Determination of Quercetin and Kaempferol in Platycladus orientalis Carbonisatus by RP - HPLC / 中国药房

ABSTRACT

OBJECTIVE:To determinate the contents of quercetin and kaempferol in Platycladus orientalis carbonisatus by RP - HPLC.METHODS:The determination was performed on Lichrospher C_(18)(250 mm?4.6 mm,5?m) with mobile phase consisted of methanol-0.2%phosphoric acid solution(5545) at a flow rate of 1.0 mL?min~(-1).The column temperature was set at 25℃and the detection wavelength was set at 368 nm.RESULTS:The linear ranges of quercetin and kaempferol were 0.24～1.68?g(r =0.999 9) and 0.052～0.624?g(r = 0.999 8),respectively.The average recovery rates of quercetin and kaempferol were 100.91%(RSD = 0.80%,n = 6)and 99.59%(RSD = 1.07%,n = 6),respectively.CONCLUSION:The method is simple,accurate,reproducible,and applicable for the quality control of Platycladus orientalis carbonisatus.
