OBJECTIVE: To detect the concentration of high-sensitivity cardiac troponin T (hs-cTnT) in healthy children aged 0-14 years by electrochemiluminescence immunoassay (ECLIA), so as to explore the differences in different ages and genders. The aim of this study is to establish the reference interval for hs-cTnT in healthy children aged 0-14 years. METHODS: After screening, 3463 healthy children, including 1924 boys and 1539 girls, were selected from 4617 children aged 0-14 years. They were divided into nine groups: one day (umbilical cord blood; 'UCB'), one day (venous blood; 'VB'), 2-28 days, 29 days-<3 months, 3-<6 months, 6 months-<1 year old, 1-< 3 years old, 3-< 6 years old and 6-14 years old. A nonparametric test was used to detect the hs-cTnT concentration. The upper limit of the reference interval is the mean of the 99th percentile after bootstrap sampling. RESULTS: Hs-cTnT levels conformed to a non-Gaussian distribution. There was no significant difference in the concentration of hs-cTnT between boys and girls in the general data, but there were differences between boys and girls in the 3-<6 years old and 6-14 years old age groups. Except for UCB and 2-28 days, the concentration of hs-cTnT was significantly different in other age groups. The level of hs-cTnT in neonatal serum (2-28 days) was the highest. In other groups, it decreased gradually with age and dropped to the reference range of adults (0-14ng/L) at one-year old. The upper limit of reference interval of hs-cTnT concentration in each group was, respectively, 60.8, 78.8, 96.6, 58.6, 34.2, 16.2, 11.4, 8.0 (7.8 female), and 7.9 (7.3 female) ng/L. CONCLUSIONS: Referring to WS/T 402-2012 establishment of reference intervals for clinical laboratory testing projects and CLSI (Clinical and Laboratory Standards Institute) C28-A3 documents and the joint expert consensus of ESC (European Society of Cardiology) and ACC (American College of Cardiology) in 2007, we established the reference interval of hs-cTnT concentration in children aged 0-14 years in Chongqing Nan'an district of China which can provide certain reference value for clinical diagnosis and treatment of myocarditis and myocardial (micro) injury in children.

Revealing genomic variation of representative and diverse germplasm is the cornerstone of deploying genomics information into genetic improvement programs of species of agricultural importance. Here we report the re-sequencing of 239 japonica rice elites representing the genetic diversity of japonica germplasm in China, Japan and Korea. A total of 4.8 million SNPs and PAV of 35,634 genes were identified. The elites from Japan and Korea are closely related and relatively less diverse than those from China. A japonica rice pan-genome was constructed, and 35 Mb non-redundant novel sequences were identified, from which 1131 novel genes were predicted. Strong selection signals of genomic regions were detected on most of the chromosomes. The heading date genes Hd1 and Hd3a have been artificially selected during the breeding process. The results from this study lay the foundation for future whole genome sequences-enabled breeding in rice and provide a paradigm for other species.

Due to the poor permeability across Gram-negative bacterial membranes and the troublesome bacterial efflux mechanism, only a few GyrB/ParE inhibitors with potent activity against Gram-negative pathogens have been reported. Among them, pyrimido[4,5-b]indole derivatives represented by GP-1 demonstrated excellent broad-spectrum antibacterial activity against both Gram-positive and Gram-negative bacteria but were limited by hERG inhibition and poor pharmacokinetics profile. To improve their drug-like properties, we designed a series of novel pyrimido[4,5-b]indole derivatives based on the tricyclic scaffold of GP-1 and the C-7 moiety of acorafloxacin. These efforts have culminated in the discovery of a promising compound 18r with reduced hERG liability and an improved PK profile. Compound 18r exhibited superior broad-spectrum in vitro antibacterial activity compared to GP-1, including a variety of clinical multidrug G- pathogens, especially Acinetobacter baumannii, and the in vivo efficacy was also demonstrated in a neutropenic mouse thigh model of infection with multidrug-resistant A. baumannii.

An efficient and modular strategy was used to obtain enaminones with a wide range of functional groups via a four-component sequential reaction. This reaction proceeded under mild conditions without a catalyst in one pot. Furthermore, the products could be transformed into thiadiazoles.

PURPOSE: Mepolizumab is a human monoclonal antibody against interleukin 5 (IL-5) used to treat severe eosinophilic asthma. Several studies have evaluated the effectiveness of mepolizumab in the real world. We conducted a systematic review and meta-analysis in the context of heterogeneity among patients, clinicians, and treatment regimens to study the effectiveness of mepolizumab in the real world. METHODS: We searched the PubMed and Embase databases for real-world studies on severe asthma treatment with mepolizumab as of June 30, 2020. Exacerbations, asthma-related hospitalizations, forced expiratory volume in 1 second (FEV1), Asthma Control Questionnaire (ACQ) or Asthma Control Test (ACT), corticosteroid use, peripheral blood eosinophil counts, and the fraction of exhaled nitric oxide were selected as indicators to evaluate the effectiveness. Standardized mean differences by the Cohen method and mean differences were chosen as indicators of effect size. Cohen d values of 0.2, 0.5, and 0.8 are considered as small, medium, and large effects, respectively. We used the Dersimonian-Laird random-effect model to quantify pooled effectiveness estimates. FINDINGS: A total of 1457 patients from 13 studies were included in this review. At all time points, mepolizumab was associated with reductions in exacerbations (2.92 and 2.73 events per patient per year fewer at 6 and 12 months, respectively) and hospitalizations (0.36 events per patient per year fewer at 12 months); improvements in asthma control (ACQ scores reductions of 1.32 and 1.03 at 6 and 12 months, respectively; ACT scores increase of 6.52 at 6-12 months); slight improvements in pulmonary function (FEV1 increase of 0.23 L at 1-3 months and 6-12 months, respectively); reductions in oral corticosteroid use (9.02- and 7.68-mg decrease at 6 and 12 months, respectively); and reductions in peripheral blood eosinophil counts (decreases of 559.11 cells/µL and 599.17 cells/µL at 1-3 months and 6-12 months, respectively) and fraction of exhaled nitric oxide (13-ppb reduction at 6-12 months). IMPLICATIONS: Our study suggests that mepolizumab is associated with improvements in several clinically meaningful real-world outcomes. This study is a supplement to and extension of the efficacy of randomized controlled trials of mepolizumab.

A new iso-C14 [Val2, Val7] surfactin isoform (1) together with eight known ones (2-9), was isolated from the culture of a mushroom derived bacterium, Bacillus halotolerans DMG-7-2. The structures of them were mainly elucidated by NMR and MS data, and the NMR data of 5 also was reported for the first time. The absolute configuration of 1 was determined by Marfey's analysis (for amino acid residues) and the 13C NMR calculation of the two plausible epimers of 1 (for fatty acid). Compounds 1-9 showed moderate cytotoxicity against two human cancer cell lines (A549, MCF-7) and mice microglial BV2 cells, the IC50 values ranged from 8.91 to 33.00 µM, and the IC50 values of the positive control 5-FU were 99.94, 71.49 and 0.12 µM, respectively.

Strong nonspecific protein/cell adhesion on conducting polymer (CP)-based bioelectronic devices can cause an increase in the impedance or the malfunction of the devices. Incorporating oligo(ethylene glycol) or zwitterionic functionalities with CPs has demonstrated superior performance in the reduction of nonspecific adhesion. However, there is no report on the evaluation of the antifouling stability of oligo(ethylene glycol) and zwitterion-functionalized CPs under electrical stimulation as a simulation of the real situation of device operation. Moreover, there is a lack of understanding of the correlation between the molecular structure of antifouling CPs and the antifouling and electrochemical stabilities of the CP-based electrodes. To address the aforementioned issue, we fabricated a platform with antifouling poly(3,4-ethylenedioxythiophene) (PEDOT) featuring tri(ethylene glycol), tetra(ethylene glycol), sulfobetaine, or phosphorylcholine (PEDOT-PC) to evaluate the stability of the antifouling/electrochemical properties of antifouling PEDOTs before and after electrical stimulation. The results reveal that the PEDOT-PC electrode not only exhibits good electrochemical stability, low impedance, and small voltage excursion, but also shows excellent resistance toward proteins and HAPI microglial cells, as a cell model of inflammation, after the electrical stimulation. The stable antifouling/electrochemical properties of zwitterionic PEDOT-PC may aid its diverse applications in bioelectronic devices in the future.

Five new peptaibols, longibramides A-E (1-5) with 11 amino acid residues, were isolated from a fungus Trichoderma longibrachiatum Rifai DMG-3-1-1, which was isolated from a mushroom Clitocybe nebularis (Batsch) P. Kumm collected from coniferous forest in the subboreal area of northeast China. The structures of longibramides A-E were determined by their spectroscopic data (NMR and MS-MS spectra), their absolute configurations were determined by X-ray diffractions and Marfey's analyses. The X-ray diffractions of longibramides A, B, and the similar CD spectra of A-E showed that they all had α-helix conformations. Longibramides B and E showed moderate cytotoxicities against BV2 and MCF-7 cells and also showed some inhibitory effects against methicillin-resistant Staphylococcus aureus MRSA T144. L-trans-Hyp was not commonly found in natural peptaibols, which was the 6th or 10th amino acid residue in longibramides C-E. The X-ray diffractions of longibramides A and B afforded the accuracy conformations of their secondary structures, which maybe help to interpret the structure-activity relationships of the family of peptaibols in the future.

BACKGROUND: Identifying the loci and dissecting the genetic architecture underlying wheat yield- and quality-related traits are essential for wheat breeding. A genome-wide association study was conducted using a high-density 90 K SNP array to analyze the yield- and quality-related traits of 543 bread wheat varieties. RESULTS: A total of 11,140 polymorphic SNPs were distributed on 21 chromosomes, including 270 significant SNPs associated with 25 yield- and quality-related traits. Additionally, 638 putative candidate genes were detected near the significant SNPs based on BLUP data, including three (TraesCS7A01G482000, TraesCS4B01G343700, and TraesCS6B01G295400) related to spikelet number per spike, diameter of the first internode, and grain volume. The three candidate genes were further analyzed using stage- and tissue- specific gene expression data derived from an RNA-seq analysis. These genes are promising candidates for enhancing yield- and quality-related traits in wheat. CONCLUSIONS: The results of this study provide a new insight to understand the genetic basis of wheat yield and quality. Furthermore, the markers detected in this study may be applicable for marker-assisted selection in wheat breeding programs.

We report a novel FeS2 loaded large SiO2 sphere (diameter of 3 cm) via double immobilization with (3-mercaptopropyl)trimethoxysilane (KH590) as a highly efficient, stable, and tweezers-recoverable heterogeneous Fenton-like catalyst (FeS2/KH590/SiO2@KH590). Notably, as-prepared composite catalyst exhibits a significant recycling improvement (10 runs) over free FeS2 powder (3 runs). The outstanding recyclability and stability of FeS2/KH590/SiO2@KH590 can be attributed to the passivation of FeS2 nanoparticles on SiO2 surface by KH590 and subsequent prevention of Fe2+ leaching. Overall, our work provides a new avenue towards fabricating composite Fenton-like catalyst with high stability, enhanced recyclability and the advantage of easy separation.

Dysregulated expression of microRNAs (miRNAs or miRs) has been implicated in the pathophysiology of type 2 diabetes mellitus (T2DM). However, their underlying role in the complication of detrusor fibrosis remains poorly understood. Therefore, this study aimed to examine the potential functional relevance of miR-363 in detrusor fibrosis of rats with streptozotocin (STZ)-induced T2DM through the predicted target gene collagen type I alpha 2 (Col1a2). Immunohistochemical analysis found an increase in the positive expression of collagen type III alpha 1 (Col3a1) and Col1a2 in detrusor tissues, where miR-363 expression was decreased. Next, gain- and loss-of-function experiments were performed to clarify the effects of miR-363 and Col1a2 on the activities of bladder detrusor cells. Of note, binding affinity between miR-363 and Col1a2 was verified by a dual-luciferase reporter gene assay and RNA immunoprecipitation (RIP) assay. Upregulated miR-363 inhibited Col1a2 expression, which led to increased expression of B-cell lymphoma 2 (Bcl-2) and Smad7 and accelerated cell viability, along with decreases in cell apoptosis and Col3a1, Bcl-2-associated X protein (Bax), transforming growth factor (TGF)-ß1, and Smad4 expressions. In conclusion, miR-363 upregulation reduces detrusor fibrosis in rats with STZ-induced T2DM through suppression of the TGF-ß1/Smad signaling pathway by targeting Col1a2. Therefore, our study provided further insights for the development of new therapeutic targets for T2DM.

The novel multidentate chelating ligands N'-(2-pyridylmethylidene)-2-(2-pyridylmethylideneamino)benzohydrazide (Hpphz) and N'-(2-salicylmethylidene)-2-(2-salicylmethylideneamino)benzohydrazide (H3sshz), which incorporate both amine and acylhydrazine Schiff base groups, were synthesized and investigated in DyIII coordination chemistry. The reactions of Hpphz and Dy(OAc)3·4H2O have yielded two {Dy2} featuring double OAc- bridges: [Dy2(H2aphz)2(OAc)4(ROH)2] [R = Me (1) and Et (2)], where the Hpphz ligands were in situ hydrolyzed into 2-amino-(2-pyridylmethylideneamino)benzohydrazide ions (H2aphz-). Besides, the reaction between H3sshz and Dy(NO)3·6H2O afforded a [Dy6(sshz)4(µ3-OH)4(µ4-O)(MeOH)4]2·17.5MeOH·2H2O cluster (3). This cluster contained two discrete {Dy6} cores, each of which consisted of a pair of {Dy3} triangular units. All the complexes displayed a single relaxation process of single-molecule magnet (SMM) behaviors under a zero dc field. Both 1 and 2 showed field-induced dual magnetic-relaxation behaviors. However, their diluted samples (1@Y and 2@Y) only showed one-step relaxation behaviors whether under a zero or applied dc field, indicating that the dual magnetic-relaxation behaviors of 1 and 2 were absent after the dilution. Combined with ab initio calculations, it could be infered that the dual magnetic-relaxation behaviors of 1 and 2 might be ascribled to the joint contributions of the single ion anisotropy and magnetic interactions. Examples of this type are rather rare in previous studies. Ab initio calculations also suggested that the discrepancy between the relaxation processes of 1 and 2 may be caused by the small difference between their magnetic interactions.

By utilizing the 2-hydroxyisophthalic acid (H3ipO) ligand, 2D metal-organic frameworks (MOFs) featuring rare Ophenol-bridged [Ln2]-magnetic building blocks (MBBs), [Ln2(ipO)2(DMF)(H2O)] [Ln = Gd (1), Dy (2); DMF = N,N-dimethylformamide], were rationally designed and synthesized. When the reaction solvents that behave as terminal ligands were changed, the coordination geometries of LnIII ions and the arrangement fashion of [Ln2]-MBBs for these MOFs were modified accordingly. Another type of 2D MOF of [Ln2(ipO)2(H2O)4]·2H2O [Ln = Gd (3), Dy (4)] was thus obtained. MOFs 1 and 3 exhibited favorable magnetocaloric effect, whose maximum -ΔSm values reach 30.0 and 31.7 J kg-1 K-1, respectively. None of the single-molecule-magnet (SMM) behavior was observed in 2. However, from 2 to 4, the change of the terminal coordinated solvents brought obvious improvement of the magnetic properties. MOF 4 showed interesting relaxation behavior, in which dual relaxation was only visible under weak direct-current fields, and its highest effective energy barrier (Ueff) reached up to 243 K. Ab initio calculations revealed the tuning mechanism of the terminal coordinated solvents. Their change optimized the arrangements of the magnetic axis of the DyIII centers in both each MBB and the whole framework, thus improving the magnetic anisotropy and magnetic interactions of the system. Significantly, within the [Dy2]-MBBs of 4, the angle made by the individual magnetic axis and Dy···Dy' line is nearly 0°. This case favoring a high SMM performance not only was scarcely achieved in discrete {Ln2}-SMMs with numerous members but also has never been observed in any MBB-based MOFs as far as we know.

Herein we report a variety of supramolecular architectures that are self-assembled by the highly charged anionic Ti4L6 (L = embonate) cages and noble-metal Ag+ ions in the presence of the different ligands, including six Ti4L6-Ag(PPh3) cages in whose structures the Ti4L6 cage catches various in situ formed [Ag(PPh3)]+ moieties by a coordination bond and one cocrystal superstructure of a Ti4L6 cage with an in situ generated [Ag2(Ph2P(CH2)5PPh2)3]2+ cage via supramolecular interactions. In addition, the third-order nonlinear-optical properties of these compounds are investigated in detail.

Potassium (K) is essential for regulating plant growth and mediating abiotic stress responses. Elucidating the biological mechanism underlying plant responses to K-deficiency is crucial for breeding new cultivars with improved K uptake and K utilization efficiency. In this study, we evaluated the extent of the genetic variation among 543 wheat accessions differing in K-deficiency tolerance at the seedling and adult plant stages. Two accessions, KN9204 and BN207, were identified as extremely tolerant and sensitive to K-deficiency, respectively. The accessions were exposed to normal and K-deficient conditions, after which their roots underwent ionomic, transcriptomic, and metabolomic analyses. Under K-deficient conditions, KN9204 exhibited stronger root growth and maintained higher K concentrations than BN207. Moreover, 19,440 transcripts and 162 metabolites were differentially abundant in the roots of both accessions according to transcriptomic and metabolomic analyses. An integrated analysis of gene expression and metabolite profiles revealed that substantially more genes, including those related to ion homeostasis, cellular reactive oxygen species homeostasis, and the glutamate metabolic pathway, were up-regulated in KN9204 than in BN207 in response to low-K stress. Accordingly, these candidate genes have unique regulatory roles affecting plant K-starvation tolerance. These findings may be useful for further clarifying the molecular changes underlying wheat root adaptations to K deprivation.

Present herein is a stepwise assembly method toward aluminum-lanthanide-based (Al-Ln) compounds. From the perspective of charge balance, polyanions are necessary to bind with Ln ions. However, the synthesis of polyanions aluminum compounds remains quite challenging. Herein, two Al4 polyanions [Al4(L)4(Cat)2]·4Hdma (AlOC-13, H3L= 2,3-dihydroxybenzoic acid, Cat = catechol, and dma = dimethylamine) and [Al4(L)4(HL)2(DMF)2]·4Hdma·0.5DMF·0.5H2O (AlOC-14, DMF = N,N-dimethylformamide) were successfully obtained under solvothermal conditions. Catechol and Hdma were generated from the in situ decarboxylation of H3L ligand and decomposition of DMF, respectively. AlOC-13 is qualified for further coordination assembly for the available vacancy coordination sites, good water solubility, and scale-up synthesis. The assembly of Al4 polyanions with equivalent Ln ions afforded a series of zigzag chain structures [LnAl4(L)4(Cat)2(DMF)2(H2O)3]·Hdma (Ln = Sm, Eu, Gd, Tb, Dy, Ho, Er, Yb) (AlOC-13-Ln). Moreover, the magnetic behavior and photoluminescence of the series of AlOC-13-Ln were also studied. AlOC-13-Dy shows obvious antiferromagnetic behavior, while AlOC-13-Tb exhibits excellent green characteristic luminescence. This study not only paves the way toward anionic aluminum clusters but also reveals their potential application in water treatment of cationic metal ions capture.

A new meroterpene, chrysomutanin (1), two new meroterpenoids (4 and 5) together with nine known ones were isolated from the diethyl sulphate (DES) mutant 3d10-01 of the marine-derived fungus Penicillium chrysogenum S-3-25. The structures of the isolated compounds were determined by their spectroscopic data, and the absolute configuration of 1 was determined by Rh2-induced electrical circular dichroism (ECD) analysis or by comparison of the measured ECD with that of the known compounds. The cytotoxic activity was preliminarily evaluated against five human cancer cell lines. HPLC-UV analysis showed that compounds 1-12 were all newly produced by the mutant, and were not detected from the initial strain S-3-25. Chrysomutanin (1) is a new member with a chain sesquiterpene unit to the family of meroterpenes. Present results confirm that DES mutagenesis strategy is an effective method to exploit the dormant metabolites of fungi.

RESUMO

A new ionic pentanuclear FeIII cluster, namely, triethylazanium tetrakis(µ2-5-amino-1,2,3,4-tetrazolido)tetrakis(µ3-4-chloro-2-{[(1H-tetrazol-1-id-5-yl)imino]methyl}phenolato)di-µ3-oxido-pentairon(III) acetonitrile monosolvate monohydrate, (C6H16N)[Fe5(C8H4ClN5O)4(CH2N5)4O2]·CH3CN·H2O, was synthesized using microvial synthesis methods and characterized by elemental analysis, FT-IR spectroscopy, single-crystal X-ray diffraction and thermogravimetric analysis. Magnetic studies reveal that the complex displays dominant antiferromagnetic intracluster interactions between the FeIII ions through the µ3-oxide bridges.

BACKGROUND: Although previous studies have reported that levocetirizine is utilized for the treatment of children with allergic rhinitis (AR), its conclusions remain inconsistent. This study aims to evaluate the efficacy and harms of levocetirizine for children with AR. METHODS: Electronic database sources will be undertaken from the beginning to the present: MEDLINE, EMBASE, The Cochrane Library, CINAHL, ACMD, PsycINFO, Chinese Biomedical Literature Database, and China National Knowledge Infrastructure. We will not apply any restrictions to language and publication status. We will only consider randomized controlled trials of levocetirizine for children with AR. Two authors will independently scan literature, select studies, and collect data. Study quality for each included trial will be assessed using Cochrane risk of bias tool, and statistical analysis will be conducted using RevMan 5.3 software. RESULTS: This study will summarize the present evidence to systematically assess the efficacy and harms of levocetirizine for children with AR. CONCLUSION: The findings of this study intent to adequately inform stakeholders or clinicians, as well as to help develop treatment guidelines. STUDY REGISTRATION NUMBER: INPLASY202040111.

Purpose: This study aimed to investigate the regulatory role and mechanism of microRNA-766 (miR-766) on cutaneous squamous cell carcinoma (CSCC) cells. Methods: The expression of miR-766 and programmed cell death 5 (PDCD5) was detected in CSCC tissues and CSCC cell lines (A431, SCL-1 and DJM-1 cells) by qRT-RCR. The proliferation, colony-forming ability, apoptosis, migration and invasion of A431 and SCL-1 cells was measured by MTT, colony formation, flow cytometry, wound healing and transwell assay, respectively. The interaction between miR-766 and PDCD5 was detected by dual-luciferase reporter gene assay. The expression of matrix metalloproteinase 2 (MMP-2), MMP-9 and PDCD5 was measured by Western blot. In addition, A431 cells were subcutaneously injected into mice, and the tumor volume and weight were measured. Results: MiR-766 was upregulated, and PDCD5 was downregulated in CSCC tissues and cells. MiR-766 significantly promoted the proliferation, migration and invasion, and inhibited the apoptosis of A431 and SCL-1 cells. MiR-766 also significantly increased the expression of MMP-2 and MMP-9 in A431 and SCL-1 cells. PDCD5 was a target gene of miR-766. PDCD5 significantly reversed the tumor-promoting effect of miR-766 on A431 and SCL-1 cells. In addition, miR-766 inhibitor inhibited the tumor growth in mice. Conclusion: MiR-766 inhibitor inhibited the proliferation, migration and invasion, and promoted the apoptosis of CSCC cells via downregulating PDCD5.
