RESUMO

Adeno-associated virus (AAV)-mediated gene delivery systems have been shown to be effective tools for gene manipulation in the inner ear. For example, hair cells (HCs) and multiple other cell types can be transduced by the local injection of AAVs into the inner ear. However, application of the AAV-mediated CRISPR/Cas9 gene-editing approach to the inner ear in adult mice has not yet been studied. Based on our previous work, we investigated several AAV serotypes in neonatal and adult mice in parallel, and found that AAV8 had the top efficiency to transduce inner HCs. We then tested the ability of Cre-expressing AAV8 to activate Cas9 in floxed-Cas9 knockin mice, and observed significant Cas9 activation in the inner ear of both neonatal and adult animals. Neither the AAV8 virus itself nor the surgical procedures used to deliver it-cochleostomy for neonatal mice and canalostomy for adult mice-caused any damage to HCs or impaired normal hearing. Our studies indicate that the local injection of AAV8-Cre can induce Cas9 activation to perform safe and efficient gene editing in the inner ear, expanding the repertoire of gene-editing tools for regulating gene expression in the inner ear as a part of efforts to rescue genetic hearing loss, initiate regeneration of HCs, or develop gene therapy techniques.

In order to develop equipment adapted to the aircraft pulse final assembly line, a vision-based aircraft transport platform system is developed. This article explores a guiding method between assembly stations which is low-cost and easy to change routes by using two-dimensional code and two complementary metal oxide semiconductor (CMOS) cameras. The two cameras installed on the front and back of the platform read the two-dimensional code containing station information to guide the platform. In the process of guiding, the theoretical position and posture of the platform at each assembly station are known, but there is a difference between the actual and theoretical values due to motion errors. To reduce the influence of the deviation on the navigation route, a localization method is proposed based on the two-dimensional images captured by the cameras. Canny edge detection is applied to the processed image to obtain the position of the two-dimensional code in the image, which can measure the angle/distance deviation of the platform. Then, the computer can locate the platform precisely by the information in the two-dimensional code and the deviation measured by the image. To verify the feasibility of the proposed method, experiments have been performed on the developed platform system. The results show that the distance and angle errors of the platform are within ±10 mm and ±0.15° respectively.

Carbon-based, all-inorganic perovskite solar cells (PSCs) have drawn enormous attention recently on account of their ungraded stability and reduced costs. However, their power conversion efficiencies (PCEs) still lag behind the ones with conventional architecture. Moreover, the high cost of FTO substrates and energy-consuming sintering process of TiO2 electron-transporting layers should be further addressed. Herein, it is demonstrated that the FTO/TiO2 substrates could be separated simply from degraded CsPbIBr2 PSCs for fabricating the new ones again, which thus reduces the production costs of resulting PSCs and makes them renewable and sustainable. Meanwhile, the characterization results reveal that there are some residual CsPbIBr2-derived species on recycled FTO/TiO2 substrates, which enable the upper CsPbIBr2 films with suppressed halide phase separation and reduced defects, the diminished work function of TiO2 layers from 4.13 to 3.89 eV, along with decreased conduction band minimum (CBM) difference of CsPbIBr2/TiO2 interface from 0.51 to 0.36 eV. Consequently, the average PCE of CsPbIBr2 PSCs is improved by 20%, from 6.51 ± 0.62% to 8.14 ± 0.63%, wherein the champion one yields the exceptional value of 9.12%. These findings provide an avenue for simultaneous performance enhancement and cost-saving of carbon-based, all-inorganic PSCs to promote their commercialization.

Self-powered photodetectors (PDs) with inorganic lead halide perovskites hold multiple traits of high sensitivity, fast response, independence from external power supply, and excellent sustainability and stability, thus holding a great promise for practical applications. However, they generally contain high-temperature-processed electron-transporting layers (ETLs) and high-cost, unstable hole-transporting layers (HTLs) coupled with noble metal electrodes, which bring significant obstacles of production cost and stability for their potential commercialization. Herein, we demonstrate the building of high-performance HTL/ETL-free, self-powered CsPbIBr2 PD with simplified architecture of fluorine-doped tin oxide (FTO)/CsPbIBr2/carbon upon interfacial modification by polyethyleneimine (PEI). The optimized PD yields a dark current of 2.03 × 10-9 A, peak responsivity (R) of 0.32 A/W, maximum specific detectivity (D*) of 3.74 × 1012 Jones, and response time of 1.21 µs. These figures of merit are far beyond those of the one prepared without PEI modification and even the PD containing TiO2 ETL. Hence, our work suggests a highly feasible route to develop self-powered PDs with significantly simplified fabrication and a reduced production cost.

BACKGROUND: Distinction in the mutational profile between the common histological types, lung adenocarcinoma (LUAD) and squamous cell lung carcinoma (LUSC) has been well-established. However, comprehensive mutation profiles of the predominant histological subtypes within LUAD and LUSC remains elusive. METHODS: We analyzed the mutational profile of 318 Chinese NSCLC patients of adenocarcinoma and squamous cell carcinoma predominant subtypes from seven hospitals using capture-based ultra-deep sequencing of 68 lung cancer-related genes. RESULTS: Of the 318 NSCLC patients, 215 were diagnosed with LUAD and 103 with LUSC. Adenocarcinoma in situ and acinar adenocarcinoma were the most predominant subtypes of LUAD. On the other hand, keratinizing squamous cell carcinoma was the most predominant subtype of LUSC. Among the LUAD subtypes, EGFR sensitizing mutations were most prevalent in the invasive lepidic subtype. More than half of the patients with preinvasive adenocarcinoma in situ, minimally invasive, acinar, micropapillary and papillary subtypes were also EGFR-mutants. Patients with colloidal, invasive mucinous, and fetal subtypes had the least number of EGFR mutations. Moreover, KRAS mutations were prevalent in patients with invasive mucinous, colloid, enteric and solid subtypes. A total of 90% of the LUSC patients harbor mutations in TP53, wherein all patients except five with nonkeratinizing were TP53 mutants. PIK3CA amplifications were most prevalent in keratinizing, followed by basaloid and nonkeratinizing subtypes. CONCLUSION: These data suggest that the mutational profiles among the predominant histological subtypes were very distinct, which provided a reliable tool to improve treatment decisions.

BACKGROUND: LRP5/6 are co-receptors in Wnt/ß-catenin pathway. Recently, we discovered multiple ß-catenin independent functions of LRP5/6 in tumor cells and in the diseased heart. Nucleoporin 37 (NUP37) is an important component of the nuclear pore complex (NPC), whose elevated expression is associated with worsened prognosis in liver cancer. Previous studies have shown that NUP37 interacted with YAP and activated YAP/TEAD signaling in liver cancer. Our preliminary findings showed a nuclear location of LRP5. We thus tested the hypothesis that LRP5 may act as a genuine regulator of YAP/TEAD signaling via modulating NUP37 in a ß-catenin-independent way. METHODS: We performed siRNA knockdown of LRP5, LRP6, or ß-catenin in liver cancer HepG2 cells to determine the effect on tumor cell proliferation. Protein expressions and interaction between LRP5 and NUP37 were determined using immunoprecipitation and western blot analyses. RESULTS: HepG2 cell proliferation was markedly inhibited by knockdown of LRP5 but not LRP6 or ß-catenin, suggesting that LRP5 has a specific, ß-catenin-independent role in inhibiting HepG2 cell proliferation. Knockdown of NUP37 by siRNA inhibited the proliferation of HepG2 cells, whereas overexpression of NUP37 reversed the decrease in cell proliferation induced by LRP5 knockdown. Immunoprecipitation assays confirmed that LRP5 bound to NUP37. Furthermore, LRP5 overexpression restored NUP37 knockdown-induced downregulation of YAP/TEAD pathway. CONCLUSIONS: LRP5 deletion attenuates cell proliferation via destabilization of NUP37, in a ß-catenin-independent manner. LRP5 therefore acts as a genuine regulator of YAP/TEAD signaling via maintaining the integrity of the NPC, and implicates a therapeutic strategy in targeting LRP5 for inhibiting liver cancer cell proliferation.

A high-performance transfer printing method using a new soluble tape which can be dissolved in acetone is proposed to be used in heterogeneous integration. Si inks array was transferred from SOI wafers onto various substrates without adhesion promoter by this new method which we refer to as the acetone soluble tape (AST) method to compare with other transfer printing methods by using thermal release tape (TRT), water soluble tape (WST) and polydimethylsiloxane (PDMS). By using the AST method, the transfer printing process does not involve interface contention between stamp/inks and inks/receiver substrate so that it maximizes the transfer printing efficiency. Experimental results present the AST method has good performances, and various alien substrates, even curvilinear surfaces, can be selected as receiver substrates by the AST method. To examine the quality of the transferred Si inks, the Si TFTs were fabricated by using the Si membrane transferred by the AST method on sapphire substrate and the devices show the good performance. All the results confirm that the AST method is an effective method in heterogeneous integration.

Long non-coding (lnc) RNAs are non-coding RNAs longer than 200 nt. lncRNAs primarily interact with mRNA, DNA, protein, and miRNA and consequently regulate gene expression at the epigenetic, transcriptional, post-transcriptional, translational, and post-translational levels in a variety of ways. They play important roles in biological processes such as chromatin remodeling, transcriptional activation, transcriptional interference, RNA processing, and mRNA translation. lncRNAs have important functions in plant growth and development; biotic and abiotic stress responses; and in regulation of cell differentiation, the cell cycle, and the occurrence of many diseases in humans and animals. In this review, we summarize the functions and mechanisms of lncRNAs in plants, humans, and animals at different regulatory levels.

Purpose: Evaluate the diagnostic accuracy of multi-parametric magnetic resonance imaging (mp-MRI) for local staging of bladder cancer (BCa). Materials and Methods: The databases of PubMed, Web of Science, Wanfang, and CNKI were searched for related literatures about BCa diagnosed by mp-MRI from January 1, 2000 to April 12, 2019. The strict inclusion and exclusion criteria were set up to extract records. The Quality Assessment of Diagnostic Accuracy Studies (QUADAS)-2 was used to evaluate quality of the candidate studies. The pooled sensitivity, specificity, positive likelihood ratio (+LR), negative likelihood ratio (-LR), and diagnostic odds ratio (DOR) were calculated to assess the diagnostic authenticity of mp-MRI. The summarized receiver operating characteristic (SROC) curve corresponding with the area under the curve (AUC) were analyzed to comprehensively evacuate the diagnostic value of mp-MRI. Results: A total of 140 studies were retrieved by computer-based searching. After quality control, 4 studies with 259 patients were enrolled for meta-analysis. The pooled results showed 0.84 of sensitivity [95% confidence interval (CI) = 0.79-0.89], 0.91 of specificity (95% CI = 0.87-0.93), 8.24 of +LR (95% CI = 4.87-13.92), 0.18 of -LR (95% CI = 0.10-0.31), 49.42 of DOR (95% CI = 19.07-128.09), and 0.946 of AUC. The Spearman correlation analysis found no threshold effect (p = 0.684). A significant heterogeneity existed among 4 included studies with sensitivity (I2 = 65.7%), specificity (I 2 = 60.0%) and diagnostic OR (I 2 = 67.5%). The Begg's test (p = 0.497) and the egger's test (p = 0.337) found no publication bias. Conclusion: mp-MRI acts a good diagnostic performance for bladder cancer. It is plausible that mpMRIs can be used as an important method for bladder cancer staging.

In this study, a multifunctional hybrid coating composed of AgBr nanoparticles (AgBrNPs) and two-dimensional molybdenum sulfide (MoS2) nanosheets (AgBr@MoS2) was constructed on Ti implant materials using an in situ growth method for the first time. With 660 nm light and visible light irradiation, the electrons were rapidly excited from the valence band of MoS2 to its conduction band, at the same time, AgBrNPs was used as a photoelectric receiver, which exhibited an enhanced photocatalytic activity due to the rapid transfer of photoelectrons from MoS2 nanosheets to AgBrNPs and the suppression of the recombination of electron-hole pairs. This contributed to the rapid production of reactive oxygen species under 660 nm light irradiation, thus the AgBr@MoS2 system killed bacteria and degraded organic matter quickly and efficiently in a short time. Meanwhile, the AgBr@MoS2 system showed excellent stability due to the strong covalent binding between S and Ag in the system, thus preventing AgBrNPs from being reduced to metal Ag.

Indium thin oxide (ITO)-free planar perovskite solar cells (PSCs) were fabricated at a low temperature (150 °C) in this work based on the transparent electrode of photolithography processed nickel/gold (Ni/Au) mesh and the high conductivity polymer, PH1000. Ultrathin Au was introduced to increase the conductivity of metal mesh, and the optimal hexagonal Ni (30 nm)/Au (10 nm) mesh (line width of 5 µm) shows a transmittance close to 80% in the visible light region and a sheet resistance lower than 16.9 Ω/sq. The conductive polymer PH1000 not only smooths the raised surface of the metal mesh but also enhances the charge collection ability of metal mesh. The fabricated PSCs have the typical planar structure (glass/Ni-Au mesh/PH1000/PEDOT:PSS/MAyFA1-yPbIxCl3-x/PCBM/BCP/Ag) and the champion PSC (0.09 cm2) obtains a power conversion efficiency (PCE) of 13.88%, negligible current hysteresis, steady current density and PCE outputs, and good process repeatability. Its photovoltaic performance and stability are comparable to the reference PSC based on the ITO electrodes (PCE = 15.70%), which demonstrates that the Ni/Au mesh transparent electrodes are a promising ITO alternative to fabricate efficient PSCs. The relatively lower performance of Ni/Au based PSC results from the relatively slower charge extraction and stronger charge recombination than the ITO based PSC. Further, we tried to fabricate the large area (1 cm2) device and achieve a PCE over 6% with negligible hysteresis and steady current density and PCE outputs. The improvements of perovskite film quality and interface modification should be an effective approach to further enhance the device performance of Ni/Au based PSCs, and the Ni/Au mesh electrode may find wider applications in PSCs and flexible devices.

Mitogen-activated protein kinase kinase kinases (MAPKKKs) are important components of MAPK cascades, which have different functions during developmental processes and stress responses. To date, there has been no systematic investigation of this gene family in the diploid cotton Gossypium arboreum L. In this study, a genome-wide survey was performed that identified 78 MAPKKK genes in G. arboreum. Phylogenetic analysis classified these genes into three subgroups: 14 belonged to ZIK, 20 to MEKK, and 44 to Raf. Chromosome location, phylogeny, and the conserved protein motifs of the MAPKKK gene family in G. arboreum were analyzed. The MAPKKK genes had a scattered genomic distribution across 13 chromosomes. The members in the same subfamily shared similar conserved motifs. The MAPKKK expression patterns were analyzed in mature leaves, stems, roots, and at different ovule developmental stages, as well as under salt and drought stresses. Transcriptome analysis showed that 76 MAPKKK genes had different transcript accumulation patterns in the tested tissues and 38 MAPKKK genes were differentially expressed in response to salt and drought stresses. These results lay the foundation for understanding the complex mechanisms behind MAPKKK-mediated developmental processes and abiotic stress-signaling transduction pathways in cotton.

Inorganic halide perovskite CsPbBr3 is emerging as one of the promising alternatives to the hybrid counterparts for optoelectronic applications owing to its upgraded stability. Yet, the inherently low solubility of a CsBr precursor material restricts the quality and especially the thickness of a solution-processed CsPbBr3 film, thus hindering the further optimization of device performance. Herein, we report a facile intermediate phase halide exchange reaction that can break the thickness limit of a solution-processed CsPbBr3 film, since it avoids the use of low-solubility CsBr. Furthermore, the CH3NH3I byproduct after halide exchange could trigger a beneficial Ostwald ripening process to promote grain coarsening in the film. Hence, the uniformly flat, pure-phase, and compact CsPbBr3 film composed of [100] preferential, microsized grains can be achieved. As a demonstration of its excellent optoelectronic features, the carbon-based, all-inorganic photodetector with such a favorable film yields a maximum photoresponsivity of 0.35 A W-1 and a specific detectivity of 1.94 × 1013 Jones coupled with a response time of 0.58 µs.

PURPOSE: Point mutations of TP53 tumour suppressor are very rare in schwannomas. We aim to characterize the frequency of exonic copy-number changes of the gene in the tumour and to examine the association between TP53 alterations, phosphorylation status of p53 protein and clinical phenotypes. METHODS: The alterations of TP53 were screened by a combination of Sanger sequencing and multiplex ligation-dependent probe amplification (MLPA) in a total of 44 vestibular schwannomas. The mutation index (MI) in a tumour was defined as the number of exons mutated/ the number of exons tested. Phosphorylation status of p53 protein was investigated by immunoblotting and immunofluorescence. RESULTS: MLPA analysis showed single and multi-exon deletion mutations of TP53 in 65.7% of the cases. Comparisons of clinical features between mutated and non-mutated patients established an association of TP53 mutations with progressive phenotypes, including an earlier formation and a larger tumour. In addition, there were significant correlations between MI and both patients' age and tumour size. The Ser 392 phosphorylation level of p53 varied among tumours, and correlation analysis revealed an age-dependent phosphorylation pattern. The majority of tumours with hyperphosphorylated p53 were from mutated and young patients, suggesting an association of Ser392 phosphorylation with the mutational status of TP53 involved in the acceleration of tumour growth in young individuals. Moreover, Ser 392 phosphorylation contributed to a nuclear accumulation of p53 in schwannona cultures with TP53 mutation. CONCLUSIONS: An interplay between the mutation status of TP53, phosphorylation patterns and tumour behaviors might be established in the disease.

OBJECTIVES: Branchio-Oto (BO) syndrome is one of the common syndromic forms of hearing loss. In this study, we aimed to characterize the clinical and genetic features of BO syndrome in two Chinese Han deaf families. METHODS: The auditory and other BO-related clinical features of Family 1809 and Family 1974 were summarized. Targeted next-generation sequencing in 144 known deafness genes was performed in the probands. Co-segregation of the pathogenic mutations and the phenotype was confirmed by Sanger sequencing in the family members. RESULTS: Interfamilial and intrafamilial variations can be observed in the clinical phenotypes of BO syndrome in Family 1809 and 1974. A novel c.1493_1494insAT (p.Ile498PhefsTer*3) mutation and a previous reported c.967-2A>G mutation in EYA1 were identified as the pathogenic cause in Family 1974 and 1809, respectively. CONCLUSION: Our results supported the heterogeneity of the genetic and phenotypic spectrum of BO syndrome. The recurrent c.967-2A>G in different ethnical groups suggested that it is a hot-spot mutation.

Nucleus accumbens-associated protein-1 (NAC1) is a transcriptional repressor encoded by the NACC1 gene, which is amplified and overexpressed in various human cancers and plays critical roles in tumor development, progression, and drug resistance. NAC1 has therefore been explored as a potential therapeutic target for managing malignant tumors. However, effective approaches for effective targeting of this nuclear protein remain elusive. In this study, we identified a core unit consisting of Met7 and Leu90 in NAC1's N-terminal domain (amino acids 1-130), which is critical for its homodimerization and stability. Furthermore, using a combination of computational analysis of the NAC1 dimerization interface and high-throughput screening (HTS) for small molecules that inhibit NAC1 homodimerization, we identified a compound (NIC3) that selectively binds to the conserved Leu-90 of NAC1 and prevents its homodimerization, leading to proteasomal NAC1 degradation. Moreover, we demonstrate that NIC3-mediated down-regulation of NAC1 protein sensitizes drug-resistant tumor cells to conventional chemotherapy and enhances the antimetastatic effect of the antiangiogenic agent bevacizumab both in vitro and in vivo These results suggest that small-molecule inhibitors of NAC1 homodimerization may effectively sensitize cancer cells to some anticancer agents and that NAC1 homodimerization could be further explored as a potential therapeutic target in the development of antineoplastic agents.

PURPOSE: Lung metastasis is an important cause of breast cancer-related deaths, in which SDF-1/CXCR4 signaling pathway plays a critical role. Single transmembrane protein LRP6 is viewed as an oncogene via activating the Wnt/ß-catenin signaling pathway. Our work aims to investigate the relationship between SDF-1/CXCR4 and LRP6 in breast cancer lung metastasis. EXPERIMENTAL DESIGN: We examined the expressions and functions of SDF-1/CXCR4 and LRP6 as well as their relationship in breast cancer in vitro and in vivo. RESULTS: LRP6 ectodomain (LRP6N) directly bound to CXCR4 and competitively prevented SDF-1 binding to CXCR4. LRP6N prevented SDF-1/CXCR4-induced metastasis to lung and prolonged survival in mice bearing breast tumors, whereas LRP6 knockdown activated SDF-1/CXCR4 signal transduction and promoted lung metastasis and tumor death. Furthermore, patients with breast cancer with high CXCR4 expression had poor prognosis, which was exacerbated by low LRP6 expression but improved by high LRP6 expression. Interestingly, a secreted LRP6N was found in the serum of mice and humans, which was downregulated by the onset of cancer metastasis in both mice bearing breast cancer as well as in patients with breast cancer. CONCLUSIONS: LRP6N might be a promising diagnostic marker for the early detection of breast cancer metastasis as well as an inhibitor of SDF-1/CXCR4-induced breast cancer metastasis. LRP6N also provides an interesting link between Wnt signaling and SDF-1/CXCR4 signaling, the two key pathways involved in cancer development.

The quality of the perovskite layer plays a key role in the performance of perovskite solar cells (PSCs). In this work, we demonstrate a facile way to improve the quality of perovskite films by using a MAI/IPA solution to post-treat the perovskite material after the film formation. XRD, SEM and AFM show that an optimal, uniform, dense and highly crystalline perovskite film is achieved after the post-treatment. The transient photocurrent and transient photovoltage also confirm that the devices after the post-treatment have both the outstanding ability of charge extraction and suppressed charge recombination. By using the post-treatment, a maximum PCE of 19.08%, which is 12% higher than that of the control device, is obtained with a Jsc of 23.12 mA cm-2, a Voc of 1.08 V and an FF of 76.03%. The results show that the MAI/IPA solution post-treatment after the perovskite film formation is a facile way to improve the film quality and the PSC performance.

Perovskite CsPbIBr2 is attracting ever-increasing attention for carbon-based, all-inorganic solar cells, owing to its well-balanced band gap and stability features. However, significant interfacial recombination of charge carriers in solar cells fabricated with this active layer, which is intrinsically associated with the unwanted conduction band misalignment between CsPbIBr2 and the commonly used TiO2 electron transport layer, has limited power conversion efficiency (PCE) values. Herein, we demonstrate successful conduction band alignment engineering at the TiO2 /CsPbIBr2 heterojunction by modifying TiO2 with CsBr clusters. Such modification triggers a beneficial increase in the conduction band minimum (CBM) of TiO2 from -4.00 to -3.81 eV and decreases the work function from 4.11 to 3.86 eV, thus promoting favorable band alignment at the heterojunction, suppressing recombination, and improving extraction and transport of charge carriers. As a result, the carbon-based, all-inorganic CsPbIBr2 solar cells exhibit over 20 % enhancement in average PCE. The champion device achieves a PCE of 10.71 %, a record among pure CsPbIBr2 -based cells, open-circuit voltage of 1.261 V, and excellent stability.

OBJECTIVE: To determine the efficacy of laparoscopic surgery combined with GnRH agonist (GnRH-a) in treating infertile patients with endometriosis (EMs), and analyze its effect on the levels of serum hormones and inflammatory factors of the patients. STUDY DESIGN: An experimental study. PLACE AND DURATION OF STUDY: The Department of Urology, Zhongda Hospital, Southeast University, Jiang Su, China, from January 2015 to March 2016. METHODOLOGY: One hundred and thirty infertile patients with EMs were randomly divided into a control group and an observation group with 65 cases in each group. Patients in the control group were treated by laparoscopic surgery, while patients in the observation group were added by the subcutaneous injection of GnRH-a, based on the treatment of the control group. Therapeutic effects, recurrence rates, and pregnancy rates between two groups were compared. Moreover, the levels of serum hormones estradiol (E2), luteinizing hormone (LH), follicle stimulating hormone (FSH), inflammatory factors interleukin 17 (IL-17), interleukin 6 (IL-6), and tumor necrosis factor- (TNF-α) between two groups after treatment were compared. RESULTS: The total effective rate and pregnancy rate in the observation group were higher than those of the control group (p=0.009 and 0.002, respectively), and the recurrence rate was lower than that of the control group (p=0.008). After treatment, levels of serum E2, LH, FSH in the observation group were lower than those of the control group (all p<0.001). Levels of serum IL-17, IL-6 and TNF-α were lower than those of the control group (all p<0.001). There was no statistically significant difference in adverse reactions between the two groups (p=0.730). CONCLUSION: Laparoscopic surgery combined with GnRH-a is a more effective treatment modality in endometriosis. It can reduce the serum levels of inflammatory factors in patients, improve their hormone level and pregnancy rate, and reduce the recurrence rate.
