RESUMO

Reduced graphene oxide (rGO) is a derivative of graphene, which has been widely used as the conductive pigment of many water-based inks and is recognized as one of the most promising graphene-based materials for large-scale and low-cost production processes. In this work, we evaluate a custom functionalised reduced graphene oxide ink (f-rGO) via inkjet-printing technology. Test line structures were designed and fabricated by the inkjet printing process using the f-rGO ink on a pretreated polyimide substrate. For the electrical characterisation of these devices, two-point (2P) and four-point (4P) probe measurements were implemented. The results showed a major effect of the number of printed passes on the resulting resistance for all ink concentrations in both 2P and 4P cases. Interesting results can be extracted by comparing the obtained multipass resistance values that results to similar effective concentration with less passes. These measurements can provide the ground to grasp the variation in resistance values due to the different ink concentrations, and printing passes and can provide a useful guide in achieving specific resistance values with adequate precision. Accompanying topography measurements have been conducted with white-light interferometry. Furthermore, thermal characterisation was carried out to evaluate the operation of the devices as temperature sensors and heaters. It has been found that ink concentration and printing passes directly influence the performance of both the temperature sensors and heaters.

Tay-Sachs and Sandhoff diseases are lethal inborn errors of acid ß-N-acetylhexosaminidase activity, characterized by lysosomal storage of GM2 ganglioside and related glycoconjugates in the nervous system. The molecular events that lead to irreversible neuronal injury accompanied by gliosis are unknown; but gene transfer, when undertaken before neurological signs are manifest, effectively rescues the acute neurodegenerative illness in Hexb-/- (Sandhoff) mice that lack ß-hexosaminidases A and B. To define determinants of therapeutic efficacy and establish a dynamic experimental platform to systematically investigate cellular pathogenesis of GM2 gangliosidosis, we generated two inducible experimental models. Reversible transgenic expression of ß-hexosaminidase directed by two promoters, mouse Hexb and human Synapsin 1 promoters, permitted progression of GM2 gangliosidosis in Sandhoff mice to be modified at pre-defined ages. A single auto-regulatory tetracycline-sensitive expression cassette controlled expression of transgenic Hexb in the brain of Hexb-/- mice and provided long-term rescue from the acute neuronopathic disorder, as well as the accompanying pathological storage of glycoconjugates and gliosis in most parts of the brain. Ultimately, late-onset brainstem and ventral spinal cord pathology occurred and was associated with increased tone in the limbs. Silencing transgenic Hexb expression in five-week-old mice induced stereotypic signs and progression of Sandhoff disease, including tremor, bradykinesia, and hind-limb paralysis. As in germline Hexb-/- mice, these neurodegenerative manifestations advanced rapidly, indicating that the pathogenesis and progression of GM2 gangliosidosis is not influenced by developmental events in the maturing nervous system.

Ammonia (NH3) toxicity, stemming from nitrification, can adversely affect aquatic life and influence the taste and odor of drinking water. This underscores the necessity for highly responsive and accurate sensors to continuously monitor NH3 levels in water, especially in complex environments, where reliable sensors have been lacking until this point. Herein, we detail the development of a sensor comprising a compact and selective analyzer with low gas consumption and a timely response based on photoacoustic spectroscopy. This, combined with an automated liquid sampling system, enables the precise detection of ammonia traces in water. The sensor system incorporates a state-of-the art quantum cascade laser as the excitation source emitting at 9 µm in resonance with the absorption line of NH3 located at 1103.46 cm-1. Our instrument demonstrated detection sensitivity at a low ppm level for the ammonia molecule with response times of less than 60 s. For the sampling system, an ammonia stripping solution was designed, resulting in a prompt full measurement cycle (6.35 min). A further evaluation of the sensor within a pilot study showed good reliability and agreement with the reference method for real water samples, confirming the potential of our NH3 analyzer for water quality monitoring applications.

Recent studies have predicted a strong increase in high harmonic emission in unbiased semiconductor superlattices due to asymmetric current flow. In parallel, an external static bias has led to orders of magnitude control of high harmonics. Here, we study how this control can affect the operation of superlattice multipliers in a range of input frequencies and powers delivered by commercially available GHz sources. We show that the strongly nonlinear behavior can lead to a very complex scenario. Furthermore, it is natural to ask what happens when we combine both asymmetry and voltage control effects. This question is answered by the simulations presented in this study. The efficiency of high-order even harmonics is increased by the combined effects. Furthermore, the development of 'petals' in high-order emission is shown to be more easily achieved, opening the possibility to very interesting fundamental physics studies and more efficient devices for the GHz-THz range.

The phosphatidylinositol 3-kinase Vps34 is part of several protein complexes. The structural organization of heterotetrameric complexes is starting to emerge, but little is known about organization of additional accessory subunits that interact with these assemblies. Combining hydrogen-deuterium exchange mass spectrometry (HDX-MS), X-ray crystallography and electron microscopy (EM), we have characterized Atg38 and its human ortholog NRBF2, accessory components of complex I consisting of Vps15-Vps34-Vps30/Atg6-Atg14 (yeast) and PIK3R4/VPS15-PIK3C3/VPS34-BECN1/Beclin 1-ATG14 (human). HDX-MS shows that Atg38 binds the Vps30-Atg14 subcomplex of complex I, using mainly its N-terminal MIT domain and bridges the coiled-coil I regions of Atg14 and Vps30 in the base of complex I. The Atg38 C-terminal domain is important for localization to the phagophore assembly site (PAS) and homodimerization. Our 2.2 Å resolution crystal structure of the Atg38 C-terminal homodimerization domain shows 2 segments of α-helices assembling into a mushroom-like asymmetric homodimer with a 4-helix cap and a parallel coiled-coil stalk. One Atg38 homodimer engages a single complex I. This is in sharp contrast to human NRBF2, which also forms a homodimer, but this homodimer can bridge 2 complex I assemblies.
