RESUMO

Evaluating the effectiveness of protected area management to help improve management outcomes is becoming an increasingly common practice. The evaluation tools developed and implemented in over 100 countries generally rely on the expert judgements of protected area managers. Despite the growing collection and use of management effectiveness evaluation data, there have been no previous attempts to measure the accuracy of these data. We measured the accuracy of managers' judgements about the conditions in their reserves by collecting independent field data. We also assessed how accurately the evaluation tool reflected managers' views by conducting semi-structured interviews with 23 protected area managers from New South Wales, Australia. We found that managers made highly accurate judgements of the extent of a common weed species, Rubus fruticosus (blackberry), but often misinterpreted the scope, scale and timeframe of the evaluation. These framing effects can lead to error being introduced into the evaluation dataset, affecting the precision of evaluations such that they cannot be reliably compared among reserves. We suggest that the wording of evaluation questions needs to be explicit about the assessment frame to minimize the influence of framing effects on management effectiveness evaluations.

Conservation practice reportedly suffers from low use of technical information. Understanding of factors that affect the influence of technical information on management decision-making is limited. We sought to identify leverage points for improved technical information dissemination in the New South Wales Parks and Wildlife Service, Australia, given the significant recent investments in monitoring and evaluation that had been made. We did so by exploring the inter-relationships between factors affecting the influence of different information types on management decisions. Results indicate that managers have a high inclination toward adaptive behavior, given they operate in an information poor environment. The most influential types of information were those that enabled interaction between information provider and recipient (e.g., staff experience and expertise). An analysis of the concordance in individuals' responses for different information types showed that neither accessibility nor organizational expectation of use was aligned with influence on decision-making. Alignment of responses also varied by work area. Raising expectations of information use or increasing access to particular types of information is therefore unlikely to result in an increase in influence on management decision-making. Rather than focussing on matching accessibility and expected use of particular information types, our results indicate that technical information uptake is best supported through existing peer networks tailored to specific work areas.

Protected area managers often fail to use empirical evidence for their management decisions, yet it is unclear whether this arises from a lack of available data, difficulty in interpreting scientific information for management application, or because managers do not value science for their decisions. To better understand the use of evidence for management decisions, we asked protected area managers in Australia what information is important when making decisions, the types of evidence they find most valuable, and the types of evidence they have for their protected areas. Managers described a complex array of information needed for management decisions, with nine different factors representing decisions about individual management issues and how to prioritize management actions. While managers reported less access to empirical evidence than other sources, this is not because they do not value it, reporting it to be the most valuable source of evidence. Instead, they make up the shortfall in empirical evidence with experience and information synthesized from multiple lines of evidence, which can provide important context for their decisions. We conclude that managers value a diversity of evidence because they face complex conservation decisions. Therefore, while empirical evidence can play an important role, alone this cannot provide all the knowledge managers need.

Interleukin (IL)-18 induces interferon (IFN)-gamma synthesis and synergizes with IL-12 in T helper type 1 (Th1) but not Th2 cell development. We report here that IL-18 receptor (IL-18R) is selectively expressed on murine Th1 but not Th2 cells. IL-18R mRNA was expressed constitutively and consistently in long-term cultured clones, as well as on newly polarized Th1 but not Th2 cells. IL-18 sustained the expression of IL-12Rbeta2 mRNA, indicating that IL-18R transmits signals that maintain Th1 development through the IL-12R complex. In turn, IL-12 upregulated IL-18R mRNA. Antibody against an IL-18R-derived peptide bound Th1 but not Th2 clones. It also labeled polarized Th1 but not Th2 cells derived from naive ovalbumin-T cell antigen receptor-alphabeta transgenic mice (D011.10). Anti-IL-18R antibody inhibited IL-18- induced IFN-gamma production by Th1 clones in vitro. In vivo, anti-IL-18R antibody reduced local inflammation and lipopolysaccharide-induced mortality in mice. This was accompanied by shifting the balance from Th1 to Th2 responses, manifest as decreased IFN-gamma and proinflammatory cytokine production and increased IL-4 and IL-5 synthesis. Therefore, these data provide a direct mechanism for the selective effect of IL-18 on Th1 but not Th2 cells. They also show that the synergistic effect of IL-12 and IL-18 on Th1 development may be due to the reciprocal upregulation of their receptors. Furthermore, IL-18R is a cell surface marker distinguishing Th1 from Th2 cells and may be a therapeutic target.

Management effectiveness evaluation has been recognized as an important mechanism for both reporting on and improving protected area management. The Convention on Biological Diversity's program of work on protected areas calls on all countries to implement such systems. In 2004, the first whole of system assessment of park management effectiveness, based on the IUCN-WCPA Management Effectiveness Evaluation Framework, was undertaken in New South Wales, Australia as part of a State of the Parks reporting requirement. This article describes the development of the State of the Parks assessment tool, its elements, and how it addresses the management effectiveness difficulties associated with assessments conducted across an extensive and diverse range of park types. The importance of engaging staff, at all levels, throughout the process is highlighted, as well as the adjustments made to the assessment tool based on staff feedback. While some results are presented, the main purpose of the article is to identify and discuss important procedural and methodological considerations. These include balancing quantitative and qualitative assessment approaches, achieving a comprehensive understanding of the management processes, and responding to any problems associated with assessments.

Interleukin-12 is a cytokine that plays a central role in mediating cell mediated immunity via enhancement of a TH1 cell response. IL-12, unusually for a cytokine, has a heterodimeric structure made up of 35 kDa and 40 kDa subunits. The aim of this study was to produce and characterize monoclonal antibodies to recombinant human IL-12. Twenty-two monoclonal antibodies to IL-12 were successfully produced and subunit specificity determined using recombinant human IL-12 and chimeric murine/human IL-12. All antibodies were shown to react with the p40 subunit by ELISA and immunoblotting with three of the MAbs being found to cross-react with murine IL-12. Using two individual bioassays for IL-12, seven of the MAbs were shown to neutralize biological activity of IL-12. Ten of the antibodies were found to be of use in immunocytochemistry, reacting with LPS-stimulated peripheral blood monocytes. The approaches and difficulties encountered in characterizing antibodies to a heterodimeric cytokine are discussed together with possible reasons for the failure to generate antibodies to the p35 subunit.

Runoff and sewage discharge from land developments can cause significant changes in water quality of coastal waters, resulting in coral degradation. Coastal waters around Phuket, Thailand are influenced by numerous sewage outfalls associated with rapid tourism development. Water quality and biological monitoring around the Phuket region was undertaken to quantify water quality and biotic characteristics at various distances from sewage outfalls. The surveys revealed strong gradients in water quality and biotic characteristics associated with tourism concentration levels as well as seasonal variability. Water and reef quality tended to decrease with increasing tourist intensity, but improved with increasing distance from sewage discharge within each of the three study locations. In addition, the effect of wastewater discharge was not localised around the source of pollution, but appeared to be transported to non-developed sites by currents, and exacerbated in the wet season.

Against a backdrop of rising sea temperatures and ocean acidification which pose global threats to coral reefs, excess nutrients and turbidity continue to be significant stressors at regional and local scales. Because interventions usually require local data on pollution impacts, we measured ecological responses to sewage discharges in Surin Marine Park, Thailand. Wastewater disposal significantly increased inorganic nutrients and turbidity levels, and this degradation in water quality resulted in substantial ecological shifts in the form of (i) increased macroalgal density and species richness, (ii) lower cover of hard corals, and (iii) significant declines in fish abundance. Thus, the effects of nutrient pollution and turbidity can cascade across several levels of ecological organization to change key properties of the benthos and fish on coral reefs. Maintenance or restoration of ecological reef health requires improved wastewater management and run-off control for reefs to deliver their valuable ecosystems services.

Increased water-dependent development and utilization have led to significant environmental and hydrological degradation of the Tarim River in western China and its dependent ecosystems. Between the 1950s and 1970s, 350 km of the lower reaches were drained and between 1960 and 1980 the water-table fell from between -2 and -3 m to between -8 and -10 m. Subsequently, riparian ecosystems were seriously degraded. In 2000, the Chinese government launched a program to restore the lower reaches of the river. Four environmental flows of 1034 x 10(6) m(3) were released from 2000 to 2002. This paper interprets and discusses the ecological significance of changes following the releases and identifies the relationship between water-table dynamics and vegetation responses. Short-term objectives for river restoration are proposed with possible monitoring parameters suggested.

In China's west since the 1950s large-scale ecosystem degeneration has occurred through water diversions for agricultural purposes. Since 2000, 1038 x 10(6) m(3) of water have been released into the Tarim River with the result that water reached the terminal Taitema Lake for the first time in 30 years. This environmental flow raised water-table levels along 350 km of the river. To assess the response of the water-table, a comparison "pristine" site is compared with downstream monitored sites. The results show huge changes in water-table levels. The study verifies that the water-table is extremely responsive to environmental flows, that strong internal similarities exist along the length of the river, and that the effect on the water-table and hence likely riparian vegetation recovery can be tentatively predicted. The actual impacts of the restoration strategy are less than those originally expected politically, socially and within the scientific community. We make recommendations on more effective release strategies.

This laboratory exercise uses a simple preparation and a straightforward protocol to illustrate many of the basic principles of vascular biology covered in an introductory physiology course. The design of this laboratory allows students to actively participate in an exercise demonstrating the regulation of arterial tone by endothelial and extrinsic factors. In addition, this hands-on laboratory allows students to gather data using well-known basic biomedical research techniques. Specifically, students are introduced to an isolated organ-chamber technique that is widely used to study cellular mechanisms of many tissues including vascular smooth muscle contraction and dilation. On the basis of student evaluations, participation in the experiments and interpreting data reinforce lecture materials on smooth muscle and endothelial cell function and illustrate mechanisms regulating vascular tone. Students come away with a greater understanding of vascular biology, a deeper appreciation of integrative physiology, and an understanding of the process of conducting tissue-chamber experiments.

OBJECTIVE: To describe a patient who developed hepatic failure, Stevens-Johnson syndrome (SJS), and died after receiving amoxicillin/clavulanate therapy. CASE SUMMARY: A 37-year-old white man without significant past medical history received a 10-day course of amoxicillin/clavulanate for treatment of pneumonia. Thirty-two days after starting amoxicillin/clavulanate, he developed jaundice, rash, pruritus, and increasing fatigue. On further evaluation, with the exclusion of toxicity from other drugs or diseases, the time course to development of cholestatic jaundice correlated with the use of amoxicillin/clavulanate. The patient consequently died with progressive hepatic failure, renal failure, and SJS. DISCUSSION: Hepatic injury has been reported with amoxicillin/clavulanate. Signs and symptoms of jaundice and pruritus may appear up to to six weeks after stopping therapy. Most cases of liver injury have been benign and reversible on discontinuation of the amoxicillin/clavulanate. Reported hepatic reactions have been mainly cholestatic, with some mixed cholestatic/hepatocellular liver function test abnormalities. CONCLUSIONS: Clinicians should be aware of amoxicillin/clavulanate as a drug capable of causing hepatitis with eventual systemic dysfunction. While recovery is usually complete following withdrawal of the drug, in patients with rash associated with hepatic dysfunction, renal insufficiency, or other unusual symptoms, earlier consideration of initiating systemic steroids or liver transplantation referral, in hopes of avoiding progressive systemic response, might be worthwhile.

T1/ST2L, an IL-1 receptor homologue, is selectively expressed on murine Th2 cells and specific anti-ST2L antibodies can profoundly modulate the Th1/Th2 balance in vivo. Naive CD4+ T cells do not express ST2L but do so on activation with specific antigen in the presence of IL-4 or when stimulated with low doses of antigen in the absence of exogenously added IL-4. Similarly enhanced ST2L expression occurred after stimulation of Th2 cells with antigen or the mitogen ConA in the presence of APC. Restimulation of Th2 cells in the presence of IFN-gamma led to a decreased expression of ST2L to below basal levels. Conversely, Th2 cells cultured with IL-4 led to increased ST2L expression. The reduced expression of ST2L in response to high doses of antigen is also reversed by the neutralization of IFN-gamma. Using an ST2L promoter/luciferase reporter gene construct, we show that the distal but not proximal ST2L promoter is responsible for specific gene expression in Th2 cells. IL-4 enhances, whereas IFN-gamma suppresses ST2L expression via direct modulation of the distal promoter of the ST2L gene. These data provide a mechanistic explanation for the selective expression of ST2L on Th2 cells.
