RESUMO

The aim of the present study was to identify natural compounds that bear significant anti­tumor activity. Thus, the effects of 63 small molecules that were isolated from traditional Chinese medicinal herbs on A549 human non­small cell lung cancer (NSCLC) and MCF­7 breast cancer cells were examined. It was found that ursolic acid (UA), a natural pentacyclic triterpenoid, exerted significant inhibitory effect on these cells. Further experiments revealed that UA inhibited the proliferation of various lung cancer cells, including the NSCLC cells, H460, H1975, A549, H1299 and H520, the human small cell lung cancer (SCLC) cells, H82 and H446, and murine Lewis lung carcinoma (LLC) cells. UA induced the apoptosis and autophagy of NSCLC cells. The inhibition of the mammalian target of rapamycin (mTOR) signaling pathway, but not the activation of the extracellular signal­regulated kinase 1/2 (ERK1/2) signaling pathway contributed to the UA­induced autophagy of NSCLC cells. Moreover, the inhibition of autophagy by chloroquine (CQ) or siRNA for autophagy­related gene 5 (ATG5) enhanced the UA­induced inhibition of cell proliferation and promotion of apoptosis, indicating that UA­induced autophagy is a pro­survival mechanism in NSCLC cells. On the whole, these findings suggest that combination treatment with autophagy inhibitors may be a novel strategy with which enhance the antitumor activity of UA in lung cancer.

Atrial fibrillation (AF) increases the risk of ischemic stroke and systemic arterial embolism. However, the risk factors or predictors of stroke in AF patients have not been clarified. Therefore, it is necessary to find effective diagnostic and therapeutic targets. Two datasets were downloaded from the Gene Expression Omnibus (GEO) database. Differently expressed genes (DEGs) were identified between samples of atrial fibrillation without stroke and atrial fibrillation with stroke. Enrichment analysis of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) by Gene Set Enrichment Analysis (GSEA), construction and analysis of protein-protein interaction (PPI) network and significant module, and the receiver operator characteristic (ROC) curve analysis were performed. A total of 524 DEGs were common to both datasets. Analysis of KEGG pathways indicated that the top canonical pathways associated with DEGs were ubiquitin-mediated proteolysis, endocytosis, spliceosome, and so on. Ten hub genes (SMURF2, CDC42, UBE3A, RBBP6, CDC5L, NEDD4L, UBE2D2, UBE2B, UBE2I, and MAPK1) were identified from the PPI network and were significantly associated with a diagnosis of atrial fibrillation and stroke (AFST). In summary, a total of 524 DEGs and 10 hub genes were identified between samples of atrial fibrillation without stroke and atrial fibrillation with stroke. These genes may serve as the target of early diagnosis or treatment of AF complicated by stroke.

Four types of middle-pressure chromatogram isolated gels are evaluated for adsorption or desorption characteristics of ginsenosides from Panax ginseng. Among them, SP207SS and SP2MGS were selected for dynamic investigations based on their static adsorption or desorption capacity of total ginsenoside. Their adsorption kinetics was better explained by pseudosecond-order model and isotherms were preferably fitted to Langmuir model. Dynamic breakthrough experiments indicated an optimum sample loading speed of 4 bed volume/h for either SP207SS or SP2MGS. Desorption speed was determined to be 2 bed volume/h according to desorption amount of total ginsenoside in their effluents. Eight ginsenosides were identified and quantified by high performance liquid chromatography-triple quadropole-mass spectrometry in total ginsenoside extract and different fractions during stepwise dynamic elution. For SP207SS, 27.62% of loaded ginsenosides was detected in 40% ethanol fraction, while 59.12% of them were found in 60% ethanol fraction. As on SP2MGS, the number went to 53.71 and 44.43%, respectively. Recovery rate of ginsenosides were calculated to 78.65% for SP207SS and 89.53% for SP2MGS, respectively. Intriguingly, content of Rg1 and Re in 40% ethanol fraction from SP207SS became 20.1 and 18.6 times higher than that in total ginsenoside extract by one-step elution, which could be leveraged for the facile enrichment of these two ginsenosides from natural sources.

BACKGROUND: How the oncoprotein epidermal growth factor receptor (EGFR) evades proteolytic degradation and accumulates in non-small cell lung cancer (NSCLC) remains unclear, and ubiquitin pathway genes (UPGs) that are critical to NSCLC needs to be systematically identified. METHODS: A total of 696 UPGs (including E1, E2, E3, and deubiquitinases) were silenced by small interfering RNA (siRNA) library in NSCLC cells, the candidates were verified, and their significance was evaluated in patients with NSCLC. The effects of a candidate gene on EGFR were investigated in vitro and in vivo. FINDINGS: We report 31 candidates that are required for cell proliferation, with the E2 ubiquitin conjugase CDC34 as the most significant one. CDC34 is elevated in tumor tissues in 76 of 114 (66.7%) NSCLCs and inversely associated with prognosis, is higher in smoker patients than nonsmoker patients, and is induced by tobacco carcinogens in normal human lung epithelial cells. Forced expression of CDC34 promotes, whereas knockdown of CDC34 inhibits, NSCLC cell proliferation in vitro and in vivo. CDC34 competes with c-Cbl to bind Y1045 to inhibit polyubiquitination and degradation of EGFR. In EGFR-L858R and EGFR-T790M/Del (exon 19)-driven lung tumor growth in mouse models, knockdown of CDC34 significantly inhibits tumor formation. INTERPRETATION: These results demonstrate that an E2 enzyme is capable of competing with E3 ligase to stabilize substrates, and CDC34 represents an attractive therapeutic target for NSCLCs. The purpose of this study was to investigate the presence of endolymphatic hydrops (EH) in both affected and unaffected ears of patients with pantonal unilateral idiopathic sudden sensorineural hearing loss (ISSNHL) using three-dimensional fluid-attenuated inversion recovery magnetic resonance imaging (3D-FLAIR MRI) and further evaluate the significance of EH in this disorder. Twenty-seven ISSHL patients were enrolled in this study. 3D-FLAIR MRI was performed 24 h after intratympanic injection of gadolinium-diethylenetriaminepentaacetic acid (Gd-DPTA). The incidences of EH in the affected ears and contralateral unaffected ears were compared and the correlations of EH with vertigo or prognosis were analyzed using the Chi-square test. The results showed that the incidence of EH was 68.0% (17/25) in the affected ears and 34.8% (8/23) in the unaffected ears. There was a statistically significant difference between affected ears and unaffected ears in regard to the incidence of EH (P<0.05). There were no significant correlations of EH with vertigo (P=1.000) or with prognosis (P=0.359) in the affected ears. In conclusion, there is EH in the inner ear of patients with pantonal ISSNHL; EH is not related to vertigo, a concomitant symptom of ISSNHL, and the prognosis of this condition. The presence of EH may be a secondary reaction following the impairment of the inner ears with pantonal ISSNHL.

MAIN CONCLUSION: ACOS5, OsACOS12 and PpACOS6 are all capable of fatty acyl-CoA synthetase activity but exhibit different substrate preferences. The transcriptional regulation of ACOS for sporopollenin synthesis appears to have been conserved in Physcomitrella, rice and Arabidopsis during evolution. Sporopollenin is the major constituent of spore and pollen exines. In Arabidopsis, acyl-CoA synthetase 5 (ACOS5) is an essential enzyme for sporopollenin synthesis, and its orthologues are PpACOS6 from the moss Physcomitrella and OsACOS12 from monocot rice. However, knowledge regarding the evolutionary conservation and divergence of the ACOS gene in sporopollenin synthesis remains limited. In this study, we analysed the function and regulation of PpACOS6 and OsACOS12. A complementation test showed that OsACOS12 driven by the ACOS5 promoter could partially restore the male fertility of the acos5 mutant in Arabidopsis, while PpACOS6 did not rescue the acos5 phenotype. ACOS5, PpACOS6 and OsACOS12 all complemented the acyl-CoA synthetase-deficient yeast strain (YB525) phenotype, although they exhibited different substrate preferences. To understand the conservation of sporopollenin synthesis regulation, we constructed two constructs with ACOS5 driven by the OsACOS12 or PpACOS6 promoter. Both constructs could restore the fertility of acos5 plants. The MYB transcription factor MS188 from Arabidopsis directly regulates ACOS5. We found that MS188 could also bind the promoters of OsACOS12 and PpACOS6 and activate the genes driven by the promoters, suggesting that the transcriptional regulation of these genes was similar to that of ACOS5. These results show that the ACOS gene promoter region from Physcomitrella, rice and Arabidopsis has been functionally conserved during evolution, while the chain lengths of fatty acid-derived monomers of sporopollenin vary in different plant species.

The sporopollenin precursors, as a general constituent of sexine, are synthesized in the tapetum and deposited on the pollen surface after transportation and processing. The polyketide synthase condenses the acyl-CoA into a hydroxyalkyl α-pyrone, which is predicted to be a component of the sporopollenin precursors. In this study, we found that the rice POLYKETIDE SYNTHASE 1 (OsPKS1) was the orthologue of Arabidopsis POLYKETIDE SYNTHASE A/LESS ADHESIVE POLLEN 6 (PKSA/LAP6) through sequence alignment. The OsPKS1 knockout mutants obtained by Crispr-Cas9-mediated editing exhibited a complete male sterile phenotype. Cytological observations revealed that abnormal bacula deposition and ubisch body structures for sexine formation led to pollen rupture in ospks1. The expression analysis showed that the OsPKS1 was highly expressed in tapetal cells and anther locules from stage 9 to stage 11 during anther development in rice. Subcellular localization demonstrated that the OsPKS1 protein was preferentially localized to the ER. The genomic sequence of OsPKS1 driven by the PKSA/LAP6 promoter restored the sexine pattern of Arabidopsis pksa/lap6. These results indicated that OsPKS1 is required for sexine layer formation in rice and functionally conserved in the sporopollenin synthesis pathway.

To systematically unveil transcription factors (TFs) that are critical to lung carcinogenesis, here we conducted a genome-wide lethality screening in non-small cell lung cancer (NSCLC) cells and reported that among the 1530 TFs tested, 21 genes were required for NSCLC cell proliferation and were negatively or positively associated with overall survival (OS) of patients with NSCLC. These included 11 potential tumor suppressing genes (AFF3, AhR, AR, CBFA2T3, CHD4, KANK2, NR3C2, PTEN, PRDM16, RB1, and STK11) and 10 potential oncogenic TFs (BARX1, DLX6, ELF3, EN1, ETV1, FOXE1, HOXB7, IRX4, IRX5, and SALL1). The expression levels of IRX5 were positively associated with OS of smoker and inversely associated with OS of non-smoker patients with lung adenocarcinoma. We showed that tobacco carcinogen benzo(a)pyrene (BaP) induced upregulation of IRX5 in lung epithelial cells, and Cyclin D1 was a downstream target of IRX5. Furthermore, silencing of IRX5 by lentivirus mediated transfection of short hairpin RNA significantly inhibited tumor growth in nude mice. These results indicate that tobacco smoke can modulate TFs to facilitate lung carcinogenesis, and inhibition of IRX5 may have therapeutic potentials in NSCLCs.

Cisplatin is a main compound for human hepatocellular carcinoma (HCC) chemotherapies, but it has certain cytotoxicity during applications. To release that, combining with other drugs are being as a regular plan in clinic. In our present study, we are focusing on one of active monomers extracted from Anemone Raddeana Regel, Raddeanin A (RA), which is on behalf of the same character like cisplatin in the tumor remedies. In order to investigate whether combination usage of RA and cisplatin can be priority to the later drug's effect development and its toxicity reduction in HCC, both of two drugs were treated 24 h or 48 h in QGY-7703 cells for estimating their abilities in tumor cell proliferation inhibition. Results show RA makes synergistic functions with cisplatin after measuring and analyzing their combination index (CI) values. Meanwhile it can strengthen cisplatin's effect through arresting the tumor cells in G0/G1 cycle and further promoting their apoptosis. Interestingly, the molecule signals correlated to tumor cell apoptosis containing both of p53 and bax are simultaneously activated, but bcl-2 and survivin are all depressed in mRNA level. Meanwhile, combining usage with RA can even raise the intracellular productions of reactive oxygen species (ROS). All these consequences reflect RA plays an important role in enhancing the therapeutic effect of cisplatin in HCC. This finding may guide for the drug usage of cisplatin in clinic practice.

The spatiotemporal dynamics of Bt protein in soil and the change of soil nutrients in rhizosphere soil, root surface soil and soils at 0-20, 20-40 and 40-60 cm were measured in greenhouse experiments. Two Bt corns, 5422Bt1 and 5422CBCL, and their near isogenic non-Bt variety 5422 were grown for 90 days and the crop residues were retained to soil. Results showed that 1.59 and 2.78 ng x g(-1) Bt protein were detected in the rhizosphere soil with Bt corns 5422Bt1 and 5422CBCL immediately after harvest. However, there were only trace amounts of Bt protein (< 0.5 ng x g(-1)) were detected in root surface soil after 90 days and in bulk soil in the two Bt corn treatments after 30, 60 and 90 days. When corn residues returned to soil, Bt protein declined rapidly within 3 days and only trace amounts of Bt protein were measured after 7 days. There were no sig- nificant differences in organic matter, available nutrient (alkaline hydrolytic N, available P, available K) or total nutrient (total N, total P, total K) in root surface soils and soils at 0-20 cm, 20-40 cm and 40-60 cm among the Bt and non-Bt corns after 90 days. Sixty days after returning crop residues of 5422Btl to soil, the contents of organic matter and total N increased and the content of available K reduced significantly in the 0-20 cm soil depth. There were no significant differences in any other parameter at the 0-20 cm depth, neither for any parameter in the 20-40 cm and 40-60 cm soil depths compared to those in the non-Bt corn 5422 treatment. There were no significant differences in soil nutrient contents in Bt corn 5422CBCL treatment compared to those in non-Bt corn 5422 treatment except that available phosphorus content was reduced in root surface soils, and total P content increased at the 0-20 cm soil depth after 90 days. When crop residues of Bt corn 5422 CBCL were returned to soil, only available P content in the 0-20 cm soil layer was evidently higher compared to the soil receiving crop residues of non-Bt corn 5422. Results suggested that Bt protein released from root and crop residues of Bt corns would not accumulate in soil, and growing Bt corns and returning crop residues to soil would have no significant effect on soil nutrients in general.

Au was used as anode in some kind of organic electroluminescent devices. Sometimes transparent Au electrodes are required, which means that the thickness of Au electrode should be as thin as possible. Therefore, two metals together forming an electrode become a choice. In the present paper, translucent Au/Al layer was inserted to anode side, and OLED device with the structure of ITO/Al (16 nm)/Au (10 nm)/TPD (30 nm)/AlQ (30 nm)/LiF (0.5 nm)/Al was prepared. There is a spectral narrowing phenomenon on the device ITO/TPD (30 nm)/AlQ (30 nm)/LiF (0. 5 nm)/Al, and through analysis and experiment it was found that this phenomenon comes from selective permeability to light of Au thin film rather than the microcavity effect. The device maintains wide viewing angle, without the angular dependence. And the color purity of device with Au thin film is improved.

Inserting metal thin layer into organic electroluminescent device often brings some unexpected effect. By inserting 5 nm Au thin layer between MoO3 and TPD, we prepared the OLED device with the structure of ITO/MoO3(5 nm)/Au(5 nm)/ TPD/AlQ/LiF/Al. The luminous efficiency is improved as compared to the device without the Au. By analysis, we believe that the Au between MoO3 and TPD formed a small trap for hole to reduce the current density in device. At the same time, since the light transmittance of green light (AlQ) of 5 nm Au is greater than 80%, the brightness of the device was not significantly affected. That is the reasons for the improved luminous efficiency. This work provides some new ideas and experimental evidence for how to improve the luminous efficiency of OLED devices.

Hybrid nanomaterials comprising phosphotungstic acid (PTA) and MIL-101(Cr) were prepared through one-pot synthesis and post-modification methods and then were used as adsorbents of dibenzothiophene (DBT) from simulated diesel fuels. Samples obtained by different ways (encapsulation and impregnation) were characterized by nitrogen adsorption, transmission electron microscope (TEM), X-ray diffraction (XRD), Fourier transform infrared spectrum (FT-IR) and series of adsorption experiments. The equilibrium adsorption capacities of PTA@MIL-101(Cr) illustrated that the direct introduction of PTA into MIL-101(Cr) during synthesis resulted in a 10.7% increase compared with MIL-101(Cr). However, porous hybrid adsorbent PTA/MIL-101(Cr) prepared via post-modification method exhibited lower adsorption capacity than virgin MIL-101(Cr). The theoretical maximum adsorption capacity (Q0) of PTA@MIL-101(Cr) is 136.5mg S/g adsorbent, 4.2 times of MIL-101(Cr). Even in competitive adsorption between aromatic compounds, which possess strong affinity with MOFs, and DBT, PTA@MIL-101(Cr) and MIL-101(Cr) remained their effectiveness in removal of DBT in the system. Based on these results, it can be presumed that MIL-101(Cr), modified properly, can be used as a promising adsorbent for eliminating aromatics and S-compounds in commercial fuels simultaneously.

To develop high-efficient biofertilizer, an environmental stress-tolerant phosphate-solubilizing microorganism (PSM) was isolated from agricultural wastes compost, and then applied to spent mushroom substrate (SMS). The isolate FL7 was identified as Pichia farinose with resistance against multiple environmental stresses, including 5-45°C temperature, 3-10 pH range, 0-23% (w/v) NaCl and 0-6M ammonium ion. Under the optimized cultivation condition, 852.8 mg/l total organic acids can be produced and pH can be reduced to 3.8 after 60 h, meanwhile, the soluble phosphate content reached 816.16 mg/l. The P. farinose was used to convert SMS to a phosphate biofertilizer through a semi-solid fermentation (SSF) process. After fermentation of 10 days, cell density can be increased to 5.6 × 10(8)CFU/g in biomass and pH in this medium can be decreased to 4.0. SMS biofertilizer produced by P. farinose significantly improved the growth of soybean in pot experiments, demonstrating a tremendous potential in agricultural application.

In the present work, the photoluminescence (PL) and electroluminescence (EL) characteristics of Tris[2-(2,4-difluorophenyl)pyridine]iridium(III) (Ir(Fppy)3) doped poly(n-vinylcarbazole) (PVK) with different doping concentrations were investigated. And a blue phosphorescent organic light-emitting diode (OLED) with the structures of ITO/PEDOT : PSS/PVK : Ir(Fppy)3/BCP/Alq3/LiF/Al was fabricated. The experimental results show that the luminescence performances of devices are different as the doping concentration of Ir(Fppy)3 is different. When the doping concentration of Ir(Fppy)3 is lower, the luminescence of PVK can be found in EL spectra. When the doping concentration is too high, concentration quenching may occur. As the doping concentration is suitable, the luminescence of PVK can not be found, only the luminescence of Ir(Fppy)3 can be found in EL spectra. It is concluded that the device with doping concentration of 4% has the best photoelectric performance according to its current density-voltage-luminance curve.

In the present work, in order to improve electron injection and transport at the interface of the hole blocking layer (HBL) and the electron transport layer (ETL) in the hole-domain solution processed phosphorescent organic light emitting devices (PhOLEDs), the mixed interface layer (MIL) was fabricated by partially co-doping hole blocking material 2,9-dimethyl-4,7-diphenyl-1,10-phenanthroline (BCP) and electron transport material tris(8-quinolinolato) aluminum (Alq3) between HBL/ ETL. The MIL thickness was kept at 10nm, while the doping ratio of these two materials varied. Under a given electric field, the devices with the MIL at any mixed ratios all show much higher luminance and current density than those with a typical interface. For example, the luminance power and current density at 10 V for a typical device are 1.03 microW and 5.13 mA x cm(-2), while in case of mixed interface are 3.64 microW and 18.1 mA x cm(-2), respectively. From data results and theoretical analysis, the possible derivation of these improvements is considered to be the reduced electron accumulation at the interface resulting from the reduced electron injection energy barrier and lowered transport mobility by BCP material, which leads to an increase in electron amount in the emission layer and therefore the higher luminance and current density.

Spent mushroom substrate (SMS) was treated with dilute sulfuric acid followed by cellulase and xylanase treatment to produce hydrolysates that could be used as the basis for media for the production of value added products. A L9 (3(4)) orthogonal experiment was performed to optimize the acid treatment process. Pretreatment with 6% (w/w) dilute sulfuric acid at 120°C for 120 min provided the highest reducing sugar yield of 267.57 g/kg SMS. No furfural was detected in the hydrolysates. Exposure to 20PFU of cellulase and 200 XU of xylanase per gram of pretreated SMS at 40°C resulted in the release of 79.85 g/kg or reducing sugars per kg acid pretreated SMS. The dilute sulfuric acid could be recycled to process fresh SMS four times. SMS hydrolysates neutralized with ammonium hydroxide, sodium hydroxide, or calcium hydroxide could be used as the carbon source for cultivation of Lactococcus lactis subsp. lactis W28 and a cell density of 2.9×10(11)CFU/mL could be obtained. The results provide a foundation for the development of value-added products based on SMS.

The complete mol-ecule of the title compound, C(12)H(10)ClO(2)PS, is generated by crystallographic mirror symmetry, with the P, S and Cl atoms lying on the mirror plane. The resulting PO(2)SCl tetra-hedron is significantly distorted [O-P-O = 96.79 (9)°]. The crystal packing exhibits no directional inter-actions.

Three organic devices with different quantum well period were fabricated. The potential barrier layer and well layer for electrons were made of N,N'-diphenyl-N,N'-bis(1-napthyl)-1,1'-biphenyl 1-4,4'-diamine(NPB) and 4,4,N,N'-dicarbazolebiphenyl(CBP). The photoluminescence quenching of these three devices under changed reverse voltages were studied. Results showed that photoluminescence quenching of NPB layer occurs more quickly than that of CBP layer in the authors' devices. This is because that the effective electric field in NPB layer is higher than that in CBP layer. The excitons in NPB and CBP layer were easily to be dissociated when the quantum well period increased under the same reverse voltage. Since these three devices are type II quantum well structure, the excitons in these devices are not very stable.

To identify novel biomarkers of metastasis of colorectal cancer (CRC), we developed an orthotopic implantation model of murine CRC and selected in vivo M5, a subclone of the SW480 CRC cell line with enhanced potential for metastasis to the liver. We compared the differences in the gene expression profiles between M5 and SW480 cells using gene expression profiling. We found that expression of special AT-rich sequence-binding protein 2 (SATB2) was down-regulated in M5 cells. Immunohistochemical analysis of 146 colorectal tumour samples showed that underexpression of SATB2 was strongly correlated with poor prognosis, tumour invasion, lymph node metastasis, distant metastasis, and Dukes' classification for CRC. Univariate and multivariate survival analyses further showed that SATB2 expression was a potential favourable prognostic factor for CRC. These results demonstrated not only that SATB2 is a potential novel prognostic factor for CRC, but also that selection of a highly metastatic clone of SW480 in vivo coupled with gene expression profiling is a powerful approach to identifying prognostic markers for CRC.
