RESUMO

The approximate process range for preparing the Cu-Cr-Zr alloy by selective laser melting (SLM) was determined by ANSYS simulation, and the influence of the SLM process parameters on the comprehensive properties of the SLM-formed alloy was studied by the design of experiments. The Cu-Cr-Zr alloy with optimum strength and hardness was prepared with high efficiency by optimizing the process parameters for SLM (i.e., laser power, scanning speed, and hatching distance). It is experimentally shown that tensile strength and hardness of the SLM alloy are increased by increasing laser power and decreasing scanning speed, whereas they are initially increased and then decreased by increasing the hatching distance. Moreover, strength, roughness and hardness of the SLM alloy are optimized when laser power is 460 W, scanning speed is 700 mm/s and hatching distance is 0.06 mm. The optimized properties of the SLM alloy are a tensile strength of 153.5 MPa, hardness of 119 HV, roughness of 31.384 µm and relative density of 91.62%.

Underactive bladder or detrusor underactivity (DUA), that is, not being able to micturate, has received less attention with little research and remains unknown or limited on pathological causes and treatments as opposed to overactive bladder, although the syndrome may pose a risk of urinary infections or life-threatening kidney damage. Here, we present an integrated expandable electronic and optoelectronic complex that behaves as a single body with the elastic, time-dynamic urinary bladder with substantial volume changes up to ~300%. The system configuration of the electronics validated by the theoretical model allows conformal, seamless integration onto the urinary bladder without a glue or suture, enabling precise monitoring with various electrical components for real-time status and efficient optogenetic manipulation for urination at the desired time. In vivo experiments using diabetic DUA models demonstrate the possibility for practical uses of high-fidelity electronics in clinical trials associated with the bladder and other elastic organs.

Wearable and implantable bio-integrated electronics have started to gain momentum because of their essential role in improving the quality of life for various patients and healthy individuals. However, their continuous operation is often limited by traditional battery technologies with a limited lifespan, creating a significant challenge for their development. Thus, it is highly desirable to harvest biomechanical energies from human motion for self-powered bio-integrated functional devices. Piezoelectric energy harvesters are ideal candidates to achieve this goal by converting biomechanical energy to electric energy. Because of their applications on soft and highly deformable tissues of the human body, these devices also need to be mechanically flexible and stretchable, thus posing a significant challenge. Effective methods to address the challenge include the exploration of new stretchable piezoelectric materials (e.g., hybrid composite material) and stretchable structures (e.g., buckled shapes, serpentine mesh layouts, kirigami designs, among others). This review presents an overview of the recent developments in new intrinsically stretchable piezoelectric materials and rigid inorganic piezoelectric materials with novel stretchable structures for flexible and stretchable piezoelectric sensors and energy harvesters. Following the discussion of theoretical modeling of the piezoelectric materials to convert mechanical deformations into electrical signals, the representative applications of stretchable piezoelectric materials and structures in wearable and implantable devices are briefly summarized. The present limitations and future research directions of flexible and stretchable piezoelectric devices are then discussed.

Traditional Chinese Medicine has been widely used in China and is regarded as the most commonly used treatment. As a natural plant used in traditional Chinese Medicine, Basil has various functions associated with a number of its components. There are many compositions in basil including polysaccharides, naphtha, steroids, flavone, coumarins, vitamins, and so on. Among these, polysaccharides play a significant role in based therapeutics. The article summarizes that basil polysaccharides have a lot of biological activities and pharmacological applications, such as their antitumor activity, antioxidant activity, anti-aging activity, immunity enhancement effect, hypolipidemic and anti-atherosclerotic effects, antibacterial effect, treatment of diabetes mellitus, and so on. This review summarized the extraction method, purification method, compositions, pharmacological applications, molecular weight, biological activities, and prospects of basil polysaccharides, providing a basis for further study of basil and basil polysaccharides.

Porous CuO/SnO2 nanofibers were prepared by co-dissolution and electrospinning, followed by annealing, and compounded with different amounts of carbon nanotubes (CNTs, 0-5 wt%). The performance of the composite based sensor was tested in different concentrations of H2S range from 0.1 to 0.5 ppm. The results showed that the gas sensor based CuO/SnO2 doped by CNTs (CNTs-CuO/SnO2) has a low optimum operating temperature, 40 °C, good behavior in detecting low concentration H2S, short response and recovery time (8.3 and 11.5 s, respectively). And at the same time, compared with other gases, 3 wt% CNTs-CuO/SnO2 composite nanomaterial had excellent selectivity to H2S at low concentrations. The prepared gas sensor had great advantages in detecting low concentration H2S.

The concrete relationship between the process parameters and nanoproduct properties is an important challenge for applying nanotechnology to produce functional nanomaterials. In this study, the relationships between series of process parameters and the medicated nanofibers' diameter were investigated. With an electrospinnable solution of hydroxypropyl methylcellulose (HPMC) and ketoprofen as the core fluid, four kinds of nanofibers were prepared with ethanol as a sheath fluid and under the variable applied voltages. Based on these nanofibers, a series of relationships between the process parameters and the nanofibers' diameters (D) were disclosed, such as with the height of the Taylor cone (H, D = 125 + 363H), with the angle of the Taylor cone (ɑ, D = 1576 - 19ɑ), with the length of the straight fluid jet (L, D = 285 + 209L), and with the spreading angle of the instable region (θ, D = 2342 - 43θ). In vitro dissolution tests verified that the smaller the diameters, the faster ketoprofen (KET) was released from the HPMC nanofibers. These concrete process-property relationships should provide a way to achieve new knowledge about the electrostatic energy-fluid interactions, and to meanwhile improve researchers' capability to optimize the coaxial process conditions to achieve the desired nanoproducts.

Flexible electronic systems have received increasing attention in the past few decades because of their wide-ranging applications that include the flexible display, eyelike digital camera, skin electronics, and intelligent surgical gloves, among many other health monitoring devices. As one of the most widely used technologies to integrate rigid functional devices with elastomeric substrates for the manufacturing of flexible electronic devices, transfer printing technology has been extensively studied. Though primarily relying on reversible interfacial adhesion, a variety of advanced transfer printing methods have been proposed and demonstrated. In this review, we first summarize the characteristics of a few representative methods of transfer printing. Next, we will introduce successful demonstrations of each method in flexible electronic devices. Moreover, the potential challenges and future development opportunities for transfer printing will then be briefly discussed.

OBJECTIVE: To investigate the effects of Pinggan Prescription (, PGP) on hypertension by the associated methods of metabonomic and pharmacodynamic. METHODS: A total of 32 male spontaneously hypertensive rats (SHRs) were randomly divided into two groups by using the random number table method: a treatment group (n=18) and a model group (n=14). The Wistar rats (n=14) were used as the normal group. Different prescription were used to intervene three groups: the treatment group in which PGP extract was administered orally at a dose of 18.336 g/kg (PGP/body weight), and the model group in which physiological saline was administered at the equivalent dose. The same treatment was applied to the normal group as the model group. The blood pressure was measured by tail-cuff method, and pharmacodynamic indexes including cyclic adenosine monophosphate (cAMP) and angiotensin II (Ang II) were tested by enzyme-linked immunosorbent assay. The plasma samples from three groups were detected by gas chromatography-mass spectrometry (GC-MS). RESULTS: Compared with the model group, blood pressure of treatment group was obviously reduced after continuous curing with PGP (P<0.01). The pharmacodynamic results illustrated that the content of Ang II increased with the raised blood pressure and the cAMP expressed the converse trend. After curing with PGP, the content of Ang II decreased, the difference between model group and treatment group was significant (P<0.01), and the cAMP expressed the converse trend. Five potential biomarkers were identified, including arachidonic acid, hexadecanoic acid, elaidic acid, octadecanedioic acid and 9,12-octadecadienoic acid. These metabolites had shown significantly changes as followed: arachidonic acid, hexadecanoic acid and elaidic acid were significantly higher and octadecanedioic acid and 9,12-octadecadienoic acid were lowered in the model group than those in the normal group. After the treatment of PGP, the metabolites had the trends of returning to normal along with the reduced blood pressure. CONCLUSIONS: PGP intervention for hypertension played a major role in the metabolism of arachidonic acid and linoleic acid. Metabonomic with pharmacodynamic methods could be potentially powerful tools to investigate the mechanism of Chinese medicine.

RESUMO

Four new gastrodin derivatives containing a trans-cinnamoyl unit (1-4) and nine known compounds (5-13) were isolated from the rhizomes of Gastrodia elata Blume. All these compounds were evaluated for their neuroprotective effects against 6-hydroxydopamine-induced cell death, and compounds 7 and 12 showed potent activities with EC50 values of 10.5 and 10.2 µM, respectively.

One new triterpenoid saponin (1), as well as six known ones (2-7), were isolated from the ethanol extract of the thorns of Gleditsia sinensis. Their structures were elucidated by extensive spectroscopic analysis in conjunction with chemical evidence. Cytotoxic activity of compounds 1-6 was evaluated against human breast cancer MCF 7 cells in vitro by the MTT method. Our results revealed moderate activities for compounds 1-6 with IC50 values of 18.43, 30.47, 18.46, 10.02, 30.76, and 17.32 µM, respectively. Furthermore, compounds 1, 3, 4, and 6 induced apoptosis in MCF 7 cell, with 1 and 6 causing late apoptosis of MCF 7 cells, while 3 and 4 acting oppositely.

Our previous studies have shown that Uncaria has an important role in lowering blood pressure, but its intervention mechanism has not been clarified completely in the metabolic level. Therefore, in this study, a combination method of HPLC-TOF/MS-based metabolomics and multivariate statistical analyses was employed to explore the mechanism and evaluate the antihypertensive effect of Uncaria. Serum samples were analyzed and identified by HPLC-TOF/MS, while the acquired data was further processed by partial least squares discriminant analysis (PLS-DA) and orthogonal partial least squares discriminant analysis (OPLS-DA) to discover the perturbed metabolites. A clear cluster among the different groups was obtained, and 7 significantly changed potential biomarkers were screened out. These biomarkers were mainly associated with lipid metabolism (dihydroceramide, ceramide, PC, LysoPC, and TXA2) and vitamin and amino acids metabolism (nicotinamide riboside, 5-HTP). The result indicated that Uncaria could decrease the blood pressure effectively, partially by regulating the above biomarkers and metabolic pathways. Analyzing and verifying the specific biomarkers, further understanding of the therapeutic mechanism and antihypertensive effect of Uncaria was acquired. Metabolomics provided a new insight into estimate of the therapeutic effect and dissection of the potential mechanisms of traditional Chinese medicine (TCM) in treating hypertension.

Insulin resistance (IR) plays a central role in the development of several metabolic diseases, which leads to increased morbidity and mortality rates, in addition to soaring health-care costs. Deep sea water (DSW) and fucoidans (FPS) have drawn much attention in recent years because of their potential medical and pharmaceutical applications. This study investigated the effects and mechanisms of combination treatment of DSW and FPS in improving IR in HepG2 hepatocytes induced by a high glucose concentration. The results elucidated that co-treatment with DSW and FPS could synergistically repress hepatic glucose production and increase the glycogen level in IR-HepG2 cells. In addition, they stimulated the phosphorylation levels of the components of the insulin signaling pathway, including tyrosine phosphorylation of IRS-1, and serine phosphorylation of Akt and GSK-3ß. Furthermore, they increased the phosphorylation of AMPK and ACC, which in turn decreased the intracellular triglyceride level. Taken together, these results suggested that co-treatment with DSW and FPS had a greater improving effect than DSW or FPS alone on IR. They might attenuate IR by targeting Akt/GSK-3ß and AMPK pathways. These results may have some implications in the treatment of metabolic diseases.

The present study aimed to investigate the intervention mechanisms of Rhizoma Coptidis using spontaneously hypertensive rats. A serum metabolomics analysis was performed with high performance liquid chromatography­quadrupole/time of flight mass spectrometer in positive mode. The obtained data were further analyzed by principal component and partial least­squares discriminant analysis to reveal differentiating metabolites. The pattern of metabolites in the serum after Rhizoma Coptidis exhibited distinct alterations. A total of 10 potential biomarkers were significantly altered in the serum and may be associated with the underlying mechanism. Alterations were primarily associated with phospholipid metabolism, fatty acid metabolism, amino acid metabolism and arachidonic acid metabolism. In addition, biochemical alterations in potential biomarkers were associated with inflammation, nitric oxide production, platelet aggregation and endothelial function. By analyzing and verifying the specific biomarkers, metabolomics may be helpful to further understand the underlying therapeutic mechanism of Rhizoma Coptidis. Metabolomics is a powerful tool used to investigate the therapeutic effects of herbal medicine with multiple targets.

Glycyrrhiza polysaccharide liposome (GPSL) was prepared by reverse-phase evaporation method and optimized with response surface methodology. As a result, the optimum preparation conditions were as follows: the ratio of soybean phosphatide to Glycyrrhiza polysaccharide (GPS) of 24:1, temperature of drug incubation of 46°C, the ultrasound time of 16min. Confirmation experiments and transmission electron micrograph (SEM) exhibited the entrapment efficiency of liposome as 78.33±0.25%, with spherical shape and uniform sizes. Furthermore, proliferation assay, allogeneic mixed lymphocyte reaction and supernatant cytokines of chBM-DCs were performed by MTT and ELISA methods to investigate the immune-modulating effects of GPSL and GPS. The results showed that GPSL could significantly promote the proliferation of immature chBM-DCs, enhance the ability of mature chBM-DCs to induce T-cell proliferation and modulate the mature chBM-DCs to secret cytokines such as IL-2, IFN-γ and IL-10. These evidences indicated that the immune-modulating activity of GPS was significantly improved after encapsulated with liposome.

RESUMO

One new tirucallane-type nortriterpenoid Nortirucallane A (1), together with Chrysoeriol (2) and Isorhamnetin (3), was isolated from the methylene chloride part of Lonicerae japonicae flos. Their structures were elucidated by the detailed analysis of comprehensive spectroscopic data. Compound 3 was isolated from the genus of Lonicera for the first time. The significance of 1 for the study of phytochemical taxonomy was discussed.

One new flavanocoumarin (1), as well as six known flavonoids (2-7), was isolated from the ethyl acetate extract of the thorns of Gleditsia sinensis. The structures of these compounds were elucidated by extensive spectroscopic measurements and comparison with data reported in literatures. Cytotoxic activities of compounds 1-6 were evaluated against human liver cancer SK-hep-1 cells in vitro by the MTT method, with compound 1 displaying moderate activity (IC50 of 62.53 µM). Furthermore, compound 1 could increase the number of apoptosis cells in a concentration-dependent manner.

Diaphragma juglandis Fructus has been known for its effect on tonifying kidney in traditional Uighur medicine. During our ongoing studies on the discovery of biologically active compounds from folk medicine, a pair of flavanonol glycoside isomers named (2S,3S)-taxifolin-3-O-α-d-arabinofuranoside (1) and (2S,3S)-taxifolin-3-O-α-l-arabinofuranoside (2) were isolated, among them, 1 was a new compound. It is the first report of taxifolin-3-O-arabinofuranosides in both α-d and α-l configuration forms in genus Juglans. Their structures were determined on the basis of spectroscopic analysis and chemical evidence, in combination with circular dichroism.

Breast cancer type 2, early onset susceptibility gene (BRCA2) is a major component of the homologous recombination DNA repair pathway. It acts as a tumor suppressor whose function is often lost in cancers. Patients with specific mutations in the BRCA2 gene often display discrete clinical, histopathological, and molecular features. However, a subset of sporadic cancers has wild type BRCA2 and display defects in the homology-directed repair pathway, which is the hallmark of 'BRCAness.' The mechanisms by which BRCAness arises are not well understood but post-transcriptional regulation of BRCA2 gene expression by microRNAs (miRNAs) may contribute to this phenotype. Here, we examine the post-transcriptional effects that some members of the six-miRNA cluster known as the miR-17/92 cluster have on the abundance of BRCA2's messenger RNA (mRNA) and protein. We discuss two interactions involving the miR-19a and miR-19b members of the cluster and the 3'UTR of BRCA2's mRNA. We investigated these miRNA:mRNA interactions in 15 cell lines derived from pancreatic, breast, colon, and kidney tissue. We show that over-expression of these two miRNAs results in a concomitant decrease of BRCA2's mRNA and protein expression in a subset of the tested cell lines. Additionally, using luciferase reporter assays we identified direct interactions between miR-19a/miR-19b and a miRNA response element (MRE) in BRCA2's 3'UTR. Our results suggest that BRCA2 is subject to a complex post-transcriptional regulatory program that has specific dependencies on the genetic and phenotypic background of cell types.

Objective: To study the chemical constituents of the methylene chloride extract of Lonicerae Japonicae Flos. Methods: The compounds were isolated and purified by silica gel column chromatography,Sephadex LH-20 gel column chromatography and recrystallization. The structures were elucidated by the physical and chemical properties,MS,1H-NMR and13C-NMR spectroscopy. Results: Nine compounds were isolated and their structures were identified as ß-sitosterol( 1),benzoic acid( 2),5-hydroxy-7,3',4'-trimethoxyflavone( 3),tetrapedic acid B( 4),tricin( 5),hydnocarpin D( 6),6,7,10-trihydroxy-8-octadecenoic acid( 7),5'-methoxyhydnocarpin-D( 8) and daucosterol( 9). Conclusion: Compound 2,4,7,8 are isolated from this plant,compounds 2,7,8 are isolated from Caprifoliaceae family for the first time.

RESUMO

A new polysaccharide named PRG was isolated from the fruiting bodies of Phellinus ribis by hot water extraction, ethanol precipitation, anion-exchange and gel-filtration chromatography. PRG was homogeneous, with a molecular weight of 5.16 × 10(3)Da, as determined by high-performance size-exclusion chromatography-multiangle laser light scattering analysis. Its structural characteristics were investigated and elucidated by methylation analysis, partial acid hydrolysis, gas-liquid chromatography mass spectrometry, Fourier transform infrared and nuclear magnetic resonance spectroscopy. Based on obtained data, PRG was found to be a ß-d-glucan containing a (1 → 3)-linked backbone, with a branch of two (1 → 6)-linked and one terminal glucoses substituting at the C-6 position every three residues, along the main chain. PRG exhibited neurotrophic activity, which significantly promoted the neurite outgrowth of the nerve growth factor-stimulated PC12 cells, suggesting that it might be a potential candidate for the treatment of neurodegenerative diseases.
