AIM: To define the computed tomography (CT) features common in skeletal muscle metastases and their prevalence and to identify the most commonly associated primary malignancy and the most common muscle groups in which skeletal muscle metastases are found. METHODS AND MATERIALS: Institutional review board (IRB) waiver for informed consent was obtained. A retrospective review was conducted of CT examinations from a single, large, academic centre picture archiving and communication system (PACS) database, performed from August 2009 to July 2013. All 10,426 examinations and 8,524 unique patients reviewed had a confirmed diagnosis of malignancy. The CT reports were screened manually to identify disease involving the skeletal muscles. Images of the 60 initial studies identified were then reviewed. Cases that showed direct invasion of the tumour into the skeletal muscles, and follow-up studies of the same patient were excluded. The 27 included cases were classified under five distinct patterns. RESULTS: In the present study, the prevalence for skeletal muscle metastasis was 0.33% across all malignancies. The most common primary involved was breast cancer (25%). The most common pattern was focal intramuscular mass with homogeneous contrast enhancement. The most common sites of skeletal muscle metastasis were in the abdomen (43%) and thorax (33%) musculature. CONCLUSION: Breast cancer was the most commonly associated primary malignancy and a focal, homogeneously enhancing intramuscular mass was the most common presentation. Although skeletal muscle metastasis remains a rare entity, its incidence rate should increase with the increased usage of whole-body PET CT for cancer staging.

Here we present a simple yet efficient analytical method for sensing ultratrace levels of Hg2+ ions by highly water soluble CdS quantum dots functionalized with thiourea as a probe. The bluish photoluminescence emission of the probe responded to a systematic linear photoluminescence quenching in the presence of increasing concentration of Hg2+ ions. The photoluminescence quenching by Hg2+ ions was attributed to agglomeration of the quantum dots, which has been confirmed by zeta potential measurements. The sensitivity (31.38 L/mg) and LoD (0.11 µg/L) of Hg2+ ion detection by our method are two folds improved with respect to the existing data of CdS as sensor. The improved detection is attributable to synthesis of less than 3 nm diameter CdS quantum dots which rendered very high water solubility and hence facilitated better interaction with Hg2+ ions. The detection of Hg2+ ion was free from most interfering cations and anions, except for minor interference from Cu2+ and Pb2+ corresponding their concentrations expected in ground water. Further, the scope for visual detection of Hg2+ was explored, which revealed naked eye recognizable photoluminescence quenching of the probe treated 0.3 mg/L of Hg2+ ion when excited by a light source of 365 nm. The suitability of our probe to analyze Hg2+ in real samples has been demonstrated by Hg2+ spike analysis in groundwater and river water samples.

BACKGROUND: There is a genetic contribution to the risk of suicide, but sparse prior research on the genetics of suicidal ideation. METHODS: Active and passive suicidal ideation were assessed in a Sri Lankan population-based twin registry (n = 3906 twins) and a matched non-twin sample (n = 2016). Logistic regression models were used to examine associations with socio-demographic factors, environmental exposures and psychiatric symptoms. The heritability of suicidal ideation was assessed using structural equation modelling. RESULTS: The lifetime prevalence of any suicidal ideation was 13.0% (11.7-14.3%) for men; 21.8% (20.3-23.2%) for women, with no significant difference between twins and non-twins. Factors that predicted suicidal ideation included female gender, termination of marital relationship, low education level, urban residence, losing a parent whilst young, low standard of living and stressful life events in the preceding 12 months. Suicidal ideation was strongly associated with depression, but also with abnormal fatigue and alcohol and tobacco use. The best fitting structural equation model indicated a substantial contribution from genetic factors (57%; CI 47-66) and from non-shared environmental factors (43%; CI 34-53) in both men and women. In women this genetic component was largely mediated through depression, but in men there was a significant heritable component to suicidal ideation that was independent of depression. CONCLUSIONS: These are the first results to show a genetic contribution to suicidal ideation that is independent of depression outside of a high-income country. These phenomena may be generalizable, because previous research highlights similarities between the aetiology of mental disorders in Sri Lanka and higher-income countries.

Detection of ultratrace levels of aqueous uranyl ions without using sophisticated analytical instrumentation and a tedious sample preparation method is a challenge for environmental monitoring and mitigation. Here we present a novel yet simple analytical method for highly sensitive and specific detection of uranyl ions via photoluminescence quenching of CdS quantum dots. We have demonstrated a new approach for synthesizing highly water-soluble and strong photoluminescence-emitting CdS quantum dots (i.e., CdS-MAA and CdS-MAA-TU) of sizes less than 3 nm. The structural, morphological, and optical properties of both the batches of CdS quantum dots were thoroughly characterized by XRD, high-resolution transmission electron microscopy (HRTEM), zeta potential, UV-visible absorption, and photoluminescence spectroscopy. Compared to the batch of CdS quantum dots prepared by capping with only mercaptoacetic acid (CdS-MAA), the batch prepared by capping with mercaptoacetic acid and thiourea in tandem (CdS-MAA-TU) exhibited higher quantum yield= 16.64 ± 1.02%, and more importantly, CdS-MAA-TU exhibited significantly a higher order of photoluminescence quenching responses when treated with ultratrace concentrations of uranyl ions. Under the optimized conditions, the sensitivity of detecting uranyl ion by CdS-MAA-TU was several folds better (0.316 L/ µg) than that of CdS-MAA (0.0053 (L/µg/), as determined from their respective Stern-Volmer plots. Qualitatively, CdS-MAA-TU probe can be used for visual detection of uranyl ions of concentration greater than 5 µg/L. However, the instrumental method of analysis based on photoluminescence spectroscopy confirmed the feasibility for quantitative analysis of ultratrace concentrations of uranyl ions as implied from a very low limit of detection (LoD = 0.07 µg/L) and limit of quantification (LoQ = and 0.231 µg/L). Systematic studies revealed very high selectivity for uranyl ion detection, though minor interference from Cu(2+), Pb(2+), Hg(2+), CO3(2-), and SO4(2-) was found. The recovery analysis performed by spiking uranyl ions (0.5 µg/L to 10.0 µg/L) in groundwater and river water samples, confirmed the robustness of the as-developed CdS-MAA-TU QDs for detecting ultratrace levels of uranyl ions in real water sample matrix. The very simple and effective strategy reported here should facilitate developing reliable sensors for detecting uranyl ion contamination in drinking water.

BACKGROUND: Self-harm is a significant public health concern in the UK. This is reflected in the recent addition to the English Public Health Outcomes Framework of rates of attendance at Emergency Departments (EDs) following self-harm. However there is currently no source of data to measure this outcome. Routinely available data for inpatient admissions following self-harm miss the majority of cases presenting to services. We aimed to investigate (i) if a dataset of ED presentations could be produced using a combination of routinely collected clinical and administrative data and (ii) to validate this dataset against another one produced using methods similar to those used in previous studies. METHODS: Using the Clinical Record Interactive Search system, the electronic health records (EHRs) used in four EDs were linked to Hospital Episode Statistics to create a dataset of attendances following self-harm. This dataset was compared with an audit dataset of ED attendances created by manual searching of ED records. The proportion of total cases detected by each dataset was compared. RESULTS: There were 1932 attendances detected by the EHR dataset and 1906 by the audit. The EHR and audit datasets detected 77% and 76 of all attendances respectively and both detected 82% of individual patients. There were no differences in terms of age, sex, ethnicity or marital status between those detected and those missed using the EHR method. Both datasets revealed more than double the number of self-harm incidents than could be identified from inpatient admission records. CONCLUSIONS: It was possible to use routinely collected EHR data to create a dataset of attendances at EDs following self-harm. The dataset detected the same proportion of attendances and individuals as the audit dataset, proved more comprehensive than the use of inpatient admission records, and did not show a systematic bias in those cases it missed.

A cross sectional observational study was conducted to assess knowledge, attitude and practices about leprosy among leprosy patients in six districts of West Bengal. Total patients selected for the study were 300; of them 185 patients were from three high prevalent districts and 115 from three low prevalent districts of West Bengal. 56.33% patients were male and 43.67% were female. Most of the patients (85.67%) belonged to Hindu community and 60% from socially backward group. 64.33% patients lived below poverty line. Thirty five percentage of patients had correct knowledge that leprosy is caused by a bacteria. Patients from high prevalent districts (41.62%) have better knowledge than those from low prevalent areas (26.09%). Difference was found to be statistically significant (p = 0.006). Correct knowledge about spread of leprosy through cough & sneezing, of the patients from high prevalent districts (30.81%) was more than those from low prevalent districts (14.78%) (p = 0.001). 74.05% patients from high prevalent districts could tell one or other forms of clinical presentation of a leprosy patients, while 56.52% from low prevalent areas could mention it correctly (p = 0.01). About infectiousness, duration of treatment, complications, patients from high prevalent districts showed better knowledge that those from low prevalent districts. Similarly, Attitude of the patients towards leprosy was found to be more adverse in low prevalent areas. 90% patients have idea that leprosy was curable, but only 51.67% patients heard about MDT. Place of residence (high prevalent districts) & level of education (secondary & above) attributed to better knowledge score of the patients, whereas Place of residence (high prevalent districts) & age (younger age group) attributed to better attitude score of the patients.

BACKGROUND: Competitive sports training causes structural and conductive system changes manifesting by various electrocardiographic alterations. We undertook this study to assess the prevalence of abnormal ECG in trained Indian athletes and correlate it with the nature of sports training, that is endurance or strength training. METHODS: We evaluated a standard resting, lying 12 lead Electrocardiogram (ECG) in 66 actively training Indian athletes. Standard diagnostic criteria were used to define various morphological ECG abnormalities. RESULTS: 33/66 (50%) of the athletes were undertaking endurance training while the other 33 (50%) were involved in a strength-training regimen. Overall 54/66 (81%) sportsmen had significant ECG changes. 68% of these changes were considered as normal training related features, while the remaining 32% were considered abnormal. There were seven common training related ECG changes-Sinus Bradycardia (21%), Sinus Arrhythmia (16%), 1st degree Atrioventricular Heart Block (6%), Type 1 2nd-degree Atrioventicular Heart Block (3%), Incomplete Right bundle branch block (RBBB) (24%), Early Repolarization (42%), Left Ventricular Hypertrophy (LVH) (14%); while three abnormal ECG changes--T-wave inversion (13%), RBBB(4%), Right ventricular hypertrophy (RVH) with strain (29%) were noted. Early repolarization (commonest change), sinus bradycardia, and incomplete RBBB were the commoner features noticed, with a significantly higher presence in the endurance trained athletes. CONCLUSION: A high proportion of athletes undergoing competitive level sports training are likely to have abnormal ECG recordings. Majority of these are benign, and related to the physiological adaptation to the extreme levels of exertion. These changes are commoner during endurance training (running) than strength training (weightlifting).

Dipeptidyl peptidase-4 (DPP-4) inhibitors belong to a prominent group of pharmaceutical agents that are used in the governance of type 2 diabetes mellitus (T2DM). They exert their antidiabetic effects by inhibiting the incretin hormones like glucagon-like peptide-1 and glucose-dependent insulinotropic polypeptide which, play a pivotal role in the regulation of blood glucose homoeostasis in our body. DPP-4 inhibitors have emerged as an important class of oral antidiabetic drugs for the treatment of T2DM. Surprisingly, only a few 2D-QSAR studies have been reported on DPP-4 inhibitors. Here, fragment-based QSAR (Laplacian-modified Bayesian modelling and Recursive partitioning (RP) approaches have been utilized on a dataset of 108 DPP-4 inhibitors to achieve a deeper understanding of the association among their molecular structures. The Bayesian analysis demonstrated satisfactory ROC values for the training as well as the test sets. Meanwhile, the RP analysis resulted in decision tree 3 with 2 leaves (Tree 3: 2 leaves). This present study is an effort to get an insight into the pivotal fragments modulating DPP-4 inhibition.

OBJECTIVE: This study describes the incidence of psychosis in unemployed people and determines whether unemployment has a greater impact on the development of psychosis amongst Black minority groups than White groups. METHOD: Patients with a first diagnosis of Research Diagnostic Criteria psychosis, in a defined area of London from 1998 to 2004, were identified. Crude and standardised incidence rates of psychosis amongst unemployed people for each ethnic group were calculated. Poisson regression modelling tested for interactions between unemployment and ethnicity. RESULTS: Hundred cases occurred amongst employed people and 78 cases occurred amongst the unemployed people. When standardised to the employed White population of the area, White unemployed people had a standardised incidence ratio (SIR) of 11.7 (95% CI 6.4-19.7), Black Caribbean people had a SIR of 60.1(95% CI 39.3-88) and Black African people had a SIR of 40.7 (95% CI 25.8-61.1). There was no interaction however between ethnicity and unemployment (Likelihood ratio test P = 0.54). CONCLUSION: Rates of psychosis are high amongst unemployed people in south London and extremely high amongst Black Caribbean and Black African unemployed people. There was no evidence however that the minority groups were particularly sensitive to the stresses, limitations or meaning of unemployment.

In this paper, opposition-based harmony search has been applied for the optimal design of linear phase FIR filters. RGA, PSO, and DE have also been adopted for the sake of comparison. The original harmony search algorithm is chosen as the parent one, and opposition-based approach is applied. During the initialization, randomly generated population of solutions is chosen, opposite solutions are also considered, and the fitter one is selected as a priori guess. In harmony memory, each such solution passes through memory consideration rule, pitch adjustment rule, and then opposition-based reinitialization generation jumping, which gives the optimum result corresponding to the least error fitness in multidimensional search space of FIR filter design. Incorporation of different control parameters in the basic HS algorithm results in the balancing of exploration and exploitation of search space. Low pass, high pass, band pass, and band stop FIR filters are designed with the proposed OHS and other aforementioned algorithms individually for comparative optimization performance. A comparison of simulation results reveals the optimization efficacy of the OHS over the other optimization techniques for the solution of the multimodal, nondifferentiable, nonlinear, and constrained FIR filter design problems.

The toxicity effect due to chronic exposure of ZnO nanoparticles (NPs) was systematically studied by repeatedly treating different lower concentrations of ZnO nanoparticles with culture media of E. coli strain. The chronic exposure of ZnO NPs of concentrations below minimum inhibitory concentration (MIC) exhibited higher toxicity than the single exposure of higher concentrations. Most striking result was 57% inhibition of growth corresponding to chronic exposure of 0.06 mg/mL of ZnO NPs which was two folds more than that exhibited by single exposure of 0.30 mg/mL ZnO NPs. The toxicity of ZnO NPs in E. coli was studied in the light of formation of reactive oxygen species (ROS), measured as malondialdehyde (MDA) equivalent by thiobarbituric acid-ROS (TBARS) assay, and effect of Zn dissolution from ZnO NPs. Higher inhibition of growth for the chronic exposure batches were correlated with higher ROS generation, which subsequently contributed to cause membrane lipid peroxidation, confirmed from observation of cell wall deformation by scanning electron microscopy study and energy dispersive X-ray analysis showed adherence of ZnO NPs on cell wall. The possibility of membrane lipid peroxidation was addressed by revealing in vitro oxidation of oleic acid, which is a monounsaturated fatty acid. Further in this study we have shown that the dissolution of ZnO NPs at pH 7.4 was not significant to cause Zn-induced toxicity.

BACKGROUND: The excess mortality following first-contact psychosis is well recognized. However, the causes of death in a complete incidence cohort and mortality patterns over time compared with the general population are unknown. METHOD: All 2723 patients who presented for the first time with psychosis in three defined catchment areas of the U.K. in London (1965-2004, n=2056), Nottingham (1997-1999, n=203) and Dumfries and Galloway (1979-1998, n=464) were traced after a mean of 11.5 years follow-up and death certificates were obtained. Data analysis was by indirect standardization. RESULTS: The overall standardized mortality ratio (SMR) for first-contact psychosis was 184 [95% confidence interval (CI) 167-202]. Most deaths (84.2%, 374/444) were from natural causes, although suicide had the highest SMR (1165, 95% CI 873-1524). Diseases of the respiratory system and infectious diseases had the highest SMR of the natural causes of death (232, 95% CI 183-291). The risk of death from diseases of the circulatory system was also elevated compared with the general population (SMR 139, 95% CI 117-164) whereas there was no such difference for neoplasms (SMR 111, 95% CI 86-141). There was strong evidence that the mortality gap compared with the general population for all causes of death (p<0.001) and all natural causes (p=0.01) increased over the four decades of the study. There was weak evidence that cardiovascular deaths may be increasing relative to the general population (p=0.07). CONCLUSIONS: People with first-contact psychosis have an overall mortality risk that is nearly double that of the general population. Most excess deaths are from natural causes. The widening of the mortality gap over the last four decades should be of concern to all clinicians involved in delivering healthcare.

Coal is primarily a fuel material but lately it has been utilized as an adsorbent for removing toxic metal ions. However, its usage for removing organic pollutants is not well studied. We report here a systematic study on the use of coal samples of varying carbon contents as adsorbents for removing Basic Blue 41 as a model cationic dye. The coal samples were collected from coal mines and were thoroughly characterized. The concentrations of carbon, hydrogen, oxygen, nitrogen and sulphur contents were measured by CHNS analyzer. The concentrations of aluminum, silicon, sulphur, titanium and iron were determined by EDXRF, which corresponded to silicon dioxide (quartz) and aluminium silicate (kaolinite) as the major mineral inclusions, corroborated by XRD results and micrographs showing elemental maps determined from SEM-EDAX. The coal samples with low carbon content revealed higher adsorption capacity (qe âˆ¼ 8.0-9.3 mg/g) of Basic Blue dye at optimized adsorbent dose (2 mg/mL), pH 9 and contact time (120 min). The adsorption kinetic studies satisfied pseudo second order model and the intra-particle diffusion of the dye was evident. The dye adsorption followed Langmuir adsorption isotherm, and the qmax values ranged between 17 and 30 mg/g for low carbon content coal. The FT-IR, Brunauer-Emmett-Teller (BET) surface area and zeta potential results of the coal samples could explain the adsorption phenomenon of cationic dye. The kinetic and thermodynamic studies revealed that the adsorption of Basic Blue 41 dye was based on chemisorptions mechanism.

The present investigation was carried out to find an efficient chemically assisted procedure for enucleation of goat oocytes related to handmade cloning (HMC) technique. After 22-h in vitro maturation, oocytes were incubated with 0.5 µg/ml demecolcine for 2 h. Cumulus cells were removed by pipetting and vortexing in 0.5 mg/ml hyaluronidase, and zona pellucida were digested with pronase. Oocytes with extrusion cones were subjected to oriented bisection. One-third of the cytoplasm with the extrusion cone was removed with a micro blade. The remaining cytoplasts were used as recipients in HMC. Goat foetal fibroblasts were used as nuclear donors. The overall efficiency measured as the number of cytoplasts obtained per total number of oocytes used was significantly (p < 0.05) higher in chemically assisted handmade enucleation (CAHE) than oriented handmade enucleation without demecolcine (OHE) (80.02 ± 1.292% vs. 72.9 ± 1.00%, respectively, mean ± SEM). The reconstructed and activated embryos were cultured in embryo development medium (EDM) for 7 days. Fusion, cleavage and blastocyst development rate were 71.63 ± 1.95%, 92.94 ± 0.91% and 23.78 ± 3.33% (mean ± SEM), respectively which did not differ significantly from those achieved with random handmade enucleation and OHE. In conclusion, chemically assisted enucleation is a highly efficient and reliable enucleation method for goat HMC which eliminates the need of expensive equipment (inverted fluorescence microscope) and potentially harmful chromatin staining and ultraviolet (UV) irradiation for cytoplast selection.

BACKGROUND: MicroRNAs play an important role in health and disease. TGF-ß signaling, upregulated by HIV Tat, and in chronic airway diseases and smokers upregulates miR-145-5p to suppress cystic fibrosis transmembrane conductance regulator (CFTR). CFTR suppression in chronic airway diseases like Cystic Fibrosis, COPD and smokers has been associated with suppressed MCC and recurrent lung infections and inflammation. This can explain the emergence of recurrent lung infections and inflammation in people living with HIV. METHODS: Tat-induced aberrant microRNAome was identified by miRNA expression analysis. microRNA mimics and antagomirs were used to validate the identified miRNAs involved in Tat mediated CFTR mRNA suppression. CRISPR-based editing of the miRNA target sites in CFTR 3'UTR was used to determine rescue of CFTR mRNA and function in airway epithelial cell lines and in primary human bronchial epithelial cells exposed to TGF-ß and Tat. FINDINGS: HIV Tat upregulates miR-145-5p and miR-509-3p. The two miRNAs demonstrate co-operative effects in suppressing CFTR. CRISPR-based editing of the miRNA target site preserves CFTR mRNA and function in airway epithelial cells INTERPRETATION: Given the important roles of TGF-ß signaling and the multitude of genes regulated by miRNAs, we demonstrate that CRISPR-based gene-specific microRNA antagonism approach can preserve CFTR mRNA and function in the context of HIV Tat and TGF-ß signaling without suppressing expression of other genes regulated by miR-145-5p.

In the Hamiltonian adaptive resolution simulation method (H-AdResS) it is possible to simulate coexisting atomistic (AT) and ideal gas representations of a physical system that belong to different subdomains within the simulation box. The Hamiltonian includes a field that bridges both models by smoothly switching on (off) the intermolecular potential as particles enter (leave) the AT region. In practice, external one-body forces are calculated and applied to enforce a reference density throughout the simulation box, and the resulting external potential adds up to the Hamiltonian. This procedure suggests an apparent dependence of the final Hamiltonian on the system's thermodynamic state that challenges the method's statistical mechanics consistency. In this paper, we explicitly include an external potential that depends on the switching function. Hence, we build a grand canonical potential for this inhomogeneous system to find the equivalence between H-AdResS and density functional theory (DFT). We thus verify that the external potential inducing a constant density profile is equal to the system's excess chemical potential. Given DFT's one-to-one correspondence between external potential and equilibrium density, we find that a Hamiltonian description of the system is compatible with the numerical implementation based on enforcing the reference density across the simulation box. In the second part of the manuscript, we focus on assessing our approach's convergence and computing efficiency concerning various model parameters, including sample size and solute concentrations. To this aim, we compute the excess chemical potential of water, aqueous urea solutions and Lennard-Jones (LJ) mixtures. The results' convergence and accuracy are convincing in all cases, thus emphasising the method's robustness and capabilities.

Heavy metal distribution in medicinal plants is gaining importance not only as an alternative medicine, but also for possible concern due to effects of metal toxicity. The present study has been focused on emphasizing the heavy metal status and bioaccumulation factors of V, Mn, Fe, Co, Cu, Zn, Se (essential metals) and Cr, Ni, Cd, As and Pb (potentially toxic metals) in medicinal plants grown under two different environmental conditions e.g., near to Khetri copper mine and those in fertile soils of Haridwar, both in India, using Instrumental Neutron Activation Analysis (relative method) and Atomic Absorption Spectrometry. The copper levels in the medicinal plants from Khetri were found to be 3-4 folds higher (31.6-76.5 mg kg(-1)) than those from Haridwar samples (7.40-15.3 mg kg(-1)), which is correlated with very high copper levels (763 mg kg(-1)) in Khetri soil. Among various heavy metals, Cr (2.60-5.92 mg kg(-1)), Cd (1.47-2.97 mg kg(-1)) and Pb (3.97-6.63 mg kg(-1)) are also higher in concentration in the medicinal plants from Khetri. The essential metals like Mn (36.4-69.3 mg kg(-1)), Fe (192-601 mg kg(-1)), Zn (24.9-49.9 mg kg(-1)) and Se (0.13-0.91 mg kg(-1)) and potentially toxic metals like Ni (3.09-9.01 mg kg(-1)) and As (0.41-2.09 mg kg(-1)) did not show much variations in concentration in the medicinal plants from both Khetri and Haridwar. The medicinal plants from Khetri, e.g., Ocimum sanctum, Cassia fistula, Withania somnifera and Azadirachta Indica were found rich in Ca and Mg contents while Aloe barbadensis showed moderately high Ca and Mg. Higher levels of Ca-Mg were found to correlate with Zn (except Azadirachta Indica). The bioaccumulation factors (BAFS) of the heavy metals were estimated to understand the soil-to-plant transfer pattern of the heavy metals. Significantly lower BAF values of Cu and Cr were found in the medicinal plants from Khetri, indicating majority fraction of these metals are precipitated and were immobilized species unsuitable for plant uptake. Overall, Withania somnifera (Ashwagandha) showed very high metal bioaccumulation.

RESUMO

Clustered regularly interspaced palindromic repeats (CRISPR) technique plays a vital role in preclinical modelling of many respiratory diseases. Diseases such as chronic obstructive pulmonary disease (COPD), asthma, acute tracheal bronchitis, pneumonia, tuberculosis, lung cancer, and influenza infection continue to significantly impact human health. CRISPR associated (Cas) proteins, isolated from the immune system of prokaryotes, are one component of a very useful technique to manipulate gene sequences or editing and gene expression with significant implications for respiratory research in the field of molecular biology. CRISPR technology is a promising tool that is easily adaptable for specific editing of DNA sequences of interest with a goal towards modifying or eliminating gene function. Among its many potential applications, CRISPR can be applied to correcting genetic defects as well as for therapeutic approaches for treatment. This review elucidates recent advances in CRISPR-Cas technology in airway diseases.

Little is known about genital mucosal immune response to chlamydial infection in women with or without sequelae (Chlamydia positive women with or without fertility disorders as infertility and multiple spontaneous abortions). Cervical lymphocytes were stimulated with chlamydial EBs and cytokine secretion was determined by ELISA, RT-PCR and ELISPOT assays. Stimulated cervical cells from women with fertility disorders (FD) secrete significantly (P<0.05) higher levels of IL-1beta, IL-6, IL-8 and IL-10 and cells from fertile women secrete significantly higher levels of IL-12 and IFN-gamma compared to other groups. RT-PCR analysis showed similar results for IFN-gamma and IL-12. For IL-10 and IL-4, mRNA expression levels were significantly higher (P<0.05) in cells obtained from women with FD compared to other groups. Results for ELISPOT assay were similar as those of RT-PCR. The results suggest that cytokine secretion profile of cervical cells may decide whether infection does not hamper fertility or will develop fertility disorder.
