In this study, three SBR reactors R1, R2, and R3 were set up and operated using (A/O)3-SBR gradient aeration, (A/O)3-SBR constant aeration, and the conventional (A/O)-SBR mode, respectively. The nutrient removal performance and aerobic granular sludge characteristics under these aeration modes were explored using real municipal wastewater as the influent matrix. The experimental results revealed that for the R1, R2, and R3 particles during the stable period, the average removal rate of COD was 88.68%, 89.05%, and 88.96%, respectively, the average removal rate of TN was 76.97%, 71.99%, and 64.92%, respectively, the average removal rate of TP was 96.28%, 85.05%, and 78.97%, respectively, and the proportion of denitrifying phosphorus accumulating bacteria to phosphorus accumulating bacteria was 25.52%, 19.60%, and 12.77%, respectively. The results showed that the operation mode of anaerobic, aerobic, and anoxic was more conducive to the enrichment of denitrifying phosphorus accumulating bacteria (DPAOs), and that the gradient aeration was more enriched than the constant aeration mode, which is of great significance to low-intensity municipal domestic sewage treatment with an insufficient carbon source. At the same time, the dissolved oxygen in the aeration section of R1 was reduced step-by-step, which improved the simultaneous nitrification and denitrification rates of particles and the utilization rate of the internal carbon source, which was beneficial for the efficient removal of TN. The particle size of the three groups of reactors was 727.368, 815.072, and 895.041 µm respectively. As the transfer rate of the matrix decreased with particle size, the microorganisms in R2 and R3 may have caused anaerobic respiration to release harmful gas, thus damaging the particle structure, such that the particles in R2 and R3 were less dense than those in R1. In addition, the PN/PS values of R1, R2, and R3 were 6.31, 5.63, and 4.83, respectively, and the EPS content (in terms of VSS) was 103.97, 92.22, and 76.98 mg·g-1, respectively, at the time of particle stabilization, which revealed that the mode of intermittent gradient aeration was beneficial to stimulate the secretion of EPS. This was especially the case for the secretion of PN, which increased the PN/PS value, enhanced the cell hydrophobicity, and made the particles dense and stable.

BACKGROUND & AIMS: p53 mutations occur frequently in human HCC. Activation of the mammalian target of rapamycin (mTOR) pathway is also associated with HCC. However, it is still unknown whether these changes together initiate HCC and can be targeted as a potential therapeutic strategy. METHODS: We generated mouse models in which mTOR was hyperactivated by loss of tuberous sclerosis complex 1 (Tsc1) with or without p53 haplodeficiency. Primary cells were isolated from mouse livers. Oncogenic signalling was assessed in vitro and in vivo, with or without targeted inhibition of a single molecule or multiple molecules. Transcriptional profiling was used to identify biomarkers predictive of HCC. Human HCC materials were used to corroborate the findings from mouse models. RESULTS: p53 haploinsufficiency facilitates mTOR signalling via the PTEN/PI3K/Akt axis, promoting HCC tumorigenesis and lung metastasis. Inhibition of PI3K/Akt reduced mTOR activity, which effectively enhanced the anticancer effort of an mTOR inhibitor. ATP-binding cassette subfamily C member 4 (Abcc4) was found to be responsible for p53 haploinsufficiency- and Tsc1 loss-driven HCC tumorigenesis. Moreover, in clinical HCC samples, Abcc4 was specifically identified an aggressive subtype. The mTOR inhibitor rapamycin significantly reduced hepatocarcinogenesis triggered by Tsc1 loss and p53 haploinsufficiency in vivo, as well as the biomarker Abcc4. CONCLUSIONS: Our data advance the current understanding of the activation of the PTEN/PI3K/Akt/mTOR axis and its downstream target Abcc4 in hepatocarcinogenesis driven by p53 reduction and Tsc1 loss. Targeting mTOR, an unexpected vulnerability in p53 (haplo)deficiency HCC, can be exploited therapeutically to treat Abcc4-positive patients with HCC. LAY SUMMARY: Tsc1 loss facilitates the p53 (haplo)insufficiency-mediated activation of the PTEN/Akt/mTOR axis, leading to the elevated expression of Abcc4 to drive HCC tumorigenesis and metastasis in mice. Inhibition of mTOR protects against p53 haploinsufficiency and Tsc1 loss-triggered tumour-promoting activity, providing a new approach for treating an aggressive subtype of HCC exhibiting high Abcc4 expression.

PURPOSE: Hepatocellular carcinoma (HCC) is a common malignant cancer and the third leading cause of death worldwide. The molecular mechanism of HCC remains unclear. Recent studies have demonstrated that the ubiquitin-proteasome system (UPS) is associated with HCC. Ubqln2, a member of the UPS, is abnormally expressed in HCC. However, whether Ubqln2 is associated with HCC prognosis remains unknown. PATIENTS AND METHODS: We analyzed the associations between overall survival and various risk factors in 355 HCC tissue samples obtained from the Cancer Genomic Atlas (TCGA) database at the mRNA level and in 166 HCC tissue samples from Southwest Hospital at the protein level. qRCR was used to determinate Ubqln2 expression in cancer and noncancerous tissues. The association between Ubqln2 and Ki-67 was analyzed by immunohistochemistry. The association between Ubqln2 expression and survival was analyzed using Kaplan-Meier curve and Cox proportional hazards models. A nomogram was used to predict the impact of Ubqln2 on prognosis. Mutated genes were analyzed to determine the potential mechanism. RESULTS: Ubqln2 highly expressed in HCC tissues. The Ubqln2 mRNA level had significant relations with UICC tumor stage (P = .022), UICC stage (P = .034) and resection potential (P = .017). Concordantly, the Ubqln2 protein was closely associated with tumor size (P = .005), UICC stage (P = .012), and recurrence (P = .009). Ubqln2 was highly expressed in HCC and positively associated with poor survival. The nomogram precisely predicted the prognosis of HCC patients with high or low Ubqln2 expression. A genomic waterfall plot suggested that Ubqln2 expression was closely associated with mutated CTNNB1. CONCLUSION: Our findings reveal that Ubqln2, an independent risk factor for HCC, is a potential prognostic marker in HCC patients. Ubqln2 expression is positively associated with mutated CTNNB1.

When ultraviolet (UV) absorption spectroscopy technology is used for nitric oxide (NO) detection, the background noise will directly affect the accuracy of concentration inversion, especially in low concentrations. Traditional processing methods attempt to eliminate background noise, which damages the absorption spectrum characteristics. However, stochastic resonance (SR) can utilize the noise to extract a weak characteristic signal. This paper reports a monostable stochastic resonance (MSR) model for processing an UV NO absorption spectrum. By analyzing the characteristics of UV absorption spectrum of NO, the evaluation indexes were constructed, thereby an adaptive MSR method was designed for parameter optimization. The numerical simulation confirmed the absorbance peak can be amplified and spectral signal-to-noise ratio (SNR) can be in the stable range of the proposed method, when noise intensity increased. Finally, this experiment obtained a NO detection limit (3σ) of 1.456 ppm and the maximum relative deviation of concentration is 6.32% by this proposed method, which is satisfactory for processing of the UV NO absorption spectrum.

The recent discovery of the interfacial superconductivity (SC) of the Bi2Te3/Fe1+yTe heterostructure has attracted extensive studies due to its potential as a novel platform for trapping and controlling Majorana fermions. Here we present studies of another topological insulator (TI)/Fe1+yTe heterostructure, Sb2Te3/Fe1+yTe, which also has an interfacial 2-dimensional SC. The results of transport measurements support that reduction of the excess Fe concentration of the Fe1+yTe layer not only increases the fluctuation of its antiferromagnetic (AFM) order but also enhances the quality of the SC of this heterostructure system. On the other hand, the interfacial SC of this heterostructure was found to have a wider-ranging TI-layer thickness dependence than that of the Bi2Te3/Fe1+yTe heterostructure, which is believed to be attributed to the much higher bulk conductivity of Sb2Te3 that enhances indirect coupling between its top and bottom topological surface states (TSSs). Our results provide evidence of the interplay among the AFM order, itinerant carries from the TSSs, and the induced interfacial SC of the TI/Fe1+yTe heterostructure system.

BACKGROUND: ET-26 hydrochloride is a novel intravenous anesthetic, approved for clinical trials, that produces a desirable sedative-hypnotic effect with stable myocardial performance and mild adrenocortical suppression in rats and beagle dogs. The objective of this study was to assess the absorption, distribution, metabolism, and excretion of ET-26 hydrochloride. METHODS: Hepatocytes from human, monkey, dog, rat, and mouse were used to determine the metabolites of ET-26 hydrochloride. Distribution and excretion were assessed in rats and pharmacokinetic studies were performed in beagle dogs. RESULTS: The metabolic pathway and proposed structure of metabolites were fully assessed resulting from the biotransformation reactions of hydrolysis, dehydrogenation, demethylation and glucuronic acid conjugation. The main distribution of the drug was in fat (15067 ± 801 ng/ml) and liver (13647 ± 1126 ng/ml), and the kidney was the primary excretion route (4.47%-11.94%). The Cmax after injection with 1.045 mg/kg, 2.09 mg/kg, and 4.18 mg/kg was 1476.5 ± 138.9 ng/ml, 2846.1 ± 223.3 ng/ml, and 6233.3 ± 238.9 ng/ml, respectively. The t1/2 of the drug was similar across dose groups at 74.8 ± 10.8 min to 81.4 ± 4.2 min. The AUC0-t values were 30208.1 ± 2026.5 min*ng/ml, 62712.8 ± 1808.3 min*ng/ml, and 130465.2 ± 7457.4 min*ng/ml, respectively. CONCLUSION: The metabolic pathway and the proposed structure of metabolites for ET-26 hydrochloride were fully assessed. The majority of distribution for ET-26 hydrochloride occurs in the fat and liver, while the primary route of excretion for ET-26 hydrochloride is through the kidney. In dogs, pharmacokinetic features of ET-26 hydrochloride had a linear relationship with dosage.

A bare optical fiber-based biosensor is proposed for measuring the refractive index of different liquids and the binding kinetics of biomolecules to the sensor surface. This optical fiber sensor is based on the Kretschmann's configuration to attain total internal reflection (TIR) for surface plasmon resonance (SPR) excitation. One end of the bare optical fiber is coated with a gold film. By guiding the light source from the other end into the optical fiber, the light is reflected from the gold-deposited end and the surface evanescent wave is excited in the gold film-transparent material interface. Methanol and ethanol solutions with different refractive indices are used for measuring the corresponding changes in the peak values of the spectra and calculating the corresponding sensitivities. These values are experimentally determined to be in the order of 10-4~10-5 refractive index unit (RIU). Binding of proteins onto the sensor surface is also monitored in real time to obtain the binding kinetics. We believe that, in the future, this optical fiber sensor can serve as a useful biosensor for in situ measurement of allergens, antibody-antigen interactions, and even circulating tumor cells in the blood.

To study the in situ doping effect upon monotonically increasing dopant concentration, a Bi2Te3 layer doped with Fe up to ~6.9% along the growth direction was fabricated by the molecular beam epitaxy (MBE) technique. Its resistance versus temperature curve displays a superconductivity transition at about 12.3 K. Detailed structural and chemical analysis via X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and energy-dispersive X-ray spectroscopy (EDS) reveal that this layer consists of two types of unexpected Fe-Te nanostructures: one is FeTe thin layer formed near the surface, and the other is FeTe2 nanorod embedded in the Bi2Te3 layer. Based on the results of further electrical and magnetotransport studies, it is likely that the observed superconductivity originates from the interface between the FeTe nanostructure and the neighboring Bi2Te3 layer. We have addressed the formation mechanisms of the observed nanostructures, which is attributed to the strong reaction between Fe and Te atoms during the growth process. The findings of this study also provide an unusual approach to synthesizing nanostructures via heavy doping if the dopant element is strongly reactive with an element in the host matrix.

Studies have shown that hypoxia can induce cytoskeletal injury and remodeling through the activation of the RhoA/ROCK signaling pathway by hypoxia-inducible factor-1α (HIF-1α). Our previous study confirmed that CAPZA1 can modulate EMT by regulating actin cytoskeleton remodeling. However, the relationship between HIF-1α and CAPZA1 has not been illustrated. Therefore, this study aimed to investigate the mechanism by which hypoxia induces the remodeling of the actin cytoskeleton by regulating CAPZA1 in hepatocellular carcinoma (HCC) cells. In the present study, we showed that the low expression of CAPZA1 promotes HCC cell invasion and migration in vitro and in vivo by regulating actin cytoskeleton remodeling to drive EMT. Furthermore, we found that the combination of PIP2 and CAPZA1 enables CAPZA1 to be released from the barbed end of F-actin, which in turn drives the remodeling of the actin cytoskeleton. Finally, we confirmed that hypoxia increases PIP2 levels and its binding to CAPZA1 in HCC cells via the HIF-1α/RhoA/ROCK1 pathway. Thus, CAPZA1 and PIP2 could be therapeutic targets to inhibit the invasion and migration promoted by hypoxia in HCC cells.

During the atmospheric detection process in open space, the excessive phase noise is introduced into the signal, due to the atmospheric turbulence, which causes the intensity and phase fluctuation. In the previous study, a spectral data processing method based on the co-frequency and dual-wave has been used to reduce the influence of the scintillation noise from the atmospheric turbulence in open space, while the influence of the phase noise remains to be solved. So the wavelength modulated signal is theoretically analyzed at first. On studying the relationship between the dual-waves in one cycle to eliminate the phase fluctuation and reduce the phase fluctuation caused by the atmospheric turbulence, a new method of the spectral phase correction for the open space atmospheric detection has been proposed. An atmospheric detection experiment on the phase correction in the open space based on co-frequency and dual-wave has been carried out. The results show that the maximum fluctuation of the spectral signal processed with this method is 1.06%, while the power spectral density fluctuation is suppressed below 50Hz, and the Allan analysis result is 8.8 × 10-8(1s). Compared with the traditional concentration inversion method using 2f-wavelength modulation and the classical light intensity elimination, the proposed phase correction method can effectively reduce the fluctuation of random noise caused by the short-term atmospheric turbulence and the laser flashing to improve the stability of the concentration measurement, which has practical engineering value.

BACKGROUND: In this study, we investigated the clinical significance of endobronchial ultrasound elastography for differentiating malignant and benign intrathoracic lymph nodes. METHODS: A meta-analysis was performed to evaluate the sensitivity and specificity of endobronchial ultrasound elastography in diagnosing intrathoracic lymph nodes. Publications before October 1, 2017, were included for analysis. Sensitivity, specificity, and other variables were pooled using the bivariate mixed-effects regression model. RESULTS: Seven studies met the inclusion criteria and were included. The pooled sensitivity, specificity, positive likelihood ratio, negative likelihood ratio, and diagnostic odds ratio was 0.93 (95% confidence interval [CI], 0.85 to 0.97), 0.85 (95% CI, 0.78 to 0.90), 6.3 (95% CI, 4.2 to 9.2), 0.08 (95% CI, 0.04 to 0.18), and 74 (95% CI, 33 to 168), respectively. The summary receiver operating characteristic curve was 0.93 (95% CI, 0.91 to 0.95). CONCLUSIONS: The results revealed endobronchial ultrasound elastography is a new technique with high sensitivity and specificity. It has a fine performance in diagnosing intrathoracic lymph nodes.

Developing high-performance film dielectrics for capacitive energy storage has been a great challenge for modern electrical devices. Despite good results obtained in lead titanate-based dielectrics, lead-free alternatives are strongly desirable due to environmental concerns. Here we demonstrate that giant energy densities of ~70 J cm-3, together with high efficiency as well as excellent cycling and thermal stability, can be achieved in lead-free bismuth ferrite-strontium titanate solid-solution films through domain engineering. It is revealed that the incorporation of strontium titanate transforms the ferroelectric micro-domains of bismuth ferrite into highly-dynamic polar nano-regions, resulting in a ferroelectric to relaxor-ferroelectric transition with concurrently improved energy density and efficiency. Additionally, the introduction of strontium titanate greatly improves the electrical insulation and breakdown strength of the films by suppressing the formation of oxygen vacancies. This work opens up a feasible and propagable route, i.e., domain engineering, to systematically develop new lead-free dielectrics for energy storage.

Perilla frutescens is one of 60 kinds of food and medicine plants in the initial directory announced by health ministry of China. With the development of Perilla domain in recent , the breeding and application of good varieties has become the main bottleneck of its development. This study reported that applied to the system selection, add to marker-assisted method to breed perilla varieties. Through the whole genome sequencing and consistency matching, annotated the mutation locus according to genome data, and comparison analysis with Perilla common variants database, finally selected 30 non-synonymous mutation SNPs used as characteristic markers of Zhongyan Feishu No.1. those SNP marker were used as chosen standard of Perilla varieties. Finally breeding new perilla variety Zhongyan Feishu No.1, which possess to characters of the leaf and seed dual-used, high yield, high resistance, and could used to green fertilizer. The Zhongyan Feishu No.1 acquired the plant new varieties identification of Beijing city , the identification numbers is 2016054. Marker assisted identification guide new varieties breeding in plants, which can provide a new reference for breeding of medicinal plants.

BACKGROUND: Surgical margin is an important prognostic factor in hepatectomy for patients with hepatocellular carcinoma (HCC). But the extent of surgical margins is still controversial. Our study was designed to systematically evaluate the prognosis of different width of resection margin. METHODS: We conducted comprehensive searches of electronic databases including PubMed, MEDLINE, EMBASE, Cochrane, and the ISI Web of Science for relevant studies. A meta-analysis was performed by RevMan 5.3 software. RESULTS: A total of 7 studies comprising 1932 patients were included. The patients with wider surgical margin were significantly higher than those with narrow surgical margin on 3-year overall survival (odds ratio [OR]: 1.58, 95% confidence interval (95% CI): 1.21-2.06, P = .0008), 5-year overall survival (OR: 1.76, 95% CI: 1.20-2.59, P = .004), 1-year disease-free survival (DFS)/recurrence-free survival (RFS) (OR: 1.43, 95% CI: 1.12-1.82, P = .005), 3-year DFS/RFS (OR: 1.66, 95% CI: 1.35-2.03, P < .00001), and 5-year DFS/RFS (OR: 1.69, 95% CI: 1.37-2.08, P < .00001). There was no significant difference in the 1-year overall survival rate for the 2 groups (OR: 1.24, 95% CI: 0.89-1.72, P = .20). CONCLUSION: In comparison with the narrow surgical margin group (<1 cm), the wide surgical margin (≥1 cm) can significantly improve the prognosis in patients with HCC.

Chinese medicine prescriptions are a type of medical documents written by doctors after they understand the patients' conditions for syndrome differentiation. Chinese medicine prescriptions are also the basis for pharmacy personnel to dispense medicines and guide patients to use drugs. It has the legal, technical and economic significances. Chinese medicine prescriptions contain such information of names, quantity and usage. Whether the names of drugs in Chinese medicine prescriptions are standardized or not is directly related to the safety and efficacy of the drugs. At present, nonstandard clinical prescriptions are frequently seen. With "Chinese medicine prescription", "names of drug in Chinese medicine prescription" and "standards of Chinese medicine prescription" as key words, the author searched CNKI, Wanfang and other databases, and consulted nearly 100 literatures, so as to summarize current names of drugs in traditional Chinese medicine prescription, analyze the reasons, and give suggestions, in the expectation of standardizing the names of drugs used in traditional Chinese medicine prescriptions.

BACKGROUND: The integrative analysis of microRNA and mRNA expression profiles can elucidate microRNA-targeted gene function. We used this technique to elucidate insights into the immunological pathology of non-alcoholic fatty liver disease (NAFLD). METHODS: We analyzed differentially expressed microRNA and mRNA expression profiles of CD4+ T lymphocytes from the liver and mesenteric lymph nodes (MLNs) of mice with NAFLD using microarrays and RNA sequencing. Normal mice were used as controls. The target genes of microRNAs were predicted by TargetScan. Integrative analysis showed that the mRNAs were overlapped with microRNAs. Furthermore, the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were performed to predict the key genes and pathways. Then, 16 microRNAs and 10 mRNAs were validated by qRT-PCR. RESULTS: Microarray analysis suggested that 170 microRNAs were significantly de-regulated in CD4+ T lymphocytes from the liver between the two groups. Eighty mRNAs corresponded with microRNA targeted genes. KEGG analysis indicated that the MAPK pathway was consistently augmented in the liver of NAFLD mice. miR-23b, let-7e, miR-128 and miR-130b possibly played significant parts in the MAPK pathways. Furthermore, between the two groups, 237 microRNAs were significantly de-regulated in CD4+ T lymphocytes from MLNs. 38 mRNAs coincided with microRNA target genes. The metabolic pathway was consistently enriched in the MLNs of NAFLD mice. miR-206-3p, miR-181a-5p, miR-29c-3p and miR-30d-5p likely play important roles in the regulation of metabolic pathways. CONCLUSION: The results of this study presented a new perspective on the application of integrative analysis to identify complex regulation means involved in the immunological pathogenesis of NAFLD.

Hepatic stellate cells (HSCs) induce immune privilege and promote hepatocellular carcinoma (HCC) by suppressing the immune system. On the other hand, galectin-1 and miRNA-22 (miR-22) are dysregulated in HCC and serve as prognostic indicators for patients. In this study, therefore, we measured galectin-1 and miR-22 expression in HSCs isolated from HCC tissues (Ca-HSCs), and in normal liver tissues (N-HSCs) as a control. We also investigated the apoptosis rate among T cells and the production of cytokines (IFN-γ and IL-10) in HSCs co-cultured with T cells. And we used immunohistochemical staining to tested for correlation between galectin-1 expression, CD3 expression and clinicopathological features in 162 HCC patients. Our results showed that galectin-1 expression was much higher in Ca-HSCs than in N-HSCs. Overexpression of galectin-1 promoted HSC-induced T cell apoptosis and cytokine production (IFN-γ and IL-10), while miR-22 expression inhibited it. Galectin-1 expression correlated negatively with miR-22 expression in HSCs. High galectin-1 and low CD3 expression levels were associated with poor prognosis in HCC patients. These results suggest that the immunosuppressive microenvironment promoted by HSC-derived galectin-1 in HCC can be inhibited by miR-22. Galectin-1 and miR-22 could potentially serve as prognostic markers and therapeutic targets in HCC.

It needs on-line monitoring of ammonia concentration on dairy feedlot to disclose ammonia emissions characteristics accurately for reducing ammonia emissions and improving the ecological environment. The on-line monitoring system for ammonia concentration has been designed based on Tunable Diode Laser Absorption Spectroscopy (TDLAS) technology combining with long open-path technology, then the study has been carried out with inverse dispersion technique and the system. The ammonia concentration in-situ has been detected and ammonia emission rules have been analyzed on a dairy feedlot in Baoding in autumn and winter of 2013. The monitoring indicated that the peak of ammonia concentration was 6.11 x 10(-6) in autumn, and that was 6.56 x 10(-6) in winter. The concentration results show that the variation of ammonia concentration had an obvious diurnal periodicity, and the general characteristic of diurnal variation was that the concentration was low in the daytime and was high at night. The ammonia emissions characteristic was obtained with inverse dispersion model that the peak of ammonia emissions velocity appeared at noon. The emission velocity was from 1.48 kg/head/hr to 130.6 kg/head/hr in autumn, and it was from 0.004 5 kg/head/hr to 43.32 kg/head/hr in winter which was lower than that in autumn. The results demonstrated ammonia emissions had certain seasonal differences in dairy feedlot scale. In conclusion, the ammonia concentration was detected with optical technology, and the ammonia emissions results were acquired by inverse dispersion model analysis with large range, high sensitivity, quick response without gas sampling. Thus, it's an effective method for ammonia emissions monitoring in dairy feedlot that provides technical support for scientific breeding.

BACKGROUND: Survivin is an oncoprotein silenced in normal mature tissues but reactivated in serous ovarian cancer (SOC). Although transcriptional activation is assumed for its overexpression, the long 3'-untranslated region (3'-UTR) in survivin gene, which contains many alternate polyadenylation (APA) sites, implies a propensity for posttranscriptional control and therefore was the aim of our study. METHODS: The abundance of the coding region, the proximal and the distal region of survivin mRNA 3'-UTR, was evaluated by real-time polymerase chain reaction (PCR) in SOC samples, cell lines, and normal fallopian tube (NFT) tissues. The APA sites were confirmed by rapid amplification of cDNA 3' ends and DNA sequencing. Real-time PCR were used to screen survivin-targeting microRNAs (miRNAs) that were inversely correlated with survivin. The expression of an inversely correlated miRNA was restored by pre-miRNA transfection or induction with a genotoxic agent to test its inhibitory effect on survivin overexpression. RESULTS: Varying degrees of APA were observed in SOC by comparing the abundance of the proximal and the distal region of survivin 3'-UTR, and changes of 3'-UTR correlated significantly with survivin expression (r = 0.708, P< 0.01). The main APA sites are proved at 1197 and 1673 of survivin 3'-UTR by DNA sequencing. Higher level of 3'-UTR proximal region than coding region was observed in NFT, as well as in SOC and cell lines. Among the survivin-targeting miRNAs, only a few highly expressed miRNAs were inversely correlated with survivin levels, and they mainly targeted the distal part of the 3'-UTR. However, in ovarian cancer cells, restoration of an inversely correlated miRNA (miR-34c) showed little effect on survivin expression. CONCLUSIONS: In NFT tissues, survivin is not transcriptionally silenced but regulate posttranscriptionally. In SOC, aberrant APA leads to the shortening of survivin 3'-UTR which enables it to escape the negative regulation of miRNAs and is responsible for survivin up-regulation.

Extensive use of pesticides has a significant impact on the environment. Carbaryl, whose residues stay in the surface water, is one of the most widely used broad spectrum insecticides in agriculture. It is important to understand carbaryl spectral characteristics and detection methods. The characteristic of excitation-emission three-dimensional spectra of carbaryl is studied. By changing the concentration of methanol in methanol-water binary solvent, the impact of methanol-water mixture on three-dimensional fluorescence spectra of carbaryl is discussed. The results show that the characteristic excitation-emission spectra of carbaryl is single peak, the range of the excitation wavelength and emission wavelength are: 244~304 and 300~350 nm respectively, the maximum excitation/emission peak located at 280 and 335 nm. With increasing the content of methanol in methanol-water binary solvent mixture, there is no obviously spectra shift of three dimensional fluorescence spectra of carbaryl. However, the intensity of fluorescence is nonlinear dependent on the content of methanol, mainly due to the specific properties of binary mixed solvent.
