RESUMO

We report a metal-, acid-, and base-free 2-(bromomethyl)naphthalene (2-BMN)-promoted organocatalytic Cloke-Wilson rearrangement of chain doubly activated cyclopropanes for the construction of 2,3-dihydrofurans via a carbocation-initiated tandem intramolecular ring-opening/recyclization process. The strategy is especially suitable for the construction of furan units in complex molecules, providing a solution to the problem of heavy-metal residues in dihydrofuran-containing drugs synthesized by traditional metal-based protocols. Thus, it is of potential interest in synthetic and medicinal chemistry.

As a potential anode material for lithium-ion batteries (LIBs), metal tin shows a high specific capacity. However, its inherent "volume effect" may easily turn tin-based electrode materials into powder and make them fall off in the cycle process, eventually leading to the reduction of the specific capacity, rate and cycle performance of the batteries. Considering the "volume effect" of tin, this study proposes to construct a carbon coating and three-dimensional graphene network to obtain a "double confinement" of metal tin, so as to improve the cycle and rate performance of the composite. This excellent construction can stabilize the tin and prevent its agglomeration during heat treatment and its pulverization during cycling, improving the electrochemical properties of tin-based composites. When the optimized composite material of C@Sn/NSGr-7.5 was used as an anode material in LIB, it maintained a specific capacity of about 667 mAh g-1 after 150 cycles at the current density of 0.1 A g-1 and exhibited a good cycle performance. It also displayed a good rate performance with a capability of 663 mAh g-1, 516 mAh g-1, 389 mAh g-1, 290 mAh g-1, 209 mAh g-1 and 141 mAh g-1 at 0.1 A g-1, 0.2 A g-1, 0.5 A g-1, 1 A g-1, 2 A g-1 and 5 A g-1, respectively. Furthermore, it delivered certain capacitance characteristics, which could improve the specific capacity of the battery. The above results showed that this is an effective method to obtain high-performance tin-based anode materials, which is of great significance for the development of new anode materials for LIBs.

BACKGROUND: Due to the broad application of next-generation sequencing, the molecular diagnosis of genetic disorders in pediatric neurology is no longer an unachievable goal. However, treatments for neurological genetic disorders in children remain primarily symptomatic. On the other hand, with the continuous evolution of therapeutic viral vectors, gene therapy is becoming a clinical reality. From this perspective, we wrote this review to illustrate the current state regarding viral-mediated gene therapy in childhood neurological disorders. DATA SOURCES: We searched databases, including PubMed and Google Scholar, using the keywords "adenovirus vector," "lentivirus vector," and "AAV" for gene therapy, and "immunoreaction induced by gene therapy vectors," "administration routes of gene therapy vectors," and "gene therapy" with "NCL," "SMA," "DMD," "congenital myopathy," "MPS" "leukodystrophy," or "pediatric metabolic disorders". We also screened the database of ClinicalTrials.gov using the keywords "gene therapy for children" and then filtered the results with the ones aimed at neurological disorders. The time range of the search procedure was from the inception of the databases to the present. RESULTS: We presented the characteristics of commonly used viral vectors for gene therapy for pediatric neurological disorders and summarized their merits and drawbacks, the administration routes of each vector, the research progress, and the clinical application status of viral-mediated gene therapy on pediatric neurological disorders. CONCLUSIONS: Viral-mediated gene therapy is on the brink of broad clinical application. Viral-mediated gene therapy will dramatically change the treatment pattern of childhood neurological disorders, and many children with incurable diseases will meet the dawn of a cure. Nevertheless, the vectors must be optimized for better safety and efficacy.

The collective oscillation of free electrons at the nanoscale surface of gold nanostructures is closely modulated by tuning the size, shape/morphology, phase, composition, hybridization, assembly, and nanopatterning, along with the surroundings of the plasmonic surface located at a dielectric interface with air, liquid, and solid. This review first introduces the physical origin of the intrinsic optical properties of gold nanostructures and further summarizes stimuli-responsive changes in optical properties, metal-field-enhanced optical signals, luminescence spectral shaping, chiroptical response, and photogenerated hot carriers. The current success in the landscape of nanoscience and nanotechnology mainly originates from the abundant optical properties of gold nanostructures in the thermodynamically stable face-centered cubic (fcc) phase. It has been further extended by crystal phase engineering to prepare thermodynamically unfavorable phases (e.g., kinetically stable) and heterophases to modulate their intriguing phase-dependent optical properties. A broad range of promising applications, including but not limited to full-color displays, solar energy harvesting, photochemical reactions, optical sensing, and microscopic/biomedical imaging, have fostered parallel research on the multitude of physical effects occurring in gold nanostructures. Anthropogenic and geological factors play an essential role in the variability of groundwater quality, resulting in a weak spatial dependence of groundwater trace elements. Thus, it is an essential study to investigate the factors affecting groundwater quality and its spatial abundance of trace elements (including As, B, and other metalloids). In this study, samples are obtained from a Permian sandstone fracture aquifer in a coal mining area. A multivariate statistical analysis, hydrogeochemistry modeling, and spatial autocorrelation analysis were used to analyze the data. The results showed that Moran index was positive for all trace elements, which had good spatial autocorrelation. The Local indicators of spatial association (LISA) indicated that trace elements were clustered. The hydrogeochemical modeling results indicated that the precipitation and stability of iron-phase minerals, such as rhodochrosite and arsenic (As) absorption on the surface of iron-phase minerals in the aquifer, may limit concentrations in the southern region. The spatial autocorrelations of both As and Boron (B) were positive (high-high) in the western areas, indicating that As contamination occurred from both natural geological causes and human coal mining activities. In contrast, B contamination was mainly linked to the influence of human agricultural or industrial activities. Over 96% of the groundwater concentrations of As (10 µg/L) and B (300 µg/L) in the study area exceeded World Health Organization (WHO) limits. Overall, the results of this work could help decision-makers involved in regional water quality management visualize disperse zones where specific anthropogenic and geological processes may threaten groundwater quality.

The selective capture of C2H6 from C2H6/C2H4 mixtures is of critical importance to realize the efficient one-step purification of C2H4 but remains challenging due to their similar properties and smaller quadrupole moment of C2H6 that usually result in C2H4-preferring adsorption. Herein, we reported two isostructural pillared-layer metal-organic frameworks, ZUL-C3 and ZUL-C4, which were constructed by mixed polycycloalkane-type ligands. Their low-polar pore environment along with more accessible low-polar C-H binding sites on the pore surface are conducive to generate more van der Waals interactions with C2H6 while the carboxylic groups distributed at four corners of pores form stronger and more dipolar interactions with C2H6, cooperatively resulting in a good C2H6/C2H4 uptake ratio of 1.50 for ZUL-C3 and 1.72 for ZUL-C4 in static adsorption experiments and a high C2H4 (>99.99% purity) productivity of 10.1 L/kg for ZUL-C3 and 14.6 L/kg for ZUL-C4 from an equimolar C2H6/C2H4 mixture in breakthrough experiments.

Sodium storage batteries are one of the ever-increasing next-generation large-scale energy storage systems owing to the abundant resources and low cost. However, their viability is severely hampered by dendrite-related hazards on anodes. Herein, a novel ultra-thin (8 µm) exterior-nonporous separator composed with honeycomb-structured fibers was prepared for homogeneous Na deposition and suppressed dendrite penetration. The unhindered ion transmission was greatly benefited from honeycomb-structured fibers with huge electrolyte uptake (376.7%) and the polymer's inherent transport ability. Additionally, polar polymer chains consisting of polyethersulfone and polyvinylidene customize the highly aggregated solvation structure of electrolytes via substantial solvent immobilization, facilitating ion-conductivity-enhanced inorganic-rich solid-electrolyte interphase with remarkably interface endurance. With the reliable mechanical strength of separator, the assembled sodium-ion full cell delivers significantly improved energy density and high safety enabling resisting stable operation under cutting and rolling. The as-prepared separator can further be generalized to lithium-based batteries for which apparent dendrite inhibition and cyclability are accessible, and demonstrates its potential for practical application. Environmental heavy atoms can further enhance the room temperature phosphorescence (RTP) emissions of gadolinium-labeled hematoporphyrin monomethyl ether (Gd-HMME) by way of the external heavy atom effect (HAE). However, the macroscopic phosphorescence intensity covered the intrinsic effect of the environmental heavy atoms. In this study, a method of separating the external HAE from the total is performed, and a quantity to describe the intrinsic nature of external HAE is defined. The environmental Gd3+ concentration evolution of the phosphorescent transition rate (kP) is obtained by correlated absorption, emission, and time-resolved spectroscopy. The kP increases linearly with environmental Gd3+ concentration, while the intercept kP0 coincides with that of the internal HAE. The slope κ could be calculated, and it is a quantity free of the Gd3+ concentration and only relies on the type of environmental heavy atoms. In addition, the environmental Lu3+ exhibits similar functionality to Gd3+ in external HAE. Environmental Pd2+ quenches the phosphorescence intensity macroscopically, while it enhances the HAE intrinsically. Our method provides an alternative insight into the intrinsic nature of environmental heavy atoms.

In this paper, the physicochemical properties, antioxidant and anti-aging abilities of three new Tremella hydrocolloids were studied. The physicochemical properties were characterized by Fourier transform infrared spectroscopy, differential scanning calorimeter, X-ray diffractometry etc. The antioxidant activities of Tremella hydrocolloids were determined by ABTS radical, DPPH radical scavenging activity. The anti-aging ability of Tremella hydrocolloids was also investigated by using the organism model of Caenorhabditis elegans (C. elegans). The results showed that the ES-THD displayed the highest radical scavenging capacity and the best anti-aging abilities. The ability of ES-THD to scavenge ABTS radicals reached 100 % at 2 mg/mL, the ability of ES-THD to scavenge DPPH radicals reached 45.55 %. Compared with the control group, the average lifespan was 29.17 % longer fed with ES-THD. These results provide the evidence basis for the use of Tremella hydrocolloids as food texture modifiers, antioxidants, and anti-aging agents in the food industries.

Cadmium (Cd) is a ubiquitous toxic metal and environmental pollutant. Increasing studies have shown that Cd exposure increases the incidence of various endocrine system diseases, including thyrotoxicity reflected by thyroid structural damage and endocrine toxicity. However, the observed outcomes are complex and conflicting, leading to the mechanism of Cd-induced thyrotoxicity remaining obscure. In this study, 4-week-old male C57BL/6 mice were given 2 or 7 mg/kg Cadmium Chloride (CdCl2) intragastrically for 4 and 8 weeks, and the Cd-mediated thyrotoxicity was evaluated by determining alterations in thyroid structure and endocrine function, and alterations of oxidant stress, apoptosis, and pyroptosis. Our data showed that Cd exposure could reduce body weight and induce thyrotoxicity by impairing thyroid follicular morphology and endocrine function, accompanied by elevated oxidative stress and apoptosis, macrophage infiltration, and inflammatory cytokine secretion. Importantly, Cd significantly promoted thyroid follicular cell pyroptosis by increasing Nlrp3, Asc, Caspase-1, Gsdmd, IL-1ß, and IL-18 expression. Mechanistical analysis suggested that Cd treatment could inhibit antioxidant pathway by downregulating antioxidant response protein, Nrf2, and upregulating its negative feedback regulator, Keap1. Collectively, our in vivo findings suggest that Cd exposure could facilitate thyroid follicular cell pyroptosis by inhibiting Nrf2/Keap1 signaling, thereby disrupting thyroid tissue structure and endocrine function, which offers novel insights into the Cd-mediated detrimental consequences on thyroid homeostasis.

Background: High mobility group box protein 1 (HMGB1) is considered as a kind of sterile inflammatory mediators, which is an overexpression in patients with unexplained recurrent spontaneous abortion (URSA). Specific targeting effect of aspirin on HMGB1 has been revealed. Our previous studies have explored the application of HMGB1 as a therapeutic target of aspirin in URSA disease of mice model and human, but the dynamic process of aspirin downregulating HMGB1 concentration has not been demonstrated. Methods: From December 2018 to November 2020, women with URSA (n = 91) and control women (n = 90) with no history of recurrent abortion or adverse pregnancy were included in the Reproductive Medicine Center of the First Affiliated Hospital of Anhui Medical University. ELISA was applied to detect the concentrations of HMGB1 and IFN-γ in the peripheral blood. Thirty-one URSA patients were monitored for low-dose aspirin treatment (2 and 4 weeks), the changes of HMGB1 and IFN-γ concentrations in peripheral blood of URSA patients before and after using aspirin were compared, and pregnancy outcomes after aspirin treatment were followed up. Results: The levels of HMGB1 in peripheral blood were significantly higher in URSA patients compared with controls, decreasing trends of HMGB1 and IFN-γ concentrations in plasma of URSA patients were observed after treatment with low-dose aspirin continuously, and the expression of HMGB1 was positively correlated with IFN-γ. There were no birth abnormalities in the babies of the URSA patients treated with aspirin. Conclusions: High levels of HMGB1 may be one of the pathogenesis of URSA. Low-dose aspirin may provide protective effect on the HMGB1-triggered URSA.

Fully inorganic perovskite cesium lead triiodide (CsPbI3) has garnered much attention from researcher for photovoltaic application because of its excellent thermal stability compared with the inorganic-organic hybrid counterparts, along with the potential to serve as the top cell in tandem devices with silicon solar cell. However, the active α-phase cubic CsPbI3 spontaneously tends to transform into the non-perovskite δ-CsPbI3 when subjected to ambient condition. This work proposes an effective method to fabricate high-quality and stable α-phase cubic CsPbI3 films via phosphorus pentachloride (PCl5) additive, in which the PCl5 acts as colloidal binder for modulation crystallization dynamics of perovskites, resulting in high-quality film and a significantly suppressed phase transition. Finally, highly stable CsPbI3 perovskite solar cells can be achieved with a power conversion efficiency up to 17.85%, and long-term stability in N2 filled glove box.

Motility is one of the most critical features to evaluate sperm quality. As longitudinal rolling of human sperm has long been ignored until recently, its detailed dynamics and cellular biological mechanisms are still largely unknown. Here we report an optical-tweezers-based method to evaluate the chirality and frequency of sperm rotation. According to the intensity distribution patterns of off-focus micron-size particles, we established a method to judge the orientation of the sperm head along the optical axis in the optical trap. Together with the rotation direction of the projection of the sperm head, the chirality of longitudinal rolling of sperm can be measured without the application of three-dimensional tracking techniques or complex optical design. By video tracking optically trapped sperm cells from different patients, both rolling chirality and rolling frequency were analyzed. In this study, all the vertically trapped human sperm cells adopt a right-hand longitudinal rolling. The orientation and rolling frequency but not the rolling chirality of sperm in the optical trap are affected by the trap height. The rotation analysis method developed in this study may have clinical potential for sperm quality evaluation.

Maltodextrin (MD), the hydrolyzed starch product, is a promising alternative ingredient to improve the quality of starch-based foods. The effects of MD on the physicochemical, microstructural, and cooking properties of sweet potato starch (SPS) noodles, as well as the mechanism of SPS-MD interactions, are discussed. Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) results indicated that MD at a suitable concentration can improve the ordered structure of SPS-MD gels. The cooking loss showed lower values of 1.47-2.16% at 0.5-2.0 wt% MD. For the texture properties, an increase in hardness and chewiness occurred at first with the addition of MD, followed by a decreasing trend, showing a maximum value at 2.0 wt% of MD. The pasting and thermal results verified the increased stability of the starch granules with MD < 3 wt%. Additionally, SPS formed a solid-like gel with MD, and the main interaction forces between SPS and MD were hydrogen bonding. The scanning electron microscopy results revealed that the higher concentrations of MD (>3 wt%) loosened the gel structure and markedly increased the pore size. These results help us to better understand the interaction mechanism of the SPS-MD complex and facilitate the development of SPS-based gel products.

This study investigated the effects of low molecular weight organic acids (citric acid and malic acid) on the migration properties of nickel in soil. A reclaimed soil sample was obtained from the Panyi Mine in Huainan City, China. The effects of adding different concentrations of Ni, citric acid (CA) and malic acid (MA) were assessed on the migration and transformation of soil Ni forms. The results showed: (1) An increase in soil Ni activity with increasing Ni concentrations. (2) An increased proportion of exchangeable forms of Ni in soil with increased malic acid and citric acid concentrations, effectively promoting Ni mobility. In addition, the active Ni fraction in reclaimed soil increased significantly with increasing concentrations of citric and malic acid. The nickel activation effect of citric acid was found to be higher than that of malic acid. (3) The activation effect of organic acids on Ni weakened with aging, exhibiting a gradual transformation from the loosely bound form of Ni, to the strongly bound form. The results of this study provide a theoretical basis for improving the effectiveness and efficiency of the phytoremediation techniques used for the treatment of Ni-polluted soils.

Heterogeneous Brønsted acidic catalysts such as phosphoric acids are the conventional activators for organic transformations. However, the photocatalytic performance of these catalysts is still rarely explored. Herein, a novel Zr-based metal-organic framework Zr-MOF-P with phosphoric acids as a heterogeneous photocatalyst has been fabricated, which shows high selectivity and reactivity towards the photo-oxidation of sulfides under white light illumination. A mechanism study indicates that the selective oxygenation of sulfides occurs with triplet oxygen rather than common reactive oxygen species (ROS). When Zr-MOF-P is irradiated, the hydroxyl group of phosphoric acid is converted into oxygen radical, which takes an electron from the sulfides, and then the activated substrates react with the triplet oxygen to form sulfoxides, avoiding the destruction of the catalysts and endowing the reaction with high substrate compatibility and fine recyclability.

The accurate estimation of the mass and center of gravity (CG) position is key to vehicle dynamics modeling. The perturbation of key parameters in vehicle dynamics models can result in a reduction of accurate vehicle control and may even cause serious traffic accidents. A dual robust embedded cubature Kalman filter (RECKF) algorithm, which takes into account unknown measurement noise, is proposed for the joint estimation of mass and CG position. First, the mass parameters are identified based on directly obtained longitudinal forces in the distributed drive electric vehicle tires using the whole vehicle longitudinal dynamics model and the RECKF. Then, the CG is estimated with the RECKF using the mass estimation results and the vertical vehicle model. Finally, different virtual tests show that, compared with the cubature Kalman algorithm, the RECKF reduces the root mean square error of mass and CG by at least 7.4%, and 2.9%, respectively.

Aims: Abnormal changes in cardiac function have been reported in menopausal women, but there are few clinical studies on this topic. Erxian decoction (EXD) is a classic prescription that is widely used in the treatment of female menopausal diseases. The purpose of this study was to investigate the dynamic evolution of cardiac function and glucose and lipid metabolism in ovariectomized (OVX) rats and the intervention effect of EXD. Materials and Methods: The OVX climacteric rat model was established by bilateral ovariectomy. After successful modeling, the rats were randomly divided into four groups: the sham operation (SHAM) group (equal volumes of purified water), OVX group (equal volumes of purified water), estradiol (E2) group (1.8 × 10-4 g/kg), and EXD group (9 g/kg). Each group of rats was treated for 16 weeks. At the 4th, 8th, 12th, and 16th weeks after treatment, the cardiac function of the rats in each group was evaluated by ultrasound. The coaxial method was used to measure blood pressure (BP). Serum endothelin-1 (ET-1) and angiotensin-2 (Ang II) levels were determined by the enzyme-linked immunosorbent assay (ELISA). The strip method was used to measure fasting blood glucose (FBG). The serum total cholesterol (TC) and triglyceride (TG) levels of rats were measured with the oxidase method. Direct methods were used to measure serum high-density lipoprotein (HDL-C) and low-density lipoprotein (LDL-C) levels. At week 16 of dosing, serum E2 levels were determined by E2 radioimmunoassay. The myocardium and uterus of the rats in each group were stained with HE (hematoxylin-eosin). The ultrastructure of the rat myocardium was observed by electron microscopy. Results: After the 16th week of treatment, the serum E2 level decreased (P < 0.05), and the uterus was atrophied in OVX rats. The cardiac ejection fraction (EF%) decreased at 4 weeks after treatment, and systolic and diastolic function decreased after 12 weeks. After the 16th week, the EF%, which reflects the "pump" function of the heart, decreased significantly (P < 0.05 or P < 0.01). At the 4th, 8th, 12th, and 16th weeks of treatment, the systolic blood pressure (SBP), diastolic blood pressure (DBP), and mean pressure (MBP) of the rats in the OVX group increased with time (P < 0.05 or P < 0.01). The serum ET-1 and Ang II levels of rats in the OVX group increased (P < 0.05 or P < 0.01). In the OVX group, FBG was increased (P < 0.05 or P < 0.01), and blood lipids, especially LDL-C, were significantly increased (P < 0.05 or P < 0.01). After the 16th week of treatment, the myocardial tissue of OVX rats showed obvious pathological changes. EXD significantly increased serum E2 levels (P < 0.01), decreased ET-1 and Ang II levels (P < 0.01), reduced the cardiac function risk factors BP, FBG, and blood lipids, and significantly improved cardiac function and structural changes in OVX rats (P < 0.05 or P < 0.01). Conclusions: EXD can improve abnormal cardiac structure and function in OVX rats.

Melatonin (MT) can improve the effect of cryopreservation on oocytes by suppressing oxidative stress and maintaining the permeability of the oolemma. In this study, MT was firstly applied to human oocytes' cryopreservation to explore the effect of prolonged cryopreservation on developmental competence and its role. Collected in vitro-matured human oocytes were cryopreserved in MT-containing or MT-free medium for 0 and 6 months; after warming, viable oocytes were assessed for developmental viability, intracellular protein expression, mitochondrial function, and oxidation-antioxidant system. Meanwhile, fresh oocytes were set as the control. The results showed that with the extension of cryopreservation time, the developmental competence of oocytes gradually declined, accompanied by the down-regulation of most mitochondrial function-related proteins, the reduction in ATP and GSH production, the increase in ROS accumulation, and the aggravation of the imbalance of ROS/GSH in oocytes. However, the participation of MT seemed to effectively mitigate these negative effects. Therefore, we speculate that melatonin may maintain normal ATP production and ROS/GSH balance in cryopreserved oocytes by protecting mitochondrial function and inhibiting oxidative damage, thereby effectively maintaining the developmental competence of human oocytes in prolonged cryopreservation.

Early diagnosis of drug susceptibility for tuberculosis (TB) patients could guide the timely initiation of effective treatment. We evaluated a novel multiplex xMAP TIER (Tuberculosis-Isoniazid-Ethambutol-Rifampicin) assay based on the Luminex xMAP system to detect first-line anti-tuberculous drug resistance. Deoxyribonucleic acid samples from 353 Mycobacterium tuberculosis clinical isolates were amplified by multiplex polymerase chain reaction, followed by hybridization and analysis through the xMAP system. Compared with the broth microdilution method, the sensitivity and specificity of the xMAP TIER assay for detecting resistance was 94.9% (95%CI, 90.0-99.8%) and 98.9% (95%CI, 97.7-100.0%) for rifampicin; 89.1% (95%CI, 83.9-94.3%) and 100.0% (95%CI, 100.0-100.0%) for isoniazid; 82.1% (95% CI, 68.0-96.3%) and 99.7% (95% CI, 99.0-100.0%) for ethambutol. With DNA sequencing as the reference standard, the sensitivity and specificity of xMAP TIER for detecting resistance were 95.0% (95% CI, 90.2-99.8%) and 99.6% (95% CI, 98.9-100.0%) for rifampicin; 96.9% (95% CI, 93.8-99.9%) and 100.0% (95% CI, 100.0-100.0%) for isoniazid; 86.1% (95% CI, 74.8-97.4%) and 100.0% (95% CI, 100.0-100.0%) for ethambutol. The results achieved showed that the xMAP TIER assay had good performance for detecting first-line anti-tuberculosis drug resistance, and it has the potential to diagnose drug-resistant tuberculosis more accurately due to the addition of more optimal design primers and probes on open architecture xMAP system.
