RESUMO

The measuring accuracy of the fiber optic gyroscope (FOG) for weak signals under very short sampling time is significantly impacted by the quantization error, impeding its application in high-speed measurement and real-time control. In this work, we propose and implement a double-electrode-pair multifunction integrated-optic circuit (MIOC), which contains an additional pair of short electrodes besides the conventional electrode-pair. Taking advantage of the better modulating precision of the additional electrode-pair, the digital feedback is more refined and the quantization error in the FOG output is significantly suppressed. The driving circuits and the control scheme of the proposed MIOC are specially designed for FOGs. Experimental results show that the resolution for extremely small angular rates at short smoothing times is significantly improved. This work provides the potential of the applications in high-speed measuring and controlling systems for high-precision FOGs.

RESUMO

Metal hydroxides nanosheets of atomic thickness have attracted much interest due to their great potentials in catalysis, energy storage devices and so on. But the lack of the efficient synthesis of 2D nanosheets has critically impeded their practical applications. Herein, we develop a general water-induced self-exfoliation (WISE) strategy to achieve the fast synthesis of metal hydroxide ultrathin nanosheets with alomost single-layer atoms thickness in a large scale. In a typical process of layered cobalt hydroxide (LCH) nanosheets, the pre-synthesized cobalt acetate hydroxide precursor is directly exfoliated to form nanosheets under the attack of H2O in a few seconds. The water-induced self-exfoliation mechanism has also been proposed based on the analysis of the designed alcohol-mediated slow-down process. In addition, the used solutions and effluents can be recycled making WISE a green, efficient and surfactants-free method. Furthermore, this general strategy can also be applied to synthesize other layerd metal hydroxides nanosheets.

BACKGROUND: Despite the worldwide prevalence of dermatophyte infections, only a few genes are reported to be related to dermatophyte infections. In addition, the mechanism by which different ecological dermatophytes infection leads to varying intensity of inflammation remains unclear. OBJECTIVES: To investigate the mechanism of varying intensity of skin inflammation caused by different ecological dermatophytes infection. METHODS: We infected HaCaT cells with anthropophilic and geophilic dermatophytes to mimic various ecological dermatophyte infections. RNA-sequencing (RNA-seq) was employed to identify the change in the gene expression of HaCaT cells. To verify the expression of differentially expressed genes (DEGs), we selected 18 HaCaT cells genes to conduct qPCR experiments. In addition, immunoblotting was conducted to validate key genes from the MAPK signaling pathway. RESULTS: After HaCaT cells were infected with the anthropophilic Trichophyton rubrum (T. rubrum) and the geophilic Microsporum gypseum (M. gypseum), 118 and 619 differentially expressed genes were identified in HaCaT cells respectively. These genes may provide a clue as to how keratinocytes respond to anthropophilic and geophilic dermatophytes. We also found that JUN may play a critical role in keratinocytes infected with M. gypseum. CONCLUSIONS: Differential gene expression in HaCaT cells may account for the various clinical presentation caused by anthropophilic and geophilic dermatophytes infections. In addition, the intense inflammatory reaction of M. gypseum infection may be triggered by activating the JNK-JUN signaling pathway.

Decarboxylative Csp3-N coupling reactions have been developed through electrochemical oxidation of amino acids. The reaction proceeds via anodic oxidative decarboxylation of carboxylic acids to form stabilized carbocations, which are trapped by azoles or amides to construct C-N bonds. This method avoids the preactivation of carboxylic acids and the use of expensive transition-metals and external chemical oxidants.

An anoxic-aerobic membrane bioreactor (AO-MBR), an anaerobic side-stream reactor (ASSR) coupled MBR (A-MBR), and an MBR with ASSR packed with carriers (AP-MBR) were operated parallelly to investigate biological nutrient removal, microbial community structure and mass balance of nutrients in sludge reduction systems. Compared to AO-MBR, A-MBR and AP-MBR were both efficient in COD and NH4+-N removal, had significantly higher nitrogen removal, reduced sludge production by 35.0% and 45.9%, but deteriorated biological phosphorus removal. Nitrosomonadaceae and Nitrospira were major bacteria responsible for ammonium and nitrite oxidation in the three systems. Inserting ASSR and packing carriers both favored denitrifying bacteria enrichment and organic substances release, and thus resulted in higher nitrate uptake rate (NUR) in the anoxic tank. Higher endogenous NUR in ASSR than in anoxic tank also indicated that ASSR and carriers both accelerated sludge decay. Denitrification and sludge reduction occurred in ASSR played important roles in biological nutrient removal.

A new coordination polymer (H2 bpy)0.5 ⋅[(UO2 )1.5 (ipa)2 (H2 O)] (1) (H2 ipa=isophthalic acid, bpy=4,4'-bipyridine) was synthesized by hydrothermal condition. It was characterized by IR spectroscopy, elemental analysis, TG-DTA analysis, and powder X-ray diffraction. Analysis of single-crystal X-ray diffraction results showed that the title compound exhibited a double chain bridged by the different uranyl ions and ipa2- ligands. Through the hydrogen bond interactions and π⋅⋅⋅π stacking interactions, the double chains were assembled into the three-dimensional supramolecular framework. Furthermore, the compound can be used as a promising bifunctional luminescence sensor for detecting and identifying Fe3+ and tetracycline hydrochloride antibiotic molecules with high selectivity and sensitivity in aqueous solutions. Moreover, the luminescent sensing mechanisms for different analytes were proposed. Moreover, the electronic properties of title compound were explored by density functional theory (DFT) calculations. The sensor system has been successfully applied for the detection of Fe3+ and tetracycline hydrochloride with high recovery percentages and low relative standard deviation in real river water samples.

Recent experiments have demonstrated the existence of vortex or flux-closure domains in ferroelectric nanostructures, which are attractive to develop high-density data storage and novel configurable electronic devices. However, it remains challenging to stabilize in-plane vortex or flux-closure domains in ferroelectric film for the absence of a lateral geometry confinement. Based on a 3D phase field model, here we show that stabilization of isolated or interacting in-plane vortices in ferroelectric film can be achieved via applying a mechanical tip-force. The formation of such dipole vortices is caused by a conjoint effect of the tip-force-induced depolarization effect and in-plane strain. The effects of factors like film thickness, misfit strain, tip force and temperature on the vortex formation are systematically revealed and summarized as phase diagrams. The interaction between tip-induced vortices is also investigated. It is found that as the two tips get closer than the critical distance, the two initially isolated vortices become coupled, with identical or opposite chirality, depending on the distance between the two tips. A maximum data storage density of isolated in-plane vortices in ferroelectric thin film is estimated to be ~1 Tb in-2. Our work thus demonstrates a mechanical strategy to stabilize dipole vortices, and provides a comprehensive insight into the characteristics of ferroelectric film under a mechanical tip force.

Dissolved organic matter (DOM) is an important parameter that reflects soil fertility and quality. In this study, the effects of intercropping perennial ryegrass and straw mulch on the content, composition and spectral characteristics of soil DOM in orchards in arid and semi-arid areas were investigated by three-dimensional fluorescence excitation-emission matrix (EEM) combined with parallel factor analysis (PARAFAC). Three treatments were applied in a 6-year-old apple orchard on the Loess Plateau: (1) clean tillage (CT); (2) intercropping perennial ryegrass mulch (RE); (3) intercropping straw mulch (CS). Soil samples (0-10, 10-20, 20-40, 40-60, 60-80, 80-100 cm) were collected from different treatments. The total soil organic carbon (SOC) and dissolved organic carbon (DOC) content were ranked as CS> RE> CT, and decreased with soil depth. In addition, the vertical distribution of DOC with different soil depth was caused by organic matter input but not leaching. Three humic-like components and two protein-like components were identified by EEM-PARAFAC. Fluorescence spectra showed that humic-like compounds were the dominant fractions in soil DOM in the CS treatment. Fluorescence Index (FI) indicated that the proportion of plant residues and soil organic matter derived DOM ranked as CT> RE> CS and gradually decreased with soil depth. Our results highlight the key role of orchard intercropping mulch in improving the content and compositions of soil DOM through different forms of organic inputs and provide new theoretical support for the evaluation of soil fertility in orchards.

BACKGROUND: For the evaluation results of skin sensitivity, such as clinical parameters, stinging test records and biophysical assessments dates might be impacted by many factors, the influence factors need to be further explored, and the skin sensitivity evaluation process and methodology needed distinction and normalization. In this study, we investigated the changes of sensitive skin indexes and lactic acid stinging test results in different seasons, facial regions, skin photo-type, and living habits. METHODS: Twenty-four healthy subjects had completed this study. Lactic acid stinging test was performed in different seasons. Transepidermal water loss (TEWL), skin hydration, sebum secretion, and pH were measured in an environment-controlled room. Correlations between stinging responses, skin biophysical parameters, and sensitive skin inducements in different seasons were statistically analyzed. RESULTS: Skin TEWL, hydration, sebum secretion, and pH values on different facial parts were various. Two-way correlation analysis between the results of lactic acid stinging test in different seasons and the sensitivity factors showed differences between summer, autumn, and winter. The mean scores of lactic acid stinging test increased in autumn. Linear regression analysis of skin sensitivity factors in type III and type IV photobiology skin found that the frequency of sleeping time and eating spicy food in the past of week could infect the sensitive skin evaluation dates statistically (P < .05). DISCUSSION/CONCLUSIONS: Skin sensitivity assessment results were impacted by seasonal transformation, living habits and customs, and facial regions. These indicted that we should consider above interfering factors when evaluated the skin sensitivity for getting more precise dates.

In the present study, four copper-based metal-organic frameworks were facilely prepared to study the effects of the substituent groups of the ligand on the enzyme-like properties of these frameworks. It was found that all of them were capable of catalyzing the reaction between 3,3',5,5'-tetramethylbenzidine (TMB) and H2O2; this demonstrated their enzyme-like properties. Moreover, the enzyme-like catalytic properties of all the Cu-MOFs obtained herein were studied in detail. Interestingly, the results indicate that the four Cu-MOFs should be addressed as different enzyme mimics although the chemical structures of their ligands are quite similar. Among these four MOFs, the Cu-MOF with the -NO2 group (Cu-MOF (ii)) exhibits highest catalytic activity at neutral pH, which would be beneficial for its application in real biological samples. Moreover, its catalytic activity should be ascribed to the generation of oxygen induced by Cu-MOF (ii)-catalyzed H2O2 decomposition; this accordingly indicates that this MOF should be called a catalase mimic. Under optimized conditions, the Cu-MOF (ii) was applied to develop a (GSH) colorimetric assay for glutathione (GSH). In addition, GSH detection in serum was performed, and satisfactory results were obtained. Therefore, a simple, sensitive and selective colorimetric assay based on the MOF catalase mimic for the detection of GSH was developed.

Long non-coding RNAs (lncRNAs) have emerged as key regulators of cellular progress in lung adenocarcinoma. In this study, to identify cancer-related lncRNAs and genes, we screened for those lncRNAs that were differentially expressed in lung adenocarcinoma, which revealed LINC00628 overexpression and low expression of laminin subunit alpha 3 (LAMA3). This was further validated in the cancerous tissues from patients diagnosed with lung adenocarcinoma. Thereafter, we explored the functional relevance of LINC00628 and LAMA3 in lung adenocarcinoma by analyzing the recruitment of DNA methyltransferase (DNMT) and the cellular processes of lung adenocarcinoma cells following treatments that induced LINC00628 overexpression or LINC00628 silencing or with 5-azacytidine (5-Aza, a DNMT inhibitor). The results showed that LINC00628 silencing decreased cell proliferation, migration, and invasion as well as the drug resistance of lung adenocarcinoma cells to vincristine (VCR). The results were opposite in the cells with LAMA3 demethylation induced by 5-Aza treatment. Further research indicated that LINC00628 recruited DNMT1, DNMT3A, and DNMT3B to promote the methylation of LAMA3 promoter, thereby decreasing its expression. Moreover, an in vivo experiment was performed in nude mice to assess the tumor growth ability and drug resistance of human lung adenocarcinoma cells. It was observed that LINC00628 silencing or 5-Aza treatment inhibited the in vivo tumor growth ability of the human lung adenocarcinoma cells and reduced their resistance to VCR. Altogether, our results provide evidence of a mechanism by which LINC00628 silencing exerts an inhibitory role in lung adenocarcinoma by modulating the DNA methylation of LAMA3, indicative of a novel molecular target for treatment of lung adenocarcinoma patients showing resistance to VCR.

The effect of nanoscale zero-valent iron (NZVI) and magnetite nanoparticles (Fe3O4 NPs) on anaerobic digestion (AD) performance was investigated through a series of 100-day semi-continuous mesophilic anaerobic digestions. The results indicated that biogas production had increased by 24.44% and 21.66% with the addition of 0.5 g/L Fe3O4 NPs and 1.0 g/L NZVI, respectively. Besides, the abundance of five widespread antibiotic resistance genes (ARGs) (ermF, ermA, ermT, aac(6')-IB, blaOXA-1) was also studied. The decrease in abundance of aac(6')-IB and blaOXA-1 was observed during the AD process with an average removal rate of 95.69% and 44.82%, respectively. Most of the ARGs, especially ermA and ermT, were less abundant in NZVI group compared with control group. The overall results suggested that the addition of NZVI and Fe3O4 NPs contributed to a better sludge anaerobic digestion performance, and NZVI was beneficial to the removal of some ARGs.

The release and nuclear translocation of the intracellular domain of NOTCH receptor (NICD) is the prerequisite for Notch signaling mediated transcriptional activation. NICD is subjected to various post-translational modifications including ubiquitination. Here we surprisingly found NUMB proteins stabilize the intracellular domain of NOTCH1 receptor (N1ICD) by regulating the ubiquitin-proteasome machinery, which is independent of NUMB's role in modulating endocytosis. BAP1, a deubiquitinating enzyme (DUB), was further identified as a positive N1ICD regulator, and NUMB facilitates the association between N1ICD and BAP1 to stabilize N1ICD. Intriguingly, BAP1 stabilizes N1ICD independent of its DUB activity but relying on the BRCA1-inhibiting function. BAP1 strengthens Notch signaling and maintains stem-like properties of cortical neural progenitor cells. Thus, NUMB enhances Notch signaling by regulating the ubiquitinating activity of the BAP1-BRCA1 complex.

Previous studies have reported that liver X receptor (LXR), ATP­binding cassette sub­family G number 1 (ABCG1) and ATP­binding cassette transporter number 1 (ABCA1), which are associated with cholesterol metabolism, may be associated with the development and progression of breast cancer. The expression levels of LXR­ß, ABCA1 and ABCG1 in triple­negative breast cancer (TNBC) tissues and in non­cancerous mammary tissues were observed by immunohistochemistry, quantum dot­based immunohistochemistry, western blot analysis and reverse transcription­quantitative polymerase chain reaction. The present study identified that the expression of ABCA1 in TNBC tissues was higher than that in non­cancerous mammary tissues. A high expression of ABCA1 in the TNBC tissues was significantly associated with the histological grade. However, no significant differences were identified between the expression levels of LXR­ß and ABCG1 in the TNBC tissues compared with the non­cancerous mammary tissues. Therefore, the findings of this study suggest that ABCA1 is a specific marker for TNBC.

Thaumarchaeota are responsible for a significant fraction of ammonia oxidation in the oceans and in soils that range from alkaline to acidic. However, the adaptive mechanisms underpinning their habitat expansion remain poorly understood. Here we show that expansion into acidic soils and the high pressures of the hadopelagic zone of the oceans is tightly linked to the acquisition of a variant of the energy-yielding ATPases via horizontal transfer. Whereas the ATPase genealogy of neutrophilic Thaumarchaeota is congruent with their organismal genealogy inferred from concatenated conserved proteins, a common clade of V-type ATPases unites phylogenetically distinct clades of acidophilic/acid-tolerant and piezophilic/piezotolerant species. A presumptive function of pumping cytoplasmic protons at low pH is consistent with the experimentally observed increased expression of the V-ATPase in an acid-tolerant thaumarchaeote at low pH. Consistently, heterologous expression of the thaumarchaeotal V-ATPase significantly increased the growth rate of E. coli at low pH. Its adaptive significance to growth in ocean trenches may relate to pressure-related changes in membrane structure in which this complex molecular machine must function. Together, our findings reveal that the habitat expansion of Thaumarchaeota is tightly correlated with extensive horizontal transfer of atp operons.

An anoxic/oxic membrane bioreactor (AO-MBR), an anaerobic side-stream reactor (ASSR) coupled MBRs (A-MBRs), an A-MBR with carriers packed in ASSR (AP-MBR) and an AP-MBR with sludge ultrasonicated before ASSR (AUP-MBR) were operated for 261 d to investigate effects of ASSR, packing carriers and ultrasonication on sludge reduction and microbial population. Sludge reduction efficiencies of A-MBR, AP-MBR and AUP-MBR were 36.2%, 46.4% and 51.4%, respectively. Packing carriers and ultrasonication both enhanced hydrolysis by stimulating activities of α-glucosidase and protease, while uncoupling metabolism was enhanced greatly by packing carriers but slightly by ultrasonication. Linear discriminant analysis of effect size (LEfSe) results showed that packing carriers promoted the growth of hydrolytic and fermentative bacteria in bulk sludge, and enriched anaerobes and fermentative bacteria on the surface of carriers. Ultrasonication screened ultrasonication-resistant bacteria, and created an anaerobic environment beneficial to hydrolytic and fermentative bacteria.

Sepsis, defined as life-threatening tissue damage and organ dysfunction caused by a dysregulated host response to infection, is a critical disease which imposes global health burden. Sepsis-induced organ dysfunction, including circulatory and cardiac dysfunction, hepatic dysfunction, renal dysfunction, etc., contributes to high mortality and long-term disability of sepsis patients. Altered inflammatory response, ROS and reactive aldehyde stress, mitochondrial dysfunction, and programmed cell death pathways (necrosis, apoptosis, and autophagy) have been demonstrated to play crucial roles in septic organ dysfunction. Unfortunately, except for infection control and supportive therapies, no specific therapy exists for sepsis. New specific therapeutic targets are highly warranted. Emerging studies suggested a role of potential therapeutic target of ALDH2, a tetrameric enzyme located in mitochondria to detoxify aldehydes, in septic organ dysfunction. In this article, we will review the presentations and pathophysiology of septic organ dysfunction, as well as summarize and discuss the recent insights regarding ALDH2 in sepsis.

Autophagy has been associated with a variety of diseases especially aging. Human dermal fibroblasts (HDFs) can internalize and then degrade elastin, collagen and advanced glycation end products (AGEs) in lysosomes, which plays prominent roles in extracellular matrix homeostasis and AGEs removal in the dermis. Although autophagy has been reported to be decreased in photoaged fibroblasts, the underlying mechanism and its relevance to photoaging remain elusive. Here, we showed that GFP-LC3 puncta per cell, LC3Ⅰ/Ⅱ conversion and p62 expression were significantly increased, whereas beclin1 expression was not altered in UVA-induced photoaged fibroblasts compared with non-photoaged control. Moreover, autophagic flux was not significantly affected by chloroquine treatment, but was remarkably induced by rapamycin treatment in photoaged fibroblasts, suggesting that UVA-induced photoaging might inhibit autophagy at the degradation stage. Further lysosomal function studies demonstrated that degradation of formed autophagosomes, LC3Ⅱprotein and DQ-Green BSA was all dramatically decreased in photoaged fibroblasts. LysoSensor yellow/blue DND 160 staining and flow cytometry assays demonstrated that photoaging obviously attenuated lysosomal acidification. Also, decreased expression of cathepsin B, L and D was found in photoaged fibroblasts. These data suggest that lowered lysosomal acidity and decreased cathepsins expression might contribute to the inhibition of autophagic degradation, which might be crucial in the development of photoaging through impairing intracellular degradation.

Needle-induced cavitation (NIC) locally probes the elastic and fracture properties of soft materials, such as gels and biological tissues. Current NIC protocols tend to overestimate properties when compared to traditional techniques. New NIC methods are needed in order to address this issue. NIC measurements consist of two distinct processes, namely (1) the needle insertion process and (2) the cavitation process. The cavitation process is hypothesized to be highly dependent on the initial needle insertion process due to the influence of residual strain below the needle. Retracting the needle before pressurization to a state in which a cylindrical, tube-like fracture is left below the needle tip is experimentally demonstrated to reduce the impact of residual strain on NIC. Verification of the critical cavitation pressure equation in this new geometry is necessary before implementing this retraction NIC protocol. Complementary modeling shows that the change in initial geometry has little effect on the critical cavitation pressure. Together, these measurements demonstrate that needle retraction is a viable experimental protocol for reducing the influence of residual strain, thus enabling the confident measurement of local elastic and fracture properties in soft gels and tissues.
