OBJECTIVE: Breast cancer has been proven as the most common malignancy influencing the health of females. This research aimed to clarify the effects of high-mobility group box 3 (HMGB3)-small interfere RNA (HMGB3-siRNA) on the proliferation of breast cancer cells. MATERIALS AND METHODS: HMGB3-mimic and HMGB3-siRNA lentiviral vectors were structured. The above lentiviral vectors were then transfected into normal breast cells (MCF10A) and breast cancer cells (MDA-MB-231). Cell counting kit-8 (CCK-8) analysis was employed to assess proliferative viabilities of cells. The formation of the mammosphere in breast cancer cells was examined using mammosphere-forming assay. The mRNA expression of Nanog, Sox2, and OCT-4 genes was evaluated using quantitative real time-PCR (qRT-PCR). CD44 positive/CD24 negative (CD44+/CD24-) cell levels were evaluated using flow cytometry assay. The correlation between HMGB3 and hypoxia-inducible factor 1α (HIF1α) was analyzed using Linear-Regression analysis. The interaction between HMGB3 and HIF1α expression was determined using the Dual-Luciferase assay. RESULTS: HMGB3 expression was remarkably enhanced in breast cancer cells compared to that in normal cells (p<0.05). HMGB3-siRNA significantly decreased the proliferative activity and remarkably suppressed the mammosphere formation compared to that in single MDA-MB-231 cells (p<0.05). HMGB3-siRNA remarkably reduced Nanog, SOX2, and OCT-4 and significantly enhanced CD44+/CD24- cells compared to single MDA-MB-231 cells (p<0.05). HMGB3-siRNA significantly weakened the expression of HIF1α in MDA-MB-231 cells compared to single MDA-MB-231 cells (p<0.05). HMGB3 was positively correlated with HIF1α expression (p<0.05). There was an interaction between HMGB expression and HIF1α expression. CONCLUSIONS: HMGB3 small interfering RNA suppressed the formation of mammosphere in MDA-MB-231 cells by downregulating the expression of HIF1α.

RESUMO

Surgical margin principle is one of the central surgical principles for head and neck cancers. Negative surgical margin is the primary purpose of surgery for head and neck cancers. This paper is based on the current clinical application of surgical margins of head and neck cancers, and it is proposed that surgical margins of head and neck cancers may be classified into three types: surface margin, deep margin, and tissue interface margin in consideration of surgical exposure of tumors. The classification of surgical margins can make the clinical application and the research of surgical margins of head and neck cancers more systematic and clear.

RESUMO

Most of the common dizziness/vertigo diseases in otology clinic are peripheral vertigo diseases, and while about 20% -30% of all vertigo diseases are caused by central nervous system diseases. Pseudo vestibular neuritis, acute ischemic stroke with audiovestibular loss, and central paroxysmal positional vertigo are the three types of central "malignant vertigo" diseases which are most easily misdiagnosed by otologists. This article described the clinical characteristics and bedside physical performance of these three diseases, and summarized the differential diagnosis points for clinical reference.

Objective: To evaluate the prevalence and risk factors of aortic valve calcification among the elderly (≥65 years old) resident of Wuxi city, Jiangsu province. Methods: The household registration population aged ≥65 years old in Wuxi city was selected as the research subject by stratified sampling method from August 2017 to December 2018. Echocardiography was performed to assess the aortic valve calcification, and the participants were divided into calcification group and non-calcification group. Multivariate logistic regression analysis was used to explore the related risk factors of aortic valve calcification. Results: The age of the respondents was (73.6±7.1) years old, of which 48.8% (461 cases) were males.The prevalence rate of aortic valve calcification was 22.0% (208/944) in the elderly (≥ 65 years old) residents in Wuxi city. The prevalence rate in 65-69 years old, 70-74 years old, 75-79 years old, 80-84 years old and ≥85 years old was 16.7% (58/347),16.7% (41/245),16.2% (26/161),23.3% (24/103), and 67.0% (59/88),respectively. There were significant differences in age, weight, abdominal circumference, hip circumference, high-salt diets, exercise, hypertension, hyperlipidemia, diabetes, coronary heart disease, cerebrovascular disease, and carotid atherosclerosis between the non-calcified group (736 cases) and the calcified group (208 cases) (P<0.01 or 0.05).Multivariate logistic regression analysis showed that age (OR=1.077, 95%CI 1.053-1.101, P<0.001), diabetes mellitus (OR=1.697, 95%CI 1.174-2.453, P=0.005), and coronary heart disease (OR=1.964, 95%CI 1.378-2.799, P<0.001) were the risk factors of aortic valve calcification. Conclusions: The prevalence of aortic valve calcification in the elderly (≥65 years old) residents in Wuxi city of Jiangsu province increases with aging. Age, diabetes mellitus and coronary heart disease are the risk factors of aortic valve calcification in this population cohort.

Objective:To investigate the clinical features of LC-BPPV with direction-fixed horizontal nystagmus in the primary Roll test. Method:Nine patients of LC-BPPV with bilateral direction-fixed horizontal nystagmus in the first Roll test were treated by repositioning maneuvers after judging the affected side and the prognosis was observed. Result:The affected side of eight patients was the side of the apogeotropic nystagmus. Barbecue or Gufoni maneuvers was effective for reposition. The other one had no conversion of nystagmus and could not judge the affected side. Conclusion:The nystagmus performance of patients with LC-BPPV is complicated. It is necessary to carefully analyze the nystagmus performance of each position. Combining with bow and lean test, lying down test and null plane, the position of the otolith is inferred. Comprehensive use of the Barbecue or Gufoni maneuvers, forced side lying on the affected side, mastoid sniper, shaking head method is effective to complete the reposition treatment.

Objective: To understand status of amphetaminetype stimulants (ATS) use among residents aged 15-64 in a border city of Yunnan province. Methods: Using the stratified cluster random sampling method, a total of 3 130 residents were recruited through both anonymous questionnaire and interview, regarding their health-related behaviour and ATS use. Results: Among 3 130 residents aged 15-64 years in this city, the overall prevalence rates of ATS use were 4.0% (126/3 130) in the lifetime and 2.6% (82/3 130) in the past one year, while the prevalence of ATS use disorder in the past year was 2.3% (73/3 130). The prevalence rates of lifetime ATS use and in the past year were 7.5% (108/1 443) and 5.1% (73/1 443) in the high epidemic area, 7.3% (122/1 682) and 4.8%(80/1 682) in males, 5.2% (118/2 260) and 3.4% (77/2 260) in 18-45 age group, 4.6%(63/1 361) and 3.4% (46/1 361) in the ones having had elementary school education, 10.3% (50/487) and 8.6% (42/487) in unmarried group, 17.1% (19/111) and 12.6% (14/111) in either divorced, widowed or separated group, 4.8% (108/2 256) and 3.2% (72/2 256) in the farmers group, 6.0% (99/1 643) and 4.4% (73/1 643) in the non-religious groups (neither Buddhism nor Christianity), 15.2% (97/639) and 11.9% (76/639) in groups with negative hobbies which were 1.3% (29/2 314) and 0.3%(6/2 314) higher than the ones with positive hobbies, 8.5% (84/992) and 6.3% (62/992) in the ones negatively managing the stress which were 2.0% (42/2 138) and 0.9% (20/2 138) higher than the ones that treating the stress in a positive way. Conclusions: The prevalence rates of ATS use and ATS use disorder in the past one year appeared high among those residents aged 15-64 years in the border city of Yunnan province. Prevention and intervention programs should be carried out for the target groups as soon as possible.

Epigenetic mutations caused by pollutants are possibly linked to many diseases. Benzo(a)pyrene (BaP) is one of the most representative air pollutants and has aroused wide concern because of its strong carcinogenicity. The reproductive toxicity induced by BaP has been identified, but little is known about the characteristics of the methylation changes induced by BaP. In this study, a methylated DNA immunoprecipitation sequencing method was used to detect the methylation of sperm DNA of rats exposed to BaP. Compared with the respective genes in normal rats, there were 3227 hypomethylated genes and 828 hypermethylated genes after BaP exposure. Gene ontology enrichment analysis reported that differentially methylated genes (DMGs) were enriched in the localization, single-multicellular organism process and plasma membrane. Kyoto Encyclopedia of Genes and Genomes pathway analysis showed that the DMGs were significantly enriched in the Ras signalling pathway, Rap1 signalling pathway, pancreatic secretion and neuroactive ligand-receptor interaction. DisGeNET disease spectrum analysis showed that DMGs were associated with infertility and certain genetic diseases. Further research needs to be done to explore whether these abnormal methylation are transgenerational.

Objective:This study aims to investigated the clinical significance of room tilt illusion(RTI) in screening for central vertigo in otology clinic. Method:A retrospective study of 8 RTI cases out of 656 patients with dizziness and/or vertigo. The characteristics of the symptoms, signs, physical examination, and other examinations of RTI patients were analyzed. Result:①Patients with RTI complaints accounted for 1.2% of all patients with dizziness and/or vertigo in the same period. Of all 8 patients, 2 were with acute vestibular syndrome, 5 were with episodic vestibular syndrome, and one had no dizziness and/or vertigo attack. ②Two was diagnosed with acute cerebellar infarction, and 3 was diagnosed with posterior circulation transient ischemic attacks, 2 were diagnosed with definite vestibular migraine, and one was unable to identify the cause. ③All 8 patients had a sensation of environment tilting, which was episodic(single or repeated), transient(seconds to minutes), in the coronal plane, and with tilt angle 30-180°. ④Seven patients had dizziness and/or vertigo for minutes to hours after a transient RTI, and one patient had no other symptoms after RTI. ⑤All patients had no decrease in vHIT gain, 6 patients had abnormal oculomotor function, and one patient had mild horizontal semicircular canal paresis. Conclusion:Patients with RTI complaints mainly had central dizziness and vertigo diseases, and once are found in otology clinic, should be alert to the possibility of suffering from central diseases.

Objective: To survey the conduction and evaluate the effectiveness of extracorporeal membrane oxygenation (ECMO) therapy in pediatric intensive care unit (PICU) in China mainland. Methods: In a questionnaire-based survey, we retrospectively reviewed the application of ECMO in children's hospital and general hospital in China mainland to summarize and analyze the categories of diseases and prognosis of children treated with ECMO therapy. Results: By December 31, 2017, a total of 23 hospitals using ECMO, including 22 tertiary referral hospitals and 1 secondary hospital, among which 16 were children's hospitals and 7 were general hospitals. Thirty-seven ECMO equipment was available. A total of 518 patients treated with ECMO, within whom 323 (62.4%) successfully weaned from ECMO and 262 (50.6%) survived to discharge. Among 375 pediatric patients, 233 (62.1%) were successfully weaned from ECMO and 186 (49.6%) survived to discharge. Among 143 newborn patients, 90 (62.9%) successfully weaned from ECMO, 76 (53.1%) survived to discharge. ECMO was applied in veno-arterial (VA) mode to 501 (96.7%) patients, veno-venous (VV) mode to 14 (2.7%) patients, and VV-VA conversion mode to 3 (0.6%) patients. Sixty-nine patients required extracorporeal cardiopulmonary resuscitation (ECPR), including 20 newborn patients (29.0%) and 38 pediatric patients (71.0%), who were all with cardiovascular disease. Neonatal respiratory distress syndrome (26/61), persistent pulmonary hypertension of the newborn (PPHN) (12/61), and meconium aspiration syndrome (MAS) (11/61) are the most common pulmonary diseases in newborn patients; among whom, infants with PPHN had highest survival rate (10/12), followed by MAS (9/11). Among newborn patients with cardiovascular diseases, those who admitted were after surgery for congenital cardiac disease were the most common (54/82), while those with septic shock had the highest survival rate (2/3). In pediatric pulmonary diseases, acute respiratory distress syndrome was the most common (42/93), while plastic bronchitis was with the highest survival rate (4/4), followed by viral pneumonia (13/16). Among pediatric cardiovascular diseases, congenital cardiac defect was the most common (124/282), while fulminant myocarditis had the highest survival rate (54/77). Conclusion: The application of ECMO as a rescue therapy for children with severe cardiopulmonary failure has dramatically developed in China mainland.

OBJECTIVE: Grape seed proanthocyanidin extract (GSPE), as one of the most popular natural drug extracted from the grape, has been reported to improve endothelial function and arteriosclerosis. However, little is known about the influence of GSPE on hypertension and vascular remodeling. Profilin-1, an Actin-binding protein, is closely involved in the remodeling of large vessels in ouabain-induced hypertension. To date, there is no effective prevention or treatment in place for the high incidence of ischemic stroke associated with hypertension. In this study, we aimed to determine the role of GSPE via inhibition Profilin-1 in ischemic cerebral cortices of ouabain-hypertension rats and potentially provide a new target to prevent stroke associated with hypertension. MATERIALS AND METHODS: The blood pressure of male Sprague-Dawley (SD) rats was measured during a period of ouabain-induced hypertension. The expression of Profilin-1, vascular endothelial growth factor (VEGF) and endothelial nitric oxide synthase (eNOS) in the cerebral cortex were determined by quantitative Real Time-PCR (qRT-PCR) and Western blot. Histopathological and behavioral tests were also conducted. RESULTS: Blood pressure elevation started at week 5 and reached clinical standards for hypertension at week 8. GSPE was proved to suppress Profilin-1 and VEGF levels through inhibition of Profilin-1-protein kinase B (AKT)-hypoxia inducible factor-1α (HIF-1α) signal pathway and promote eNOS expression. Moreover, the histopathological and ethiological improvement was observed in GSPE over-expression and Profilin-1 inhibition groups. CONCLUSIONS: We detected that GSPE could improve cerebral vascular damage through inhibiting Profilin-1 in an ouabain-induced hypertension model.

OBJECTIVE: To investigate whether miR-374b can promote the differentiation of MSCs into osteoblasts by mediating PTEN, thus promoting fracture healing. MATERIALS AND METHODS: Primary cultured mouse mesenchymal stem cells were obtained for following experiments. Flow cytometry was used to determine the expression of MSCs surface antigens to identify the purity. Alizarin red staining was used to detect whether MSCs could differentiate into osteoblasts. QRT-PCR was used to detect the expression of osteogenic marker genes as well as miR-374b and PTEN in bone marrow-derived MSCs from fractured mice model. ALP activity detection kit was used to detect ALP activity in cells. Changes of osteogenic proteins in cells were evaluated by Western blot. Bioinformatics methods were used to predict the binding sites between miR-374 and target genes. Luciferase reporter assay was used to confirm whether miR-374 could bind to the target gene. RESULTS: Under normal culture, MSCs grew into a long fusiform shape on the 4th day. After induced in the osteogenic induction medium for seven days, calcified nodules appeared. The results of the detection of MSCs surface antigen markers showed that CD90 was 99.12%, and CD45 was 0.23%. Alizarin red staining showed that MSCs possess the ability to differentiate into osteogenic. The expression level of MiR-374b and PTEN increased significantly in the early stage of fracture in mice, but no significant difference was observed at a later stage. After overexpression of miR-374b, the cell ALP activity, the expression of osteogenesis-related genes and osteogenesis-related proteins was significantly increased, while after knocking out miR-374b, the opposite result was observed. The result of luciferase reporting assay showed that miR-374b can bind to PTEN. As mentioned above, overexpression of miR-374b resulted in upregulation of osteogenic-related genes and proteins, while over-expression of both PTEN and miR-374b could partly reverse the outcomes. CONCLUSIONS: miR-374b can promote osteogenic differentiation of MSCs by degrading PTEN, thereby promoting fracture healing.

Objective:To understand the expression of LINC00460 in laryngeal squamous cell carcinoma (LSCC), and to discuss the role of LINC00460 in carcinogenesis and development of laryngeal squamous cell carcinoma. Method:The expression of LINC00460 in 68 laryngeal squamous cell carcinoma tissues and paired adjacent normal tissues were examined by real-time PCR. The relationship between the expression levels of LINC00460 and the clinicopathological features including clinical stage, histological grade, pathological type, and lymph node metastasis in LSCC was analyzed according to the clinical data. Result:The expression of LINC00460 was significantly upregulated in the LSCC tissue compared with that of adjacent normal mucosal tissue (P=0.006). There were no statistical differences of the quantity of LINC00460 expression among supraglottic, glottic and subglottic LSCC (P>0.05). Moreover, LINC00460 had no significant changes in poorly differentiated LSCC when compared with that of well and moderately differentiated LSCC (P>0.05). Furthermore, the expression of LINC00460 in LSCC with lymph node metastasis had no significant changes when compared with that without of lymph node metastasis (P>0.05). Notably, LINC00460 expression in T1+T2 stages patients were significantly lower than T3+T4 stages (P<0.05). Conclusiont:Upregulation of long non-coding RNA LINC00460 might contribute to the carcinogenesis and development of LSCC, furthermore play important biological function.

The maximum content of cation vacancies in perovskites determines the maximum adjustment to the physical properties. By using the classical criterion of the tolerance factor, we put forward a new method to obtain the maximum content of cation vacancies in perovskites. The effective ionic radius was calculated using the weighted average method, and the limit of tolerance factors was used to determine the maximum value of the cation vacancies. The calculated results of the La1-xMnO3 and La0.75Sr0.25Mn1-yO3 materials coincide with experimental results. For application, [(A1)1-x(A2)x]1-z1(B1B2)X3 or [(A1)1-x(A2)x](B1B2)1-z2X3 (X = O, F, Cl, Br, I) inorganic and hybrid organic-inorganic perovskite materials with vacancies at the A or B sites were predicted successfully, which provides a new method to improve the physical properties of perovskite materials.

AIMS: To evaluate the effect of ultraviolet (UV) disinfection on antibiotic-resistant Escherichia coli. METHODS AND RESULTS: Antibiotic-resistant E. coli strains were isolated from a wastewater treatment plant and subjected to UV disinfection. The effect of UV disinfection on the antibiotic resistance profiles and the antibiotic resistance genes (ARGs) of antibiotic-resistant E. coli was evaluated by a combination of antibiotic susceptibility analysis and molecular methods. Results indicated that multiple-antibiotic-resistant (MAR) E. coli were more resistant at low UV doses and required a higher UV dose (20 mJ cm-2 ) to enter the tailing phase compared with those of antibiotic-sensitive E. coli (8 mJ cm-2 ). UV disinfection caused a selective change in the inhibition zone diameters of surviving antibiotic-resistant E. coli and a slight damage to ARGs. The inhibition zone diameters of the strains resistant to antibiotics were more difficult to alter than those susceptible to antibiotics because of the existence and persistence of corresponding ARGs. CONCLUSIONS: The resistance of MAR bacteria to UV disinfection at low UV doses and the changes in inhibition zone diameters could potentially contribute to the selection of antibiotic-resistant bacteria in wastewater treatment after UV disinfection. The risk of spread of antibiotic resistance still exists owing to the persistence of ARGs. SIGNIFICANCE AND IMPACT OF THE STUDY: Our study highlights the acquisition of other methods to control the spread of ARGs.

Objective:To investigate the expression of LINC00261 in laryngeal carcinoma and to discuss their relevance and the roles in carcinogenesis and development of laryngeal carcinoma. Method:The expressions of LINC00261 in laryngeal carcinoma tissue and paired adjacent normal tissue was determined by real-time PCR. The relationship between the expressions of LINC00261 and the clinic pathological characteristics including clinical stage, pathological type, histological grade and lymph node metastasis in LSCC was analysed according to the clinical data. Result:①The expression of LINC00261 was significantly decreased in the LSCC tissue compared with the normal laryngeal tissue(P<0.01).②In clinical stage grouping, there were no statistical differences of the quantity of LINC00261 expression among supraglottic, glottic and subglottic LSCC(P>0.05).In histological differentiation grouping, LINC00261 had no significant changes in poorly differentiated LSCC compared with the well and moderately differentiated LSCC(P>0.05). In histological grade grouping, the expression of LINC00261 in T1+T2 stages was significantly higher than T3+T4 stages(P<0.05). Moreover, the expression of LINC00261 in LSCC with lymph node metastasis was significantly lower than that without of lymph node metastasis(P<0.05). Conclusion:Down regulation of LINC00261 in laryngeal carcinoma may contribute to the carcinogenesis and development of laryngeal carcinoma. The decreased expression of LINC00261 maybe relative to T term degree and lymphamatic metastasis of laryngeal carcinoma.

Objective: To summarize clinical experience of using extracorporeal membrane oxygenation (ECMO) in rescuing children with acute fulminant myocarditis (AFM). Method: Data of 12 children with acute fulminate myocarditis (6 boys and 6 girls, median age 8.3 (0.6, 13.0) years, median weight 33.1 (6, 61) kg) who were rescued with ECMO in Children's Hospital, Zhejiang University from September 2009 to August 2015 were analyzed retrospectively. The analysis focused on the intervene timing of ECMO for the cardiogenic shock and hypoperfusion caused by heart failure and(or) lethal arrhythmia and the essentials of ECMO cardiopulmonary resuscitation(ECPR) for cardiac arrest in pediatric AFM were summarized. Result: The median ECMO duration was 110(22, 240) h. Ten cases survived and 2 were dead of the total of 12 patients. Six ECPR patients survived and 2 were dead in the total of 8 ECPR patients. The complication of 10 survivors were cannula site bleeding (3 cases), hypernatremia and intracranial hemorrhage (1 case), limping (1 case), hoarse voice (1 case), and cerebral injury (1 case). Conclusion: The key points of improving ECMO rescuing outcome for the AFM children are grasping the ECMO intervene timing and training skilled ECMO team. For ECPR patients, keeping effective chest compressions resuscitation is the key to achieve survival and improve the quality of life.

OBJECTIVE: To review the use of non-open chest extracorporeal membrane oxygenation (ECMO) in pediatric intensive care unit (PICU) in China. METHOD: The survey was conducted in 28 tertiary hospitals in China mainland from March to October 2015. All children <18 years of age have been supported with non-open chest ECMO in PICU were reviewed.Patient demographics, diagnosis, indication for ECMO, details of ECMO support, complications, and patient survival were analyzed. All the patients were divided according to age into pediatric patients (age>28 d) and neonatal patients (age 0-28 d). For non-normally distributed measurement data, two groups were compared using independent samples of the Mann Whitney U test and for categorical data constitute ratio were compared by χ(2) test or Fisher's exact test. RESULT: A total of 63 patients received non-open chest ECMO support during this study, including 51 pediatric patients and 12 neonates. For 51 pediatric patients, their mean age was 55.5 (15.0-117.0) months, and mean weight was 17.5 (10.0-32.9) kg. Cardiac failure was the primary indication in 28 patients, respiratory failure in 21 patients, and both cardiac and respiratory in 2 patients. Patients with cardiac disease had a lower mortality rate compared with cases with respiratory disease (21%(6/28) vs. 67% (14/21), χ(2)=9.145, P=0.002). The average length of ECMO run was 112.0 (74.5-175.2) h, and 96.7(76.2-139.5)h for cardiac patients, 149.0(78.9-241.0)h for patients with respiratory disease. There were no significant difference between patients with cardiac disease and patients with respiratory disease in ECMO support time (Z=1.476, P=0.140). Forty-two patients (82%) were decanulated from ECMO successfully, and thirty-one (61%) patients survived to hospital discharge. The most common complications during ECMO run were bleeding, hemolysis and disfunction of oxygenation. Of the 25 (49%) survivors whom we followed up, 8 (17%) experienced obvious sequelae, and 5 (10%) had neurologic problems. Of twelve neonates, their mean weight was(3.2±0.5)kg. The primary cause of ECMO was neonatal respiratory distress syndrome(7 cases). All of the neonatal patients were treated with veno-arterial (VA)-ECMO. The mean duration of ECMO support was 88.4 (45.50-110.25) h. Seven patients were decanulated from ECMO successfully, five survived to hospital discharge. CONCLUSION: ECMO support can significantly improve the prognosis of pediatric and neonatal patients with refractory respiratory and cardiac failure. More efforts are needed on patient selection, experienced team establishment and ECMO therapy technology improvement need further improvement in China in the future.

OBJECTIVE: To construct human coagulation factor Ⅸ mini-gene (Mini-hF9) and some nonsense mutants, detect the levels of the Mini-hF9 mRNA, and analyze the molecular mechanism of microRNA125 regulating F9 gene with nonsense mutation. METHODS: Three nonsense mutants were obtained by using PCR mutagenesis to analyze the mechanism of plasma thromboplastin component recognition. The Mini-hF9 gene mRNA levels were detected by Real-time PCR in mammalian cells co-transfected with nonsense mutant expression vectors and miR-125 mimics. RESULTS: Mini-hF9 gene was constructed successfully and cloned into the mammalian expression vector. The only normal transcript was detected in cells transfected with the Mini-hF9 gene expression vectors. Three nonsense mutants, M1 (nt 34 G>T in Exon 7), M2 (nt 52 G>T in Exon 7) and M3 (nt 85 G>T in Exon 7), were obtained by using PCR mutagenesis. The levels of the Mini-hF9 mRNA decreased to 14.1% (t=15.464, P=0.004) in M1 and 22.4% (t=15.755, P=0.004) in M2 mutants after transfection, respectively. It was proved to be caused by nonsense-mediated mRNA decay (NMD) in CHX experiment. The levels of Mini-hF9 mRNA increased to 1.70 times (t=-4.883, P=0.039) and 2.40 times (t=-17.537, P=0.003) in M1 mutant after miR-125a or miR-125b mimics treatment, respectively. The levels of Mini-hF9 mRNA increased to 2.02 times (t=-19.264, P=0.003) and 2.07 times (t=-9.158, P=0.012) in M2 mutant after miR-125a or miR-125b mimics treatment, respectively. CONCLUSION: Nonsense mutant location is a key determinant for triggering NMD. MicroRNA125 could improve the stability of Mini-hF9 mRNA in M1 and M2 mutants by repressing NMD. MicroRNA125, a short non-coding RNA molecule, could be a potential therapeutic target in conditions caused by the NMD pathway.
