RESUMO

Modern food systems facilitate rapid dispersal of pests and pathogens through multiple pathways. The complexity of spread dynamics and data inadequacy make it challenging to model the phenomenon and also to prepare for emerging invasions. We present a generic framework to study the spatio-temporal spread of invasive species as a multi-scale propagation process over a time-varying network accounting for climate, biology, seasonal production, trade and demographic information. Machine learning techniques are used in a novel manner to capture model variability and analyse parameter sensitivity. We applied the framework to understand the spread of a devastating pest of tomato, Tuta absoluta, in South and Southeast Asia, a region at the frontier of its current range. Analysis with respect to historical invasion records suggests that even with modest self-mediated spread capabilities, the pest can quickly expand its range through domestic city-to-city vegetable trade. Our models forecast that within 5-7 years, Tuta absoluta will invade all major vegetable growing areas of mainland Southeast Asia assuming unmitigated spread. Monitoring high-consumption areas can help in early detection, and targeted interventions at major production areas can effectively reduce the rate of spread.

The tomato leafminer, Tuta absoluta, is a major pest of tomato crops worldwide. This study surveyed the resistance of T. absoluta populations from four regions in Brazil to pyrethroid insecticides, the frequencies of L1014F, T929I and M918T Na channel mutations, and the role of detoxification metabolism in the resistance. Resistance ratios varied from 1- to 11-times among populations and insecticides, but control failure likelihood assays showed that all pyrethroids assessed exhibited no efficacy at all (and thus, 98-100% control failure likelihood) against all T. absoluta populations. The activity of glutathione S-transferase and cytochrome P450-mediated N-demethylation in biochemical assays was significantly correlated with the level of resistance to deltamethrin and permethrin suggesting that these enzymes may play a role in resistance. TaqMan assays were used to screen for the presence of knockdown resistance (kdr) mutations and revealed that the L1014F kdr mutation was fixed in all populations and associated with two super-kdr mutations, M918T and particularly T929I, at high frequency. Altogether, results suggest that control failures are because of mutations in the domain II of the sodium channel, as a prevailing mechanism of resistance to pyrethroids in populations of T. absoluta in Brazil. But, enhanced cytochrome P450-dependent monooxygenases and GST activities also play an important role in the resistance of some populations, which reinforce that pyrethroids must not be used overall to control T. absoluta.

Tomato pinworm, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) is the major pest of tomato crops in Pakistan. Insecticides are commonly used for the management of this insect-pest. To develop a better insecticide resistance management strategy and evaluate the risk of resistance evolution, a field collected population of the tomato pinworm was selected with flubendiamide in the laboratory. We investigated the genetics of flubendiamide resistance and concentration-mortality response to other insecticides by selecting a field strain of tomato pinworm with commercial flubendiamide formulation. Tuta absoluta was reciprocally crossed with resistant strain (Fluben-R) and was selected up to 13 generations, while F1 progeny was back-crossed with resistant parent (Fluben-R). The results of LC50 and Resistance Ratio (RR) demonstrated a higher resistance developed in field and laboratory-selected strains (G2 and G13, respectively). Field-collected and laboratory-selected (Fluben-R) strains demonstrated higher intensity of concentration-mortality response against chlorantraniliprole, thiamethoxam, permethrin, abamectin and tebufenozide compared to susceptible ones. Based on the overlapping of 95% FL, it demonstrated significant differences, revealing that it was not sex linked (autosomal) with no maternal effects. The backcross analysis of the F1´ resistant parent resulting in significant differences at all concentrations suggests that resistance is controlled by more than one factor; the null hypothesis was rejected and inheritance was under polygenic control. Resistance progression from 38 to 550 folds demonstrated that T. absoluta can develop a higher level of resistance to flubendiamide. Concentration-mortality response experiments demonstrated that the LC50 of some tested insecticides was higher for field-collected and laboratory-selected strains, suggesting that resistance mechanisms should be studied at a molecular level for better understanding. These results could be helpful to design resistance management strategies against the tomato pinworm.

BACKGROUND: Bemisia tabaci (Hemiptera: Aleyrodidae) represents one of the greatest threats to agricultural crops. Chemical control is the primary tool used in integrated pest management (IPM) programs. However, release of the predator Nesidiocoris tenuis (Hemiptera: Miridae) on tomato plants is a highly recommended control tactic. The objective of this study was to evaluate the efficacy of a commercial borax plus citrus oil (BCO) product against B. tabaci in the presence and absence of N. tenuis. The synthetic insecticide lambda-cyhalothrin was used as a positive control. We also evaluated the sublethal effects of BCO on the behavior and predation rate of N. tenuis. RESULTS: Our results demonstrated that BCO, alone and at its maximum recommended field rate for B. tabaci, was not effective in controlling the pest under laboratory conditions. Application of BCO simultaneous with N. tenuis release did not reduce the increase in the B. tabaci population. Effective control of B. tabaci was achieved using only N. tenuis. However, synthetic lambda-cyhalothrin pyrethroid, used here as a control, caused high pest mortality and led to on-site extinction of N. tenuis, which did not occur for insects exposed to BCO. Lambda-cyhalothrin and BCO significantly affected the foraging behavior of N. tenuis, reducing the predation rate, especially following exposure to lambda-cyhalothrin. CONCLUSION: The insecticide lambda-cyhalothrin achieved satisfactory results in suppressing B. tabaci, but was harmful to N. tenuis. Additionally, lambda-cyhalothrin and BCO affected predator behavior. © 2020 Society of Chemical Industry.

This work aimed at identifying plant compounds with insecticidal activity against Diaphania hyalinata (L.) (Lepidoptera: Pyralidae), Musca domestica (L.) (Diptera: Muscidae), Periplaneta americana (L.) (Blattodea: Blattidae) and Rhyzopertha dominica (F.) (Coleoptera: Bostrichidae). The plant species used were: basil (Ocimum selloi Benth.), rue (Ruta graveolens L.), lion's ear (Leonotis nepetaefolia L.), Jimson weed (Datura stramonium L.), 'baleeira' herb (Cordia verbenaceae L.), mint (Mentha piperita L.), wild balsam apple (Mormodica charantia L.) and billy goat weed (Ageratum conyzoides L.). Firstly, the insecticidal activities of hexane and ethanol plant extracts were evaluated against adults of R. dominica. Among them, only the hexane extract of A. conyzoides showed insecticidal activity. The hexane extract of this plant species was therefore fractionated by silica gel column chromatography to isolate and purify its bioactive chemical constituents. Three compounds were identified using IR spectra, (1)H NMR, (13)C NMR, HMBC and NOE after gel chromatography: 5,6,7,8,3', 4', 5'-heptamethoxyflavone, 5,6,7,8,3'-pentamethoxy-4', 5'-methylenedioxyflavone and coumarin. The complete assignment of (13)C NMR to 5,6,7,8,3'-pentamethoxy-4', 5'-methylenedioxyflavone was successfully made for the first time. 5,6,7,8,3'-Pentamethoxy-4', 5'-methylenedioxyflavone did not show any insecticidal activity against the four insect species tested. 5,6,7,8,3', 4', 5'-Heptamethoxyflavone showed low activity against D. hyalinata and R. dominica and was not toxic to M. domestica or P. americana. In contrast, coumarin showed insecticidal activity against all four insect pest species tested, with the following order of susceptibility: R. dominica < P. americana < D. hyalinata < M. domestica after 24 h exposure.

BACKGROUND: Phthalic and anthranilic diamides comprise a new insecticide class recently registered in Brazil to control Lepidoptera such as Tuta absoluta (Meyrick). Therefore, the baseline of susceptibility was determined for eight representative field populations of this species to establish a resistance monitoring programme. The potential for cross-resistance as well as detoxification metabolism was assessed in order to fine-tune the resistance management programme. RESULTS: Brazilian populations were very susceptible to chlorantraniliprole (LC50 values varied from 3.17 to 29.64 µg AI L(-1) ), cyantraniliprole (LC50 values varied from 8.61 to 28.95 µg AI L(-1) ) and flubendiamide (LC50 values varied from 94 to 230 µg AI L(-1) ), with respective resistance ratios of 9.33-, 3.36- and 2.45-fold between most susceptible and tolerant populations. Anthranilic diamides showed significant correlations between log LC50 values among themselves, suggesting a high risk of cross-resistance. However, the log LC50 values of T. absoluta to phthalic diamide did not show any correlation with anthranilic diamides. Cytochrome- P450-dependent monooxygenase activity showed a weak correlation with log LC50 values of T. absoluta populations to anthranilic diamides, which suggests a potential route for evolving resistance to anthranilic diamides. CONCLUSION: The diamides were highly effective against T. absoluta, with populations showing a homogeneous response to them. Cross-resistance is very likely between anthranilic diamides in T. absoluta. Populations of this pest may evolve resistance by increasing cytochrome- P450-dependent monooxygenases.

The introduction of an agricultural pest species into a new environment is a potential threat to agroecosystems of the invaded area. The phytosanitary concern is even greater if the introduced pest's phenotype expresses traits that will impair the management of that species. The invasive tomato borer, Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae), is one such species and the characterization of the insecticide resistance prevailing in the area of origin is important to guide management efforts in new areas of introduction. The spinosad is one the main insecticides currently used in Brazil for control of the tomato borer; Brazil is the likely source of the introduction of the tomato borer into Europe. For this reason, spinosad resistance in Brazilian populations of this species was characterized. Spinosad resistance has been reported in Brazilian field populations of this pest species, and one resistant population that was used in this study was subjected to an additional seven generations of selection for spinosad resistance reaching levels over 180,000-fold. Inheritance studies indicated that spinosad resistance is monogenic, incompletely recessive and autosomal with high heritability (h(2) = 0.71). Spinosad resistance was unstable without selection pressure with a negative rate of change in the resistance level ( = -0.51) indicating an associated adaptive cost. Esterases and cytochrome P450-dependent monooxygenases titration decreased with spinosad selection, indicating that these detoxification enzymes are not the underlying resistance mechanism. Furthermore, the cross-resistance spectrum was restricted to the insecticide spinetoram, another spinosyn, suggesting that altered target site may be the mechanism involved. Therefore, the suspension of spinosyn use against the tomato borer would be a useful component in spinosad resistance management for this species. Spinosad use against this species in introduced areas should be carefully monitored to prevent rapid selection of high levels of resistance and the potential for its spread to new areas.
