RESUMO

Nitrogen (N) resorption is an important pathway of N conservation, contributing to an important proportion of plant N requirement. However, whether the ratio of N resorption to N requirement may be affected by environmental factors, mycorrhizal types or atmospheric CO2 concentration remains unclear. Here, we conducted a meta-analysis on the impacts of environmental factors and mycorrhizal types on this ratio. We found this ratio in ectomycorrhizal (EM) trees decreased with mean annual precipitation, mean annual temperature, soil total N content and atmospheric CO2 concentration and was significantly lower than that in arbuscular mycorrhizal (AM) trees. An in situ 15 N tracing experiment further confirmed that AM trees have a stronger reliance on N resorption than EM trees. Our study suggests that AM and EM trees potentially have different strategies for alleviation of progressive N limitation, highlighting the necessity of incorporating plant mycorrhizal types into Earth System Models.

The link between above- and belowground communities is a key uncertainty in drought and rewetting effects on forest carbon (C) cycle. In young beech model ecosystems and mature naturally dry pine forest exposed to 15-yr-long irrigation, we performed 13C pulse labeling experiments, one during drought and one 2 wk after rewetting, tracing tree assimilates into rhizosphere communities. The 13C pulses applied in tree crowns reached soil microbial communities of the young and mature forests one and 4 d later, respectively. Drought decreased the transfer of labeled assimilates relative to the irrigation treatment. The 13C label in phospholipid fatty acids (PLFAs) indicated greater drought reduction of assimilate incorporation by fungi (-85%) than by gram-positive (-43%) and gram-negative bacteria (-58%). 13C label incorporation was more strongly reduced for PLFAs (cell membrane) than for microbial cytoplasm extracted by chloroform. This suggests that fresh rhizodeposits are predominantly used for osmoregulation or storage under drought, at the expense of new cell formation. Two weeks after rewetting, 13C enrichment in PLFAs was greater in previously dry than in continuously moist soils. Drought and rewetting effects were greater in beech systems than in pine forest. Belowground C allocation and rhizosphere communities are highly resilient to drought.

Whether nitrogen (N) availability will limit plant growth and removal of atmospheric CO2 by the terrestrial biosphere this century is controversial. Studies have suggested that N could progressively limit plant growth, as trees and soils accumulate N in slowly cycling biomass pools in response to increases in carbon sequestration. However, a question remains over whether longer-term (decadal to century) feedbacks between climate, CO2 and plant N uptake could emerge to reduce ecosystem-level N limitations. The symbioses between plants and microbes can help plants to acquire N from the soil or from the atmosphere via biological N2 fixation-the pathway through which N can be rapidly brought into ecosystems and thereby partially or completely alleviate N limitation on plant productivity. Here we present measurements of plant N isotope composition (δ15 N) in a peat core that dates to 15,000 cal. year BP to ascertain ecosystem-level N cycling responses to rising atmospheric CO2 concentrations. We find that pre-industrial increases in global atmospheric CO2 concentrations corresponded with a decrease in the δ15 N of both Sphagnum moss and Ericaceae when constrained for climatic factors. A modern experiment demonstrates that the δ15 N of Sphagnum decreases with increasing N2 -fixation rates. These findings suggest that plant-microbe symbioses that facilitate N acquisition are, over the long term, enhanced under rising atmospheric CO2 concentrations, highlighting an ecosystem-level feedback mechanism whereby N constraints on terrestrial carbon storage can be overcome.

Drought alters carbon (C) allocation within trees, thereby impairing tree growth. Recovery of root and leaf functioning and prioritized C supply to sink tissues after drought may compensate for drought-induced reduction of assimilation and growth. It remains unclear if C allocation to sink tissues during and following drought is controlled by altered sink metabolic activities or by the availability of new assimilates. Understanding such mechanisms is required to predict forests' resilience to a changing climate. We investigated the impact of drought and drought release on C allocation in a 100-y-old Scots pine forest. We applied 13CO2 pulse labeling to naturally dry control and long-term irrigated trees and tracked the fate of the label in above- and belowground C pools and fluxes. Allocation of new assimilates belowground was ca. 53% lower under nonirrigated conditions. A short rainfall event, which led to a temporary increase in the soil water content (SWC) in the topsoil, strongly increased the amounts of C transported belowground in the nonirrigated plots to values comparable to those in the irrigated plots. This switch in allocation patterns was congruent with a tipping point at around 15% SWC in the response of the respiratory activity of soil microbes. These results indicate that the metabolic sink activity in the rhizosphere and its modulation by soil moisture can drive C allocation within adult trees and ecosystems. Even a subtle increase in soil moisture can lead to a rapid recovery of belowground functions that in turn affects the direction of C transport in trees.

Soil microbial biomass and microbial stoichiometric ratios are important for understanding carbon and nutrient cycling in terrestrial ecosystems. Here, we compiled data from 12245 observations of soil microbial biomass from 1626 published studies to map global patterns of microbial biomass carbon (MBC), microbial biomass nitrogen (MBN), microbial biomass phosphorus (MBP), and their stoichiometry using a random forest model. Concentrations of MBC, MBN, and MBP were most closely linked to soil organic carbon, while climatic factors were most important for stoichiometry in microbial biomass ratios. Modeled seasonal MBC concentrations peaked in summer in tundra and in boreal forests, but in autumn in subtropical and in tropical biomes. The global mean MBC/MBN, MBC/MBP, and MBN/MBP ratios were estimated to be 10, 48, and 6.7, respectively, at 0-30 cm soil depth. The highest concentrations, stocks, and microbial C/N/P ratios were found at high latitudes in tundra and boreal forests, probably due to the higher soil organic matter content, greater fungal abundance, and lower nutrient availability in colder than in warmer biomes. At 30-100 cm soil depth, concentrations of MBC, MBN, and MBP were highest in temperate forests. The MBC/MBP ratio showed greater flexibility at the global scale than did the MBC/MBN ratio, possibly reflecting physiological adaptations and microbial community shifts with latitude. The results of this study are important for understanding C, N, and P cycling at the global scale, as well as for developing soil C-cycling models including soil microbial C, N, and P as important parameters.

In this work, a convenient and dual-signal readout optical sensing platform for the sensitively and selectively determination of beta-glucosidase (ß-Glu) activity was reported using protein-inorganic hybrid nanoflowers [BSA-Cu3(PO4)2·3H2O] possessing peroxidase-mimicking activity. The nanoflowers (NFs) were facilely synthesized through a self-assembled synthesis strategy at room temperature. The as-prepared NFs could catalytically convert the colorless and non-fluorescent Amplex Red into colored and highly fluorescent resorufin in the presence of hydrogen peroxide via electron transfer process. ß-Glu could hydrolyze cyanogenic glycoside, using amygdalin (Amy) as a model, into cyanide ions (CN-), which can subsequently efficiently suppress the catalytic activity of NFs, accompanied with the fluorescence decrease and the color fading. The concentration of CN- was controlled by ß-Glu-triggered enzymatic reaction of Amy. Thus, a sensing system was established for fluorescent and visual determination of ß-Glu activity. Under the optimum conditions, the present fluorescent and visual bimodal sensing platform exhibited good sensitivity for ß-Glu activity assay with a detection limit of 0.33 U·L-1. The sensing platform was further applied to determinate ß-Glu in real samples and satisfactory results were attained. Additionally, the optical sensing system can potentially be a promising candidate for ß-Glu inhibitors screening.

Above and belowground compartments in ecosystems are closely coupled on daily to annual timescales. In mature forests, this interlinkage and how it is impacted by drought is still poorly understood. Here, we pulse-labelled 100-year-old trees with 13 CO2 within a 15-year-long irrigation experiment in a naturally dry pine forest to quantify how drought regime affects the transfer and use of assimilates from trees to the rhizosphere and associated microbial communities. It took 4 days until new 13 C-labelled assimilates were allocated to the rhizosphere. One year later, the 13 C signal of the 3-h long pulse labelling was still detectable in stem and soil respiration, which provides evidence that parts of the assimilates are stored in trees before they are used for metabolic processes in the rhizosphere. Irrigation removing the natural water stress reduced the mean C residence time from canopy uptake until soil respiration from 89 to 40 days. Moreover, irrigation increased the amount of assimilates transferred to and respired in the soil within the first 10 days by 370%. A small precipitation event rewetting surface soils altered this pattern rapidly and reduced the effect size to +35%. Microbial biomass incorporated 46 ± 5% and 31 ± 7% of the C used in the rhizosphere in the dry control and irrigation treatment respectively. Mapping the spatial distribution of soil-respired 13 CO2 around the 10 pulse-labelled trees showed that tree rhizospheres extended laterally 2.8 times beyond tree canopies, implying that there is a strong overlap of the rhizosphere among adjacent trees. Irrigation increased the rhizosphere area by 60%, which gives evidence of a long-term acclimation of trees and their rhizosphere to the drought regime. The moisture-sensitive transfer and use of C in the rhizosphere has consequences for C allocation within trees, soil microbial communities and soil carbon storage.

Altered freeze-thaw cycle (FTC) patterns due to global climate change may affect nitrogen (N) cycling in terrestrial ecosystems. However, the general responses of soil N pools and fluxes to different FTC patterns are still poorly understood. Here, we compiled data of 1519 observations from 63 studies and conducted a meta-analysis of the responses of 17 variables involved in terrestrial N pools and fluxes to FTC. Results showed that under FTC treatment, soil NH4+ , NO3- , NO3- leaching, and N2 O emission significantly increased by 18.5%, 18.3%, 66.9%, and 144.9%, respectively; and soil total N (TN) and microbial biomass N (MBN) significantly decreased by 26.2% and 4.7%, respectively; while net N mineralization or nitrification rates did not change. Temperate and cropland ecosystems with relatively high soil nutrient contents were more responsive to FTC than alpine and arctic tundra ecosystems with rapid microbial acclimation. Therefore, altered FTC patterns (such as increased duration of FTC, temperature of freeze, amplitude of freeze, and frequency of FTC) due to global climate warming would enhance the release of inorganic N and the losses of N via leaching and N2 O emissions. Results of this meta-analysis help better understand the responses of N cycling to FTC and the relationships between FTC patterns and N pools and N fluxes.

Increasing atmospheric reactive nitrogen (N) deposition due to human activities could change N cycling in terrestrial ecosystems. However, the differences between the fates of deposited NH4+ and NO3- are still not fully understood. Here, we investigated the fates of deposited NH4+ and NO3-, respectively, via the application of 15 NH4 NO3 and NH415 NO3 in a temperate forest ecosystem. Results showed that at 410 days after tracer application, most 15NH4+ was immobilized in litter layer (50 ± 2%), while a considerable amount of 15NO3- penetrated into 0-5 cm mineral soil (42 ± 2%), indicating that litter layer and 0-5 cm mineral soil were the major N sinks of NH4+ and NO3-, respectively. Broad-leaved trees assimilated more 15 N under NH415 NO3 treatment compared to under 15 NH4 NO3 treatment, indicating their preference for NO3--N. At 410 days after tracer application, 16 ± 4% added 15 N was found in aboveground biomass under 15NO3- treatment, which was twice more than that under 15NH4+ treatment (6 ± 1%). At the same time, approximately 80% added 15 N was recovered in soil and plants under both treatments, which suggested that this forest had high potential for retention of deposited N. These results provided evidence that there were great differences between the fates of deposited NH4+ and NO3-, which could help us better understand the mechanisms and capability of forest ecosystems as a sink of reactive nitrogen.

Nitrogen cycling in terrestrial ecosystems is critical for biodiversity, vegetation productivity and biogeochemical cycling. However, little is known about the response of functional nitrogen cycle genes to global change factors in soils under different land uses. Here, we conducted a multiple hierarchical mixed effects meta-analyses of global change factors (GCFs) including warming (W+), mean altered precipitation (MAP+/-), elevated carbon dioxide concentrations (eCO2), and nitrogen addition (N+), using 2706 observations extracted from 200 peer-reviewed publications. The results showed that GCFs had significant and different effects on soil microbial communities under different types of land use. Under different land use types, such as Wetland, Tundra, Grassland, Forest, Desert and Agriculture, the richness and diversity of soil microbial communities will change accordingly due to differences in vegetation cover, soil management practices and environmental conditions. Notably, soil bacterial diversity is positively correlated with richness, but soil fungal diversity is negatively correlated with richness, when differences are driven by GCFs. For functional genes involved in nitrification, eCO2 in agricultural soils and the interaction of N+ with other GCFs in grassland soils stimulate an increase in the abundance of the AOA-amoA gene. In agricultural soil, MAP+ increases the abundance of nifH. W+ in agricultural soils and N+ in grassland soils decreased the abundance of nifH. The abundance of the genes nirS and nirK, involved in denitrification, was mainly negatively affected by W+ and positively affected by eCO2 in agricultural soil, but negatively affected by N+ in grassland soil. This meta-analysis was important for subsequent research related to global climate change. Considering data limitations, it is recommended to conduct multiple long-term integrated observational experiments to establish a scientific basis for addressing global changes in this context.

While freeze-thaw cycle (FTC) can influence greenhouse gas emissions, the specific greenhouse gas that responds most strongly to FTC, as well as the underlying mechanisms, remain unclear. Here, we conducted a meta-analysis to explore the responses of global warming potential (GWP) and the fluxes of CO2 and N2O to FTC. Our results showed that FTC treatment significantly increased GWP, N2O flux, cumulative GWP, and cumulative N2O emissions by 23.1 %, 53.2 %, 14.5 %, and 164.6 %, respectively, but did not affect CO2 flux, indicating that the enhanced GWP during the FTC period may be primarily due to the contribution of N2O flux rather than CO2 flux. The responses of GWP (+68.6 %), CO2 (+21.0 %), and N2O fluxes (+136.3 %) in croplands was higher than those in other ecosystems, exhibiting a strong dependence on ecosystem types. The effect size of FTC treatment on greenhouse gas emissions escalated with decreasing freezing temperature and diminished with increasing FTC frequency. Moreover, mean annual temperature (MAT) and FTC patterns were key factors influencing GWP during the FTC period. These findings provide critical insights into the variations in greenhouse gas emissions due to FTC and its influencing factors, allowing for more accurate predictions of the future impact of global climate change on GWP.

Soil stoichiometry of carbon (C), nitrogen (N), and phosphorus (P) are indicators for nutrient balance. Shrub encroachment into grasslands could change nutrient concentrations and stoichiometry in soils, but the general patterns remain unclear. With a meta-analysis of a global dataset covering 344 observations from 68 studies, we examined the responses of grassland soil C:N:P stoichiometry to shrub encroachment under various environmental conditions. Our results show that: 1) Shrub encroachment significantly increased the concentrations of soil C (+29 %), N (+25 %), P (+20 %), C:N (+5 %), C:P (+12 %), and N:P (+6 %). The magnitude of such effects varied with climate, soil texture, and soil layer. 2) Increases in SOC and TN concentrations mainly occurred in Mediterranean and very humid climate zones. Soil C:P and N:P decreased in semi-humid climate zone after shrub encroachment. 3) The increases in SOC and TN concentrations and in the C:N, C:P, and N:P ratios after shrub encroachment were greater in the topsoil than in deeper soil layers. 4) Both finest-textured soil (clay) and coarsest-textured soil (sand) are beneficial for increase of soil nutrient concentrations following shrub encroachment. 5) The magnitude of the change in soil C:N was negatively correlated with the duration of shrub encroachment, due to greater increases in soil TN than in SOC concentrations with longer durations of encroachment. Our results indicate that soil stoichiometric shifts in shrub-encroached grasslands are relatively sensitive to environmental factors, including soil texture, soil pH, and climate. These findings help us to better understand the effects of shrub encroachment on biogeochemical cycling, functioning, and services in grasslands across a broad range of spatio-temporal scales.

Acquired Immune Deficiency Syndrome (AIDS) has become one of the most severe public health issues and nowadays around 38 million people are living with the human immunodeficiency virus (HIV). Ensuring healthy lives and promoting well-being is one of 17 United Nations Sustainable Development Goals. Here, we used the Markov chain matrix and geospatial clustering to comprehensively quantify the trends of the AIDS epidemic at the provincial administrate level in the mainland of China from 2005 to 2017. The Geographically Weighted Regression (GWR) model was further adopted to explore four groups of potential influencing factors (i.e., economy, traffic and transportation, medical care, and education) of the AIDS incidence rate in 2017 and their spatially distributed patterns. Results showed that the AIDS prevalence in southeastern China had been dominant and become prevalent in the past decade. The AIDS intensity level had been increasing between 2008 and 2011 but been gradually decreasing afterward. The analysis of the Markov chain matrix indicated that the AIDS epidemic has been generally in control on the Chinese mainland. The economic development was closely related to the rate of AIDS incidence on the Chinese mainland. The GWR result further suggested that medical care and the education effects on AIDS incidence rate can vary with different regions, but significant conclusions cannot be directly demonstrated. Our findings contribute an analytical framework of understanding AIDS epidemic trends and spatial variability of potential underlying factors throughout a complex extent to customize scientific prevention.

As an important factor regulating soil carbon cycle, freeze-thaw cycle significantly affects winter soil respiration in temperate regions. However, few in situ studies have been carried out to evaluate the effect of freeze-thaw cycle on soil respiration. Here, a field experiment was conducted to explore the response of winter soil respiration to freeze-thaw cycle and the underlying mechanisms in larch and Chinese pine plantation forests in a mid-temperate region. These results indicated that CO2 emissions during the freeze-thaw period accounted for 18.89-18.94% and 0.79-1.00% of the cumulative winter CO2 emissions and the annual soil CO2 emissions, respectively. Soil respiration rates during the thawing phase were 1.54-3.95 times higher than those during the freezing phase, which was mainly due to the increase of soil microbial biomass upon thawing. This effect declined during the second freeze-thaw cycle compared to the first freeze-thaw cycle due to the exhaustion of resources for microbes. The different responses of soil CO2 flux to freeze-thaw cycle between the two types of forests were mainly because of the difference in the thickness of litter layer, which plays an important role in regulating soil temperature and enzyme activity. These results suggest the intensity and frequency of freeze-thaw cycle strongly affect soil carbon emissions during the freeze-thaw cycle period. Therefore, these factors should be considered in laboratory studies and model simulations under climate change scenarios.

During the decomposition process of soil organic carbon (SOC), microbial products such as microbial necromass and microbial metabolites may form an important stable carbon (C) pool, called microbially derived C, which has different decomposition patterns from plant-derived C. However, current Earth System Models do not simulate this microbially derived C pool separately. Here, we incorporated the microbial necromass pool to the first-order kinetic model and the Michaelis-Menten model, respectively, and validated model behaviors against previous observation data from the decomposition experiments of 13C-labeled necromass. Our models showed better performance than existing models and the Michaelis-Menten model was better than the first-order kinetic model. Microbial necromass C was estimated to be 10-27% of total SOC in the study soils by our models and therefore should not be ignored. This study provides a novel modification to process-based models for better simulation of soil organic C under the context of global changes.

The role of carbon (C) and nutrient uptake, allocation, storage and especially their interactions in survival and recovery of trees under increased frequencies and intensities of drought events is not well understood. A full factorial experiment with four soil water content regimes ranging from extreme drought to well-watered conditions and two fertilization levels was carried out. We aimed to investigate whether nutrient addition mitigates drought effects on downy oak (Quercus pubescens Willd.) and whether storage pools of non-structural carbohydrates (NSC) are modified to enhance survival after 2.5 years of drought and recovery after drought relief. Physiological traits, such as photosynthesis, predawn leaf water potential as well as tissue biomass together with pools and dynamics of NSC and nutrients at the whole-tree level were investigated. Our results showed that fertilization played a minor role in saplings' physiological processes to cope with drought and drought relief, but reduced sapling mortality during extreme drought. Irrespective of nutrient supply, Q. pubescens showed increased soluble sugar concentration in all tissues with increasing drought intensity, mostly because of starch degradation. After 28 days of drought relief, tissue sugar concentrations decreased, reaching comparable values to those of well-watered plants. Only during the recovery process from extreme drought, root NSC concentration strongly declined, leading to an almost complete NSC depletion after 28 days of rewetting, simultaneously with new leaves flushing. These findings suggest that extreme drought can lead to root C exhaustion. After drought relief, the repair and regrowth of organs can even exacerbate the root C depletion. We concluded that under future climate conditions with repeated drought events, the insufficient and lagged C replenishment in roots might eventually lead to C starvation and further mortality.

Nitrous oxide (N2O) is an important greenhouse gas and is involved in the destruction of ozone layer. However, the underlying mechanisms of the high soil N2O emission during the freeze-thaw (FT) period are still unclear. Here, we conducted a mesocosm study with high frequency in situ measurements to explore the responses of soil microbes to the FT cycles and their influences on soil N2O emission. We found the high N2O emission rate during the FT period was mainly due to the release of substrates, the maintenance of high enzyme activities at the freezing stage, and the fast recovery of microbial biomass nitrogen (MBN) and high microbial activities at the thawing stage. Physical isolation of previously produced N2O was an important mechanism for the higher N2O flux at the thawing stage. With increasing numbers of the FT cycles, MBN at the thawing stage remained stable and potential dehydrogenase activities at the thawing stage also remained stable after the first eight FT cycles and only declined during the last two cycles, suggesting the sustainability of the biological mechanisms. Our study suggests that although MBN declined, microbial enzymes could maintain high activities at a few degrees Celsius below zero in this temperate forest soil and produce high N2O fluxes even at the freezing stage, which were trapped under the ice layer and released at the thawing stage, resulting in high soil N2O emission during the FT period.

To demonstrate the responses of plant (Pakchoi) and soil to poly-γ-glutamic acid (γ-PGA) is essential to better understand the pathways of the promotional effect of γ-PGA on plant growth. In this study, the effects of γ-PGA on soil nutrient availability, plant nutrient uptake ability, plant metabolism and its distribution in a plant-soil system were tested using labeled γ-PGA synthesized from 13C1-15N-L-glutamic acid (L-Glu). γ-PGA significantly improved plant uptake of nitrogen (N), phosphorus (P), and potassium (K) and hence increased plant biomass. γ-PGA greatly strengthened the plant nutrient uptake capacity through enhancing both root biomass and activity. γ-PGA affected carbon (C) and N metabolism in plant which was evidenced with increased soluble sugar contents and decreased nitrate and free amino acids contents. About 26.5% of the γ-PGA-N uptake during the first 24 h, after γ-PGA application, was in the form of intact organic molecular. At plant harvest, 29.7% and 59.4% of γ-PGA-15N was recovered in plant and soil, respectively, with a 5.64% of plant N nutrition being derived from γ-PGA-N. The improved plant nutrient uptake capacity and soil nutrient availability by γ-PGA may partly explain the promotional effect of γ-PGA, however, the underlying reason may be closely related to L-Glu.

Microbes are widely distributed in soils and play a very important role in nutrient cycling and ecosystem services. To understand the biogeographic distribution of forest soil bacteria, we collected 115 soil samples in typical forest ecosystems across eastern China to investigate their bacterial community compositions using Illumina MiSeq high throughput sequencing based on 16S rRNA. We obtained 4,667,656 sequences totally and more than 70% of these sequences were classified into five dominant groups, i.e., Actinobacteria, Acidobacteria, Alphaproteobacteria, Verrucomicrobia, and Planctomycetes (relative abundance >5%). The bacterial diversity showed a parabola shape along latitude and the maximum diversity appeared at latitudes between 33.50°N and 40°N, an area characterized by warm-temperate zones and moderate temperature, neutral soil pH and high substrate availability (soil C and N) from dominant deciduous broad-leaved forests. Pairwise dissimilarity matrix in bacterial community composition showed that bacterial community structure had regional similarity and the latitude of 30°N could be used as the dividing line between southern and northern forest soils. Soil properties and climate conditions (MAT and MAP) greatly accounted for the differences in the soil bacterial structure. Among all soil parameters determined, soil pH predominantly affected the diversity and composition of the bacterial community, and soil pH = 5 probably could be used as a threshold below which soil bacterial diversity might decline and soil bacterial community structure might change significantly. Moreover, soil exchangeable cations, especially Ca(2+) (ECa(2+)) and some other soil variables were also closely related to bacterial community structure. The selected environmental variables (21.11%) explained more of the bacterial community variation than geographic distance (15.88%), indicating that the edaphic properties and environmental factors played a more important role than geographic dispersal limitation in determining the bacterial community structure in Chinese forest soils.
