RESUMO

There is a critical shortage of blood available for transfusion in many low- and middle-income countries. The consequences of this scarcity are dire, resulting in uncounted morbidity and mortality from trauma, obstetric hemorrhage, and pediatric anemias, among numerous other conditions. The process of collecting blood from a donor to administering it to a patient involves many facets from donor availability to blood processing to blood delivery. Each step faces particular challenges in low- and middle-income countries. Optimizing existing strategies and introducing new approaches will be imperative to ensure a safe and sufficient blood supply worldwide.

A balanced, diverse gut microbiota is vital for animal health. The microbial population is shaped by multiple factors including genetic background and environment, but other determinants remain controversial. Numerous studies suggest that the dominant factor is genetic background while others emphasize the environmental factors. Here, we bred asexual hybridization queens (AHQs) of honeybees through nutritional crossbreeding (laid in Apis mellifera colony but bred in Apis cerana colony), sequenced their gut microbiome, and compared it with normally bred sister queens to determine the primary factor shaping the gut microbiota. Our results showed that the dominant genera in the gut microbiota of AHQs were Brevundimonas, Bombella, and Lactobacillus, and its microbial community was more related to A. mellifera queens. The AHQs had a moderate number of different bacterial species and diversity, but total bacterial numbers were low. There were more significant taxa identified in the comparison between AHQ and A. cerana queen according to LEfSe analysis results. The only genetic-specific taxon we figured out was Brevundimonas. The growth of core bacterial abundance showed different characteristics among different queen groups in the first week after emerging. Collectively, this study suggested that the genetic background played a more dominant role than environmental factors in shaping the gut microbiota of honeybee queen and the microbiota of midgut was more sensitive than that of rectum to this impact.

Background: Recently, a new coronavirus has been rapidly spreading from Wuhan, China. Forecasting the number of infections scientifically and effectively is of great significance to the allocation of medical resources and the improvement of rescue efficiency. Methods: The number of new coronavirus infections was characterized by "small data, poor information" in the short term. The grey prediction model provides an effective method to study the prediction problem of "small data, poor information". Based on the order optimization of NHGM(1,1,k), this paper uses particle swarm optimization algorithm to optimize the background value, and obtains a new improved grey prediction model called GM(1,1|r,c,u). Results: Through MATLAB simulation, the comprehensive percentage error of GM(1,1|r,c,u), NHGM(1,1,k), UGM(1,1), DGM(1,1) are 2.4440%, 11.7372%, 11.6882% and 59.9265% respectively, so the new model has the best prediction performance. The new coronavirus infections was predicted by the new model. Conclusion: The number of new coronavirus infections in China increased continuously in the next two weeks, and the final infections was nearly 100 thousand. Based on the prediction results, this paper puts forward specific suggestions.

Synthesis of multiple stimuli-responsive magnetic nanomaterials in a green way remains as a big challenge currently. Herein, temperature-responsive elastin-like polypeptides (ELPs) were designed to involve in the biomimetic mineralization and successfully prepared magnetic nanoparticles (MNPs) (named ELPs-MNPs) with multiple responsiveness (temperature, magnetic, and biomimetic silicification responsiveness) in one pot. ELPs-MNPs were identified as cubic nanomaterials with an average size of about 32 nm and in line with the classic ferromagnetic behavior. Interestingly, ELPs-MNPs show clearly lower critical solution temperature phase behavior with a transition temperature of 36 °C. Moreover, ELPs-MNPs can spontaneously trigger the biosilicification of tetramethyl orthosilicate (TMOS) to entrap themselves into silicon oxide as proved by the Fourier transform infrared spectra (FTIR) and elemental mapping of transmission electron microscopy (TEM), with an average size of about 62 nm. The possible role of ELPs in the biomimetic preparation of the multiple stimuli-responsive MNPs was also addressed. The proposed novel and simple one-pot strategy to synthesize multifunctional nanomaterials with higher effectiveness is the first report for preparing MNPs with multiple stimuli response. This strategy conforms to the concept of green chemistry and will pave a new way for the design of smart biomaterials, which may have great potentials for different fields.

Identifying ways to deal with the challenges presented by aging is an urgent task, as we are facing an aging society. External factors such as diet, exercise and drug therapy have proven to be major elements in controlling healthy aging and prolonging life expectancy. More recently, the intestinal microbiota has also become a key factor in the anti-aging process. As the intestinal microbiota changes with aging, an imbalance in intestinal microorganisms can lead to many age-related degenerative diseases and unhealthy aging. This paper reviews recent research progress on the relationship between intestinal microorganisms and anti-aging effects, focusing on the changes and beneficial effects of intestinal microorganisms under dietary intervention, exercise and drug intervention. In addition, bacteriotherapy has been used to prevent frailty and unhealthy aging. Most of these anti-aging approaches improve the aging process and age-related diseases by regulating the homeostasis of intestinal flora and promoting a healthy intestinal environment. Intervention practices based on intestinal microorganisms show great potential in the field of anti-aging medicine.

PURPOSE: To develop a novel resin for provisional prostheses using hyperbranched polyurethane acrylate (HBPUA) and triethylene glycol dimethacrylate (TEGDMA) with promising mechanical properties and low volumetric shrinkage. METHODS: Four groups including TIH3-0 (100 wt% TEGDMA), TIH3-30 (30 wt% HBPUA + 70 wt% TEGDMA), TIH3-60 (60 wt% HBPUA + 40 wt% TEGDMA), and TB-60 (60 wt% bisphenol A-glycidyl dimethacrylate + 40 wt% TEGDMA) were prepared and commercial Luxatemp (DMG) was used for comparison. Fourier transform infrared spectroscopy and gel permeation chromatography were used for material characterization. Mechanical properties including microhardness, flexural strength, flexural modulus, and load energy were measured before and after water immersion. Physical properties measurement included weight changes, solubility, water absorption, surface hydrophobicity, and volumetric shrinkage. Finally, biocompatibility was evaluated using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay. RESULTS: The number- and weight-average molecular weights of the HBPUA were approximately 870 and 1480, respectively. The addition of HBPUA to TEGDMA increased the mechanical strength considerably. Although the weight changes and water absorption of TIH3-60 were higher than those of Luxatemp, the microhardness, flexural strength, flexural modulus, load energy, solubility, shrinkage, and biocompatibility of TIH3-60 were either comparable or superior to those of Luxatemp. CONCLUSION: Based on the findings of the present study, TIH3-60 has potential for development as a new provisional material.

This paper is the first in the two-part series quantitatively modelling human grasp functionality and understanding the way human grasp objects. The aim is to investigate the thumb movement behavior influenced by object shapes, sizes, and relative positions. Ten subjects were requested to grasp six objects (3 shapes × 2 sizes) in 27 different relative positions (3 X deviation × 3 Y deviation × 3 Z deviation). Thumb postures were investigated to each specific joint. The relative position (X, Y, and Z deviation) significantly affects thumb opposition rotation (Rot) and flexion (interphalangeal (IP) and metacarpo-phalangeal (MCP)), while the object property (object shape and size) significantly affects thumb abduction/adduction (ABD) motion. Based on the F value, the Y deviation has the primary effects on thumb motion. When the Y deviation changing from proximal to distal, thumb opposition rotation (Rot) and flexion (IP and MCP joint) angles were increased and decreased, respectively. For principal component analysis (PCA) results, thumb grasp behavior can be accurately reconstructed by first two principal components (PCs) which variance explanation ratio reached 93.8% and described by the inverse and homodromous coordination movement between thumb opposition and IP flexion. This paper provides a more comprehensive understanding of thumb grasp behavior. The postural synergies can reproduce the anthropomorphic motion, reduce the robot hardware, and control dimensionality. All of these provide a more accurate and general basis for the design and control of the bionic thumb and novel wearable assistant robot, thumb function assessment, and rehabilitation.

Zinc-finger protein 304 (ZNF304) plays a critical role in silencing genes through transcription, regulating cell survival, proliferation, apoptosis, and differentiation during development. However, the roles of transcription factor ZNF304 and its clinical significance in clear cell renal carcinoma (ccRCC) remain unclear. In this study, we found that the expression of ZNF304 was downregulated in ccRCC tissues. Lower levels of ZNF304 were correlated with poor survival. Downregulation of ZNF304 promoted ccRCC cell growth in vitro, whereas overexpression of ZNF304 inhibited growth. Our results indicated that miR-183-5p/FOXO4 mediated ZNF304 regulation of cell growth. Interestingly, we revealed that ZNF304 promoted FOXO4 expression in ccRCC cells. Mechanistically, ZNF304 binds to miR-183 promoter and inhibits miR-183-5p transcription. Furthermore, the expression of miR-183-5p wes increased in ccRCC tissues, and the upregulation of miR-183-5p was related to the poor prognosis of ccRCC patients. miR-183-5p upregulation repressed the expression of FOXO4 and promoted ccRCC progression. These results demonstrated that ZNF304/miR-183-5p/FOXO4 axis played essential role in promoting ccRCC progression, which suggests that disruption of this axis may be a potential therapeutic target in ccRCC.

BACKGROUND: The microbiota of the female reproductive tract is increasingly recognized as playing fundamental roles in animal reproduction. To explore the relative contribution of reproductive tract microbiomes to egg production in chickens, we investigated the microbiota in multiple reproductive and digestive tract sites from 128 female layer (egg-producing) chickens in comparable environments. RESULTS: We identified substantial differences between the diversity, composition, and predicted function of site-associated microbiota. Differences in reproductive tract microbiota were more strongly associated with egg production than those in the digestive tract. We identified 4 reproductive tract microbial species, Bacteroides fragilis, Bacteroides salanitronis, Bacteroides barnesiae, and Clostridium leptum, that were related to immune function and potentially contribute to enhanced egg production. CONCLUSIONS: These findings provide insights into the diverse microbiota characteristics of reproductive and digestive tracts and may help in designing strategies for controlling and manipulating chicken reproductive tract microbiota to improve egg production.

OBJECTIVE: To compare clinical efficacy between anatomical locking plate (ALP) and ordinary steel plate (OSP) in treating closed calcaneal fractures with SandersⅡ and Ⅲ. METHODS: From May 2016 to May 2018, 68 patients with closed Sanders typeⅡ and Ⅲ calcaneal fractures were retrospectively analyzed, and were divided into anatomical locking plate group (ALP group) and ordinary steel plate group (OSP group) according to two kinds of plate fixation, and 34 patients in each group. In ALP group, there were 21 males and 13 females aged from 20 to 63 years old with average of (35.16±8.45) years old; 14 patients were typeⅡand 20 patients were type Ⅲaccording to Sanders classification;treated with ALP. In OSP group, there were 20 males and 14 females aged from 19 to 63 years old with average of (35.05±8.39) years old;19 patients were typeⅡand 15 patients were type Ⅲ according to Sanders classification;treated with OSP. Operative time, intraoperative blood loss and complications between two groups were observed and compared;preoperative and postoperative Böhler angle and gissane angle were also compared;American Orthopaedic Foot & Ankle Society (AOFAS) ankle and hind foot scores, foot and ankle disability index (FADI) scores were applied to evaluate clinical effect. RESULTS: All patients were followed up from 11 to 14 months with an average of (12.06±0.81) months. There were no statistical differences in opertive time, intraoperative blood loss, incision infection and refracture rate in complications between two groups (P>0.05);while there was significant difference in the number of screw loosening (P<0.05). Böhler angle and Gissane angle in ALP group at 6 and 12 months after opertaion were higher than that of OSP group (P<0.05), and the degree of improvement of Böhler angle and Gissane angle in ALP group were also higher than that of OSP group (P<0.05). Postopertaive AOFAS score and FADI score at 6 and 12 months in ALP group were higher than that of OSP group (P<0.05), while no statistical difference in AOFAS grading between two groups(P>0.05). CONCLUSION: Compared with OSP, ALP in treating SandersⅡ and Ⅲ calcaneal fractures could achieve better therapeutic effect, avoid screw loosening, reduce complications, and improve limb function in further.

Global climate change is a major threat to biodiversity, which may increase the extinction risk of rare species, particularly those like Ostrya rehderiana Chun (Betulaceae) with very few remaining extant wild individuals. We aimed to estimate the potential distribution of O. rehderiana under climate change and to analyze possible relevant climatic factors. Maximum entropy (Maxent) was employed to model the potential distribution of O. rehderiana under present and future climate scenarios. Suitable habitat areas in different periods and the main contributing climate factors were identified using species distribution models. The minimum temperature in winter and precipitation seasonality were the principal climatic factors influencing the establishment of O. rehderiana. The proportion of high potential distribution area in China was 3.91% and would further shrink significantly under changing climate, especially reduce by 97% under high radiative forcing. The extinction risk of O. rehderiana would still be extraordinarily high under future climate scenarios. The Tianmu and Luoxiao Mountains would be the only potential refugia for O. rehderiana in the future. Special conservation efforts are urgently required to rescue extremely endangered species as O. rehderiana. We propose priorities for the conservation region and suggestions for conservation management strategies.

Monosodium urate (MSU) crystals, the etiological agent of gout, are formed in joints and periarticular tissues due to long-lasting hyperuricemia. Although MSU crystal-triggered NLRP3 inflammasome activation and interleukin 1ß (IL-1ß) release are known to have key roles in gouty arthritis, recent studies revealed that MSU crystal-induced necrosis also plays a critical role in this process. However, it remains unknown what forms of necrosis have been induced and whether combined cell death inhibitors can block such necrosis. Here, we showed that MSU crystal-induced necrosis in murine macrophages was not dependent on NLRP3 inflammasome activation, as neither genetic deletion nor pharmacological blockade of the NLRP3 pathway inhibited the necrosis. Although many cell death pathways (such as ferroptosis and pyroptosis) inhibitors or reactive oxygen species inhibitors did not have any suppressive effects, necroptosis pathway inhibitors GSK'872 (RIPK3 inhibitor), and GW806742X (MLKL inhibitor) dose-dependently inhibited MSU crystal-induced necrosis. Moreover, a triple combination of GSK'872, GW806742X, and IDN-6556 (pan-caspase inhibitor) displayed enhanced inhibition of the necrosis, which was further fortified by the addition of MCC950 (NLRP3 inhibitor), suggesting that multiple cell death pathways might have been triggered by MSU crystals. Baicalin, a previously identified inhibitor of NLRP3, inhibited MSU crystal-induced inflammasome activation and suppressed the necrosis in macrophages. Besides, baicalin gavage significantly ameliorated MSU crystal-induced peritonitis in mice. Altogether, our data indicate that MSU crystals induce NLRP3-independent necrosis, which can be inhibited by combined inhibitors for multiple signaling pathways, highlighting a new avenue for the treatment of gouty arthritis.

The ancestor of ß-1,3-xylanases (AncXyl09) were reconstructed by the optimized ancestral sequences reconstruction strategy to solve the poor catalytic performances of existing ß-1,3-xylanases. The results showed that the half-life at 50 °C was 65.08 h, indicating good thermostability. The large number of hydrogen bonds and the disulfide bonds were the major attributes related with the thermal stability of Anxyl09. Interestingly, AncXyl09 could hydrolyze lichen besides the original substrate of ß-1, 3-xylan, which is the first reported ß-1,3-xylanase with substrate promiscuity. Moreover, the hydrolytic products are mainly disaccharides, the content of ß-1,3-xylobiose and lichoridiose more than 70% as determined by high performance liquid chromatography (HPLC), which could significantly facilitate the separation and purification of oligosaccharides. The successful design of AncXyl09 was the representative of the semi-rationally engineered ß-1, 3-xylanase, which will shield a new light on the ß-1,3-xylanase engineering, active oligosaccharide preparation and marine algae resource utilization.

OBJECTIVE: To investigate the correlation between short-term imaging techniques and long-term function using receiver operating characteristic (ROC) curve after tibia fracture surgery. METHODS: Seventy patients with tibia fracture admitted to our hospital were selected as the study subjects, and were divided into a < 60 years old group (n = 40) and a ≥ 60 years old group (n = 30), a removed internal fixation group (n = 26) and a retained internal fixation group (n = 44). The short-term imaging results, long-term function, the degree of pain, the range of ankle joint motion, and the subjective satisfaction of patient care were scored and compared among the four groups. The influencing factors of subjective satisfaction of patients were analyzed using logistic regression analysis. RESULTS: There were marked differences in the scores of Jakim, Garland & Werley scoring system, and Kofoed's scale and subjective satisfaction between the < 60 years old group and the ≥ 60 years old group, and between the removed internal fixation group and the retained internal fixation group (P < 0.05), but there was no remarkable difference in visual analog scale (VAS) scores (P > 0.05). Spearman's correlation analysis revealed a correlation between the short-term imaging manifestations and the long-term function in tibia fracture patients, and the correlation was remarkable in patients aged < 60 years without internal fixation (P < 0.05). Logistic correlation analysis showed that Kofoed's scale scores were significantly correlated with patients' subjective satisfaction (P < 0.05). CONCLUSION: There is a correlation between early imaging and long-term function in tibia fracture patients, and the correlation was remarkable in patients aged < 60 years and without internal fixation. The range of joint motion and degree of pain are the primary influencing factors for evaluation of patients' subjective satisfaction.

OBJECTIVE: To investigate the clinical values of magnetic resonance imaging (MRI), computed tomography (CT) and X-ray in differential diagnosis of benign and malignant bone metastases. METHODS: A total of 80 patients with bone metastases admitted to our hospital were selected as the study subjects. X-ray, CT and MRI examinations were performed, respectively. The pathological examination results were taken as the gold standard to analyze the lesion sites of metastatic tumors in 80 subjects. The diagnostic sensitivity, specificity and accuracy of X-ray, CT and MRI examinations were calculated and compared. RESULTS: Among the 80 patients, 71 cases were diagnosed as malignant bone metastases and 9 cases as benign lesions according to pathological examination. The diagnostic sensitivity, specificity and accuracy of X-ray, CT and MRI examinations were 63.38%, 33.33% and 60.00%, 84.51%, 66.67% and 82.50%, and 90.14%, 77.78% and 88.75%, respectively. MRI was superior to X-ray in the sensitivity, specificity and accuracy of differential diagnosis of benign and malignant bone metastases (P < 0.05). MRI was superior to CT, but there was no significant difference between the two groups (P < 0.05). The diagnostic sensitivity, specificity and accuracy of CT + MRI were significantly higher than those of any single detection method (P < 0.05). CONCLUSION: MRI, CT and X-ray exhibit a good value in differential diagnosis of benign and malignant bone metastases. Overall, MRI is superior to CT and X-ray in the screening effects, and the combined detection methods are more satisfactory. It is recommended that CT should be used for primary screening, and CT + MRI should be implemented for a enhanced diagnosis.

Vermicompost was used as an additive in swine manure composting to investigate the expression of bacterial functional genes on nutrients biotransformation. Three treatments with vermicompost compositions of 10%, 20%, and 30% in swine manure were set up. Raw manure was used as the control. The thermophilic period increased to 12 days, the NH4+ -N/NO3- -N ratio decreased to 0.85, and the germination index (GI) increased to 166% after vermicompost addition. Furthermore, higher relative abundances of Firmicutes were observed in the substrate during the initial stages of experiment. The abundance of the dominant phylum Proteobacteria and its related pathogenic genera Acinetobacter and Stenotrophomonas decreased in the thermophilic stage while the potentially beneficial genera Actinomadura and Chryseolinea increased. The expression of primary functional genes associated with the metabolism of carbohydrates, amino acids, xenobiotics, and fatty acids was enhanced during the thermophilic phase. Besides, most dominant genera showed strengthened correlations with NO3--N and GI, which were the strongest environmental factors for bacterial communities. Network analysis revealed a new metabolic pathway associated with dominant genera Pseudomonas, Acinetobacter, Stenotrophomonas, and Oceanobacter, whose abundance increased with vermicompost addition. Collectively, the results of this study indicate that vermicompost can promote composting efficiency by increasing the potentially beneficial bacteria, decreasing pathogenic bacteria, and enhancing the metabolic capacity of bacterial communities.

OBJECTIVE: To assess the awareness, attitude and barriers of colorectal cancer screening among high-risk populations in China. DESIGN: A cross-sectional study was employed. SETTING: This study was conducted in nine hospitals in Hunan province, China. PARTICIPANTS: Individuals with a high-risk for colorectal cancer were interviewed using a pretested structured questionnaire. PRIMARY AND SECONDARY OUTCOME MEASURES: Knowledge, attitude towards colorectal cancer screening, sociodemographic factors associated with screening knowledge and behaviour and barriers of colorectal cancer screening. RESULTS: This study included 684 participants. The mean knowledge score was 11.86/24 (SD 4.84). But over 70% of them held a positive attitude towards screening. Only 13.3% had undergone colorectal cancer screening. Independent factors related to knowledge were education level of college or above, working as a white collar, higher income, having health insurance, having seen a doctor in the past year and with a high perceived risk (p<0.05). Factors independently associated with screening behaviour included personal history of colorectal disease, having seen a doctor in the past year, previous discussion of colorectal cancer screening, high perceived risk and better knowledge (p<0.05). Main reasons for not undergoing screening were no symptoms or discomfort (71.1%), never having thought of the disease or screening (67.4%) and no doctor advised me (29.8%). CONCLUSION: In China, the majority of high-risk people had deficient knowledge and had never undergone colorectal cancer screening. But most of them held a positive attitude towards the benefits of colorectal cancer screening. This has promising implications to design targeted educational campaigns and establish screening programmes to improve colorectal cancer awareness and screening participation. Healthcare professionals should advise high-risk individuals to participate in screening and inform them about cancer risk.

RESUMO

The development of biotechnology and the in-depth research on disease mechanisms have led to increased application of enzymes in the treatment of diseases. In addition, enzymes have shown great potential in drug manufacturing, particularly in production of non-natural organic compounds, due to the advantages of mild reaction conditions, high catalytic efficiency, high specificity, high selectivity and few side reactions. Moreover, the application of genetic engineering, chemical modification of enzymes and immobilization technologies have further improved the function of enzymes. This review summarized the advances of using enzymes as drugs for disease treatment or as catalysts for drug manufacturing, followed by discussing challenges, potential solutions and future perspectives on the application of enzymes in the medical and pharmaceutical field.

A comprehensive transcriptomic survey of pigs can provide a mechanistic understanding of tissue specialization processes underlying economically valuable traits and accelerate their use as a biomedical model. Here we characterize four transcript types (lncRNAs, TUCPs, miRNAs, and circRNAs) and protein-coding genes in 31 adult pig tissues and two cell lines. We uncover the transcriptomic variability among 47 skeletal muscles, and six adipose depots linked to their different origins, metabolism, cell composition, physical activity, and mitochondrial pathways. We perform comparative analysis of the transcriptomes of seven tissues from pigs and nine other vertebrates to reveal that evolutionary divergence in transcription potentially contributes to lineage-specific biology. Long-range promoter-enhancer interaction analysis in subcutaneous adipose tissues across species suggests evolutionarily stable transcription patterns likely attributable to redundant enhancers buffering gene expression patterns against perturbations, thereby conferring robustness during speciation. This study can facilitate adoption of the pig as a biomedical model for human biology and disease and uncovers the molecular bases of valuable traits.

MicroRNA-124 (miR-124) has been predicted as a tumor suppressor in esophageal squamous cell carcinoma (ESCC). However, factors contributing to miR-124 reduction remain unclear. Circular RNAs (circRNAs) are a new family of non-coding RNAs with gene regulatory potential via interacting with miRNAs. We predicted three circRNAs, including CircRNA_14359, CircRNA_2646, and CircRNA_129, that could interact with miR-124 by bioinformatics analysis and determined their expressions in ESCC tissues and adjacent normal tissues. We found that CircRNA_2646 was up-regulated in ESCC, negatively correlated with the expression of miR-124 and positively associated with TNM stage and lymph node metastasis of ESCC. Luciferase reporter assay showed that CircRNA_2646 interacted with miR-124 in ESCC Eca109 and TE-1 cells. Moreover, ectopical overexpression of CircRNA_2646 accelerated cell proliferation, migration, invasion, and epithelial-to-mesenchymal transition (EMT), but restoration of miR-124 abrogated these functions and promoted Bcl-2-dependent cell apoptosis. Furthermore, it was found that the oncogene Proteolipid Protein 2 (PLP2) was the target gene of miR-124. In Eca109 and TE-1 cells, restoration of miR-124 decreased the level of PLP2 and inhibited PLP2-induced cell proliferation, migration, invasion, and EMT, but enhanced cell apoptosis. The in vivo study confirmed that CircRNA_2646 promoted ESCC development by repressing miR-124 and activating PLP2. Taken together, we identified that CircRNA_2646 functioned as an inhibitor in miR-124 signaling pathway in ESCC for carcinogenesis and could be a promising target for ESCC therapy.
