RESUMO

In eukaryotes, the genome does not emerge in a specific shape but rather as a hierarchial bundle within the nucleus. This multifaceted genome organization consists of multiresolution cellular structures, such as chromosome territories, compartments, and topologically associating domains, which are frequently defined by architecture, design proteins including CTCF and cohesin, and chromatin loops. This review briefly discusses the advances in understanding the basic rules of control, chromatin folding, and functional areas in early embryogenesis. With the use of chromosome capture techniques, the latest advancements in technologies for visualizing chromatin interactions come close to revealing 3D genome formation frameworks with incredible detail throughout all genomic levels, including at single-cell resolution. The possibility of detecting variations in chromatin architecture might open up new opportunities for disease diagnosis and prevention, infertility treatments, therapeutic approaches, desired exploration, and many other application scenarios.

Background and Objectives: Oral lichen planus (OLP) is an autoimmune, mucocutaneous, oral potentially malignant disorder (OPMD), which characteristically manifests with chronic, recalcitrant lesions, with frequent flare-ups and remissions. The precise etiopathogenesis of OLP is still debatable, although it is believed to be a T-cell-mediated disorder of an unidentified antigen. Despite the availability of various treatments, no cure for OLP exists due to its recalcitrant nature and idiopathic etiology. Platelet-rich plasma (PRP) has antioxidant, anti-inflammatory, and immunomodulatory properties, in addition to its regulatory action on keratinocyte differentiation and proliferation. These salient properties substantiate the possible role of PRP in the treatment of OLP. Our systematic review focuses on assessing the therapeutic potential of PRP as a treatment modality in OLP. Materials and Methods: We conducted a detailed literature search for studies assessing PRP as a therapeutic regimen in OLP, using the Google Scholar and PubMed/MEDLINE search engines. The search was limited to studies published from January 2000 to January 2023 and included a combination of Medical Subject Heading (MeSH) terms. ROBVIS analysis was carried out for the assessment of publication bias. Descriptive statistics were performed using Microsoft Excel. Results: This systematic review included five articles that met the inclusion criteria. Most of the included studies demonstrated that PRP treatment considerably ameliorated both objective and subjective symptoms in OLP subjects, with comparable efficacy to the standard corticosteroid treatment. Further, PRP therapy offers the added benefit of minimal adverse effects and recurrences. Conclusion: This systematic review suggests that PRP has significant therapeutic potential for treating OLP. However, further research with larger sample sizes is imperative to corroborate these findings.

The availability and continuous supply of black soldier fly larvae (BSFL) is crucial for efficient operation of a BSF biowaste recycling facility. Its rearing performance was for the first time investigated in Pakistan under outdoor ambient weather conditions. Comparison of the findings with the BSF rearing performance of Indonesia's facility highlights the life stages needing special attention. In Pakistan, mean BSF emergence, hatching and survival rate of 58.8% (SD 15.2), 44.5% (SD 21.8) and 91.4% (SD 1.68) were achieved respectively. A positive significant correlation was found between the number of emerged flies and prepupae (R = 0.75) and the number of eggs produced and hatched (R = 0.92). On average, BSF took 49.5 days (SD 3.20) to complete one life cycle (LC) under ambient temperature and relative humidity (RH) between (22-35 °C) and (24.7-89.3%) respectively. The mean duration of eclosion, pre-oviposition, egg hatching, larval feeding and pupation was 15.6 days (SD 1.6), 3.5 days (SD 0.5), 3 days (SD 0.6), 22 days (SD 2.5), 3.8 days (SD 1.2) respectively. In Pakistan, the LC duration was longer with a smaller number of eggs/fly, lower BSF emergence and hatching rates as compared to Indonesia. BSF tolerated the semiarid weather conditions of Pakistan, successfully developed into all instars and completed all LCs under observation. It is suggested to provide controlled environmental conditions at the nursery stage to improve BSFL rearing performance for sustainable biowaste management.

BACKGROUND: Global efforts toward the development and deployment of a vaccine for COVID-19 are rapidly advancing. To achieve herd immunity, widespread administration of vaccines is required, which necessitates significant cooperation from the general public. As such, it is crucial that governments and public health agencies understand public sentiments toward vaccines, which can help guide educational campaigns and other targeted policy interventions. OBJECTIVE: The aim of this study was to develop and apply an artificial intelligence-based approach to analyze public sentiments on social media in the United Kingdom and the United States toward COVID-19 vaccines to better understand the public attitude and concerns regarding COVID-19 vaccines. METHODS: Over 300,000 social media posts related to COVID-19 vaccines were extracted, including 23,571 Facebook posts from the United Kingdom and 144,864 from the United States, along with 40,268 tweets from the United Kingdom and 98,385 from the United States from March 1 to November 22, 2020. We used natural language processing and deep learning-based techniques to predict average sentiments, sentiment trends, and topics of discussion. These factors were analyzed longitudinally and geospatially, and manual reading of randomly selected posts on points of interest helped identify underlying themes and validated insights from the analysis. RESULTS: Overall averaged positive, negative, and neutral sentiments were at 58%, 22%, and 17% in the United Kingdom, compared to 56%, 24%, and 18% in the United States, respectively. Public optimism over vaccine development, effectiveness, and trials as well as concerns over their safety, economic viability, and corporation control were identified. We compared our findings to those of nationwide surveys in both countries and found them to correlate broadly. CONCLUSIONS: Artificial intelligence-enabled social media analysis should be considered for adoption by institutions and governments alongside surveys and other conventional methods of assessing public attitude. Such analyses could enable real-time assessment, at scale, of public confidence and trust in COVID-19 vaccines, help address the concerns of vaccine sceptics, and help develop more effective policies and communication strategies to maximize uptake.

Arrays of TiO2 nanotubes (TiO2 NTs) with grassy surfaces were observed on titanium foil anodised at 60 V in fluorinated ethylene glycol (EG) with added hydrogen peroxide (H2O2). The grassy surface was generated by the chemical etching and dissolution of the surface of the TiO2 NTs walls, which was accelerated by the temperature increase on the addition of H2O2 . Upon annealing at 600 °C, the grassy part of the TiO2 NTs was found to consist of mostly anatase TiO2 whereas the bottom part of the anodic oxide comprised a mixture of anatase and rutile TiO2. The TiO2 NTs were then used to reduce hexavalent chromium (Cr(VI)) under ultraviolet radiation. They exhibited a rather efficient photocatalytic effect, with 100% removal of Cr(VI) after 30 min of irradiation. The fast removal of Cr(VI) was due to the anatase dominance at the grassy part of the TiO2 NTs as well as the higher surface area the structure may have. This work provides a novel insight into the photocatalytic reduction of Cr(VI) on grassy anatase TiO2 NTs.

Dynamic, in vivo evaluations of knee mechanics are important for understanding knee injury and repair, and developing successful treatments. Computational models have been used with in vivo experiments to quantify joint mechanics, but they are typically not predictive. The current study presents a novel integrated approach with high-speed stereo radiography, musculoskeletal modeling, and finite element (FE) modeling for evaluation of subject-specific, in vivo knee mechanics in a healthy subject performing a seated knee extension and weight-bearing lunge. Whole-body motion capture, ground reaction forces, and radiography-based kinematics were used to drive musculoskeletal and predictive FE models for load-controlled simulation of in vivo knee mechanics. A predictive simulation of knee mechanics was developed in four stages: (1) in vivo measurements of one subject performing a lunge and a seated knee extension, (2) rigid-body musculoskeletal modeling to determine muscle forces, (3) FE simulation of knee extension for knee-ligament calibration, and (4) predictive FE simulation of a lunge. FE models predicted knee contact and ligament mechanics and evaluated the impact of cruciate ligament properties on joint kinematics and loading. Calibrated model kinematics demonstrated good agreement to the experimental motion with root-mean-square differences of tibiofemoral flexion-extension <3 deg, internal-external <4 deg, and anterior-posterior <2 mm. Ligament reference strain and attachment locations were the most critical properties in the calibration process. The current work advances previous in vivo knee modeling through simulation of dynamic activities, modeling of subject-specific knee behavior, and development of a load-controlled knee model.

In recent years, lipid metabolism has garnered significant attention as it provides the necessary building blocks required to sustain tumor growth and serves as an alternative fuel source for ATP generation. Fatty acid synthase (FASN) functions as a central regulator of lipid metabolism and plays a critical role in the growth and survival of tumors with lipogenic phenotypes. Accumulating evidence has shown that it is capable of rewiring tumor cells for greater energy flexibility to attain their high energy requirements. This multi-enzyme protein is capable of modulating the function of subcellular organelles for optimal function under different conditions. Apart from lipid metabolism, FASN has functional roles in other cellular processes such as glycolysis and amino acid metabolism. These pivotal roles of FASN in lipid metabolism make it an attractive target in the clinic with several new inhibitors currently being tested in early clinical trials. This article aims to present the current evidence on the emergence of FASN as a target in human malignancies.

BACKGROUND: Periodontal diseases are prevalent among adult populations. Its diagnosis depends mainly on clinical findings supported by radiographic examinations. In previous decades, cone beam computed tomography has been introduced to the dental field. The aim of this study was to address the diagnostic efficacy of cone-beam computed tomographic (CBCT) imaging in periodontics based on a systematic search and analysis of the literature using the hierarchical efficacy model. METHODS: A systematic search of electronic databases such as PubMed, Scopus, Web of Science, and Cochrane was conducted in February 2019 to identify studies addressing the efficacy of CBCT imaging in Periodontics. The identified studies were subjected to pre-identified inclusion criteria followed by an analysis using a hierarchical model of efficacy (model) designed for an appraisal of the literature on diagnostic imaging modality. Four examiners performed the eligibility and quality assessment of relevant studies and consensus was reached in cases where disagreement occurred. RESULTS: The search resulted in 64 studies. Of these, 34 publications were allocated to the relevant level of efficacy and quality assessments wherever applicable. The overall diagnostic accuracy of the included studies showed a low or moderate risk of bias and applicability concerns in the use of CBCT. In addition, CBCT is accurate in identifying periodontal defects when compared to other modalities. The studies on the level of patient outcomes agreed that CBCT is a reliable tool for the assessment of outcomes after the treatment of periodontal defects. CONCLUSION: CBCT was found to be beneficial and accurate in cases of infra-bony defects and furcation involvements.

BACKGROUND: Patellofemoral complications may cause pain and discomfort, sometimes leading to revision surgery for total knee arthroplasty patients, and patellar implant design has an impact on function of the reconstructed knee. The purpose of this in vivo biomechanics study was to understand the kinematic, functional, strength, and patient-reported outcome data of patients with anatomic and dome patellar implants. METHODS: Satisfactory age-matched, gender-matched, and body mass index-matched patients who underwent rotating-platform total knee arthroplasty from one joint replacement system with either dome (n = 16) or anatomic (n = 16) patellar components were tested in a human motion laboratory using high-speed stereoradiography during an unweighted seated knee extension and a weight-bearing lunge activity. Patellar kinematics, range of motion, strength, and patient-reported outcomes were compared between subjects with anatomic or dome component geometry. RESULTS: Both groups of patients achieved similar functional knee range of motion and reported similar outcomes and satisfaction. On average, patients with the anatomic component had 36% greater extensor strength compared with dome subjects. Patients with anatomic patellar components demonstrated significantly greater flexion of the patella relative to the femur and lower external rotation during the weighted lunge activity. CONCLUSIONS: Relative to the modified dome geometry, patients with anatomic patellar geometry achieved greater patellar flexion which may better replicate normal patellar motion. Patients with anatomic implants may regain more extensor strength compared to patients with dome implants due to geometric differences in the patellar component designs.

Antibiotic resistance in bacterial species is opening new avenues to search for alternative modes of antimicrobial treatment, medicinal plant extracts being one among them. The aim of this study was to access the possibility of medicinal plant extract from Shih in the manufacture of pharmaceutical preparations for oral hygiene specifically for the prevention and treatment of dental caries due to Streptococcus mutans. Antimicrobial effects of crude organic extract of Shih on S. mutans isolated from the saliva were examined by taking S. mutans with culture media only (-ve control); S. mutans treated with the antibiotic gentamicin (+ve control) and S. mutans treated with Shih. Minimal Inhibitory Concentration (MIC) and Minimal Bactericidal Concentration (MBC) were Determination by Iodonitrotetrazolium chloride (INT) colorimetric assay Time-kill dynamic assay was performed using broth microdilution method. The metabolic reason behind the bacteriostatic and bactericidal effect were studied by measuring the glucose utilization by the microbes, pH as a measure of acid production, nucleic acids quantitation to check the DNA status and inhibition of water-insoluble glucan synthesis were undertaken. Shih MIC for S. mutans was at 2.5 mg/ml and MBC was 4 mg/ml. S. mutans bacterial population started reclining within 60 min of incubation with Shih at MBC. Utilization of added glucose was very high at MIC due to bacteria overcoming the stress, whereas at MBC its utilization was less. Accordingly pH also became acidic to 2.9 with MIC and 4.03 with MBC. There was a great degree of inhibition in the formation of nucleic acids indicating this crude extract interferes with DNA replication. Inhibition of glucan synthesis was to the tune of 45% as compared to control. Thus we conclude that Shih has potentially effective antibacterial activity hence it can be proposed as a potentially effective antiplaque and anticariogenic agent in the form of mouth wash or gum paint. However, the cytotoxicity of the extract needs to be evaluated in in-vitro and in-vivo conditions before it is considered as a safe antiplaque and anticariogenic agent.

Research on thermoelectrics has seen a huge resurgence since the early 1990s. The ability of tuning a material's electrical and thermal transport behavior upon nanostructuring has led to this revival. Nevertheless, thermoelectric performances of nanowires and related materials lag far behind those achieved with thin-film superlattices and quantum dot-based materials. This is despite the fact that nanowires offer many distinct advantages in enhancing the thermoelectric performances of materials. The simplicity of the strategy is the first and foremost advantage. For example, control of the nanowire diameters and their surface roughnesses will aid in enhancing their thermoelectric performances. Another major advantage is the possibility of obtaining high thermoelectric performances using simpler nanowire chemistries (e.g., elemental and binary compound semiconductors), paving the way for the fabrication of thermoelectric modules inexpensively from non-toxic elements. In this context, the topical review provides an overview of the current state of nanowire-based thermoelectrics. It concludes with a discussion of the future vision of nanowire-based thermoelectrics, including the need for developing strategies aimed at the mass production of nanowires and their interface-engineered assembly into devices. This eliminates the need for trial-and-error strategies and complex chemistries for enhancing the thermoelectric performances of materials.

Modeling complex knee biomechanics is a continual challenge, which has resulted in many models of varying levels of quality, complexity, and validation. Beyond modeling healthy knees, accurately mimicking pathologic knee mechanics, such as after cruciate rupture or meniscectomy, is difficult. Experimental tests of knee laxity can provide important information about ligament engagement and overall contributions to knee stability for development of subject-specific models to accurately simulate knee motion and loading. Our objective was to provide combined experimental tests and finite-element (FE) models of natural knee laxity that are subject-specific, have one-to-one experiment to model calibration, simulate ligament engagement in agreement with literature, and are adaptable for a variety of biomechanical investigations (e.g., cartilage contact, ligament strain, in vivo kinematics). Calibration involved perturbing ligament stiffness, initial ligament strain, and attachment location until model-predicted kinematics and ligament engagement matched experimental reports. Errors between model-predicted and experimental kinematics averaged <2 deg during varus-valgus (VV) rotations, <6 deg during internal-external (IE) rotations, and <3 mm of translation during anterior-posterior (AP) displacements. Engagement of the individual ligaments agreed with literature descriptions. These results demonstrate the ability of our constraint models to be customized for multiple individuals and simultaneously call attention to the need to verify that ligament engagement is in good general agreement with literature. To facilitate further investigations of subject-specific or population based knee joint biomechanics, data collected during the experimental and modeling phases of this study are available for download by the research community.

Kaptai Lake, the largest artificial reservoir in Southeast Asia, is home to a diverse fish fauna that supports thousands of livelihoods and is distinguished by multi-species and multi-gear fisheries. In Kaptai Lake, the gear-based catch composition, catch rate and distribution pattern are little known. From August 2020 to April 2021, a nine-month study was conducted in five upazilas using direct catch assessment surveys and fishing effort surveys from four fishing gears, namely seine nets, gill nets, lift nets, and push nets. A total of 49 morpho-species from 22 families were found, with three species from the Clupeidae accounting for 93.63 % of the catch in all gear combined. The total catch composition and CPUE were higher in seine nets (75.07 %, 13.86 ± 1.8 kg/gear/trip respectively) and lower in lift nets (4.97 %, 1.01 ± 0.21 kg/gear/trip) and showed significant differences among gears, except sampling sites whereas CPUE was higher in Naniarchar for seine nets (17.29 ± 8.89 kg/gear/trip) and lower in Langadu for lift nets (0.62 ± 0.25 kg/gear/trip). Seine nets captured more species, and the number of species increased significantly as CPUE increased. Our study assessed four gears that targeted different fish species with little overlap in leading species; seine nets and gill nets primarily targeted Clupeidae (96.53 % and 41.69 %, respectively), whereas lift nets and push nets primarily targeted Cyprinidae and Palaemonidae (38.93 % and 99.37 % respectively). The observed abundance and variety of fish species captured in gill nets suggest a significant overlap in the selectivity of this fishing method with that of lift nets. Due to the varying contributions of sites and gears, the nMDS ordination pattern reveals a weak spatial variation in catch composition. According to the SIMPER results, Bagridae, Gobiidae, and Ambassidae were the most significant contributors to site grouping patterns across all gears. Furthermore, the findings indicate that the catch composition does not follow the typical pattern of spatial variation. By implementing measures to eliminate or decrease the usage of small mesh nets, there is expected to be a corresponding decrease in the capture of small fish. Additionally, this action will help mitigate the issue of overlapping selectivity among the current fishing gears. Our findings provide baseline data on the potential efficacy of gear limitation and suggest a gear-based management strategy.

BACKGROUND: Artificial intelligence (AI) holds significant promise for transforming healthcare delivery, including dentistry. However, the successful integration of AI into dental practice necessitates an understanding of dental professionals' perspectives, attitudes, and readiness to adopt AI technology. This study aimed to explore dental professionals' perceptions, attitudes, and practices regarding AI adoption in dentistry. METHODS: This cross-sectional study was conducted among 256 dental professionals using an online questionnaire. Participants were assessed for familiarity with AI technology, perceived barriers to adoption, attitudes towards AI, current usage patterns, and factors influencing adoption decisions. Data are analysed using descriptive statistics, including frequencies, percentages, means, and standard deviations. Inferential statistics, such as chi-square tests and regression analysis, were employed to examine associations between variables and identify predictors of AI adoption in dentistry. RESULTS: The study surveyed 256 dental professionals from various regions across India, primarily aged 30 to 50 years (mean age: 42.6), with a nearly equal gender split (male: 48.4%, female: 51.6%) and high educational attainment (67.8% with master's or doctoral degrees). Private practices were predominant (56.3%). The diagnostic algorithms and treatment planning software were well known (77.3% and 70.3% familiarity, respectively). Technical concerns (average score: 3.82 ± 0.68) were the main barriers to AI adoption, followed by financial considerations (average score: 3.45 ± 0.72), ethical and legal issues (average score: 3.21 ± 0.65), and organizational factors (average score: 3.67 ± 0.71). Despite these concerns, most participants had positive attitudes towards AI (70.3% agreed). Current usage varied, with diagnostic support and administrative tasks being the most common (44.5% and 82.8% usage, respectively). Perceived utility (average score: 4.12 ± 0.75) and ease of use (average score: 3.98 ± 0.69) significantly influenced adoption, as identified by regression analysis (perceived utility: ß = 0.342, p < 0.001; ease of use: ß = 0.267, p = 0.005). CONCLUSION: This study provides valuable insights into AI adoption in dentistry, highlighting the multifaceted nature of barriers and facilitators that influence dental professionals' adoption decisions. Strategies to promote AI adoption should address practical considerations, ethical concerns, and educational needs to facilitate the integration of AI technology into dental practices.

The term "artificial intelligence" (AI) has become increasingly prevalent, but it is essential to provide clarity before proceeding. AI pertains to the theory and creation of systems capable of executing tasks that typically necessitate human intelligence. Integrating artificial intelligence (AI) in pediatric dentistry has emerged as a promising avenue to enhance patient care, improve diagnostic accuracy, streamline treatment planning, and augment patient engagement. AI-driven tools such as image analysis, natural language processing, and machine learning algorithms assist in early caries detection, orthodontic treatment planning, behavior management, and personalized oral hygiene education for pediatric patients. This paper presents an overview of AI's applications in pediatric dentistry, particularly behavior management, highlighting its potential to revolutionize traditional pediatric dental practices.

Lung cancer is the leading cause of cancer deaths. Lethal pulmonary adenocarcinomas (ADC) present with frequent mutations in the EGFR. Genetically engineered murine models of lung cancer expedited comprehension of the molecular mechanisms driving tumorigenesis and drug response. Here, we systematically analyzed the evolution of tumor heterogeneity in the context of dynamic interactions occurring with the intermingled tumor microenvironment (TME) by high-resolution transcriptomics. Our effort identified vulnerable tumor-specific epithelial cells, as well as their cross-talk with niche components (endothelial cells, fibroblasts, and tumor-infiltrating immune cells), whose symbiotic interface shapes tumor aggressiveness and is almost completely abolished by treatment with Unesbulin, a tubulin binding agent that reduces B cell-specific Moloney murine leukemia virus integration site 1 (BMI-1) activity. Simultaneous magnetic resonance imaging (MRI) analysis demonstrated decreased tumor growth, setting the stage for future investigations into the potential of novel therapeutic strategies for EGFR-mutant ADCs. SIGNIFICANCE: Targeting the TME is an attractive strategy for treatment of solid tumors. Here we revealed how EGFR-mutant landscapes are affected at the single-cell resolution level during Unesbulin treatment. This novel drug, by targeting cancer cells and their interactions with crucial TME components, could be envisioned for future therapeutic advancements.

Medical and dental professionals should continue to adhere to preventive measures after COVID-19 vaccination due to their increased risk of exposure to the virus, particularly as new variants emerge that may heighten their risk perception and susceptibility. Therefore, this study aimed to explore the effects of COVID-19 vaccination on complacency to adherence to COVID-19 preventive behavioral measures and mental health among medical and dental professionals. In this cross-sectional study 410 medical and dental professionals were recruited from different medical and dental hospitals in Islamabad, Pakistan. The data was collected using a valid and reliable questionnaire comprising of three sections (socio-demographic, information of preventive behaviors performance against COVID-19 after vaccination, mental health status). A chi-square test and ordinal logistic regression were used for analysis. Post COVID-19 vaccination there was decrease in the frequency of use of hand washing, sanitizers (70.2%), and social distancing (60.5%), however greeting with a handshake (58.8%) and use of public transport (45.9%) seen upward trend among participants. Only face mask usage post-vaccination was statistically significant (p < .05) in association with age, marital status, and years of working Experience. The greatest decrease in the usage of masks after COVID-19 vaccination was seen in age group of 10-30 (41.7%) and working experience group of 0-5 years (39.7%). All the preventive behaviors are statistically significant (p < .05) associated with the mental status of the participants except online shopping and use of public transport. These results indicate the presence of vaccination-induced complacency in adherence to COVID-19 preventive behavioral measures among healthcare professionals.

OBJECTIVE: The aim of this study was to assess the incidence of micronuclei (MN) in buccal mucosal cells of professionals working in radiology area to determine the risk of stochastic effects of radiation. MATERIALS AND METHODS: All the professionals and students working in King Khalid University - College of Dentistry radiology area were included in the Risk Group (RG = 27). The Control Group (CG = 27) comprised of healthy individual matching the gender and age of the RG. Buccal mucosal scraping from all the 54 subjects of RG and CG were stained with Papanicolaou stain and observed under oil immersion lens (×100) for the presence of micronuclei (MN) in the exfoliated epithelial cells. RESULTS: There was no significant difference between the incidence of MN in RG and CG (p = >0.05) using t-test. CONCLUSION: Routine radiation protection protocol does minimize the risk of radiation induced cytotoxicity, however, screening of professionals should be carried out at regular intervals.
