RESUMO

We report unprecedented Friedel-Crafts arylation of chlorofullerenes C60Cl6 and C70Cl8 with unprotected carboxylic acids as an efficient single-step synthesis of the inherently stable water-soluble fullerene derivatives. Using this method, a series of previously unaccessible compounds was obtained without chromatographic purification in almost quantitative yields. Promising anti-HIV activity comparable to characteristics of commercial drugs was demonstrated for some of these compounds.

Numerous of pollutants threaten our planet, for instance plastic wastes causes a huge potential risk on the environment in addition to many of emergened pollutants as pharmaceutical residue in aquatic environments which affecting ecological balance and in-turn affecting human health. Accordingly, this research proposed an innovative facile, one-step synthesis of functionalized magnetic fullerene nanocomposite (FMFN) via catalytic thermal decomposition of sustainable poly (ethylene terephthalate) bottle wastes as feedstock and ferrocene as a catalyst and precursor of magnetite. Growth mechanism of FMFN was discussed and batch experiments were achieved to examine its adsorption efficiency in relation to Ciprofloxacin antibiotic. Different adsorption parameters including time, initial Ciprofloxacin concentration, and solution temperature were investigated and optimized using Response Surface Methodology (RSM) model. In addition, a study on the antibiotic adsorption process impact on the organisms of an ecosystem was conducted using E. coli DH5α, and results validated method's efficiency in overcoming problem of appearance of antibiotic-resistant microbes.

In this study, we developed an impedimetric sensor system by using molecularly cortisol imprinted acrylamide polymers on fullerene modified carbon electrode to detect cortisol in real saliva samples. The polymer layer was formed on fullerene modified screen printed carbon electrodes with the ratio of 1:2 (cortisol:acrylamide) and ammonium persulfate (APS) was used as the initiator of polymerization. The sensor surface was investigated by cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and Scanning Electron Microscopy (SEM). Performances of the electrode were determined as follows; calibration curve was calculated between 0.5 nM and 64 nM, R2 = 0.9939, LOD and LOQ were 0.14 nM and 0.44 nM, respectively. Real human saliva samples were obtained from 15 participants between 25 and 32 ages regardless of gender. The samples, which were collected at least 30 min after waking up and the new sensor method analysis were compared tandem mass spectroscopy coupled liquid chromatography system (LC-MS/MS). The sensor analysis showed a significant correlation with LC-MS/MS results (R2 = 0.9727).

In this study, novel water-soluble quaternary ammonium salts of iminofullerenes (IFQA) were synthesized by nitrene chemistry in combination with quaternization and identified as [C60(NCH2CH2NH3+·CF3COO-)4·10H2O]n by various spectroscopies. Maize and Arabidopsis seeds were used to test the bioactivity of IFQA in seed germination. Compared with the control, maize seed exposure to 50 mg/L IFQA (normal: 73.1% vs 58.7%; drought: 66.7% vs 50.0% at the second day) and Arabidopsis seed exposure to 20 mg/L IFQA (normal: 77.5% vs 58.8%; drought: 63.3% vs 36.7% at the second day) had higher germination rates and quicker germination. The results of two-dimensional gel electrophoresis combined with mass spectroscopy showed that the abundance of 21 proteins in embryo proteome of maize seeds was significantly changed (>1.5 fold). The downregulated six storage proteins and upregulated four proteins induced by IFQA for energy production and sugar metabolism indicated a faster metabolic activity of maize seed germination. The upregulated eight stress-related proteins and antioxidant enzymes suggested that the role of IFQA was to activate the metabolic processes in seed germination and also increase seed stress response. The results provide important information to understand the mechanism of seed germination enhancement by carbon nanomaterials.

Interactions between fullerene C24 and a frequently used supplement for sport activities, ephedrine (EPH), have been studied in detail by a combination of density functional theory (DFT), time dependent DFT (TD-DFT) calculations, the symmetry-adapted perturbation theory (SAPT) approach and molecular dynamics (MD) simulations. Information about interaction energies and non-covalent interactions formed between C24 and EPH have been obtained by DFT calculations. TD-DFT calculations have been used in order to obtain UV/vis spectra and to check whether the presence of the EPH molecule produces significant changes in the spectrum. The SAPT approach has been employed in order to decompose the interaction energy into components and therefore to better understand the physical origins of interaction between C24 and EPH. Last, but not least, MD simulations have been used in order to track the influence of temperature on the interactions between C24 and EPH.

Host-guest interactions are of central importance in many biological and chemical processes. However, the investigation of the formation and decomplexation of host-guest systems at the single-molecule level has been a challenging task. Here we show that the single-molecule conductance of organoplatinum(II) metallocycle hosts can be enhanced by an order of magnitude by the incorporation of a C60 guest molecule. Mechanically stretching the metallocycle-C60 junction with a scanning tunneling microscopy break junction technique causes the release of the C60 guest from the metallocycle, and consequently the conductance switches back to the free-host level. Metallocycle hosts with different shapes and cavity sizes show different degrees of flexibility to accommodate the C60 guest in response to mechanical stretching. DFT calculations provide further insights into the electronic structures and charge transport properties of the molecular junctions based on metallocycles and the metallocycle-C60 complexes.

The effect of aqueous unmodified fullerene C60 dispersions (AFD) at a concentration of 5 mg/L on the total number and metabolic activity of heterotrophic river bacteroplankton was investigated. The stimulating effect of AFD on the number of bacterioplankton and on the activity of electron transport chains in its cells is shown. It is established that, in the presence of AFD, the bactericidal activity of antibiotics decreases. The stimulating effect of fullerene on the natural heterotrophic bacterioplankton, which we discovered, casts doubt on the expediency of using AFD as a bacteriostatic agent.

RESUMO

A short metallofullere-peapod of Y2@C79N⊂[4]CHBC was constructed. The strong confinement effect from the large π-extended [4]CHBC nanoring induces molecular orientation of the wrapped Y2@C79N, which can be sensed by a Y2@C79N spin probe. The low susceptibility of the spin phase memory time (Tm) for the Y2@C79N spin was also found in a confined space.

Extensive application of fullerene nanoparticles (nC60) leads to potential environmental pollution. The acute toxic effects of nC60 have been largely investigated, but studies of behavioural and chronic toxicity at sublethal doses are still rare and the underlying molecular mechanisms remain unknown. The present study investigated behavioural and chronic toxicity of nC60 to Daphnia magna. The results showed that, in response to nC60 exposure, hopping, heartbeat frequencies and feeding ability of D. magna decreased significantly, displaying negative relationship with exposure time and dose. Chronic treatments with 0.1 mg/L or 1 mg/L nC60 for 21 days significantly reduced survival and reproduction of D. magna. These harmful effects suggested negative impacts of nC60 on aquatic ecosystems. Moreover, transcriptome analysis showed that the behavioural and chronic toxicity of nC60 to D. magna might be related to physiological functions such as cell structural repair, protein degradation, energy metabolism and reproduction. We found that nC60 accumulated in guts of D. magna, which should be responsible for the decrease of food ingestion and consequently inhibiting energy intake. Deficiency of energy not only affects behaviours but also declines reproduction in D. magna. Overall, this is the first study comprehensively considered the behavioral and chronic toxicity of nC60 to aquatic organism. The results should be helpful to better understand the ecological consequences of C60 released into water environments.

Introduction and objective: Degradation of the extracellular matrix (ECM) changes the physicochemical properties and dysregulates ECM-cell interactions, leading to several pathological conditions, such as invasive cancer. Carbon nanofilm, as a biocompatible and easy to functionalize material, could be used to mimic ECM structures, changing cancer cell behavior to perform like normal cells. Methods: Experiments were performed in vitro with HS-5 cells (as a control) and HepG2 and C3A cancer cells. An aqueous solution of fullerene C60 was used to form a nanofilm. The morphological properties of cells cultivated on C60 nanofilms were evaluated with light, confocal, electron and atomic force microscopy. The cell viability and proliferation were measured by XTT and BrdU assays. Immunoblotting and flow cytometry were used to evaluate the expression level of proliferating cell nuclear antigen and determine the number of cells in the G2/M phase. Results: All cell lines were spread on C60 nanofilms, showing a high affinity to the nanofilm surface. We found that C60 nanofilm mimicked the niche/ECM of cells, was biocompatible and non-toxic, but the mechanical signal from C60 nanofilm created an environment that affected the cell cycle and reduced cell proliferation. Conclusion: The results indicate that C60 nanofilms might be a suitable, substitute component for the niche of cancer cells. The incorporation of fullerene C60 in the ECM/niche may be an alternative treatment for hepatocellular carcinoma.

Purpose: We have previously reported that some cationic fullerene derivatives exhibited anticancer activity, and they are expected to be a potential lead compound for an anti-drug resistant cancer agent. However, they are bis-adducts and a mixture of multiple regioisomers, which cannot be readily separated due to the variability of substituent positions on the fullerene cage. To overcome this issue, we evaluated the antiproliferative activities of a set of mono-adduct derivatives and examined their structure-activity relationship. In addition, the in vivo antitumor activity of selected derivatives was also examined. Methods: Nineteen pyridinium fullerene derivatives were newly designed and synthesized in this study. Their antiproliferative activities were evaluated using several cancer cell lines including drug-resistant cells. Furthermore, in vivo antitumor activity of several derivatives was investigated in mouse xenograft model of human lung cancer. Results: The derivatives inhibited the proliferation of cancer cell lines, including cisplatin-resistant cells and doxorubicin-resistant cells. It was also shown that compound 10 (10 µM), 13 (10 µM) and cis-14 (10 µM) induced the intracellular oxidative stress. In addition, compound 13 (20 mg/kg) and cis-14 (15 mg/kg) significantly exhibited antitumor activity in mouse xenograft model of human lung cancer. Conclusion: We synthesized a novel set of mono-adduct fullerene derivatives functionalized with pyridinium groups and found that most of them show potent antiproliferative activities against cancer cell lines and some of them show significant antitumor activities in vivo. We propose that these fullerene derivatives serve as the lead compounds for a novel type of antitumor agents.

The toxicity of C60(OH)30, C70(OH)30, and C120O(OH)n fullerenols, prepared by a new original method, has been studied. This method allowed us to obtain high-purity fullerenols and eliminate the risks of synthesis of preparations containing insoluble fractions contaminated with impurities such as fullerenes not completely reacted by hydroxylation. All fullerenols were detected inside cultured cells. The MTT assay as well as the analysis of apoptosis and cell cycle showed that С60(ОН)30 and С70(OH)30 are non-toxic for cultured V79 и HeLa cells at concentrations exceeding physiological levels by an order of magnitude. С120O(OH)n caused low toxicity. Studies in Drosophila melanogaster showed that any preparations used did not result in a decreased lifespan or in behavior abnormalities in flies.

Particulate matter (PM), a widespread air pollutant, consists of a complex mixture of solid and liquid particles suspended in air. Many diseases have been linked to PM exposure, which induces an imbalance in reactive oxygen species (ROS) generated in cells, and might result in skin diseases (such as aging and atopic dermatitis). New techniques involving nanomedicine and nano-delivery systems are being rapidly developed in the medicinal field. Fullerene, a kind of nanomaterial, acts as a super radical scavenger. Lower water solubility levels limit the bio-applications of fullerene. Hence, to improve the water solubility of fullerene, while retaining its radical scavenger functions, a fullerene derivative, fullerenol C60(OH)36, was synthesized, to examine its biofunctions in PM-exposed human keratinocyte (HaCaT) cells. The PM-induced increase in ROS levels and expression of phosphorylated mitogen-activated protein kinase and Akt could be inhibited via fullerenol pre-treatment. Furthermore, the expression of inflammation-related proteins, cyclooxygenase-2, heme oxygenase-1, and prostaglandin E2 was also suppressed. Fullerenol could preserve the impaired state of skin barrier proteins (filaggrin, involucrin, repetin, and loricrin), which was attributable to PM exposure. These results suggest that fullerenol could act against PM-induced cytotoxicity via ROS scavenging and anti-inflammatory mechanisms, and the maintenance of expression of barrier proteins, and is a potential candidate compound for the treatment of skin diseases.

A simple and straightforward synthesis of diporphyrins and pentaporphyrins is reported here. The supramolecular interactions of the new porphyrin derivatives with C60 and PyC60 (a pyridyl [60]fulleropyrrolidine) were evaluated by absorption and fluorescence titrations in toluene. While no measurable modifications of the absorption and fluorescence spectra were observed upon addition of C60 to the porphyrin derivatives, the addition of PyC60 to the corresponding mono-Zn(II) porphyrins resulted in the formation of Zn(porphyrin)-PyC60 coordination complexes and the binding constants were calculated. Results show that the four free-base porphyrin units in pentaporphyrin 6 have a significant contribution in the stabilization of the 6-PyC60 complex. The crystal and molecular features of the pentaporphyrin Zn5 were unveiled using single-crystal X-ray diffraction studies.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (MTB), remains the top fatal infection continuing to threat public health, and the present detection method for MTB is facing great challenges with the global TB burden. In response to this issue, a novel electrochemical DNA biosensor was developed for detecting the IS6110 fragment within MTB. For the first time, the nanohybrid of gold nanoparticles decorated fullerene nanoparticles/nitrogen-doped graphene nanosheet (Au-nano-C60/NGS) directly served as a new signal tag to generate signal response without additional redox molecules and subsequently labeled with signal probes (SPs) to form tracer label to achieve signal amplification. Additionally, a biotin-avidin system was introduced to immobilize abundant capture probes (CPs), further improving the sensitivity of the proposed biosensor. After a typical sandwich hybridization, the proposed electrochemical DNA biosensor was incubated with tetraoctylammonium bromide (TOAB), which was used as a booster to induce the intrinsic redox activity of the tracer label, resulting in a discriminating current response. The proposed electrochemical DNA biosensor shows a broad linear range for MTB determination from 10 fM to 10 nM with a low limit of detection (LOD) of 3 fM. In addition, this proposed biosensor not only distinguishes mismatched DNA sequence, but also differentiates MTB from other pathogenic agents. More importantly, it has been preliminarily applied in clinical detection and displayed excellent ability to identify the PCR products of clinical samples. There is great potential for this developed method to be used in early diagnosis and monitor of TB.

In this protocol, a photoelectrochemical (PEC) biosensor based on methylene blue (MB) sensitized fullerene (C60) was designed for ultrasensitive DNA detection. First, C60 nanoparticles (C60 NPs) as photoactive material was modified on the electrode surface to obtain an initial photocurrent signal. Additionally, the nicking endonuclease (Nb.BvCI enzyme)-assisted signal amplification strategy was implemented to transform limited target to plenty of products, which hybridized with DNA2 to form plentiful DNA duplex for immobilizing numerous MB as the sensitizer, thus accomplishing the effective sensitization toward C60 NPs and achieving an enhanced photocurrent for quantitatively detecting target DNA. This established PEC strategy realized the target DNA detection with the detection limit down to 3.3 fM, which paved a new avenue for DNA determination and exhibited huge application potential for other biomarkers in clinical analysis and disease diagnosis.

Herein, a novel signal-off photoelectrochemical (PEC) aptasensor was proposed for sensitive detection of thrombin on the basis of C60@C3N4 nanocomposites as quencher and Au nanoparticles (depAu) decorated perylene tetracarboxylic acid (PTCA) as sensing platform. Owing to the excellent membrane-forming of PTCA and superior conductivity of depAu, the PTCA between two depAu layers can simply and effectively produce an extremely high initial photocurrent to afford a precondition for sensitive biodetection. Thereafter, the assembly of C60@C3N4 nanocomposites on electrode via typical sandwich reaction enabled the generation of a significantly decreased photocurrent. Here, the C3N4 with high surface area not only provided massive binding sites for C60 immobilization, but also partly competed with PTCA in light absorption for producing a significantly smaller photocurrent in the presence of electron donor ascorbic acid (AA). Additionally, both the C3N4 and C60 have the poor conductivity, which could inhibit the electron transfer to achieve a further decreased photocurrent, effectively improving the sensitivity of proposed biosensor. As a result, the PEC biosensor in a "signal-off" mode showed an extremely low detection limit down to 1.5 fM, providing a sensitive and universal strategy for protein detection.

Environmental waters cover a range of water quality characteristics which could greatly affect the behavior and fate of C60 in the aquatic environment. In this study, the dispersion and stability of C60 in several environmental water matrices during a 70-day extended mixing period were investigated to better understand its environmental behavior and fate in environmental waters. Relatively stable nanoscale aggregates in water (aqu/nC60) could be formed in wastewater influent, while unstable suspensions were obtained in river water, wastewater effluent, seawater, and estuarine water. During the extended mixing under sunlight, oxygen-containing moieties were produced on the surface of the C60 aggregates, independent of the kind of environmental water matrices. Once the mixed system went under quiescent condition, aggregation and sedimentation of aqu/nC60 occurred. However, an extremely short-time disturbance could easily resuspend the C60 aggregates deposited and increase the concentration of aqu/nC60 in the overlying water column. Therefore, the effects of resuspension should be considered when investigating the environmental behavior and fate of C60.

BACKGROUND: Cisplatin (Cis-Pt) is a widely used anticancer drug but its therapeutic efficiency is limited by hemato-, cardio-, hepato-, nephro- and neurotoxicity. Complexation of Cis-Pt with C60 fullerene nanoparticle will allow to enhance the antitumor activity of the drug and to reduce its side toxic effects. AIM: To estimate the antitumor effects of С60-Cis-Pt nanocomplex in Lewis lung carcinoma (LLC) and analyze hematological toxicity in tumor-bearing mice. MATERIALS AND METHODS: Complexation of C60 fullerene and Cis-Pt molecule was studied by computer simulation. С60-Cis-Pt nanocomplex was i.p. injected to LLC-bearing mice in a total dose of 7.5 mg/kg (C60:Cis-Pt as 3.75:3.75 mg/kg). The survival of tumor-bearing mice and the relative reduction of tumor weight was recorded. Blood indices were determined using the Particle Counter PCE-210 automatic hematology analyzer. RESULTS: Computer simulation demonstrated the formation of С60-Cis-Pt nanocomplex in physiological medium and its stability due to the hydrophobic interactions. Treatment with C60-Cis-Pt nanocomplex increased survival time of LLC-bearing mice by 32%, normalized hemoglobin content (up to 100 g/l), erythrocyte and platelet count as compared to the untreated LLC-bearing mice. Tumor weight decreased by 35.5%; the mitotic index of tumor cells decreased by 78%, and apoptotic index increased by 75%. The revealed effects of the C60-Cis-Pt nanocomplex were more pronounced than the effects of Cis-Pt or C60 fullerene alone in equivalent dose. CONCLUSION: Treatment with C60-Cis-Pt nanocomplex prolonged the survival of LLC-bearing mice and reduced anemia in LLC-bearing mice.

In this article, we report the facile synthesis, self-assembly, and characterization of shape amphiphiles (BPOSS-PDI-X) based on isobutyl-functionalized polyhedral oligomeric silsesquioxane (BPOSS), perylene tetracarboxylic diimide (PDI), and (60)fullerene (C60) moieties. Firstly, an asymmetrically functionalized diblock shape amphiphile precursor (BPOSS-PDI-OH) was obtained through the one-pot reaction between perylene-3,4,9,10-tetracarboxylic dianhydride and two different amines, namely BPOSS-NH2 and 3-amino-1-propanol. It was further conjugated with C60-COOH to give a tri-block shape amphiphile (BPOSS-PDI-C60). Their chemical structures were thoroughly characterized by NMR, IR and MALDI-TOF MS spectrometry. In order to gain insights on the structure-property relationship, their self-assembly in gas phase, in solution, and in solid state were characterized using traveling wave ion mobility mass spectrometry (TWIM-MS), UV/Vis absorption, fluorescence emission spectrophotometer, and transmission electron microscopy, respectively. It was found that BPOSS-PDI-OH formed more complicated dimers than BPOSS-PDI-C60. Both samples showed unique aggregation behaviors in solution with increasing concentration, which could be attributed neither to H- nor to J-type and might be related to the discrete dimers. While BPOSS-PDI-C60 could hardly crystalize into ordered structures, BPOSS-PDI-OH could form nanobelt-shaped single crystals, which may hold potential applications in microelectronics.
