuclear cyanido-bridged localized MV complexes RuIICNRuIII,III2NCRuII have been designed and synthesized. The ultrafast time-resolved transient absorption (TA) spectroscopy experiment reveals that the MMCT rate of 1 and 2 is 0.18 × 1014 s-1 (τ = 5.7 × 10-14 s) and 0.29 × 1013 s-1 (τ = 3.46 × 10-13 s), respectively, which suggests that the MMCT rate or the lifetime of the MMCT excited state may be controlled by a slight change of the substituent group on the metal center.

We detail the rational design of bimetallic cyanide-bridged complexes [TpmRu(LD)(µ-CN)Ru(LP)Cp*][PF6]2 (Tpm = Tris(1-pyrazolyl)methane, LD = 1,10-phenanthroline (phen), 2,2'-bipyridine (bpy), 4,4'-dimethyl-2,2'-bipyridine (dbpy), LP = bis(diphenylphosphino)methane (dppm), bis(diphenylphosphino)ethane (dppe), Cp* = 1,2,3,4,5-pentamethylcyclopentadiene). The metal-to-metal charge transfer (MMCT) properties of these one-electron oxidized complexes were investigated, suggesting that the substitution of the ancillary ligand provides a strong impetus to systematically tune the MMCT properties. The investigation results indicate that all the mixed-valence complexes belong to Class II mixed-valence complexes, according to the Robin-Day classification.

At low temperatures, the liver increases glucose utilization and expresses RNA-binding motif 3 (RBM3) to cope with cold exposure. In this study, the expression of heat shock protein 70 (HSP70), Toll-like receptor 4 (TLR4), bone marrow differentiation factor 88 (MYD88), and phosphorylated nuclear factor-κB (NF-κB) was consistent with fluctuations in insulin in fasted cold-exposed mice. We also found up-regulation of peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α) in acute cold exposure with a decrease in core body temperature. RBM3 transcription and translation were activated 2 h after cold exposure. The anti-apoptotic factor Bcl-2/Bax ratio also increased, while expression of apoptosis factors: cleaved caspase-3, cleaved poly(ADP-ribose)polymerase 1 (PARP-1) and cytochrome-c (Cyt-c) was unchanged. Liver glycogen was depleted after 2 h of cold exposure, and blood glucose decreased after 4 h. Glycogen synthase kinase 3ß (GSK3ß) phosphorylation continued to increase to promote hepatic glycogen synthesis. We found a high level of protein kinase B (AKT) phosphorylation after 6 h of cold exposure. In addition, we demonstrated that after cold exposure for 2 h, in the liver, continued phosphorylation of fructose-2,6-diphosphate (PFKFB2) and decreased accumulation of glycogen intermediates fructose-1,6-diphosphate (FDP) and pyruvic acid (PA). In summary, the liver responds to cold exposure through a number of different pathways, including activation of HSP70/TLR4 signaling pathways, up-regulation of RBM3 expression, and increased glycolysis and glycogen synthesis. We propose a possible signaling pathway in which regulation of RBM3 expression by the liver affects the AKT metabolic signaling pathway. Lay summary In response to changes in ambient temperature, mice regulate global metabolism and gene expression through hormones. This study focused on the effects of environmental hypothermia on molecular pathways of glucose metabolism in the liver, which is the important metabolic organ in mice. This provides a basis for further study of mice against cold exposure damage.

OBJECTIVE: To compare the molecular epidemiology of methicillin-resistant Staphylococcus aureus (MRSA) isolates between an old, urban hospital and a new, rural hospital over the same time period. METHODS: The molecular characteristics of 398 MRSA bloodstream isolates collected between 2007 and 2013 from two hospitals in Taiwan were analyzed retrospectively; 202 isolates were from the old hospital and 196 from the new hospital (opened in 2007). RESULTS: The rate of resistance to multiple antibiotics was significantly higher in the old hospital (93%) than in the new hospital (81%) (p<0.001). Genetic community-associated MRSA carrying staphylococcal cassette chromosome (SCC) type IV or V accounted for 58% of all MRSA isolates in the new hospital, significantly higher than the rate in the old hospital (p=0.018). The rate of spa t037-SCCmec III MRSA was significantly lower in the new hospital than in the old hospital (p=0.02). A significant decreasing trend in spa t002-SCCmec II MRSA isolates was observed in the old hospital (p=0.006), while the proportion of spa t037-SCCmec III MRSA decreased significantly in the new hospital (41.7% to 26.1%, p=0.022). CONCLUSIONS: The rate of multiple antibiotic resistance and the molecular characteristics of MRSA differed significantly between the old and new hospitals and changed over time.

The two stable pairs of trimetallic compounds trans-[Cp*(dppe)Ru(µ-NC)Ru(dmap)4 (µ-CN)Ru(dppe)Cp*][PF6 ]n (1[PF6 ]n , n=2, 3; Cp*=1,2,3,4,5-pentamethylcyclopentadiene; dppe=1,2-bis-(diphenylphosphino)ethane; dmap= 4-dimethylaminopyridine) and trans-[Cp*(dppe)Ru(µ-CN)Ru(dmap)4 (µ-NC)Ru(dppe)Cp*][PF6 ]n (2[PF6 ]n , n=2, 3), which demonstrate cyanide/isocyanide isomerism, have been synthesized and fully characterized. 13+ [PF6 ]3 and 23+ [PF6 ]3 are the one-electron oxidation products of 12+ [PF6 ]2 and 22+ [PF6 ]2 , respectively. The results suggest that 1[PF6 ]3 is a class III mixed valence compound, whereas 2[PF6 ]3 might be an unusually symmetrical class II-III mixed valence compound composed of the two asymmetrical delocalized RuIII -NC-RuII mixed valence subunits.

Previous studies have shown that folate levels were decreased in patients with type 2 diabetes (T2D) and further lowered in T2D patients with cognitive impairment. However, whether folate deficiency could cause T2D and subsequent cognitive dysfunction is still unknown. The present study aimed to explore the effects of chronic folate deficiency (CFD) on glucose and lipid metabolism and cognitive function in mice. Seven-week-old mice were fed with either a CFD or control diet for 25 weeks. Serum folate was significantly reduced, whereas serum total homocysteine was significantly increased in the CFD group. Moreover, CFD induced obesity after a 6-week diet treatment, glucose intolerance and insulin resistance after a 16-week-diet treatment. In addition, CFD reduced the hepatic p-Akt/Akt ratio in response to acute insulin administration. Moreover, CFD increased serum triglyceride levels, upregulated hepatic Acc1 and Fasn mRNA expression, and downregulated hepatic Cd36 and ApoB mRNA expression. After a 24-week diet treatment, CFD induced anxiety-related activities and impairment of spatial learning and memory performance. This study demonstrates that folate deficiency could induce obesity, glucose and lipid metabolism disorders and subsequent cognitive dysfunction.

Long non-coding RNAs (lncRNAs) are designated as the transcripts longer than 200 nucleotides without protein-coding capacity. As a category of important gene regulatory factors, lncRNAs regulate the expression of target genes at epigenetic, transcription and post-transcriptional levels by various mechanisms, such as chromatin remodeling, DNA modification, transcription inhibition and RNA-RNA interactions, etc. In recent years, studies have shown that many lncRNAs can be induced by viruses or interferon (IFN) and to regulate the expression of related antiviral genes in IFN-mediated antiviral innate immune responses. In this review, we focus on the regulation of lncRNAs in IFN-mediated antiviral innate immune responses, especially in the transcription of IFN-stimulated genes (ISGs). In addition, we summarize the regulatory network of lncRNAs, IFN and ISGs. This review will provide a valuable reference for the researchers working in this field.

A trinuclear cyanide-bridged luminescent compound, trans-RuII(DMAP)4(CN)2[(PY5Me2)Mn]2[PF6]4 (1-R), and its oxidation product, ferromagnetic trans-RuIII(DMAP)4(CN)2[(PY5Me2)Mn]2-[PF6]5 (1-O), were synthesized and fully characterized. This is the first example of a molecular material showing a redox-controlled transformation between fluorescence and ferromagnetism.

An adverse intrauterine environment may be an important factor contributing to the development of type 2 diabetes in later life. The present study investigated the longitudinal effects of maternal lipopolysaccharide (LPS) exposure during the third trimester on glucose metabolism and sex hormone balance in the offspring. Pregnant mice were intraperitoneally injected with LPS (50 µg/kg) daily from gestational day (GD) 15 to GD17. Glucose tolerance test (GTT) and insulin tolerance test (ITT) were assessed at postnatal day (PND) 60 and PND120. Sex hormones, their receptors, and metabolic enzymes (aromatase) were measured in male offspring at different phases of development (PND14: juvenile; PND35: adolescence; PND60: adulthood; and PND120: middle age). LPS-exposed male offspring exhibited glucose intolerance and insulin resistance by GTT and ITT at middle age, accompanied by an increase in fasting blood glucose and reductions in serum insulin levels and hepatic phosphorylated (p) -AKT/AKT ratio. However, glucose intolerance and insulin resistance were not observed in LPS-exposed female offspring. Maternal LPS exposure upregulated hepatic aromatase proteins and mRNA levels in male offspring at all time points. At adolescence, the testosterone/estradiol ratio (T/E2) was markedly reduced in LPS-exposed male offspring. Moreover, maternal LPS exposure significantly increased hepatic estrogen receptor (ER) α expressions and decreased hepatic androgen receptor (AR) expressions in male offspring. At adulthood, maternal LPS exposure increased serum estradiol levels, decreased serum testosterone levels and elevated hepatic ERß expressions in male offspring. In conclusion, maternal LPS exposure upregulated aromatase expressions, followed by a reduction in the T/E2 ratio and an alteration in sex hormone receptor activity, which might be involved in the development of glucose metabolism disorders in middle-aged male offspring. This study provides a novel clue and direction to clarify the pathogenesis of maternal infection-related diabetes in male offspring.

The purposes of this study were to determine the prevalence of heifer clinical mastitis (HCM) and heifer subclinical mastitis (HSCM), isolate HCM-causing bacteria, and assess the association of some risk factors in Huanggang, Central China. A total of 1,374 lactating heifers from three dairy farms were examined in the present study; 22.64% of heifers were positive for mastitis, out of which, 3.86 and 18.78% were with HCM and HSCM, respectively. Of the 67 HCM samples, 91.05% were single growth, 7.46% mixed growth, and 1.49% no bacterial growth. Coagulase-negative staphylococci (CNS) accounted for 30.98% of the isolates followed by Escherichia coli (29.58%), Staphylococcus aureus (16.9%), and Streptococcus uberis (11.27%). Logistic regression analyses showed that heifer milk yield lower than 3 L had the highest prevalence of HCM compared to heifer milk yield of more than 3 L (P < 0.01), and prevalence of HCM was significantly high in heifers with the presence of teat lesions (P < 0.01). Moreover, the heifer milk yield lower than 3 L had the highest prevalence of HSCM compared to heifer milk yield of more than 3 L (P < 0.01). These two factors were significantly associated with the occurrence of heifer mastitis, which needs to be considered in the prevention and control strategies of the disease.

Plasma exchange (PE) for the treatment of ricin toxicity has not been previously reported. Here we describe the use of PE to treat children who experienced ricin toxicity after ingesting castor beans. Seven children (median age: 8.1 years) who consumed castor beans (median: 5 beans) were treated with PE. All had bradycardia and sinus arrhythmia, and most had experienced episodes of vomiting and/or diarrhea. PE settings were blood flow, 50-80 mL/min; PE rate, 600-800 mL/h; volume of exchange, 1440-1950 mL. Median time from ingestion to PE was 73 h. All clinical symptoms disappeared and vital signs rapidly returned to normal after PE; no severe organ dysfunction occurred. All children were discharged and recovered uneventfully. Concentrations of all serum biochemical parameters significantly decreased immediately after PE. Some, but not all, of these parameters were also significantly decreased at 48 and 72 h after PE compared with before PE. Our findings suggest that PE can be an effective early intervention in the treatment of ricin toxicity due to castor bean ingestion.

OBJECTIVE: To evaluate the clinical efficacy and safety of integrative medical program based on blood cooling and detoxification recipe (BCDR) in treating patients with hepatitis B virus related acute-on-chronic liver failure (HBV-ACLF) of heat-toxicity accumulation syndrome (HTAS). METHODS: Adopting randomized controlled clinical design, a total of 105 HBV-ACLF patients of HTAS were randomly assigned to the trial group (64 cases) and the control group (41 cases). Patients in the control group were treated with comprehensive Western therapy, while those in the trial group were treated with comprehensive Western therapy plus BCDR. All were treated for 8 weeks and followed up for 40 weeks. Effect and safety of the treatment were assessed, including fatality, liver functions [total bilirubin (TBIL), albumin (ALB), alanine aminotransferase (ALT), and aspartate transaminase (AST)], and prothrombin activity (PTA) after treatment and at week 48 of follow-ups. RESULTS: After 8-week treatment, there was statistical difference in the overall fatality rate (15.63% vs 34.15%), the fatality rate in the mid-term (25.0% vs 64.7%), TBIL at week 8 (64.54 +/- 79.75), AST [at week 2: (178.97 +/- 44.24) U/L vs (288.48 +/- 58.49) U/L; at week 4: (61.65 +/- 27.36) U/L vs (171.12 +/- 89.11) U/L] and PTA [at week 4: (58.30 +/- 15.29) vs (42.56 +/- 15.27); at week 6: (60.77 +/- 20.40) vs (43.08 +/- 12.79)] (all P < 0.05). At week 48 of the followup, the fatality rate of the trial group (21.88%) decreased by 17. 14% when compared with that of the control group (39.02%; P < 0.05). No obvious adverse event occurred in the two groups during the 8-week treatment period. CONCLUSION: BCDR could significantly reduce the mortality of HBV-ACLF patients.
