RESUMO

The fish acute toxicity test (TG203; OECD, 2019) is frequently used and highly embedded in hazard and risk assessment globally. The test estimates the concentration of a chemical that kills 50% of the fish (LC50) over a 96 h exposure and is considered one of the most severe scientific procedures undertaken. Over the years, discussions at the Organisation for Economic Co-operation and Development (OECD) have resulted in changes to the test which reduce the number of fish used, as well as the development of a (potential) replacement test (TG236, OECD, 2013). However, refinement of the mortality endpoint with an earlier (moribundity) endpoint was not considered feasible during the Test Guideline's (TG) last update in 2019. Several stakeholders met at a UK-based workshop to discuss how TG203 can be refined, and identified two key opportunities to reduce fish suffering: (1) application of clinical signs that predict mortality and (2) shortening the test duration. However, several aspects need to be addressed before these refinements can be adopted. TG203 has required recording of major categories of sublethal clinical signs since its conception, with the option to record more detailed signs introduced in the 2019 update. However, in the absence of guidance, differences in identification, recording and reporting of clinical signs between technicians and laboratories is likely to have generated piecemeal data of varying quality. Harmonisation of reporting templates, and training in clinical sign recognition and recording are needed to standardise clinical sign data. This is critical to enable robust data-driven detection of clinical signs that predict mortality. Discussions suggested that the 96 h duration of TG203 cannot stand up to scientific scrutiny. Feedback and data from UK contract research organisations (CROs) conducting the test were that a substantial proportion of mortalities occur in the first 24 h. Refinement of TG203 by shortening the test duration would reduce suffering (and test failure rate) but requires a mechanism to correct new results to previous 96 h LC50 data. The actions needed to implement both refinement opportunities are summarised here within a roadmap. A shift in regulatory assessment, where the 96 h LC50 is a familiar base for decisions, will also be critical.

A "reproducibility crisis" is widespread across scientific disciplines, where results and conclusions of studies are not supported by subsequent investigation. Here we provide a steroid immunoassay example where human errors generated unreproducible results and conclusions. Our study was triggered by a scientific report citing abnormally high concentrations (means of 4-79 ng L-1) of three natural sex steroids [11-ketotestosterone (11-KT), testosterone (T) and oestradiol (E2)] in water samples collected from two UK rivers over 4 years (2002-6). Furthermore, the data suggested that trout farms were a major source because reported steroid concentrations were 1.3-6 times higher downstream than upstream. We hypothesised that the reported levels were erroneous due to substances co-extracted from the water causing matrix effects (i.e. "false positives") during measurement by enzyme-linked immunoassay (EIA). Thus, in collaboration with three other groups (including the one that had conducted the 2002-6 study), we carried out field sampling and assaying to examine this hypothesis. Water samples were collected in 2010 from the same sites and prepared for assay using an analogous method [C18 solid phase extraction (SPE) followed by extract clean-up with aminopropyl SPE]. Additional quality control ("spiked" and "blank") samples were processed. Water extracts were assayed for steroids using radioimmunoassay (RIA) as well as EIA. Although there were statistically significant differences between EIA and RIA (and laboratories), there was no indication of matrix effects in the EIAs. Both the EIAs and RIAs (uncorrected for recovery) measured all three natural steroids at <0.6 ng L-1 in all river water samples, indicating that the trout farms were not a significant source of natural steroids. The differences between the two studies were considerable: E2 and T concentrations were ca. 100-fold lower and 11-KT ca. 1000-fold lower than those reported in the 2002-6 study. In the absence of evidence for any marked changes in husbandry practice (e.g. stock, diet) or environmental conditions (e.g. water flow rate) between the study periods, we concluded that calculation errors were probably made in the first (2002-6) study associated with confusion between extract and water sample concentrations. The second (2010) study also had several identified examples of calculation error (use of an incorrect standard curve; extrapolation below the minimum standard; confusion of assay dilutions during result work-up; failure to correct for loss during extraction) and an example of sample contamination. Similar and further errors have been noted in other studies. It must be recognised that assays do not provide absolute measurements and are prone to a variety of errors, so published steroid levels should be viewed with caution until independently confirmed.

Marine aquaculture has the potential to increase its contribution to the global food system and provide valuable ecosystem services, but appropriate planning, licensing and regulation systems must be in place to enable sustainable development. At present, approaches vary considerably throughout the world, and several national and regional investigations have highlighted the need for reforms if marine aquaculture is to fulfil its potential. This article aims to map and evaluate the challenges of planning and licensing for growth of sustainable marine aquaculture. Despite the range of species, production systems and circumstances, this study found a number of common themes in the literature; complicated and fragmented approaches to planning and licensing, property rights and the licence to operate, competition for space and marine spatial planning, emerging species and diversifying marine aquaculture production (seaweed production, Integrated Multi-Trophic Aquaculture [IMTA], nutrient and carbon offsetting with aquaculture, offshore aquaculture and co-location and multiuse platforms), and the need to address knowledge gaps and use of decision-support tools. Planning and licensing can be highly complicated, so the UK is used as a case study to show more detailed examples that highlight the range of challenges and uncertainty that industry, regulators and policymakers face across interacting jurisdictions. There are many complexities, but this study shows that many countries have undergone, or are undergoing, similar challenges, suggesting that lessons can be learned by sharing knowledge and experiences, even across different species and production systems, rather than having a more insular focus.

Mortality has received insufficient attention as a fish welfare topic. Here, we aim to prompt fish farming stakeholders to discuss fish mortalities in relation to welfare. Mortality in farmed fish populations is due to a variety of biotic and abiotic causes, although it is often difficult to differentiate between underlying and immediate causes of mortality. Most mortality appears to occur during episodes associated with disease outbreaks and critical periods (in development or production). Most causes of mortality can be assumed to be associated with suffering prior to death. As mortality rates in farmed fish populations are suspected to rank amongst the highest in commonly farmed vertebrate species, mortality should be a principal fish welfare issue. Long-term mortality rates can be used as a retrospective welfare performance indicator and short-term mortality rates as an operational welfare indicator. Scrutiny of mortality records and determining causes of death will enable action to be taken to avoid further preventable mortality. The welfare performance of fish farms should only be judged on levels of predictable and preventable mortality. Fish farmers will already be monitoring mortality due to commercial and legal requirements. As profitability in fish farming is directly linked to survival, confronting mortality should ultimately benefit both fish and farmers.

Previous reviews of stress, and the stress hormone cortisol, in fish have focussed on physiology, due to interest in impacts on aquaculture production. Here, we discuss cortisol in relation to fish welfare. Cortisol is a readily measured component of the primary (neuroendocrine) stress response and is relevant to fish welfare as it affects physiological and brain functions and modifies behaviour. However, we argue that cortisol has little value if welfare is viewed purely from a functional (or behavioural) perspective-the cortisol response itself is a natural, adaptive response and is not predictive of coping as downstream impacts on function and behaviour are dose-, time- and context-dependent and not predictable. Nevertheless, we argue that welfare should be considered in terms of mental health and feelings, and that stress in relation to welfare should be viewed as psychological, rather than physiological. We contend that cortisol can be used (with caution) as a tractable indicator of how fish perceive (and feel about) their environment, psychological stress and feelings in fish. Cortisol responses are directly triggered by the brain and fish studies do indicate cortisol responses to psychological stressors, i.e., those with no direct physicochemical action. We discuss the practicalities of using cortisol to ask the fish themselves how they feel about husbandry practices and the culture environment. Single time point measurements of cortisol are of little value in assessing the stress level of fish as studies need to account for diurnal and seasonal variations, and environmental and genetic factors. Areas in need of greater clarity for the use of cortisol as an indicator of fish feelings are the separation of (physiological) stress from (psychological) distress, the separation of chronic stress from acclimation, and the interactions between feelings, cortisol, mood and behaviour.

Appropriate end-points are integral to the refinement of laboratory animal experiments. Our recent experience has highlighted that ambiguity around end-points is hampering their adoption in experiments that cause severe suffering to fish. In toxicology, the term endpoint (single word) refers to the response variable to the treatment that is measured and analysed. This differs to usage within laboratory animal experimentation, where end-point (hyphenated) refers to the time-point when exposure of the animal(s) to the treatment (and suffering) ends. Within laboratory animal experimentation, standardised terminology is needed for different types of early end-point which are based on the condition of the animal(s) or progress of the experiment. We propose that those involved in regulating and conducting animal experiments consider seven distinct types of early end-point (aim, technical error, biological error, mortality, moribundity, prognostic humane, non-prognostic humane) in addition to the planned experimental end-point (i.e. maximum duration). Moribundity (not morbidity) refers to an animal in a severely debilitated state close to death. Moribundity in fish is not yet defined, so we propose identification via a lack of response to external stimuli, loss of equilibrium (i.e. loss of righting reflex), and a slow opercular ventilation rate. As these clinical signs equate to those of deep/surgical anaesthesia, this moribundity end-point cannot be considered a humane end-point as the fish is likely to be unconscious and have passed the point of maximum suffering. We believe that identification of earlier humane end-points based on clinical signs and wider recognition of other types of early end-point can reduce suffering in experiments.

PbSO4 is a key component in the charging and discharging of lead acid batteries-such as the cycling of automotive batteries. PbSO4 is a poor conductor that forms on the positive and negative electrodes during discharging and dissolves during charging of a lead acid battery. Over time, buildup of PbSO4 occurs on the electrodes, ultimately reducing the efficiency of the battery. This study aims to determine the nucleation and growth mechanisms of PbSO4 nanoparticles in various solutions to potentially reduce or control the buildup of PbSO4 on battery electrodes over time. The time dependency of particle morphology was observed using various reaction conditions. PbSO4 particles were created using premixed solutions at various times of reaction. H2O, acetone, methanol, ethanol, and isopropanol were used to stop the reaction and development of the PbSO4 particles. The structure of the nanoparticles was characterized via transmission electron microscopy, high-angle annular dark field scanning transmission electron microscopy, and selected area electron diffraction. This study provides insight into the mechanism by which PbSO4 nanoparticles form in various solutions and reveals that the degree of complexity of the solution plays a large role in the nucleation and growth of the PbSO4 nanoparticles. This insight can provide avenues to reduce unwanted buildup of PbSO4 on battery electrodes over time, which can extend battery life and performance.

BACKGROUND: Fish farmed under high intensity aquaculture conditions are subjected to unnatural environments that may cause stress. Therefore awareness of how to maintain good health and welfare of farmed fish is important. For Atlantic salmon held in sea cages, water flow, dissolved oxygen (DO) levels and temperature will fluctuate over time and the fish can at times be exposed to detrimentally low DO levels and high temperatures. This experimental study investigates primary and secondary stress responses of Atlantic salmon post smolts to long-term exposure to reduced and fluctuating DO levels and high water temperatures, mimicking situations in the sea cages. Plasma cortisol levels and cortisol release to the water were assessed as indicators of the primary stress response and intestinal barrier integrity and physiological functions as indicators of secondary responses to changes in environmental conditions. RESULTS: Plasma cortisol levels were elevated in fish exposed to low (50% and 60% saturation) DO levels and low temperature (9°C), at days 9, 29 and 48. The intestinal barrier function, measured as electrical resistance (TER) and permeability of mannitol at the end of the experiment, were reduced at 50% DO, in both proximal and distal intestine. When low DO levels were combined with high temperature (16°C), plasma cortisol levels were elevated in the cyclic 1:5 h at 85%:50% DO group and fixed 50% DO group compared to the control (85% DO) group at day 10 but not at later time points. The intestinal barrier function was clearly disturbed in the 50% DO group; TER was reduced in both intestinal regions concomitant with increased paracellular permeability in the distal region. CONCLUSIONS: This study reveals that adverse environmental conditions (low water flow, low DO levels at low and high temperature), that can occur in sea cages, elicits primary and secondary stress responses in Atlantic salmon post smolts. The intestinal barrier function was significantly affected by prolonged hypoxic stress even when no primary stress response was observed. This suggests that intestinal barrier function is a good experimental marker for evaluation of chronic stress and that it can be a valuable tool to study the impact of various husbandry conditions on health and welfare of farmed Atlantic salmon.

OBJECTIVES: Diabetes is associated with loss of capillaries and macrovessel dilation in the conjunctiva, similar to well-known vessel changes in the retina. However, little is known about the effect of diabetes on the tortuosity of vessels of the conjunctiva. The authors examined the tortuosity of conjunctival vessels in participants with and without diabetes. DESIGN: Case-control study. PARTICIPANTS AND CONTROLS: Fifty-three patients with diabetes (17 with type 1 diabetes, 36 with type 2 diabetes) and 60 controls (all aged 20-94 years). METHODS: Digital red-free images of conjunctivae were analyzed using an automated computer algorithm to identify vessel axes and to quantify vessel tortuosity. Differences in vessel tortuosity were adjusted for age, gender, blood pressure, and smoking status. MAIN OUTCOME MEASURES: Tortuosity was expressed in units of curve energy (the square of the radian angular change between subsequent locations identified by the algorithm, standardized by vessel length). RESULTS: A longer duration of diabetes was associated with a reduction in overall vessel tortuosity (-2.8%; 95% confidence interval [CI], -4.3% to -1.3% per decade). This inverse association was driven by changes in larger vessels (40 microm in width or more), whereas increased tortuosity was observed in capillary sized vessels (<25 microm, 4.0%; 95% CI, -0.2% to 8.2% per decade). Compared with controls, those with type 1 diabetes (median duration of disease, 26 years) showed a 17.9% increase (95% CI, 4.7% to -31.0%) in capillary tortuosity. Conversely, those with type 1 diabetes showed a 7% decrease (95% CI, -11.8% to -2.3%) in tortuosity among vessels 40 to 80 microm or less in size and a 26.8% decrease (95% CI, -66.2% to 12.7%) in the fewer number of vessels more than 80 microm in size compared with controls. Similar, but smaller differences were seen in those with type 2 diabetes with shorter duration of diabetes (median, 7 years). CONCLUSIONS: Macrovessel dilation associated with diabetes may result in vessel engorgement and straightening, especially among those with longer durations of disease. Increased tortuosity associated with diabetes among conjunctival capillaries mirrors established vessel changes observed in the retina. Conjunctival angiopathy associated with diabetes may contribute to susceptibility to anterior eye disease among patients with diabetes.

We present a new analysis and visualization method for studying the functional relationship between the pulse morphology of pressure signals and time or signal metrics such as heart rate, pulse pressure, and means of pressure signals, such as arterial blood pressure and central venous pressure. The pulse morphology is known to contain potentially useful clinical information, but it is difficult to study in the time domain without the aid of a tool such as the method we present here. The primary components of the method are established signal processing techniques, nonparametric regression, and an automatic beat detection algorithm. Some of the insights that can be gained from this are demonstrated through the analysis of intracranial pressure signals acquired from patients with traumatic brain injuries. The analysis indicates the point of transition from low-pressure morphology consisting of three distinct peaks to a high-pressure morphology consisting of a single peak. In addition, we demonstrate how the analysis can reveal distinctions in the relationship between morphology and several signal metrics for different patients.

OBJECTIVE: Retinovascular changes associated with diabetes have been clearly documented; changes in vessels of the conjunctiva are less well described. We examined changes in conjunctival vessel morphologic features in participants with and without diabetes. DESIGN: Case-control study. PARTICIPANTS AND CONTROLS: Fifty-three patients with diabetes (17 with type 1 diabetes, 36 with type 2 diabetes) and 60 controls (all aged 20-94 years). METHODS: Digital red-free conjunctival images were captured and an automated computer algorithm was used to derive indices that describe the morphologic features of vessels of the conjunctiva. Percentage differences in vessel indices were adjusted for age, gender, blood pressure, and smoking status. MAIN OUTCOME MEASURES: Mean vessel diameter (micrometers) and vessel density (square millimeters of vessel per square centimeter of bulbar conjunctiva). RESULTS: A strong positive association between the duration of diabetes and overall mean vessel width was observed (P<0.001), resulting from changes in larger vessels (>80 mum in width). Conversely, the duration of diabetes showed a strong inverse association with vessel area (P<0.001) that appeared to be driven by the trend observed in smaller vessels (<40 mum in width). A 25% reduction (95% confidence interval [CI], -35% to -13%; P<0.001) in vessel density in those with type 1 diabetes and a 14% reduction (95% CI, -24% to -3%; P = 0.016) in those with type 2 diabetes, compared with controls, was observed. Mean vessel widths were 11% (95% CI, 4%-17%; P = 0.001) wider in type 1 and 5% (95% CI, 0%-10%; P = 0.073) wider in those with type 2 diabetes compared with controls. The difference in magnitude of effect for type 1 and type 2 diabetes compared with controls was explained by duration of diabetes. Grade of diabetic retinopathy and elevated blood pressure showed similar but less strong associations with vessel indices. CONCLUSIONS: Loss of capillaries and macrovessel dilation in the conjunctiva associated with diabetes compares with well-known vessel changes in the retina. Associations between morphologic changes in the conjunctiva and elevated blood pressure were similar but less strong; this may show that diabetic angiopathy predominates in those with both diabetes and elevated blood pressure.

RESUMO

This paper considers the problem of automatically learning an activity-based semantic scene model from a stream of video data. A scene model is proposed that labels regions according to an identifiable activity in each region, such as entry/exit zones, junctions, paths, and stop zones. We present several unsupervised methods that learn these scene elements and present results that show the efficiency of our approach. Finally, we describe how the models can be used to support the interpretation of moving objects in a visual surveillance environment.

Artificial light at night (ALAN) is gaining recognition as having an important anthropogenic impact on the environment, yet the behavioural and physiological impacts of this stressor are largely unknown. This dearth of information is particularly true for freshwater ecosystems, which are already heavily impacted by anthropogenic pressures. Atlantic salmon (Salmo salar L.) is a species of conservation and economic importance whose ecology and behaviour is well studied, making it an ideal model species. Recent investigations have demonstrated that salmon show disrupted behaviour in response to artificial light; however, it is not yet clear which physiological processes are behind the observed behavioural modifications. Here, two novel non-invasive sampling methods were used to examine the cortisol stress response of dispersing salmon fry under different artificial lighting intensities. Fish egg and embryos were reared under differing ALAN intensities and individual measures of stress were subsequently taken from dispersing fry using static sampling, whereas population-level measures were achieved using deployed passive samplers. Dispersing fry exposed to experimental confinement showed elevated cortisol levels, indicating the capacity to mount a stress response at this early stage in ontogenesis. However, only one of the two methods for sampling cortisol used in this study indicated that ALAN may act as a stressor to dispersing salmon fry. As such, a cortisol-mediated response to light was not strongly supported. Furthermore, the efficacy of the two non-invasive methodologies used in this study is, subject to further validation, indicative of them proving useful in future ecological studies.

Interpretation of plasma cortisol levels in wild-caught fish is confounded by the stress of capture. Measurement of cortisol metabolites in fish bile could provide a method for assessing the stress level of wild fish because the time-lag for metabolism, conjugation and excretion into bile avoids the effects of sampling stress. To determine which biliary metabolite(s) to target, four Atlantic cod, Gadus morhua L., were injected with radioactive cortisol. After 22 h, the bile was collected and found to contain 30% of the injected activity. Cortisol metabolites were extracted from diluted bile samples using solid phase extraction and the radioactive metabolites separated by several different chromatographic procedures. The metabolites were predominantly present as sulfates (95%) with the remainder being glucuronidated. Chromatography split the sulfates into at least seven peaks, and acid solvolysis (which removes sulfate groups from steroids) generated four major radioactive steroids. These were identified, using microchemical reactions and re-crystallization to constant specific activity, as: 11ß,17,21-trihydroxypregn-4-ene-3,20-dione (cortisol), 3α,11ß,17,21-tetrahydroxy-5ß-pregnan-20-one (tetrahydrocortisol; THF), 3α,17,21-trihydroxy-5ß-pregnane-11,20-dione (tetrahydrocortisone; THE) and 3α,17,20ß,21-tetrahydroxy-5ß-pregnan-11-one (ß-cortolone). The last of these was the most abundant, and thus a likely target for a biliary stress assay. Studies were also carried out to determine the best method for extraction and solvolysis of sulfates. Solid phase extraction (i.e. using octadecylsilane) was found to be too unreliable for routine use. Even though the extraction efficiency could be improved by acidifying the bile, this caused premature solvolysis of sulfated steroids. Acid solvolysis of unextracted bile worked best (c. 90% converted to free steroids) on volumes that were 1 µL or lower. Aryl sulfatase digestion of unextracted bile did not work well (only 20% of radioactivity was converted to free steroids).

RESUMO

Computer-Aided Tomography Angiography (CTA) images are the standard for assessing Peripheral artery disease (PAD). This paper presents a Computer Aided Detection (CAD) and Computer Aided Measurement (CAM) system for PAD. The CAD stage detects the arterial network using a 3D region growing method and a fast 3D morphology operation. The CAM stage aims to accurately measure the artery diameters from the detected vessel centerline, compensating for the partial volume effect using Expectation Maximization (EM) and a Markov Random field (MRF). The system has been evaluated on phantom data and also applied to fifteen (15) CTA datasets, where the detection accuracy of stenosis was 88% and the measurement accuracy was with an 8% error.

Measurement of tree root systems by conventional methods is a Herculean task. The electrical capacitance method offers a rapid and non-destructive alternative, but it has largely been restricted to herbaceous species. The Dalton Model has been the main concept for understanding equivalent root circuitry; it proposed that roots were cylindrical capacitors with epidermis and xylem being the external and internal electrodes. Capacitance (C) therefore varied in proportion to root surface area (A), mass (M), length (L) and relative permittivity of the plant tissue ε(r). We used the capacitance method on forest and plantation trees (13 to circa 100 y.o.) in situ to test hypotheses derived from implicit assumptions about tree-root-soil circuitry. We concluded: C was not confounded by intermingled root systems; C was strongly related to diameter at breast height (DBH); C was less strongly related to DBH for multiple species at the same site; and C was a poor indicator of DBH, M and L across species, ages and sites. We proposed that ε(r) was proportional to root tissue density ρ and fitted a model with P < 0.05 and R(2) = 0.70 when the three immature (13 y.o.) trees were excluded. There was no significant difference (P = 0.28) between the parameters of the tree model (excluding the immature trees) and one of the same form fitted to data from bean (Vicia faba L.; R(2) = 0.55). Together, the data sets suggested (R(2) = 0.94; n = 26) that there may exist a general relationship of this form applied over two orders of magnitude of L.

Land use (and land management) change is seen as the primary factor responsible for changes in sediment and nutrient delivery to water bodies. Understanding how sediment and nutrient (or constituent) concentrations vary with land use is critical to understanding the current and future impact of land use change on aquatic ecosystems. Access to appropriate land-use based water quality data is also important for calculating reliable load estimates using water quality models. This study collated published and unpublished runoff, constituent concentration and load data for Australian catchments. Water quality data for total suspended sediments (TSS), total nitrogen (TN) and total phosphorus (TP) were collated from runoff events with a focus on catchment areas that have a single or majority of the contributing area under one land use. Where possible, information on the dissolved forms of nutrients were also collated. For each data point, information was included on the site location, land use type and condition, contributing catchment area, runoff, laboratory analyses, the number of samples collected over the hydrograph and the mean constituent concentration calculation method. A total of ∼750 entries were recorded from 514 different geographical sites covering 13 different land uses. We found that the nutrient concentrations collected using "grab" sampling (without a well defined hydrograph) were lower than for sites with gauged auto-samplers although this data set was small and no statistical analysis could be undertaken. There was no statistically significant difference (p<0.05) between data collected at plot and catchment scales for the same land use. This is most likely due to differences in land condition over-shadowing the effects of spatial scale. There was, however, a significant difference in the concentration value for constituent samples collected from sites where >90% of the catchment was represented by a single land use, compared to sites with <90% of the upstream area represented by a single land use. This highlights the need for more single land use water quality data, preferably over a range of spatial scales. Overall, the land uses with the highest median TSS concentrations were mining (∼50,000mg/l), horticulture (∼3000mg/l), dryland cropping (∼2000mg/l), cotton (∼600mg/l) and grazing on native pastures (∼300mg/l). The highest median TN concentrations are from horticulture (∼32,000µg/l), cotton (∼6500µg/l), bananas (∼2700µg/l), grazing on modified pastures (∼2200µg/l) and sugar (∼1700µg/l). For TP it is forestry (∼5800µg/l), horticulture (∼1500µg/l), bananas (∼1400µg/l), dryland cropping (∼900mg/l) and grazing on modified pastures (∼400µg/l). For the dissolved nutrient fractions, the sugarcane land use had the highest concentrations of dissolved inorganic nitrogen (DIN), dissolved organic nitrogen (DON) and dissolved organic phosphorus (DOP). Urban land use had the highest concentrations of dissolved inorganic phosphorus (DIP). This study provides modellers and catchment managers with an increased understanding of the processes involved in estimating constituent concentrations, the data available for use in modelling projects, and the conditions under which they should be applied. Areas requiring more data are also discussed.
