This study investigated the effects of dietary supplementation with Bacillus subtilis (B. subtilis) or Bacillus licheniformis (B. licheniformis) on growth performance, immunity, antioxidant capacity, short chain fatty acid (SCFA) production, and the cecal microflora in broiler chickens. In total, 360 male, 1-day-old Cobb 500 birds were randomly divided into 3 groups: the control group was fed a basal diet; the B. subtilis group was fed a basal diet supplemented with 1.5 × 109 CFU/kg B. subtilis; the B. licheniformis group was fed a basal diet supplemented with 1.5 × 109 CFU/kg B. licheniformis. Results showed that chickens supplemented with either B. subtilis or B. licheniformis had comparatively higher (P < 0.05) body weight and average daily gain, whereas no difference (P > 0.05) was observed in feed efficiency. Concentrations of serum IgA, IgY, and IgM, as well as anti-inflammatory IL-10 were significantly increased (P < 0.05), and proinflammatory IL-1ß and IL-6 were significantly decreased (P < 0.05) by B. subtilis or B. licheniformis supplementation. Moreover, chickens fed with diets supplemented by either B. subtilis or B. licheniformis had greater antioxidant capacity, indicated by the notable increases (P < 0.05) in glutathione peroxidase, superoxide dismutase, and catalase, along with decrease (P < 0.05) in malondialdehyde. Compared to the control group, levels of SCFA, excluding acetic and propionic acid, in cecal content had improved (P < 0.05) by adding B. licheniformis, and significant increase (P < 0.05) in acetic and butyric acid was observed with B. subtilis supplementation. Microbial analysis showed that both B. subtilis or B. licheniformis supplementation could increase butyrate-producing bacteria such as Alistipes and Butyricicoccus, and decrease pathogenic bacteria such as the Synergistetes and Gammaproteobacteria. In summary, dietary supplemented with B. subtilis or B. licheniformis improved growth performance, immune status, and antioxidant capacity, increased SCFA production, and modulated cecal microbiota in chickens. Moreover, B. licheniformis was more effective than B. subtilis with the same supplemental amount.

Antibiotic overuse in poultry husbandry poses a potential threat to meat safety and human health. Lauric acid (LA) is a primary medium-chain fatty acid (MCFA) with a strong antibacterial capacity. The goal of this study was to evaluate the beneficial effects of LA on the growth performance, immune responses, serum metabolism, and cecal microbiota of broiler chickens. One-day-old male Ross 308 broilers were randomly divided into 4 groups: CON, fed a basal diet; ANT, a basal diet supplemented with 75 mg/kg antibiotic; LA500, a basal diet supplemented with 500 mg/kg LA; LA1000, a basal diet supplemented with 1000 mg/kg LA. The feeding period was 42 d. The results showed that LA significantly improved broiler growth and immune functions, as evidenced by increased body weight (BW) and average daily gain (ADG), enhanced intestinal mucosal barrier, upregulated immunoglobulins (IgA, IgM, and IgY), and downregulated inflammatory cytokines (IL-1ß, IL-6, TNF-α, IL-4, and IL-10) (P < 0.05). HPLC/MS-based metabolome analysis revealed that the serum metabolites in the LA group differed from those of CON and ANT groups. LA markedly decreased the abundance of phosphatidylcholines (PCs), increased lysophosphatidylcholines (LysoPCs), and inhibited the sphingolipid metabolism pathway, indicating its capacity to modulate lipid metabolism. 16S rRNA sequencing indicated that LA significantly altered cecal microbiota composition by reducing Phascolarctobacterium, Christensenellaceae_R-7_group, and Bacteroides, and increasing Faecalibacterium and Ruminococcaceae_UCG-014 (P < 0.05). Furthermore, Spearman correlation analysis revealed that changes in metabolism and microbiota were highly correlated with the growth and immune indices; strong links were also found between lipid metabolism and microbial composition. Taken together, LA promotes broiler growth and immune functions by regulating lipid metabolism and gut microbiota. The above findings highlight the substantial potential of LA as a supplement in poultry diets and provide a new strategy to reduce antibiotic usage and improve food safety.

BACKGROUND: Rhamnolipids (RLS), well known as glycolipid biosurfactants, display low toxicity, high biodegradability, and strong antibacterial properties. This study was carried out to evaluate the use of RLS supplementation as a substitute for antibiotics, and particularly to evaluate its effects on growth performance, immunity, intestinal barrier function, and metabolome composition in broilers. RESULTS: The RLS treatment improved the growth performance, immunity, and intestinal barrier function in broilers. The 16S rRNA sequencing revealed that the genus Alistipes was the dominant genus in broilers treated by RLS. An ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF/MS)-based metabolomic analysis indicated that the sphingolipid metabolism, glycine, serine, and threonine metabolism, the gycerophospholipid metabolism, and the tryptophan metabolism were changed in broilers that were treated with RLS. CONCLUSION: l-Tryptophan may be the medium for RLS to regulate the growth and physiological metabolism. Rhamnolipids can be used as a potential alternative to antibiotics, with similar functions to antibiotics in the diet of broilers. The optimal level of supplemented RLS in the diet was 1000 mg kg-1 . © 2021 Society of Chemical Industry.

AIM: This study was conducted to investigate the effects of Clostridium butyricum in isolation or in combination with 1, 25-dihydroxyvitamin D3 in early-stage broilers. METHODS AND RESULTS: A total of 360 half male and half female Cobb broilers (1 day old) were randomly divided into four groups: Con (basal diet), Anti (basal diet+75 mg/kg chlortetracycline), Cb (basal diet+109  CFU per kg C. butyricum) and CD (basal diet+109  CFU per kg C. butyricum+25 µg/kg 1,25(OH)2 D3 ). The results were as follows: (1) Compared with Con, CD significantly increased ADG (p < 0.05). (2) Contrast with Con and Anti, Cb and CD significantly increased glutathione peroxidase and SOD in the serum and liver, and decreased malondialdehyde content in serum (p < 0.05). (3) In addition, the content of immunoglobulin (IgA, IgY and IgM) in Cb and CD birds was higher than that in Con birds (p < 0.05); the Cb supplementation decreased (p < 0.05) the contents of IL-8, IL-1ß and TNF-α than those in Con. (4) Cb and CD had lower caecal acetic and propionic content than the Anti group (p < 0.05). (5) The community richness of Con was significantly higher than that of Anti (p < 0.05). The relative abundance of Alistipes and Ruminococcaceae-UCG-014 in Cb and CD supplemented birds were lower than those in Con (p < 0.05). The relative abundant of Escherichia-Shigella in CD was higher than Con and Anti (p < 0.05). CONCLUSIONS: These data indicated that dietary C. butyricum and 1, 25-dihydroxyvitamin D3 can improve the growth performance, immunity responses, antioxidation, bone development and intestinal microflora in early-stage broilers. SIGNIFICANCE AND IMPACT OF THE STUDY: Oral administration of C. butyricum or C. butyricum combined with 1,25-dihydroxyvitamin D3 enhanced immunity and antioxidant activity in early-stage birds.

This study evaluated the effects of Bacillus coagulans (B. coagulans) as an alternative to antibiotics on growth performance, antioxidant capacity, immunity function and gut health in broilers. A total of 480 one-day-old broilers were randomly divided into 3 treatments with 8 replicates comprising 20 broilers each. The experiment lasted 42 d. Treatments included: basal diet without antibiotics (NCO); basal diet supplemented with 75 mg/kg chlortetracycline (ANT); basal diet supplemented with 5 × 109 CFU/kg B. coagulans(BC). The B. coagulans enhanced body weight (BW) and average daily gain compared with the NCO group (P < 0.05). However, there were no significant differences in average daily feed intake and feed: gain ratio (F: G) among three groups (P > 0.05). The B. coagulans significantly increased catalase, superoxide dismutase, and glutathione peroxidase levels and reduced malondialdehyde levels (P < 0.05). The serum immunoglobulins (IgA, IgM, and IgY) were significantly higher in the BC group when compared to the NCO and ANT groups (P < 0.05). The B. coagulans also markedly reduced serum levels of proinflammatory factors (IL-1ß, IL-6, and TNF-α) and enhanced anti-inflammatory factor (IL-10) concentrations compared with control group (P < 0.05). Moreover, compared with the control group, BC significantly inhibited serum xanthine oxidase activity (P < 0.05). The levels of acetic acid, propionic acid, butyrate, isobutyric acid and valerate in BC group were significantly increased on d 42 compared with the NCO and ANT groups (P < 0.05). Furthermore, BC significantly altered cecal microbiota by reducing Desulfovibrio and Parasutterella, and by increasing Alistipes and Odoribacter (P < 0.05, P < 0.05, P < 0.001, P < 0.01, respectively). In conclusion, dietary B. coagulans, when used as an alternative to antibiotics, improved body weight, average daily gain, antioxidant capacity, immunity function and gut health in broilers.

A series of 5-phenyloxazole-2-carboxylic acid derivatives were synthesized, and their structure-activity relationships (SARs) were studied. N,5-diphenyloxazole-2-carboxamides 6, 7, and 9, which mimicked ABT751, showed improved cytotoxicity compared with ABT751. Compound 9 exhibited the highest antiproliferative activities against Hela A549, and HepG2 cancer cell lines, with IC50 values of 0.78, 1.08, and 1.27 µM, respectively. Furthermore, compound 9 showed selectivity for human cancer cells over normal cells, and this selectivity was greater than those of ABT751 and colchicine. Preliminary mechanism studies suggested that compound 9 inhibited tubulin polymerization and led to cell cycle arrest at G2/M phase. Molecular docking studies indicated that compound 9 bound to the colchicine binding site of tubulin. Our findings provided insights into useful SARs for further structural modification of inhibitors of tubulin polymerization.

van der Waals (vdW) heterojunctions stacked by different two-dimensional (2D) layered materials not only exhibit the complementary effect of short plates, but also harbor novel physical phenomena. In particular, the emergence of 2D magnetic vdW materials has provided novel opportunities for the application of these materials in spintronics. However, to the best of our knowledge, to date, the spin-related transport mechanism in magnetic tunnel junctions (MTJs) based on these 2D vdW magnetic materials and the effect of pinning layers on their transport properties have not been elucidated by the non-equilibrium state theory. Herein, based on first-principles calculations, we report the spin-polarized quantum transport properties of sandwich-type vdW magnetic tunnel junctions (CrI3/h-BN/n·CrI3) comprising monolayer CrI3, a hexagonal boron nitride (h-BN) spacer layer, and n-layer CrI3 (n = 1, 2, 3, and 4). Considering the inter-layer antiferromagnetic coupling in n-layer CrI3, a few layers of CrI3 can be regarded as its own natural pinning layers. Especially, when n is equal to 3, an almost fully spin-polarized current and large tunnel magnetoresistance ratio (3600%) are obtained in the equilibrium state. Excitingly, due to different numbers of pinning layers in MTJs, the transport properties of these MTJs at positive bias voltages exhibit an interesting odd-even effect within a limited thickness of these pinning layers. Moreover, an almost perfect spin filtering effect and remarkable negative differential resistance (NDR) were observed in the MTJs where n was odd (n = 1 and 3). The observed non-equilibrium quantum transport phenomenon is explained by spin-dependent transmission coefficient at different bias voltages. Our results provide effective guidance for the experimental studies of the MTJs based on 2D magnetic vdW materials.

To investigate the bioactive compounds that contribute to the α-glucosidase inhibitory activity of rosemary, phenolics and triterpene acids were characterized and quantified using quadrupole-Orbitrap mass spectrometry and enzyme assay. Two phenolic diterpenes (carnosol and hydroxy p-quinone carnosic acid) and two triterpene acids (betulinic acid and ursolic acid) were identified as potent α-glucosidase inhibitors. Carnosol, a major diterpene in rosemary, showed significant α-glucosidase inhibitory activity with IC50 value of 12 µg mL-1, and its inhibition mode was competitive. The inhibition mechanism of carnosol on α-glucosidase was further investigated by a combination of surface plasmon resonance (SPR) spectroscopy, fluorescence quenching studies and molecular-modeling techniques. The SPR assay suggested that carnosol had a high affinity to α-glucosidase with equilibrium dissociation constant (KD) value of 72.6 M. Fluorescence quenching studies indicated that the binding between carnosol and α-glucosidase was spontaneous and mainly driven by hydrophobic forces. Molecular docking studies revealed that carnosol bound to the active site of α-glucosidase. Furthermore, the oral administration of carnosol at 30 mg kg-1 significantly reduced the postprandial blood glucose levels of normal mice. This is the first report on the α-glucosidase inhibition and hypoglycemic activity of phenolic diterpenes, and these results could facilitate the utilization of rosemary as a dietary supplement for the treatment of diabetes.

The thermophysical properties of refrigerant can be modified via adding solid materials to it. In this paper, molecular simulations and thermodynamic calculations were employed to investigate the adsorption and energy storage of ethane (R170), 1,1,1,2-tetrafluoroethane (R134a), 1,1,1-trifluoroethane (R143a), and 1,1-difluoroethane (R152a) in metal organic framework (MOF)-5 nanoparticles. The results show that the fluorine atom in the refrigerants will strengthen the adsorption of refrigerants in MOF-5. However, the fluorine-free refrigerant, R170, owns larger enthalpy difference of desorption than the other refrigerants with fluorine under high pressure. The thermal energy storage capacity of the refrigerant/MOF-5 mixture is larger than that of the pure refrigerant at low pressure. Also, the negative enhancement of the energy storage property of the mixture is found in some cases when the refrigerant experiences phase transition.

Improving the production traits and resistance against mastitis in dairy cattle is a challenge for animal scientists across the globe. The present study was designed to investigate the genetic effects of single nucleotide polymorphisms (SNPs) in Janus kinase 2 (JAK2) and diacylglycerol acyltransferase (DGAT1) genes with production and mastitis-related traits. Four SNPs in JAK2 and one in DGAT1 were analyzed through Chinese Cow's SNPs Chip-I (CCSC-I) and genotyped in a population of 312 Chinese Holsteins. Our findings demonstrated that milk fat percentage, somatic cell count (SCC), somatic cell score (SCS), serum cytokines interleukin 6 (IL-6) and interferon gamma (IFN-γ) showed significant associations (P < 0.05) with at least one or more identified SNPs. Consequently, the analysis based on haplotypes amongst the SNPs in JAK2 revealed noteworthy (P < 0.05) association with SCC and IL-6. Collectively, our results verified the pleiotropic ability of detected SNPs in bovine JAK2 and DGAT1 for milk fat percentage as well as mastitis-related traits. The significant SNPs in both the genes could serve as powerful genetic markers to minimize mastitis risk. In addition, besides SCC and SCS, the IFN-γ and IL-6 could also be used as indicators of improved genetic resistance against mastitis.

Accurate measurement of overvoltage in power grids is of great significance to study the characteristics of overvoltage and design of insulation coordination. Based on the research of D-dot voltage sensor, we designed a Dual-Differential D-dot overvoltage sensor. In order to quantify the structural parameters of the sensor, improve the performance and measurement accuracy of the sensor. The Field-Circuit Coupling method was proposed to be used in the parameter design of D-dot overvoltage sensor. The joint simulation of space electromagnetic field model and equivalent circuit model of the Dual-Differential D-dot overvoltage sensor was established with the finite element simulation software Ansoft Maxwell and circuit simulation software Simplorer. Finally, the actual sensor was manufactured. A test platform was built to verify the steady-state and transient performance of the sensor. The results show that the Dual-Differential D-dot sensor has excellent steady-state and transient performance, the error of phase and amplitude are small, and the sensor can achieve the non-contact measurement of power transmission line. Simultaneously, the rationality of the Field-Circuit Coupling method was further verified.

The search for more types of band inversion-induced topological states is of great scientific and experimental interest. Here, we proposed that the band inversion between px,y and pz orbitals can produce a topological phase transition in honeycomb lattices based on tight-binding model analyses. The corresponding topological phase diagram was mapped out in the parameter space of orbital energy and spin-orbit coupling. Specifically, the quantum anomalous Hall (QAH) effect could be achieved when ferromagnetism was introduced. Moreover, our first-principles calculations demonstrated that the two systems of half-iodinated silicene (Si2I) and one-third monolayer of bismuth epitaxially grown on the Si(111)-√3 ×√3 surface are ideal candidates for realizing the QAH effect with Curie temperatures of ∼101 K and 118 K, respectively. The underlying physical mechanism of this scheme is generally applicable, offering broader opportunities for the exploration of novel topological states and high-temperature QAH effect systems.

The recent studies of magno-assisted tunnelling in ferromagnetic van der Waals heterostructures formed by graphene and ultrathin CrBr3 films (D. Ghazaryan et al., Nat. Electron., 2018, 1, 344) offer broader opportunities for exploration of novel quantum phenomena, especially for realizing the graphene-based quantum anomalous Hall effect (QAHE). Based on first-principles approaches, we reveal that three types of graphene/CrBr3 (Gr/CrBr3) heterostructures exhibit metallic band behavior due to strong charge-transfer at the interfaces of these heterosystems. Remarkably, the pressure-induced QAHE can be achieved in Gr/CrBr3 and CrBr3/Gr/CrBr3 systems. Further low energy k·p model analyses show that the nontrivial topological properties are mainly attributed to the Rashba spin-orbit coupling (SOC), but not to the intrinsic SOC of graphene. Moreover, a multichannel device prototype is proposed in the superlattices composed of Gr/CrBr3 and normal insulator (such as hexagonal boron nitride) layers. Our work provides an experimentally feasible scheme for realizing the high-temperature and multichannel QAHE in graphene-based heterostructures.

This study was conducted to evaluate the effects of dietary supplementation with yeast extract on growth, body composition, non-specific immunity, and antioxidant status of Chinese mitten crab (Eriocheir sinensis). A total of 432 crabs (initial average weight, 4.62 ±â€¯0.11 g) were randomly distributed into four treatment groups with six replicates of 18 crabs. The crabs were fed a basal diet or the same diet supplemented with 2.5, 5, and 10 g/kg yeast extract for 8 weeks. The results showed that dietary yeast extract inclusion enhanced the edible viscera index (linear, P < 0.001), edible viscera crude protein (CP) content (linear, P = 0.025) and serum phenoloxidase (ProPO) activity (quadratic, P = 0.023) at 56 day, increased the total superoxide dismutase (T-SOD) activity at 28 day (quadratic, P = 0.037) and catelase (CAT) activity at 56 day (quadratic, P = 0.034) of edible viscera, and muscular T-SOD activity (quadratic, P = 0.020) at 56 day in Chinese mitten crab. Compared with the control group, the inclusion of 5 g/kg yeast extract in the diet increased the edible viscera index, enhanced the CAT activity of edible viscera at 56 day in Chinese mitten crab (P < 0.05). Dietary 10 g/kg yeast extract inclusion enhanced the edible viscera index at 56 day in Chinese mitten crab than that of the control group (P < 0.05). These results implied that dietary yeast extract inclusion improved the edible viscera index and crude protein content of edible viscera, enhanced serum immunity, and increased the antioxidant status of edible viscera and muscle in Chinese mitten crab, especially when it is supplemented at 5 g/kg yeast extract in the diet.

OBJECTIVE: To investigate the characteristics of recurrence of T1G3 transitional cell carcinoma (TCC) of bladder. METHODS: The clinical data of 72 patients with TCC of bladder of the stage T1, 37 being of the stage T1G3 and 35 being of the stage T1G1-2, who underwent the initial treatment by transurethral resection of bladder tumor plus post-operative bladder infusion were analyzed. RESULTS: The first, second, third, and fourth year recurrent rates of the T1G3 group were 29.7%, 40.5%, 18.9%, and 10.8% respectively, all significantly higher than those of the T1G2 group (14.3%, 34.3%, 31.4%, and 20.0% respectively, t = 1.994, P = 0.025). The rate of recurrence for only time was 10.8% in the T1G3 group (4/37), significantly lower than that of the T1G1-2 group (17/35, 48.6%, P < 0.01). The rate of recurrence for more than 3 times was 54.0% in the T1G3 group (20/37), significantly higher than that of the T1G1-2 group (6/35, 17.2%, P < 0.01). The rate of recurrent tumor at the pathological stage over T2 was 86.5% (32/37) in the T1G3 group, higher than that of the T1G1-2 group (42.8%, 15/35). The rate of partial or total cystectomy at the last recurrence in the T1G3 group was 94.6% (35/37), significantly higher than that of the T1G1-2 group (57.1%, 20/35, chi(2) = 14.00, P < 0.05). CONCLUSION: A kind of dangerous bladder cancer, TCC at the stage T1G3 is apt recur soon after the treatment with a high recurrent frequency. Most recurrent tumors evolve into invasive cancer.

OBJECTIVE: To investigate separation and purification of skeletal muscle satellite cells with improved incontinuous density Percoll gradient centrifugation technique. METHODS: The primary skeletal muscle satellite cells of New Zealand white rabbits were cultured with different adhesion time method and incontinuous density Percoll gradient centrifugation technique. The cells were observed under invert microscope and scanning electron microscope. The degree of purification was examined by celluar immunochemical stain. The growth curve was tested by thiazolyl blue assay. RESULTS: Over 90% satellite cells were harvested by incontinuous density Percoll gradient centrifugation technique, in contrast to which, only 30%-40% cells were harvested by different adhesion time. Morphological observation accorded with satellite cells. The growth curve indicated that the cells grew in a good status. CONCLUSION: The high purification satellite cells can be obtained by incontinuous density Percoll gradient centrifugation technique. It is a good method to culture seed cells for tissue engineering applications.

OBJECTIVE: To investigate the preparation method of porcine bladder acellular matrix graft (BAMG) and evaluate the feasibility of using BAMG as biomaterial scaffold to construct tissue engineering bladder. METHODS: Bladders were harvested from adult pigs at a slaughterhouse and were decellularized by the method of enzyme digestion or eradicator washing. The BAMG was then examined by optic microscope and electron microscope to confirm no cell elements remained. The structure of the BAMG was always observed through the Scanning Microscope. The skeletal muscle stem cells of New Zealand white rabbits were implanted in 96-hole-plank and cultured by BAMG extract liquid or normal culture medium. The cytotoxicity of BAMG was tested through MTT assay. New Zealand white rabbits were underwent bladder triangle area spared cystectomy and reconstructed by the heterogeneity BAMG. The animal bladders were obtained after six weeks and the tissue regeneration was examined. RESULTS: Good outcome were obtained with either of the two methods. There were no cell elements remained under the examination of optical microscope and electron microscope. The interstice could observed on BAMG through the Scanning Microscope. The relative growth rate (RGR) was 81%, 88%, 93% and 96% separately on the 2nd, the 4th, the 6th and the 8th day (t = 3.7419, P < 0.025). The cytotoxicity score was I, which indicated that BAMG had a good cytocompatibility. All rabbits were alive except one which died from diarrhea. The reconstructed bladder resumed the construction and function of the bladder. The transitional cells and muscle cells growth could be seen in the scaffold of the repaired bladder after six weeks. CONCLUSION: The porcine BAMG has good biocompatibility and can be used as ideal biomaterial scaffold of bladder tissue engineering.

OBJECTIVE: To investigate the incidence and related factors of psychic symptoms in patients with chronic prostatitis. METHODS: Patients diagnosed with chronic prostatitis were selected as research objects, their course of disease, therapeutic process and psychic symptoms were inquired and recorded carefully, clinical symptoms were evaluated, expressed prostatic secretions (EPS) were examined, and many related factors were compared with psychic symptoms. RESULTS: Among the 315 patients selected, 51.1% (161/315) had psychic symptoms. Psychic symptoms in patients with chronic prostatitis had nothing to do with course of disease, the incidence of psychic symptoms in patients with course less than 1 year, 1 to 2 years, and more than 2 years were 49.1%, 48.0%, and 56.1%, respectively; had closed relationship with therapeutic process, the incidence of psychic symptoms in patients to see doctors for the first times, the 2 to 3 times and more than 3 times were 35.3%, 43.5%, and 62.6%, with significant statistical difference (P < 0.05); had no relationship with severity of clinical symptoms, the incidence of psychic symptoms in patients with mild, media and severe clinical symptoms were 46.2%, 52.4%, and 55.9%; no statistical difference was noticed in patients with different degrees of inflammation, the incidence of psychic symptoms in patients with inflammatory and non-inflammatory prostatitis were 53.8% and 47.3%. CONCLUSION: Psychic symptoms in patients with chronic prostatitis had direct relationship with times patient seeking medical care, and prolonged therapeutic process aggravated the psychic symptoms.
