RESUMO

Experiments on white female rats showed that chronic ethanol consumption leads to activation of the NADPH - GSH dependent system of the liver, which provides protection against ethanol-induced oxidative stress. Post-treatment administration of dry extract from Vicia truncatula Fish ex Bieb. (300 mg/kg body weight) on the background of 40% ethanol increased this adaptive reaction without depletion in the system, in contrast to what was observed in rats treated with carsil. These results suggest a significant hepatoprotective effect of the dry extract during the chronic alcohol-induced injury of the liver.

RESUMO

A relationship between the vasodilating activity of flavonoids under conditions of endothelial dysfunction in experimental diabetes and their antioxidant effect is established. Flavicin and quercetin show the maximum influence on the vasodilating function of endothelium and exhibit more pronounced antioxidant activity under conditions of endothelial dysfunction.

RESUMO

The formation of nitric oxide radicals in the organs of Wistar rats was studied by the ESR method using ferrous-diethyldithiocarbamate complexes as an NO spin trap. It was found that, under the conditions of CCl4-induced hepatosis, a 4.32-5.46-fold increase in the concentrations of NO radical spin adducts in the samples of heart and liver, correspondingly, occurs. After preliminary intraperoral introduction of quercetin and hesperidin, this effect was not observed. Thus, the flavonoids quercetin and hesperidin exert a distinct antioxidant action, substantialy suppressing the hepatosis-induced increase in the intensity of NO radical generation.

RESUMO

The prophylactic hepatoprotective properties of a series of natural flavonoids (hesperidin from citrus peel, diosmine from Vicia tanuifolia Roth., and diosmine analog from Vicia trunctula) were studied in a group of white male rats with a model of acute CCl4 hepatitis. The effect was evaluated by the ability of tested substances to normalize the biochemical characteristics of the functional state of the liver, in comparison to the reference drug carsil. The most pronounced hepatoprotective action was observed for diosmine analog administered in a dose of 100 mg/kg, which was superior in some respects to the reference hepatoprotector (in equivalent doses). This is probably related to the higher bioaccessibility of diosmine analog ensured by a carbohydrate moiety of this compound, in combination with the C2-C3 double bond present in diosmetin (diosmin aglycon).

RESUMO

As shown by accumulation of malonic dialdehyde and chemiluminescence rate monitoring tetracycline activated both NADPH- and ascorbate-dependent lipid peroxidation in liver tissue of 1, 3, 12 and 24 months old rats. The most distinct induction of lipid peroxidation was observed in old animals, which appears to occur due to a decrease in efficiency of NADPH-GSH-dependent enzymatic antioxidant system. Silibore prevented the stimulating effect of tetracycline on enzymatic and nonenzymatic lipid peroxidation, being apparently involved in hepatoprotective effect.
