RESUMO

Bisphenol analogues (BPs), triclocarban (TCC), and triclosan (TCS) are well-known environmental endocrine disrupters. Many studies have characterized their occurrence in the freshwater environment. However, their environmental behaviors in the coastal marine environment remain poorly understood. Here, matched seawater and sediment samples were collected from East China Sea, and analyzed for 13 BPs (including halogenated derivatives of bisphenol A), TCC, and TCS. Cadmium (Cd2+) pollution occurring in salt-affected soils has become an increasing environmental concern in the world. Fast-growing poplars have been widely utilized for phytoremediation of soil contaminating heavy metals (HMs). However, the woody Cd2+-hyperaccumulator, Populus × canescens, is relatively salt-sensitive and therefore cannot be directly used to remediate HMs from salt-affected soils. The aim of the present study was to testify whether colonization of P. × canescens with ectomycorrhizal (EM) fungi, a strategy known to enhance salt tolerance, provides an opportunity for affordable remediation of Cd2+-polluted saline soils. Ectomycorrhization with Paxillus involutus strains facilitated Cd2+ enrichment in P. × canescens upon CdCl2 exposures (50 µM, 30 min to 24 h). The fungus-stimulated Cd2+ in roots was significantly restricted by inhibitors of plasmalemma H+-ATPases and Ca2+-permeable channels (CaPCs), but stimulated by an activator of plasmalemma H+-ATPases. NaCl (100 mM) lowered the transient and steady-state Cd2+ influx in roots and fungal mycelia. Noteworthy, P. involutus colonization partly reverted the salt suppression of Cd2+ uptake in poplar roots. EM fungus colonization upregulated transcription of plasmalemma H+-ATPases (PcHA4, 8, 11) and annexins (PcANN1, 2, 4), which might mediate Cd2+ conductance through CaPCs. EM roots retained relatively highly expressed PcHAs and PcANNs, thus facilitating Cd2+ enrichment under co-occurring stress of cadmium and salinity. We conclude that ectomycorrhization of woody hyperaccumulator species such as poplar could improve phytoremediation of Cd2+ in salt-affected areas.

To improve the high brittleness of recycled aggregate concrete containing iron ore tailings (TRAC), the feasibility of adding polypropylene fiber (PPF) to TRAC was studied by performing a compression stress-strain curve test, scanning electron microscope characterization, and a freeze-thaw cycle test. The results indicated that PPF had a beneficial impact on reducing the brittleness of TRAC. With the increase in PPF content, the peak strain increased, the elastic modulus decreased, and the peak stress and energy absorption capacity increased at first and then decreased. Furthermore, the microstructure investigation revealed that the interface friction between the PPF, aggregate, and cement matrix was the main source of energy dissipation. When the load acted on the concrete, the stress was dispersed to the fiber monofilaments, thus effectively enhancing the peak stress and peak strain of concrete and suppressing the generation and development of cracks in the concrete. In terms of freeze-thaw resistance, adding a small amount of PPF could reduce the negative effects of the freeze-thaw process on the cement matrix. On the premise of ensuring strength, the waste utilization should be as high as possible. Therefore, it was suggested that the content of PPF in fiber-reinforced tailings recycled aggregate concrete (TRAC-PP) should be 0.6%.

FLOWERING LOCUS M (FLM) is a well-known MADS-box transcription factor that is required for preventing early flowering under low temperatures in Arabidopsis thaliana. Alternative splicing of FLM is involved in the regulation of temperature-responsive flowering. However, how the basic transcript level of FLM is regulated is largely unknown. Here, we conducted forward genetic screening and identified a previously uncharacterized flowering repressor gene, UBA2c. Genetic analyses indicated that UBA2c represses flowering at least by promoting FLM transcription. We further demonstrated that UBA2c directly binds to FLM chromatin and facilitates FLM transcription by inhibiting histone H3K27 trimethylation, a histone marker related to transcriptional repression. UBA2c encodes a protein containing two putative RNA recognition motifs (RRMs) and one prion-like domain (PrLD). We found that UBA2c forms speckles in the nucleus and that both the RRMs and PrLD are required not only for forming the nuclear speckles but also for the biological function of UBA2c. These results identify a previously unknown flowering repressor and provide insights into the regulation of flowering time.

Trichodermin, a trichothecene first isolated in Trichoderma species, is a sesquiterpenoid antibiotic that exhibits significant inhibitory activity to the growth of many pathogenic fungi such as Candida albicans, Rhizoctonia solani, and Botrytis cinerea by inhibiting the peptidyl transferase involved in eukaryotic protein synthesis. Trichodermin has also been shown to selectively induce cell apoptosis in several cancer cell lines and thus can act as a potential lead compound for developing anticancer therapeutics. The biosynthetic pathway of trichodermin in Trichoderma has been identified, and most of the involved genes have been functionally characterized. An exception is TRI3, which encodes a putative acetyltransferase. Here, we report the identification of a gene cluster that contains seven genes expectedly involved in trichodermin biosynthesis (TRI3, TRI4, TRI6, TRI10, TRI11, TRI12, and TRI14) in the trichodermin-producing endophytic fungus Trichoderma taxi. As in Trichoderma brevicompactum, TRI5 is not included in the cluster. Functional analysis provides evidence that TRI3 acetylates trichodermol, the immediate precursor, to trichodermin. Disruption of TRI3 gene eliminated the inhibition to R. solani by T. taxi culture filtrates and significantly reduced the production of trichodermin but not of trichodermol. Both the inhibitory activity and the trichodermin production were restored when native TRI3 gene was reintroduced into the disruption mutant. Furthermore, a His-tag-purified TRI3 protein, expressed in Escherichia coli, was able to convert trichodermol to trichodermin in the presence of acetyl-CoA. The disruption of TRI3 also resulted in lowered expression of both the upstream biosynthesis TRI genes and the regulator genes. Our data demonstrate that T. taxi TRI3 encodes an acetyltransferase that catalyzes the esterification of the C-4 oxygen atom on trichodermol and thus plays an essential role in trichodermin biosynthesis in this fungus.

Studies have demonstrated the worldwide presence of bisphenol A (BPA) and its toxic effects on human health. BPA may exist as several structural isomers, which are byproducts in industrial BPA production. However, nearly nothing is known about the occurrence of BPA isomers in human blood and the partitioning of BPA metabolites between human serum and whole blood. In this study, BPA, BPA-sulfate (BPA-S), and BPA-glucuronide (BPA-G) were quantified in 144 pairs of serum and whole blood samples from Chinese participants. BPA was detected in 115 serum and 121 whole blood samples, with mean concentrations of 0.53 and 0.88 ng/mL, respectively. Herein, we re-explored α-cyano-4-hydroxycinnamic acid (CHCA) as a reactive matrix for selective and sensitive analysis of glutathione (GSH) by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). CHCA efficiently reacted with GSH, and the resulting CHCA-GSH conjugate was readily detected by MALDI-MS without interferences. The detection limit of the CHCA-GSH conjugate decreased to 200 pmol µL-1, which was 2 orders of magnitude lower than that of pure GSH.Forapplication, CHCA was successfully applied for the detection of GSH, present in HepG2 cell lysates. The results demonstrated detection advantages of simple, high-throughput, and selective and screening of GSH in biological samples by MALDI-MS.

Sea Island cotton (Gossypium barbadense) is the source of the world's finest fiber-quality cotton, yet relatively little is understood about genetic variations among diverse germplasms, genes underlying important traits, and the effects of pedigree selection. Here, we resequenced 336 G. barbadense accessions and identified 16 million SNPs. Phylogenetic and population structure analyses revealed two major gene pools and a third admixed subgroup derived from geographical dissemination and interbreeding. We conducted a genome-wide association study (GWAS) of 15 traits including fiber quality, yield, disease resistance, maturity, and plant architecture. The highest number of associated loci was for fiber quality, followed by disease resistance and yield. Using gene expression analyses and VIGS transgenic experiments, we confirmed the roles of five candidate genes regulating four key traits, i.e., disease resistance, fiber length, fiber strength, and lint percentage. Geographical and temporal considerations demonstrated selection for the superior fiber quality (fiber length and fiber strength), and high lint-percentage in improving G. barbadense in China. Pedigree selection breeding increased Fusarium wilt disease resistance, and separately improved fiber-quality and yield. Our work provides a foundation for understanding genomic variation and selective breeding of Sea Island cotton.

The whole ecosystem is suffering from serious plastic pollution. Automatic and accurate classification is an essential process in plastic effective recycle. In this work, we proposed an accurate approach for plastics classification using a residual network based on laser-induced breakdown spectroscopy (LIBS). To increasing efficiency, the LIBS spectral data were compressed by peak searching algorithm based on continuous wavelet, then were transformed to characteristic images for training and validation of the residual network. Acrylonitrile butadiene styrene (ABS), polyamide (PA), polymethyl methacrylate (PMMA), and polyvinyl chloride (PVC) from 13 manufacturers were used. The accuracy of the proposed method in few-shot learning was evaluated. The results show that when the number of training image data was 1, the verification accuracy of classification by plastic type under residual network still kept 100%, which was much higher than conventional classification algorithms (BP, kNN and SVM). Furthermore, the training and testing data were separated from different manufacturers to evaluate the anti-interference properties of the proposed method from various additives in plastics, where 73.34% accuracy was obtained. To demonstrate the superiority of classification accuracy in the proposed method, all the evaluations were also implemented by using conventional classification algorithm (kNN, BP, SVM algorithm). The results confirmed that the residual network has a significantly higher accuracy in plastics classification and shows great potential in plastic recycle industries for pollution mitigation.

Tumor-induced osteomalacia TIO is a rare paraneoplastic syndrome caused by excessive production of fibroblast growth factor 23 FGF23 by a tumor. Previous studies have revealed generalized mineralization defects and low areal bone mineral density aBMD in TIO. However, data on the bone microarchitecture in TIO are limited. In this study, we evaluated the microarchitecture in the peripheral distal radius and tibia and axial lumbar spine skeleton using high-resolution peripheral quantitative computed tomography HR-pQCT and trabecular bone score TBS and investigated related factors in a large cohort of Chinese patients with TIO. A total of 186 patients with TIO who had undergone dual-energy X-ray absorptiometry DXA or HR-pQCT scans were enrolled. Compared with age-, sex- and BMI-matched healthy controls, TIO patients n=113 had lower vBMD, damaged microstructure and reduced bone strength in the peripheral skeleton, especially at the tibia. The average TBS obtained from 173 patients was 1.15 ± 0.16. The proportion of patients with abnormal TBS <1.35 was higher than that with low L1-4 aBMD Z-score Z≤-2 43.9% vs. 89.6%, p < 0.001. Higher intact fibroblast growth factor 23 iFGF23, intact parathyroid hormone iPTH, alkaline phosphatase and ß-isomerized C-terminal telopeptide of type I collagen ß-CTx levels, more severe mobility impairment and a history of fracture were associated with poorer HR-pQCT parameters but not with lower TBS. However, greater height loss and longer disease duration were correlated with worse HR-pQCT parameters and TBS. Moreover, TBS was correlated with both trabecular and cortical HR-pQCT parameters in TIO. In conclusion, we revealed impaired bone microarchitecture in the axial and peripheral skeleton in a large cohort of Chinese TIO patients. HR-pQCT parameters and TBS showed promising advantages over aBMD for assessing bone impairment in patients with TIO. A longer follow-up period is needed to observe changes in bone microarchitecture after tumor resection.

PURPOSE: Low-dose rivaroxaban is often given to patients with atrial fibrillation (AF) around the world, but the rationale for its use remains unclear. We aimed to compare the efficacy and safety of standard- or low-dose rivaroxaban in patients with AF through systematic review of literature with meta-analysis. METHODS: We searched PubMed, Web of Science, EMBASE, Clinical Trials.gov, the Cochrane Library, and Bayer trial website from inception of each database until June 2020. Randomized controlled trials (RCTs) and cohort studies were included in the meta-analysis. A random-effects model was employed to calculate the pooled effect estimates. RESULTS: Two RCTs and 17 cohort studies were included in the qualitative analysis. Indirect comparison of RCTs showed no significant difference between the two rivaroxaban dosages in risk of efficacy or safety outcomes (p > 0.05). Indirect comparison of cohort studies showed a lower risk of MACE among Caucasians in standard-dose group (HR 0.779; 95% CI 0.687-0.884; p < 0.001). Bleeding outcomes did not differ significantly between the two dosage regimens in Asian or Caucasian populations, except that the standard dose was associated with higher risk of major bleeding among elderly Caucasian patients (HR 1.329; 95% CI 1.141-1.547; p < 0.001). The quality of evidence was rated ranging from very low to low for all the efficacy and safety outcomes. CONCLUSION: In Caucasians with AF, standard-dose rivaroxaban may prevent MACE significantly better than low-dose treatment. Further studies in Asians are needed to verify the advantages of the standard dose.

As a micro-/nano-sized hydrogel, polymeric microgel not only has three-dimensional (3D) molecular networks but also displays the small-size effect, which has been widely used in various fields, such as drug nanocarrier, photonic crystal, functional coating, and aqueous lubrication. In this work, a photothermal lubricating coating was prepared using polymeric/inorganic hybrid microgels and its surficial friction was deliberately modulated by the remote irradiation of near-infrared (NIR) light. Specifically, a photothermal hybrid microgel, Fe3O4@poly(N-isopropylacrylamide-co-polyacrylic acid) (Fe3O4@PNA), was first fabricated and then sprayed onto poly(ethyleneimine) (PEI)-modified substrate to form a lubricating microgel coating. At room temperature, this microgel coating was hydrophilic and achieved good hydration lubrication with relatively low friction. After the introduction of NIR light, the photothermal microgel coating converted light energy into heat energy for increasing its own temperature rapidly. Due to the thermosensitive PNA shell, the wettability of the coating was transformed to hydrophobicity above the lower critical solution temperature (LCST), resulting in a remarkable increase in friction. In other words, the surficial friction of this microgel coating could be reversibly modulated using NIR light. This work expands the application scope of microgels in the field of aqueous lubrication and introduces the functional microgels into making the smart lubricating coating for the first time. This basic research in the field of friction control may provide an efficient strategy for the design of interfacial sensing, controlled transmission, and intelligent manipulator.

With the increasing pressure of current life, fatigue caused by high-pressure work has deeply affected people and even threatened their lives. In particular, fatigue driving has become a leading cause of traffic accidents and deaths. This paper investigates electroencephalography (EEG)-based fatigue detection for driving by mining the latent information through the spatial-temporal changes in the relations between EEG channels. First, EEG data are partitioned into several segments to calculate the covariance matrices of each segment, and then we feed these matrices into a recurrent neural network to obtain high-level temporal information. Second, the covariance matrices of whole signals are leveraged to extract two kinds of spatial features, which will be fused with temporal characteristics to obtain comprehensive spatial-temporal information. Experiments on an open benchmark showed that our method achieved an excellent classification accuracy of 93.834% and performed better than several novel methods. These experimental results indicate that our method enables better reliability and feasibility in the detection of fatigued driving.

Introduction: Personality disorder (PD) and childhood traumatic experience (CTE) are well- recognized risk factors for the development of schizophrenia (SZ) and major depressive disorder (MDD). The relationship between CTE and PD is extremely close, and both conditions can affect subsequent psychiatric disorders. Little is known about the differences of these factors in patients with SZ and those with MDD. Materials and Methods: A total of 1,026 outpatients participated in the study, including 533 (51.9%) with SZ and 493 (48.1%) with MDD who were sequentially sampled. The PD traits were assessed using the Personality Diagnostic Questionnaire Fourth Edition Plus (PDQ-4+). The Child Trauma Questionnaire Short Form (CTQ-SF) was used to assess childhood adversities. The scores and associations of PDQ-4+ and CTQ-SF between patients with SZ and those with MDD were compared. Results: The MDD group exhibited more PD traits and more childhood emotional neglect than the SZ group. In patients with MDD, the correlation between PD traits and CTE was significantly higher than that in patients with SZ. Patients with SZ vs. those with MDD showed different PD traits and CTE. The schizotypal and antisocial PD traits, as well as sexual abuse and physical neglect CTE, were significantly related to SZ. In contrast, the borderline, narcissistic and avoidant PD traits, and emotional abuse/neglect CTE were significantly associated with MDD. Discussion: These findings indicated a robust relationship between CTE and PD traits. Moreover, patients with SZ or MDD, have different interactive patterns. Both CTE and PD traits have the potential to be premorbid risk factors that could be targeted for preventative interventions.

OBJECTIVE: Diabetic peripheral neuropathy (DPN) is one of the most important risk factors of diabetic foot ulcers, and early screening and treatment of DPN are crucial. The Ipswich Touch Test (IPTT) is a new method for screening for DPN and, compared with traditional methods, is more simple to operate and requires no equipment. However, the screening accuracy of IPTT in patients with DPN has not been well characterised. We aim to conduct a systematic review and meta-analysis to characterise the sensitivity and specificity of IPTT compared with traditional methods and to understand the potential screening value of IPTT. DESIGN: Systematic review and meta-analysis. DATA SOURCES: PubMed, Embase, Cochrane Library, Web of Science, China National Knowledge Infrastructure, Wanfang, Chinese Biomedical Literature Database up to 16 April 2020. METHODS: Stata V.15.1 software was used for analysis, and the screening value of IPTT in DPN was described using 10 g monofilament (10g-MF), neuropathy disability scores (NDS), Pin prick, 128 Hz tuning fork, and ankle reflex as reference standards. Sensitivity, specificity and other measures of accuracy of IPTT for screening DPN were pooled based on a quality effects model. The protocol was registered with PROSPERO (42020168420). RESULTS: Of the 441 records retrieved, 7 studies were evaluated for the screening value of IPTT. Five studies with 10g-MF as the reference standard were included in the meta-analysis, and the pooled sensitivity and specificity were 0.77 (95%CI 0.69-0.84) and 0.96(95%CI 0.93-0.98), respectively, and the area under curve was 0.897. Compared with vibration perception threshold, IPTT showed a sensitivity between 0.76 and 1, and a specificity between 0.90 and 0.97. Compared with NDS, IPTT showed a sensitivity between 0.53 and 1, and a specificity between 0.90 and 0.97. Compared with Pin prick, IPTT showed a sensitivity and specificity of 0.8 and 0.88, respectively. Compared with 128 Hz tuning fork, IPTT showed a sensitivity and specificity of 0.4 and 0.27, respectively. Compared with ankle reflex, IPTT had a sensitivity of 0.2 and a specificity of 0.97. CONCLUSIONS: IPTT shows a high degree of agreement with other commonly used screening tools for DPN screening. It can be used clinically, especially in remote areas and in primary medical institutions, and by self-monitoring patients. More high-quality studies are needed to assess and promote more effective screening practices. PROSPERO REGISTRATION NUMBER: Registration Number is CRD (42020168420).

BACKGROUND AND AIMS: The lung is the first organ to fail in sepsis. Our previous studies have proven that 2% molecular hydrogen (H2) inhalation remain a protective effect on a septic animal model via its anti-inflammatory and anti-apoptosis properties. This current research aims to observe the therapeutic effect of high concentration hydrogen (67%, HCH) on lipopolysaccharide (LPS) induced acute lung injury (ALI), and further investgate the role of Nrf2 signaling pathway. METHODS: ALI model was induced by LPS areosol inhalation. HCH were treated for 1 h at 1 and 6 h after modelling. Lung tissues and bronchoalveolar lavage fluid (BALF) were collected 4 and 24 h after the exposure of LPS. The histological scores, wet/dry weight ratios, myeloperoxidase (MPO) activity, protein content and cytokine levels in BALF, apoptosis condition of lung cells, expression of Nrf2 and NF-κB were assessed in both wild type and Nrf2-knockout mice. RESULTS: HCH Inhalation significantly alleviated LPS-induced pathological alterations of lung, and reduced the protein concentration, the wet/dry weight ratio, and the MPO activity of lung tissue. HCH Inhalation improved LPS-induced increasement in caspase-3 activity and the number of TUNEL-positive cells. HCH inhalation attenuated the LPS induced increased total cell content and polymorphonuclear granulocyte content, and pro-inflammatory cytokines, Nrf2 and NF-κB expression. HCH could not produce protective effct in Nrf2-knockout mice. CONCLUSION: HCH can effectively alleviate LPS-induced ALI, which may be related to activation of Nrf2 signaling pathway and inhibition of inflammatory response and cell apoptosis mediated by NF-κB.

Doctors' prescribing behaviors impact both medical expenses and health resources. This study aims to identify the significant determinants of prescribing behaviors of doctors, which could potentially provide theoretical evidence on how to improve prescribing decisions. A multistage, stratified, cluster, random sampling method was employed in this survey. Data were collected from Jiangsu and Shanxi provinces in China in 2018. A total of 444 doctors in public hospitals completed the self-administered questionnaires. A structural equation model based on the theory of planned behavior (TPB) was adopted for analysis. On the basis of the TPB, we constructed a model of doctors' prescribing behaviors, which explained the subjective and objective reasons for irrational prescribing behavior. Behavioral attitude, subjective norms, and perceived behavioral control could positively influence the actual behaviors, of which subjective norms impact prescribing behaviors the most. Employing the TPB helped in identifying determinants of prescribing behaviors from a new perspective. More significant policy changes and government support are required to help improve appropriate prescribing behaviors and ultimately make better prescribing decisions. This study provided a deeper understanding of this complex issue and will inform the development of a theory and evidence-based intervention for future research.

BACKGROUND: The decreased level of miR-192-5p has been reported in several kinds of cancers, including bladder, colon, ovarian, and non-small cell lung cancer. However, the expression and function of miR-192-5p in papillary thyroid carcinoma/cancer (PTC) remains unknown. OBJECTIVE: The present study aimed to explore the function and underlying mechanism of miR-192-5p in PTC development. METHODS: PTC tissues and relative normal controls from PTC patients were collected. qRT-PCR analysis was performed to measure miR-192-5p and SH3RF3 mRNA level in PTC tissues and cell lines. CCK-8 method and FCM assay were used to test cell proliferation and apoptosis in TPC-1 cells, respectively. The abilities of cell migration and invasion were detected by wound healing and transwell assays, respectively. The protein expression was evaluated by Western blot. The interaction between miR-192-5p and Src homology 3 (SH3) domain containing ring finger 3 (SH3RF3) were confirmed by dual-luciferase reporter assay. RESULTS: MiR-192-5p level was obviously decreased in PTC tissues and cell lines. Overexpression of miR-192-5p suppressed proliferation, migration, invasion, and EMT process, while induced apoptosis in TPC-1 cells. In addition, miR-192-5p negatively modulated SH3RF3 expression by binding to its 3'-untranslated region (3'UTR). Silencing SH3RF3 inhibited the migration, invasion, and EMT of TPC-1 cells. In the meantime, matrine, an alkaloid extracted from herb, exerted its anti-cancer effects in PTC cells dependent on increase in miR-192-5p expression and decrease in SH3RF3 expression. CONCLUSION: We firstly declared that miR-192-5p played a tumor suppressive role in PTC via targeting SH3RF3. Moreover, matrine exerted its anti-cancer effects in PTC via regulating miR-192-5p/SH3RF3 pathway.

Photoactuated liquid crystalline elastomer (LCE) materials are gaining much attention in many application fields, but challenges for the precise modulation of their photoresponses still exist. Researchers have explored various optical parameters, such as polarization, intensity, and wavelength, to obtain differential responses. The development of photoactuated LCE materials with wavelength-selective responsiveness is more versatile and has attracted more interest, but such LCE materials are commonly prepared by incorporating different molecular chromophores or dyes into the LCE matrices. When the surface plasmon resonance (SPR) characteristic of nanometals, which can generate strong photothermal conversion, and the difference of SPR absorption wavelength bands of different nanometals are considered, a strategy of constructing wavelength-selective actuation of LCE materials by using the SPR photothermal effect can be demonstrated, as done herein. The LCE nanocomposites doped by nanogold or nanosilver were fabricated and exhibited good SPR absorption but in different wavelength bands of the visible spectrum range. They had strong actuation under light irradiation with the wavelengths being inside their respective absorption band but could not be effectively actuated by the light beyond their respective absorption band. A smart electronic device, implementing a hierarchical structured LCE nanocomposite doped by nanogold and nanosilver in different domains as the two-switch actuator, was prepared and capable of outputting different signals in response to the different wavelength bands filtered from a light source, which released the actuator from the restriction of light scanning direction or position. Our work provides new insights for the convenient and precise photoactuation of the LCE actuators.
