Inhibition of tumor growth and inducing autophagy effects of Ophiopogon Japonicus alcohol extract on lung cancer / 中草药

ABSTRACT

Objective:

To evaluate the effect of Ophiopogon japonicus alcohol extract (OJAE) on tumor growth of lung cancer and the regulation on autophagy of lung cancer cells.

Methods:

Lewis tumor-bearing mice model was established to observe the effect of OJAE on tumor growth. The volume of tumor was observed and calculated by Visual Sonics Vevo 2100 Animal Dedicated High-frequency Color Ultrasound. To detect Ki67 and P53 protein expression in lung cancer tissue by Western blotting assay method and immunohistochemical assay method. The induction of OJAE on autophagy of A549 cells was evaluated by scanning electron microscopy, combining with Western blotting assay and RT-PCR methods.

Results:

OJAE had certain inhibitory effect on Lewis lung cancer tumor growth. It also could make the expression of Ki67 significantly reduced and the expression of P53 significantly enhanced. It could induce autophagy.

Conclusion:

OJAE has obvious inhibitory effect against lung cancer, and has the ability to induce autophagy. This study reveals the anti-lung cancer effect of OJAE may be related to its induction on autophagy, thus provides the basis for the research on anti-lung cancer effect of OJAE.
