Imaging membrane voltage from genetically defined cells offers the unique ability to report spatial and temporal dynamics of electrical signaling at cellular and circuit levels. Here, we present a general approach to engineer electrochromic fluorescence resonance energy transfer (eFRET) genetically encoded voltage indicators (GEVIs) with positive-going fluorescence response to membrane depolarization through rational manipulation of the native proton transport pathway in microbial rhodopsins. We transform the state-of-the-art eFRET GEVI Voltron into Positron, with kinetics and sensitivity equivalent to Voltron but flipped fluorescence signal polarity. We further apply this general approach to GEVIs containing different voltage sensitive rhodopsin domains and various fluorescent dye and fluorescent protein reporters.

The present paper makes a general approach to the ethical concerns that neuroscientific research suggests, especially evaluating its overreaching uses in the criminal process. Therefore, with the objective of regulating an adequate use, a proposal of ethical regulation of this matter is made, which should precede its probative use in the forensic field

Most neuroscientists would agree that for brain research to progress, we have to know which experimental manipulations have no effect as much as we must identify those that do have an effect. The dominant statistical approaches used in neuroscience rely on P values and can establish the latter but not the former. This makes non-significant findings difficult to interpret: do they support the null hypothesis or are they simply not informative? Here we show how Bayesian hypothesis testing can be used in neuroscience studies to establish both whether there is evidence of absence and whether there is absence of evidence. Through simple tutorial-style examples of Bayesian t-tests and ANOVA using the open-source project JASP, this article aims to empower neuroscientists to use this approach to provide compelling and rigorous evidence for the absence of an effect.

Individuals of African ancestry have been starkly underrepresented in the pursuit of personalized medicine for brain illnesses. The African Ancestry Neuroscience Research Initiative will seek to generate much-needed brain gene and protein expression profiles for people of African ancestry.
