RESUMO

NaH2SIP was selected as an organic ligand (NaH2SIP = 5-sulfoisophthalic acid monosodium salt). We successfully constructed a new class of lanthanide coordination polymers Ln-HS ([Ln(SIP)(DMF)(H2O)4]DMF·H2O; Ln = Eu, Tb, Sm, and Dy) by a simple solvothermal synthesis method. They exhibited excellent photoluminescence properties for Ln3+ ions, where Eu-HS and Tb-HS exhibited high quantum yields of 13.70 and 42.38%, respectively. The codoped lanthanide coordination polymers obtained by doping with different ratios of Eu3+/Tb3+ serve as excellent ratiometric thermometers with high sensitivities in the physiological temperature range, with values of 16.8, 7.0, and 14.5%·K-1, respectively. The luminescent colors of Tb0.95Eu0.05-HS and Tb0.94Eu0.06-HS exhibit variations from green to yellow to orange, achieving visualized luminescence in a narrow temperature range. The composite film material Tb0.94Eu0.06-HS@PMMA demonstrates this color variation. Next, Tb0.5Sm0.5-HS obtained by Tb3+/Sm3+ codoping was investigated. The difference in the luminescence colors visible to the naked eye at different excitation wavelengths and the change in luminescence colors occur in a very narrow temperature range. All of them show the great value of the visualized luminescence in practical anticounterfeiting, with double anticounterfeiting function and high security.

This study is aimed at examining the impact of ChatGPT on pediatric endocrine and metabolic conditions, particularly in the areas of screening and diagnosis, in both Chinese and English modes. A 40-question questionnaire covering the four most common pediatric endocrine and metabolic conditions was posed to ChatGPT in both Chinese and English three times each. Six pediatric endocrinologists evaluated the responses. ChatGPT performed better when responding to questions in English, with an unreliable rate of 7.5% compared to 27.5% for Chinese questions, indicating a more consistent response pattern in English. Among the reliable questions, the answers were more comprehensive and satisfactory in the English mode. We also found disparities in ChatGPT's performance when interacting with different target groups and diseases, with improved performance for questions posed by clinicians in English and better performance for questions related to diabetes and overweight/obesity in Chinese for both clinicians and patients. Language comprehension, providing incomprehensive answers, and errors in key data were the main contributors to the low scores, according to reviewer feedback. CONCLUSION: Despite these limitations, as ChatGPT continues to evolve and expand its network, it has significant potential as a practical and effective tool for clinical diagnosis and treatment. WHAT IS KNOWN: • The deep learning-based large-language model ChatGPT holds great promise for improving clinical practice for both physicians and patients and has the potential to increase the speed and accuracy of disease screening and diagnosis, as well as enhance the overall efficiency of the medical process. However, the reliability and appropriateness of AI model responses in specific field remains unclear. • This study focused on the reliability and appropriateness of AI model responses to straightforward and fundamental questions related to the four most prevalent pediatric endocrine and metabolic disorders, for both healthcare providers and patients, in different language scenarios. WHAT IS NEW: • The AI model performed better when responding to questions in English, with more consistent, as well as more comprehensive and satisfactory responses. In addition, we also found disparities in ChatGPT's performance when interacting with different target groups and different diseases. • Despite these limitations, as ChatGPT continues to evolve and expand its network, it has significant potential as a practical and effective tool for clinical diagnosis and treatment.

During the past decade, the average 5-year survival rate of patients with Lung adenocarcinoma (LUAD) has remained < 20%, although the targeted therapies and novel immunotherapy approaches have held promise. Epigenetic modifications could provide prognostic value as molecular biomarkers, and we aimed to identify the independent risk of m6A-related lncRNAs to establish a risk model for the clinical prediction of prognoses in women with LUAD. In this study, we first assessed 31 N6-methyladenosine (m6A)-related lncRNAs associated with overall survival. Moreover, we evaluated the expression of the oncogenic driver and the tumor immune microenvironment (TIME) in two female LUAD subtypes (clusters 1 and 2) using consensus clustering. We also found 16 m6A-related lncRNAs as the independent prognostic indicator of women with LUAD and established a risk model developed from these lncRNAs. We comprehensively investigated the correlation between the TIME and m6A-related lncRNA and found that m6A-related lncRNA may significantly affect the immune cell infiltration level in LUAD. In conclusion, our study provides evidence on the prognostic prediction in women with LUAD and may help elucidate the processes and mechanisms of m6A-regulated lncRNAs.

BACKGROUND: Encouraged by the goal of developing an effective treatment strategy for prostate cancer, this study explored the mechanism involved in metformin-mediated inhibition of AR-negative prostate cancer. METHODS: Cell behaviors of DU145 and PC3 cells were determined by CCK8 test, colony formation experiment and scratch test. Flow cytometry was used to detect cell cycle distribution. Cell autophagy was induced with metformin, and an autophagy inhibitor, 3-MA, was used to assess the level of autophagy. Detection of LC3B by immunofluorescence was conducted to determine autophagy level. Cell proliferation, autophagy and cell cycle were examined by performing Western blot. DU145 and PC3 cell lines were transfected with AMPK siRNA targeting AMPK-α1 and AMPK-α2. Tumor formation experiment was carried out to evaluate the anti-prostate cancer effect of metformin in vivo. RESULTS: The inhibitory effect of metformin on the proliferation of prostate cancer cell lines was confirmed in this study, and the mechanism of such an effect was related to autophagy and the block of cell cycle at G0/G1 phase. Metformin also induced the activation of AMPK, markedly promoted expression of LC3II, and down-regulated the expression of p62/SQSTM1. Animal experiments showed that the tumor volume of metformin group was smaller, meanwhile, the levels of p-AMPK (Thr172) and LC3B were up-regulated and the Ki-67 level was down-regulated, without abnormalities in biochemical indicators. CONCLUSION: This study found that autophagy induction might be the mechanism through which metformin suppressed the growth of AR-negative prostate cancer. Moreover, the activation of AMPK/autophagy pathway might be a therapeutically effective for treating AR-negative prostate cancer in the future.

Background: Matrix Metalloproteinases (MMPs) play an indispensable role in the initial alteration and development of PCa. We tried to generate an MMP-related prognostic signature (MMPS) in prostate cancer (PCa). Methods: TCGA-PRAD, MSKCC/GSE21032, GSE116918, GSE70769 cohorts were enrolled to assess the prognostic value of MMPs. The least absolute shrinkage and selection operator (LASSO) Cox regression was employed to generate the MMPS signature. The log-rank test and Kaplan-Meier (K-M) survival curve were applied to show the difference RFS, The receiver operating characteristic (ROC) curve and area under the ROC curve (AUC) was plotted to predict the accuracy of signature. CIBERSORT was conducted to analyze the different immune infiltration in MMPS-H and MMPS-L groups. Potential signaling pathways activated in the MMPS-H groups by Metascape. Results: MMP1, MMP7, MMP11, MMP24 and MMP26 were selected by LASSO regression and established the MMPS predict signature. The MMPS showed the high prognostic value in TCGA-PRAD training cohort (AUC=0.714) and validation cohorts (GSE116918: AUC=0.976, GSE70769: AUC=0.738, MSKCC: AUC=0.793). Pid integrin1 pathway, G2M checkpoint, and response to growth factor signaling pathways were activated in MMPS-H group, patients with the high MMPS risk score and low M2 macrophage showed the worst recurrence-free survival (RFS). Conclusion: MMPs involved and played an essential role in the tumorigenesis and biochemical recurrence in PCa patients. The MMPS signature could accurately predict the recurrence of PCa patients and validated in several cohorts.

Generic in-capillary as well as offline CE-based enzyme assays were developed for serine-ß-lactamases and metallo-ß-lactamases. The hydrolysis of benzylpenicillin to benzylpenicilloic acid was analyzed using 100 mM sodium phosphate solution, pH 6.0, as a background electrolyte. In-capillary assays employed an uncoated as well as a polyethylene oxide-coated capillary, while the offline assays employing long end and short end injection were performed in an uncoated capillary. Using procaine hydrochloride or 4-hydroxybenzoic acid as internal standard, the respective assays were validated with regard to linearity, LOD and LOQ, repeatability, precision, and accuracy. The assays were applied to the determination of the Michaelis-Menten parameters Km and Vmax of Bacillus cereus penicillinase as well as New Delhi metallo-ß-lactamase 1 and Verona integrin-encoded metallo-ß-lactamase 2. Furthermore, the inhibition of the enzymes by irreversible and competitive inhibitors was evaluated. Comparable data were obtained with all assays. The use of a simple substrate ensured broad applicability to the various types of ß-lactamases.

Considering size effect and functionalized pore interaction dyes guests and MOFs hosts, 4-aminonaphthalimide was successfully introduced into the pore of LnMOF for the first time and constructed 4-ANA⊂LnMOF luminescent composites with excellent dual-emission properties. A series of temperature-dependent luminescence test results show that 4-ANA⊂Gd4L3 can be used as a reversible ratiometric luminescent temperature sensor. The functional construction method provides ideas for the development of clear purpose novel dual-emission dye⊂LnMOF ratiometric luminescent sensors.

Two novel lanthanide metal-organic framework (Ln-MOF) luminescent sensors for the detection of picric acid have been successfully assembled. Following a function-oriented strategy, urea hydrogen-bonding functional sites were introduced into two MOF frameworks. A structural analysis indicated that the two MOFs have the exact same structure, namely 2D layers with diamond-shaped holes that are accumulated into a 3D framework through the hydrogen-bonding interactions between urea and carboxylate groups. Interestingly, only half of the urea units are involved in supporting the MOF framework through N-H⋅⋅⋅O hydrogen-bonding interactions, whereas the other half are located in the pore channel and act as empty recognition sites. Abundant N-H urea bonds are present in the inner walls of three types of interpenetrating 1D channels. Luminescence studies revealed that the two Ln-MOFs exhibit high sensitivity, good selectivity, and a fast luminescence quenching response towards picric acid. In particular, the two Ln-MOFs can be simply and quickly regenerated, and exhibit excellent recyclability. In summary, we have successfully used a function-oriented strategy to achieve multiple functions in a ligand to construct lanthanide MOF luminescent sensors for the detection of picric acid, thereby providing a potential strategy for the future development of MOF luminescent sensors with a specific target.

The major challenge to treat the clinical adverse effects of long-segment urethra is in achieving viable tissue substitution. The substituted construct's properties-such as its resilience, contraction, and ability to minimize scar-stenosis formation should be considered. In the present work, a unique polyurethane-urea (PUU) fibrous membrane is fabricated by electrospinning. Then PUU was coated by collagen and formed the elasticity hydrogel after immersed in collagen solution. Meanwhile, the cPUU hydrogel exhibited a fibrous microstructure. This cPUU hydrogel had outstanding stretching property with 404 ± 40% elongation at break compared with traditional hydrogels, which satisfied the requirement of urethra. The cPUU hydrogel also supported the adhesion and growth of bladder smooth-muscle cells (BSMCs) in natural state cell morphology. Urethral defects in New Zealand male rabbits were repaired with cPUU seeded with BSMCs in vivo. After three months, more smooth-surface area of reconstructed urethral tissues was observed in the cPUU hydrogel-BMSCs groups compared with that of the control group. The luminal patency and the incidence of complications-including calculus formation, urinary fistula, and urethral-stricture occurrence were significantly lower in the cPUU group compared with that of the control group. Hence, cPUU fibrous hydrogels are promising scaffolds for application in urological tissue engineering.

Coumarinocoumarin, one of the coumarin derivatives, is easy to synthesize and has rich modification sites. The large conjugate plane of coumarinocoumarin gives it a more excellent optical property than conventional coumarin, for example, the two-photon fluorescence property. So, the coumarinocoumarin-based probe (CCy) has been designed and synthesized, which is the first lysosomal targeting fluorescent biosensor for cysteine. This probe was prepared by a three-step procedure as a latent fluorescence probe to achieve high sensitivity and fluorescence turn-on response toward cysteine (Cys) over homocysteine (Hcy), glutathione (GSH), and other various natural amino acids under physiological conditions. Upon addition of Cys to the solution of CCy, remarkable enhancement on 520 nm of fluorescence spectra can be monitored. This probe was then successfully used for fluorescence imaging of Cys in mice organ tissues and HeLa cells, and quantitative determination has been achieved within a certain range, which proved the permeability of CCy. The concentration of Cys in animal serum was measured and the viability exceeded 80%, indicating that CCy can be a biocompatible and rapid probe for Cys in vivo. Simultaneously, its ability to detect Cys in lysosome has been validated by its lysosomal targeting.

The muscle-specific creatine kinase (CKM) A/G variants (rs8111989) have been associated with skeletal muscle performance in humans; they are correlated with physical performance and contribute to differences in the maximum oxygen uptake (VO2max) responses during power or endurance training. However, there is not enough definitive evidence to demonstrate whether the A and G allelic variants of the CKM gene rs8111989 are indeed genetic factors that can influence human physical performance. In our study, we identified 9 articles on CKM in a literature search, and conducted two meta-analyses on the CKM rs8111989 A/G allele or genotype differences between power or endurance athletes and general controls. We found that the power athletes had a significantly higher frequency of the G allele (OR, 1.14; 95% CI, 1.02-1.28, P=0.03) and GG genotype (OR, 1.54; 95% CI, 1.24-1.91, P<0.0001) compared to controls, but there was no significant difference for the endurance athletes (G allele, OR, 0.95, 95%CI, 0.85-1.06, P=0.34; GG genotype, OR, 1.00, 95%CI, 0.78-1.27, P=1.00). The results provide additional evidence to support the notion that human physical performance might be influenced by genetic profiles, especially in power sports.

Stereospecific capillary electrophoresis-based methods for the analysis of methionine sulfoxide [Met(O)]-containing pentapeptides were developed in order to investigate the reduction of Met(O)-containing peptide substrates by recombinant Aspergillus nidulans methionine sulfoxide reductase A (MsrA) as well as enzymes carrying mutations in position Glu99 and Asp134. The separation of the diastereomers of the N-acetylated, C-terminally 2,4-dinitrophenyl (Dnp)-labeled pentapeptides ac-Lys-Phe-Met(O)-Lys-Lys-Dnp, ac-Lys-Asp-Met(O)-Asn-Lys-Dnp and ac-Lys-Asn-Met(O)-Asp-Lys-Dnp was achieved in 50 mM Tris-HCl buffers containing sulfated ß-CD in fused-silica capillaries, while the diastereomer separation of ac-Lys-Asp-Met(O)-Asp-Lys-Dnp was achieved by sulfated ß-CD-mediated MEKC. The methods were validated with regard to range, linearity, accuracy, limits of detection and quantitation as well as precision. Subsequently, the substrates were incubated with wild-type MsrA and three mutants in the presence of dithiothreitol as reductant. Wild-type MsrA displayed the highest activity towards all substrates compared to the mutants. Substitution of Glu99 by Gln resulted in the mutant with the lowest activity towards all substrates except for ac-Lys-Asn-Met(O)-Asp-Lys-Dnp, while replacement Asn for Asp134 lead to a higher activity towards ac-Lys-Asp-Met(O)-Asn-Lys-Dnp compared with the Glu99 mutant. The mutant with Glu instead of Asp134 was the most active among the mutant enzymes. Molecular modeling indicated that the conserved Glu99 residue is buried in the Met-S-(O) groove, which might contribute to the correct placing of substrates and, consequently, to the catalytic activity of MsrA, while Asp134 did not form hydrogen bonds with the substrates but only within the enzyme.

A novel luminescent microporous lanthanide metal-organic framework (Ln-MOF) based on a urea-containing ligand has been successfully assembled. Structural analysis revealed that the framework features two types of 1D channels, with urea N-H bonds projecting into the pores. Luminescence studies have revealed that the Ln-MOF exhibits high sensitivity, good selectivity, and a fast luminescence quenching response towards Fe3+ , CrVI anions, and picric acid. In particular, in the detection of Cr2 O72- and picric acid, the Ln-MOF can be simply and quickly regenerated, thus exhibiting excellent recyclability. To the best of our knowledge, this is the first example of a multi-responsive luminescent Ln-MOF sensor for Fe3+ , CrVI anions, and picric acid based on a urea derivative. This Ln-MOF may potentially be used as a multi-responsive regenerable luminescent sensor for the quantitative detection of toxic and harmful substances.

Tissue engineering-based urethral replacement holds potential for repairing large segmental urethral defects, which remains a great challenge at present. This study aims to explore the potential of combining biodegradable poly(l-lactide) (PLLA)/poly(ethylene glycol) (PEG) scaffolds and human amniotic mesenchymal cells (hAMSCs) for repairing urethral defects. PLLA/PEG fibrous scaffolds with various PEG fractions were fabricated via electrospinning. The scaffolds were then seeded with hAMSCs prior to implantation in New Zealand male rabbits that had 2.0 cm-long defects in the urethras. The rabbits were randomly divided into three groups. In group A, hAMSCs were grown on PLLA/PEG scaffolds for two days and then implanted to the urethral defects. In group B, only the PLLA/PEG scaffolds were used to rebuild the rabbit urethral defect. In group C, the urethral defect was reconstructed using a regular urethral reparation technique. The repair efficacy was compared among the three groups by examining the urethral morphology, tissue reconstruction, luminal patency, and complication incidence (including calculus formation, urinary fistula, and urethral stricture) using histological evaluation and urethral radiography methods. Findings from this study indicate that hAMSCs-loaded PLLA/PEG scaffolds resulted in the best urethral defect repair in rabbits, which predicts the promising application of a tissue engineering approach for urethral repair.

A novel water-soluble colorimetric and ratiometric fluorescent probe was synthesized and applied to imaging palladium species under physiological conditions in phosphate buffered saline (PBS) containing less than 1% organic cosolvent without adding any additional reagents. Based on palladium triggered terminal propargyl ethers cleavage reaction, the probe exhibited a high selectivity and sensitivity for palladium species of all the typical oxidation states (0, +2, +4), with a low detection limit (25 nM, 2.7 µg/L) and an obvious color change. Furthermore, the probe was successfully used for ratiometric fluorescence imaging of palladium in living cells.

A merocyanine-based highly selective colorimetric and ratiometric fluorescent probe is described for Cys detection in water and diluted deproteinized human serum. Upon reaction with Cys in aqueous buffer solution, the probe showed a dramatic color change from faint yellow to pink and remarkable ratiometric fluorescence enhancement signals were also observed, which are ascribed to an intramolecular charge transfer (ICT) process. This strategy was based on modulating the merocyanine π-electron system by conjugation and removal of the acrylate group to release the chromophore group, resulting in a specific colorimetry and fluorescence response. The probe has low cytotoxicity and good cell permeability. It is readily employed for assessing the change of the intracellular Cys level.

Pyrethroids are widely used insecticides worldwide, while their on-site and rapid detection still faces technological challenges. Herein, an innovative detection mechanism was designed for deltamethrin, a typical kind of type II pyrethroids, based on a dual-emitting fluoroprobe consisting of NH2-SiQDs and Eu3+. Deltamethrin can rapidly hydrolyze into 3-phenoxybenzaldehyde (3-PBD) and react specifically with fluoroprobe, causing fluorescence quenching of SiQDs while maintaining the fluorescent stability of Eu3+. Building upon the above fluorescence-responsive principle, SiQDs@Eu3+ provided satisfactorily dual-emitting signals, realizing the highly-selective and sensitive detection of deltamethrin. Correlation between the surface structure of SiQDs and their absorption spectra was in-depth unraveled by TD-DFT calculation and FT-IR analysis. As for the analytical performance, the recovery and LOD of deltamethrin in lettuce, provided by SiQDs@Eu3+, were comparable or even superior over conventional chromatographic analysis. Meanwhile, an innovative smartphone-based optical device was developed, which greatly decreased errors caused by the previously reported smartphone-based fluorescence detection.

The on-site detection of pyrethroids, particularly type II pyrethroids, remains a challenging task in complex vegetable samples. Herein, a novel method based on naphthalimide was developed to realize the specific detection of type II pyrethroids by hydrolyzing and utilizing the compound m-phenoxybenzaldehyde (3-PBD). Hydrazine group, used as the appropriate moiety, was introduced into the fluorescent dye 1,8-naphthalimide to construct the fluoroprobe NAP. In the presence of 3-PBD, NAP displayed the prominently enhanced fluorescence and also exhibited high selectivity. This proposed method exhibited high anti-inference effects in complex media, realizing sensitive detection of 3-PBD with linear range of 2.15-800 µM and a low detection limit (LOD) of 0.64 µM. The underlying fluorescence-responsive mechanisms were in-depth elucidated by combining spectral analyses with TD-DFT theoretical calculations. Additionally, a direct and rapid hydrolysis method for deltamethrin in celery was established, achieving a high hydrolysis efficiency of >90% within 15 min. Furthermore, a portable fluorescence sensor (PFS) was developed based on high-power LEDs and photodetectors. PFS supplied a LOD of 2.23 µM for 3-PBD and exhibited comparable stability by a fluorescence spectrometer when detecting celery hydrolysate. Moreover, external power source is not required for PFS operations, thereby enabling rapid and on-site detection by transmitting data to a smartphone via bluetooth. These findings extend the academic knowledge in the field of specific pyrethroids detection and contribute to the development of on-site methods for pesticide residual analyses in food matrices.

Tetracycline hydrochloride (TCH), a prevalent antibiotic in aquaculture for treating bacterial infections, poses challenges for on-site detection. This study employed the reversed-phase microemulsion method to synthesize a uniform nano metal-organic framework (MOF) material, europium-benzene-p-dicarboxylic acid (Eu-BDC), doped with Tb3+ to form a dual-emission fluorescence probe. By leveraging the combined a-photoinduced electron-transfer (a-PET) and inner filter effect (IFE) mechanisms, high-sensitivity TCH detection in Carassius auratus and Ruditapes philippinarum was achieved. The detection range for TCH is 0.380-75 µM, with a low limit of detection (LOD) at 0.115 µM. Upon TCH binding, Eu-BDC fluorescence rapidly decreased, while Tb3+ fluorescence remained constant, establishing a ratiometric fluorescence change. Investigation into the TCH quenching mechanism on Eu-BDC was conducted using time-dependent density functional theory (TD-DFT) calculations and fluorescence quenching kinetic equations, suggesting a mixed quenching mechanism. Furthermore, a novel photoelectric conversion fluorescence detection device (FL-2) was developed and evaluated in conjunction with high-performance liquid chromatography-diode-array detection (HPLC-DAD). This is the first dedicated fluorescence device for TCH detection, showcasing superior photoelectric conversion performance and stability that reduces experimental errors associated with smartphone photography methods, presenting a promising avenue for on-site rapid TCH detection.

Immobilizing Fe-based nanoparticles on electron-rich biochar has becoming an attractive heterogeneous Fenton-like catalysts (Fe/BC) for wastewater decontamination. However, the insufficient graphitization of biochar causing low electron transfer and by slow H2O2 activation limited its application. Herein, we firstly constructed FeS/biochar composite through all-solid molten salt method (Fe/MSBCs), which can provide strong polarization force and liquid reaction environment to improve carbonization. As expected, the obtained Fe/MSBCs exhibits high surface area and fast interfacial electron transfer between FeS and biochar. More importantly, the partially oxidized FeS (001) facet facilitate H2O2 adsorption and thermodynamically easily decomposition into •OH. Such a synergistic effect endowed them excellent photo-Fenton degradation performance for methyl orange (MO) with large kinetic rate constants (0.079 min-1) and high H2O2 utilization efficiency (95.9%). This study first demonstrated the critical regulatory role of molten salt method in iron-based biochar composites, which provide an alternative for H2O2 activator in water pollutant control.
