In clinical application, a microneedle system that continuously delivers drugs is of great value for the delivery of some vaccines and hormone drugs. In this study, a controlled-release chitosan-based microneedle array (PVA/CS-MN) was designed, combining microneedle patches with drugs for controlled-release of drugs. Here we report the optimization of the preparation process of PVA/CS-MN. The appearance, morphology, mechanical properties, dissolution and swelling properties, and in vitro penetration properties of the MN arrays were characterized. The PVA/CS-MN prepared by the optimal process showed good morphology and mechanical properties. PVA/CS-MN can smoothly open microchannels on the skin and achieve controllable dissolution and swelling functions. Ascorbic acid (l-ascorbic acid) was used as a model drug to prepare a Vc-PVA/CS-MN. In vitro transdermal diffusion experiments showed that the Vc-PVA/CS-MN released about 57% of the drug within 1 h. About 66.7% of the drug was slowly released within 12 h, and a total of 92% of the drug was released after 7 days. The controllable sustained-release properties and excellent drug delivery efficiency of PVA/CS-MN provide a new option for sustained transdermal drug delivery.

With in-depth development of industrialization and urbanization in China, improving the professional skills and quality of migrant workers in the construction industry has become an important measure to optimize the labor force structure and promote the industry upgrading. Numerous studies have been carried out on this topic, and construction industrial workers with high skills level and professional quality have replaced the professional identity of migrant workers. However, the psychological cognitive mechanism of migrant workers' occupational role enhancement behavior has not been fully revealed. This study aims to construct a theoretical model of the intention to influence the industrialization of migrant workers in the construction industry based on the frameworks of the theory of planned behavior and risk perception theory, and to explore the key factors and cognitive mechanisms in their transformation into industrial workers in the construction industry. Empirical study using structural equation modeling through field collection of 383 questionnaires from migrant construction workers shows that perceived behavioral control, subjective norm, and behavioral attitude all have significant positive effects on behavioral intention, with decreasing direct effects in descending order of magnitude. Perceived behavioral control also predicts professionalization through the mediation of behavioral intentions, and the newly introduced risk perception factor in the model has a negative inhibitory effect on behavioral intentions and actual behavior. This study validates the important role of psychological intention on the industrialization of migrant workers in the construction industry, providing a new perspective to promote their transformation into industrial workers, and laying the foundation for the modern transformation and sustainable industry development.

Xinfengjiang River, the largest tributary of Dongjiang River, plays a key role in the water supply of Heyuan, Huizhou, Guangzhou and even the Pearl River urban agglomeration. It is crucial to determine the pollution status, potential ecological risk degree of heavy metals in Xinfengjiang river sediment and their influence on the abundance of fish species. In this paper, seven heavy metal concentrations in sediment from the Heyuan section of the Xinfengjiang river were investigated. The order of average concentration was: As > Zn > Pb > Cr > Cu > Cd > Hg. The average concentrations of Cd, Zn and Cu in the upper reaches of the Xinfengjiang Reservoir were significantly higher than those in the reservoir. The mean value order of Igeo was: Cd > Zn > Pb > As > Cu > Cr > Hg. Cd and As had the highest ecological risk index and the greatest threat to the ecological environment. Pearson correlation analysis and principal component analysis demonstrated that the pollution source of heavy metals such as Cu and Cd are much more likely to originate from the mine fields located in the northeast of the sampling sites. In addition, agriculture, electronic industry and domestic sewage also contributed to the concentration of heavy metals in different degrees. Redundancy analysis showed that the abundance of Cypriniformes was negatively correlated with Cu and Cd concentrations, suggesting that mining activities might indirectly affect the abundance of fish species.

This study developed, a novel polypropylene (PP) mesh combined with poly (L-lactic acid) (PLA) electrospun nanofibers loaded sirolimus (SRL). The PP mesh was combined with PLA/SRL (1/0, 1/0.01, 1/0.02; mass ratios) composed electrospun membrane characterized by FTIR spectroscopy, XPS and SEM, and evaluated for cytocompatibility in vitro. In an in vivo study, a total of 84 Sprague-Dawley rats were employed to evaluate the efficacy of the novel composite PP mesh anti-adhesion, mechanical properties and inflammation. As a results, the PLA/SRL membrane could compound with PP mesh stably and load SRL. Although tensile testing showed that the mechanical properties of composite mesh decreased in vivo, the integration strength between the tissue and mesh was still able to counteract intra-abdominal pressure. Compared with the native PP mesh group, the novel PP mesh group showed a lower score for abdominal adhesion and inflammation. More importantly, the novel PP mesh completely integrated with the abdominal wall and had sufficient mechanical strength to repair abdominal wall defects.

Many uncertainties such as variable irregular structure and complex flow conditions bring difficulties to the design of a nature-like fishway. This study defines the main factors and parameters affecting flow conditions such as permeability ratio, offset ratio, bottom slope and pool length to simplify and generalize the irregular geometry of the nature-like fishway. According to the engineering requirements of the Mopiling nature-like fishway, the effect of the above parameters of pool geometry on the flow structure is investigated through a 3D turbulent numerical simulation, and the parameter thresholds are summarized according to the optimization of the flow conditions. The results show that under the same conditions, the maximum velocity of the control section increases with the increase in permeability ratio, bottom slope and pool length, and the offset ratio has limited effect on the maximum velocity of the control section. It is recommended that when the bottom slope is 1/250 and the pool length is 10 m, the permeability ratio should not be greater than 0.30 and the offset ratio should be located between 0.15 and 0.60. When the bottom slope is adjusted to 1/200, it is recommended to control the permeability ratio below 0.20, the offset ratio between 0.30 and 0.60, and the pool length can be adjusted to 8 m. Within the above threshold range of the design parameters, the maximum velocity in the fishway can be basically controlled at about 1.0 m/s. The mainstream in the pool is clear and the flow pattern is good, which can basically satisfy the requirements of fish passing. The relevant design parameters and optimization strategies can provide reference for similar projects.

As a new transdermal drug delivery technology, bubble microneedle could achieve painless and precise drug delivery, which has attracted great attention from researchers. In order to improve the utilization rate of the drug carried by microneedle, we proposed a method for preparing a tip-loaded bubble-soluble microneedle. During the molding process of the microneedle, air bubbles were formed in the needle body, and the drug was concentrated on the needle tip. The preparation process of the bubble microneedle was optimized. The effects of foaming agent concentration, drying temperature, and solution viscosity on the forming of bubble microneedles were explored. Furthermore, the transdermal effect of the product was analyzed. The experimental results showed that the bubble microneedle forming process was stable, with the forming rate above 90% and the forming cycle shortened to about 4 h. The drug was mainly concentrated on the tip of the microneedle, with a length of 180 µm, and the length of the bubble was 250 µm. Moreover, the microneedle array can create microchannels on the mouse skin, and the needle bodies can be rapidly dissolved within 5 min. The bubble microneedle could rapidly release about 48% of the drug within 1 min and about 91% of the drug within 5 min. The bubble microstructure of the microneedle array hindered the diffusion of the drug to the substrate, which improves the utilization rate of the drug. This study provides a technical basis for the practical application of microneedle for transdermal drug delivery.

The massive food wastes pose a growing health concern for spreading of antibiotic resistance and pathogens due to food spoilage. However, little is known about these microbial hazards during collection, classification, and transportation before eventual treatment. Here, we profiled the temporal variations of antibiotic resistance genes (ARGs), pathogens, bacterial and fungal communities across four typical food wastes (vegetable, fish, meat, and rice) during storage at room temperature in summer (maximum 28-29 °C) of typical southeast city in China. A total of 171 ARGs and 32 mobile genetic elements were detected, and the absolute abundance of ARGs significantly increased by up to 126-fold with the storage time. Additionally, five bacterial pathogens containing virulence factor genes were detected, and Klebsiella pneumoniae was persistently detected throughout the storage time in all food types except rice. Moreover, fungal pathogens (e.g., Aspergillus, Penicillium, and Fusarium) were also frequently detected. Notably, animal food wastes were demonstrated to harbor higher abundance of ARGs and more types of pathogens, indicating a higher level of hazard. Mobile genetic elements and food types were demonstrated to mainly impact ARG profiles and pathogens, respectively. This work provides a comprehensive understanding of the microbial hazards associated with food waste recycling, and will contribute to optimize the food waste management to ensure biosecurity and benefit human health.

The damming of the river changes the structure of the original river ecosystem, and although fish passage plays an important role in maintaining the connectivity of the river ecosystem, the fish have difficulty finding the fish passage entrance during the upstream process. This paper studied the rheotaxis of fish under three different water flow conditions experimentally through recirculating water tanks. To better understand the response of Crucian carp (Carassius auratus) to water flow stimulation, the representative swimming trajectory, sensing success rate, attraction success rate, reaction time, and attraction time of the fish were analyzed by using a video monitoring system. The experimental results showed that fish responded differently to single-peak and lateral bimodal outflow conditions: (1) the single-peak outflow condition had a much better attraction effect than the lateral bimodal outflow condition, both in terms of sensing success rate and attraction success rate; (2) the fish swam mainly in the middle area of the lateral bimodal outflow condition, while the fish swam more evenly in the single-peak outflow condition. Therefore, setting the attraction current at the right time and near the entrance of the fish passage may help to improve the effect of fish attraction.

A meta-analysis was performed to evaluate the preoperative smoking and smoke cessation on wound healing and infection in post-surgery subjects. A systematic literature search up to January 2022 incorporated 11 trials involving 218 567 subjects after post-surgery at the beginning of the study; 176 670 were smoke cessation or non-smokers, and 41 897 were smokers. Statistical tools like the dichotomous method were used within a random or fixed-influence model to establish the odds ratio (OR) with 95% confidence intervals (CIs) to evaluate the influence of preoperative smoking and smoke cessation on wound healing and infection in post-surgery subjects. Smoke cessation or non-smokers had significantly lower postoperative wound healing problems (OR, 0.59; 95% confidence interval, 0.43-0.82, P < .001), and surgical site wound infection (OR, 0.74; 95% CI, 0.63-0.87, P < .001) compared with smokers in post-surgery subjects. Smoke cessation or non-smokers had significantly lower postoperative wound healing problems, and surgical site wound infection compared with smokers in post-surgery subjects. Furthermore, evidence is needed to confirm the outcomes.

Bisphenol A (BPA) is a widely used endocrine disruptor, which has attracted much attention due to its harmful effects on male reproduction. To investigate the interference of BPA on steroid synthesis in males, male rare minnows (Gobiocypris rarus) were exposed to 15 µg L-1 BPA for 7, 14 and 21 d. Meanwhile, a positive control group was performed with 25 ng L-1 17α-ethynyl estradiol (EE2). Results showed that BPA exposure induced lower testosterone (T) levels, while affecting the transcripts of steroidogenic gene StAR. Moreover, BPA induced abnormal germ cells proliferation in the testis in rare minnow. Transcriptome analysis showed that 354 transcripts significantly differentially expressed after BPA exposure for 14 d, several of them were enriched in the signaling pathways of cell cycle process, PPAR signaling pathway, the steroid synthesis pathway and estrogen signaling pathway. BPA significantly increased estrogen receptor (ER) levels and induced abnormal protein levels of PPARγ. BPA disrupted the StAR expression by interfering ER enrichments within StAR 5' flanking region. Additionally, our study also revealed that BPA and EE2 might have different mechanisms for interfering with steroid hormone levels and germ cells proliferation in the testis.

Epstein-Barr virus (EBV), the first identified human tumor virus, is etiologically associated with various kinds of malignant and benign diseases, accounting for 265,000 cancer incident cases and 164,000 cancer deaths in 2017. EBV prophylactic vaccine development has been gp350 centered for several decades. However, clinical studies show that gp350-centered vaccines fail to prevent EBV infection. Advances in the EBV infection mechanisms shed light on gB and gHgL, the two key components of the infection apparatus. In this study, for the first time, we utilized recombinant vesicular stomatitis virus (VSV) to display EBV gB (VSV-ΔG-gB/gB-G) or gHgL (VSV-ΔG-gHgL). In vitro studies confirmed successful virion production and glycoprotein presentation on the virion surface. In mouse models, VSV-ΔG-gB/gB-G or VSV-ΔG-gHgL elicited potent humoral responses. Neutralizing antibodies elicited by VSV-ΔG-gB/gB-G were prone to prevent B cell infection, while those elicited by VSV-ΔG-gHgL were prone to prevent epithelial cell infection. Combinatorial vaccination yields an additive effect. The ratio of endpoint neutralizing antibody titers to the endpoint total IgG titers immunized with VSV-ΔG-gHgL was approximately 1. The ratio of IgG1/IgG2a after VSV-ΔG-gB/gB-G immunization was approximately 1 in a dose-dependent, adjuvant-independent manner. Taken together, VSV-based EBV vaccines can elicit a high ratio of epithelial and B lymphocyte neutralizing antibodies, implying their unique potential as EBV prophylactic vaccine candidates. IMPORTANCE Epstein-Barr virus (EBV), one of the most common human viruses and the first identified human oncogenic virus, accounted for 265,000 cancer incident cases and 164,000 cancer deaths in 2017 as well as millions of nonmalignant disease cases. So far, no prophylactic vaccine is available to prevent EBV infection. In this study, for the first time, we reported the VSV-based EBV vaccines presenting two key components of the EBV infection apparatus, gB and gHgL. We confirmed potent antigen-specific antibody generation; these antibodies prevented EBV from infecting epithelial cells and B cells, and the IgG1/IgG2a ratio indicated balanced humoral-cellular responses. Taken together, we suggest VSV-based EBV vaccines are potent prophylactic candidates for clinical studies and help eradicate numerous EBV-associated malignant and benign diseases.

INTRODUCTION: NLRP3 inflammasome responses and gut microbiota have been shown an important role in lung cancer, however, the relationship between gut microbiota and NLRP3 inflammasome responses in lung cancer with Qi-yin deficiency remains elusive. METHODS: To investigate the effect of the traditional Chinese medicine BuFeiXiaoJiYin (BFXJY) on NLRP3 inflammasome responses and dysbiosis in lung cancer with Qi-yin deficiency, the female BALB/cA-nu mice were treated with LPS and ATP to induce inflammation, and were intragastrically treated with warm Chinese medicine and smoked with shavings to induce Qi-yin deficiency, as well as were injected with 1 × 107/ml A549 cells to simulate lung cancer. Then the three different doses of BuFeiXiaoJiYin (BFXJY) and positive control (CRID3) were used for intervention in mice for 27 consecutive days. Then, we estimated the protection effect of BFXJY on lung cancer mice with Qi-yin deficiency, through deterring tumor growth, NLRP3 inflammasome, PKC signaling, and homeostasis of gut microbiota. RESULTS: In this study, we found that BFXJY could inhibit the tumor growth in lung cancer with Qi-yin deficiency by reducing the production of IL-1ß and IL-18 and inhibiting NLRP3 inflammasome activation, which might be associated with the inhibition of PKC signaling. Furthermore, BFXJY could promote microbial diversity and balance the microbial composition changes induced by inflammation and Qi-yin deficiency in lung cancer. CONCLUSION: BuFeiXiaoJiYin ameliorates the NLRP3 inflammation response and gut microbiota in mice with lung cancer companied with Qi-yin deficiency. Our study provides a theoretical basis for the clinical development of therapeutic drugs targeting to treat lung cancer.

(+)-Sarcanan A (1a) and (-)-Sarcanan A (1b), a pair of new dihydrobenzofuran neolignan enantiomers, together with six known compounds (2-7), were isolated from the aerial parts of Sarcandra glabra. Their structures were elucidated by spectroscopic analysis, and the absolute configurations of 1a and 1b were determined by analyses of the electronic circular dichroism (ECD) data. All compounds were evaluated for their inhibitory effects on the nitric oxide (NO) production induced by lipopolysaccharide (LPS) in RAW264.7 cells, and compounds 2-4 exhibited moderate inhibition against NO production.

Bisphenol-A (BPA) has been reported to disrupt blood-testis barrier (BTB) integrity in mammals. However, its effects on fish testis sertoli cell (SC) barrier and the underlying mechanisms have been largely unknown to date. To study the SC barrier toxicity induced by BPA, male rare minnows (Gobiocypris rarus) were exposed to 15 µg L - 1 BPA for 7, 14 and 21 d. Meanwhile, a 25 ng L-1 17α-ethynyl estradiol (EE2) group was set up as the positive control. Results showed that BPA induced immune response in the testes and decreased offspring hatching rate. The biotin tracer assay showed that BPA exposure destroyed the integrity of the testis SC barrier. In addition, BPA exposure decreased the expressions of occludin, ZO-1, CX43 and N-cadherin proteins. The transcripts of CX43 and occludin were significantly decreased and SP1 recruitment in each gene promoter was repressed after BPA exposure. Moreover, the cytokines (TNFα and IL-1ß) were significantly increased while the JNK signal pathway was activated after BPA exposure. BPA also increased the matrix metalloproteinases 1 (MMP1) and MMP2 levels in the testes. In addition, estrogenic effect did not entirely explain the mechanism by which BPA disrupted the SC barrier in G. rarus testes. These results suggested that BPA disrupted the SC barrier integrity by inhibiting SP1 enrichments within CX43 and occludin 5' flanking regions through activated cytokines/JNK signaling pathway. MMPs were also involved in the disruption of SC barrier caused by BPA exposure.

2-Methyl-3-buten-2-ol (MBO232) is a potential biofuel and renewable fuel additive. In a combustion environment, the consumption of MBO232 is mainly through the reaction with a OH radical, one of the most important oxidants. Here, we predict the intricate reactions of MBO232 and OH radicals under a broad range of combustion conditions, that is, 230-2500 K and 0.01-1000 atm. The potential energy surfaces of H-abstraction and OH-addition have been investigated at the CCSD(T)/CBS//M06-2X/def2-TZVP level, and the rate constants were calculated via Rice-Ramsperger-Kassel-Marcus/master equation (RRKM/ME) theory. The decomposition reactions of the critical intermediates from the OH-addition reactions have also been studied. Our results show that OH-addition reactions are dominant below 850 K, while H-abstraction reactions, especially the channel-abstracting H atoms from the methyl groups, are more competitive at higher temperatures. We found that it is necessary to discriminate H atoms attached to the same C atom, as their abstraction rates can differ by up to 1 order of magnitude. The calculated results show good agreement with the reported experimental data. We have provided the modified Arrhenius expressions for rate constants of the dominant channels. The kinetic data determined in this work are of much value for constructing the combustion models of MBO232 and understanding the combustion kinetics and mechanism of other unsaturated alcohols.

BACKGROUND: Sphingolipids produce pleiotropic signaling pathways, and participate in the pathological mechanism of hepatocyte apoptosis and necrosis during liver injury. However, the role of glucosylceramide synthase (GCS)-key enzyme that catalyzes the first glycosylation step, in liver injury is still vague. METHODS: All experiments were conducted using 7-9-week-old pathogen-free male C57BL/6 mice. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were detected in murine models of liver disease, in addition to histological characterization of liver injuries. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the relative expression of the GCS, matrix metallopeptidase-1 (MMP-1), and tissue inhibitor of metalloproteinase-1 (TIMP-1) genes. The GCS was observed through a fluorescence microscope, and the flow cytometry was used to detect hepatocyte apoptosis. The concentrations of serum IL-4, IL-6, and IL-10 were measured using enzyme-linked immune-sorbent assay (ELISA) kit. MMP-1 and TIMP-1 protein expression was measured via western blot (WB) analysis. RESULTS: Con A is often used as a mitogen to activate T lymphocytes and promote mitosis. A single dose of Con A injected intravenously will cause a rapid increase of ALT and AST, which is accompanied by the release of cytokines that cause injury and necrosis of hepatocytes. In this study, we successfully induced acute immune hepatitis in mice by Con A. Con A administration resulted in GCS upregulation in liver tissues. Moreover, the mice in the Con A group had significantly higher levels of ALT, AST, IL-4, IL-6, IL-10 and increased hepatocyte apoptosis than the control group. In contrast, all of the aforementioned genes were significantly downregulated after the administration of a GCS siRNA or Genz-123346 (i.e., a glucosylceramide synthase inhibitor) to inhibit the GCS gene. Additionally, the histopathological changes observed herein were consistent with our ALT, AST, IL-4, IL-6, and IL-10 expression results. However, unlike this, hepatocyte apoptosis has been further increased on the basis of the Con A group. Moreover, our qRT-PCR and WB results indicated that the expression of MMP-1 in the Con A group was significantly lower than that in the control group, whereas TIMP-1 exhibited the opposite trend. Conversely, MMP-1 expression in the GCS siRNA and Genz-123346 groups was higher than that in the Con A group, whereas TIMP-1 expression was lower. CONCLUSIONS: GCS inhibition reduces Con A-induced immune-mediated liver injury in mice, which may be due to the involvement of GCS in the hepatocyte repair process after injury.

Vortex beams carrying orbital angular momentum (OAM), which feature helical wavefronts, have been regarded as an alternative degree of freedom for free-space optical (FSO) communication systems. However, in practical applications, atmospheric turbulence and limited-size receiving aperture effects will cause OAM modal degradation and seriously reduce the received power. In this paper, by controlling the radial phase distribution of conventional OAM beams, quasi-ring Airy vortex beams (QRAVBs) are successfully generated in the experiments to increase the received power under the limited-size receiving aperture conditions. By employing 72-Gbit/s 16-ary quadrature amplitude modulation (16-QAM) discrete multi-tone (DMT) signals, we successfully demonstrate free-space data transmission with QRAVBs in the experiments. Moreover, the transmission performance of QRAVBs under atmospheric turbulence is also evaluated. Comparing with conventional OAM beam and Bessel beam, the obtained results show that QRAVBs can achieve higher received power and better BER performance under limited-size receiving aperture and atmospheric turbulence conditions.

We present a spectrum-tunable fiber Bragg grating (FBG) based on a unique compress-twist deformation mode of non-rigid origami. By applying axial displacement on the FBG-bonded Kresling-ori, a non-uniform strain field emerges. The mechanics-induced non-uniform strain can shift the wavelength of an apodized unchirped FBG and/or transform an apodized unchirped FBG to a chirped one. For the spectrum-shaping mode, the bandwidth of the FBG was tuned from 0.32 nm up to 2.9 nm, measured at the -6dB level. For the wavelength-shift mode, a maximum wavelength shift of 0.6 nm can be achieved.

BACKGROUND: To observe the effects of triptolide (TP) on the proliferation, migration, invasion, and epithelial-mesenchymal transition (EMT) of glioma cells, and to explore the possible mechanisms of phenotypic changes in EMT. METHODS: The U87 and U251 glioma cell lines were treated TP. The Cell Counting Kit-8 (CCK-8) method was used to detect the half-maximal inhibitory concentration (IC50) of TP in these two cell lines and the inhibition of cell proliferation at the IC50 concentration. The wound-healing experiment and Transwell invasion assay were used to detect the cells' migration and invasion abilities, respectively. Using western blot protocol, the expression levels of the EMT markers were analyzed, and the levels of the autophagy markers were also detected. The pEGFP-C2-LC3B plasmid was transfected into glioma cells, and the effect of TP on autophagy was detected by immunofluorescence. A subcutaneous tumor model in nude mice was established to observe the effect of TP on cell proliferation in vivo, and immunohistochemistry (IHC) was used to detect the expression levels of EMT markers in mouse tumor tissues. RESULTS: TP significantly inhibited the proliferation of U87 and U251 cells in a dose- and time-dependent manner. TP had a significant inhibitory effect on the migration and invasion of U87 and U251 cells. Western blot showed that TP reversed the process of EMT in glioma cells, which was evidenced by the upregulated expression of the epithelial marker E-cadherin, and the downregulated expression of the mesenchymal markers N-cadherin, Vimentin, ZEB1, Snail, and Slug. TP increased autophagy in glioma cells, increased the LC3B II/I ratio, and upregulated Beclin-1 and Atg-7 expression. Immunofluorescence showed that the number of autophagosomes increased significantly after TP was applied to cells. In the nude mouse subcutaneous tumor model, experiments revealed an inhibitory effect of TP on glioma cell proliferation in vivo. IHC confirmed that the expression of E-cadherin was upregulated in mouse tumor tissues, while the expression levels of N-Cadherin and Vimentin were downregulated. CONCLUSIONS: TP can inhibit glioma cell proliferation, migration, and invasion, and reverse EMT progression. The possible mechanism of EMT reversal in glioma cells is that TP induces autophagy.
