RESUMO

We present a numerically exact steady-state inchworm Monte Carlo method for nonequilibrium quantum impurity models. Rather than propagating an initial state to long times, the method is directly formulated in the steady state. This eliminates any need to traverse the transient dynamics and grants access to a much larger range of parameter regimes at vastly reduced computational costs. We benchmark the method on equilibrium Green's functions of quantum dots in the noninteracting limit and in the unitary limit of the Kondo regime. We then consider correlated materials described with dynamical mean field theory and driven away from equilibrium by a bias voltage. We show that the response of a correlated material to a bias voltage differs qualitatively from the splitting of the Kondo resonance observed in bias-driven quantum dots.

RESUMO

A group of seven-week-old albino mice of both genders were orally administered with a suspension of 25 mg Ti-6Al-4Nb/ml of saline/kg body weight and evaluated in comparison with a control group of animals treated with saline. Evaluation of both the groups was conducted through behavioral tests (Rota rod, open field, novel object and light dark box test), blood biochemical tests [complete blood count and selected serum parameters ([cholesterol, high-density lipoproteins, low-density lipoproteins, creatinine and triglycerides)] and on the basis of measured concentration of antioxidant metabolites (superoxide dismutase, catalase and lipid peroxidation) in vital organs (brain, heart, liver, kidney and lungs). Based upon the results of these tests, it has been found that the applied dose of Ti-6Al-4Nb alloy powder has not effect on physical and neurological outcome of these animals. However, it can increase low-density lipoprotein concentrations as well as disturb the H2O2 and lipid peroxidation associated metabolic pathways, especially in male albino mice. Whereas all other hematological indices and antioxidative stress parameters were unaffected.

RESUMO

We demonstrate how to identify which physical processes dominate the low-energy spectral functions of correlated electron systems. We obtain an unambiguous classification through an analysis of the equation of motion for the electron self-energy in its charge, spin, and particle-particle representations. Our procedure is then employed to clarify the controversial physics responsible for the appearance of the pseudogap in correlated systems. We illustrate our method by examining the attractive and repulsive Hubbard model in two dimensions. In the latter, spin fluctuations are identified as the origin of the pseudogap, and we also explain why d-wave pairing fluctuations play a marginal role in suppressing the low-energy spectral weight, independent of their actual strength.

RESUMO

Insects are incredibly successful and diverse organisms, but they also pose a significant threat to agricultural crops, causing potential losses of up to US$470 billion. Among these pests, Plutella xylostella (Linnaeus), a devastating insect that attacks cruciferous vegetables, alone results in monetary losses of around US$4-5 billion worldwide. While insecticides have effectively protected plants under field conditions, their use comes with various environmental and mammalian hazards. Additionally, insects are developing resistance to commonly used insecticides, rendering management strategies less effective. Arthropods employ a range of behavioral and biochemical mechanisms to cope with harmful chemicals, which contribute to the development of resistance. Understanding these mechanisms is crucial for addressing the issue of resistance. It is imperative to integrate strategies that can delay the development of resistance and enhance the efficiency of insecticides. Therefore, we present an overview of insecticide resistance in insects, focusing on P. xylostella, to provide insights into the current resistance status of this pest and propose tactics that can improve the effectiveness of insecticides.

RESUMO

This study investigated the anti-arthritic potential of novel mannich-based derivatives of 2-mercaptobenzimidazole (AK7 and AK9) in rats. The compounds were characterized by NMR and FTIR spectroscopies and their acute anti-inflammatory effects were measured by carrageenan (CRG)-induced paw edema model. The most potent doses of AK7 and AK9 were subsequently evaluated in the complete Freund's adjuvant (CFA)-induced inflammatory arthritis model. AK7 and AK9 inhibited CRG-induced inflammation in a dose-dependent fashion and a similar reduction in CFA-induced paw inflammation was observed. Moreover, X-ray and histopathological analyses of AK7-treated animals displayed normal joint structure whereas AK9, despite of its anti-inflammatory effects, failed to protect against cartilage destruction. Interestingly, biochemical analysis revealed a better safety profile for AK7 than for AK9 and methotrexate. Both compounds suppressed mRNA levels of pro-inflammatory mediators (IRAK1, NF-κB1, TNF-α, IL1B) while only AK7 reduced the transcript levels of interstitial collagenase (MMP1). Molecular docking analysis of AK7 and AK9 with TNF-α and MMP1 also supported the experimental data. These findings clearly highlight the beneficial effects of AK7 in the prevention and/or treatment of inflammatory arthritis.

RESUMO

Climate change has been discussed as to exert shifts in geographical range of plants, animals or insect species by increasing, reducing or shifting its appropriate climatic habitat. Globally, Pakistan has been ranked at 5th position on the list of countries most vulnerable to climate change in 2020. Climate change has resulted in the losses of biodiversity and alteration in ecosystem as a result of depletion of natural habitats of species in Pakistan as well as in the world. Ants have been regarded as indicators of environmental change and ecosystem processes. Brachyponera nigrita (Emery, 1895) was reported for the first time from Pakistan (Pothwar region). Objective of our studies was to model geographic distribution of newly recorded ant species, B. nigrita based on two representative concentration pathways (RCP) (RCP4.5 and RCP8.5) for 2050s using maximum entropy model (Maxent) in Pakistan. In modeling procedure, 21occurrence records and 8 variables namely Bio4 (Temperature seasonality), Bio8 (Mean temperature of wettest quarter), Bio10 (Mean temperature of warmest quarter), Bio12 (Annual precipitation), Bio13 (Precipitation of wettest month), Bio15 (Precipitation seasonality), Bio17 (Precipitation of driest quarter) and Bio18 (Precipitation of warmest quarter) were used to determine the current and future distributions. Performance of the model was evaluated using AUC (area under curves) values, partial ROC, omission rates (E = 5%) and AICc (Model complexity). The results showed the average AUC value of the model was 0.930, which indicated that the accuracy of the model was excellent. The jackknife test also showed that Bio4, Bio18, Bio17 and Bio15 contributed 98% for the prediction of potential distribution of the species as compared to all other variables. Maxent results indicated that distribution area of B. nigrita under future predicted bioclimatics 2050 (RCP 4.5 and RCP8.5) would be increased in various localities of Pakistan as compared to its current distribution. In Pothwar region, moderately suitable and highly suitable areas of this species would increase by 505.932321km2and 572.118421km2as compared to current distribution under 2050 (RCP 4.5), while under 2050 (RCP 8.5), there would be an increase of 6427.2576km2and 3765.140493km2 respectively in moderately suitable and highly suitable areas of B. nigrita. This species was associated with termites, collembolans and larval stages of different insects. White eggs, creamy white pupae and many workers of this species were observed in a variety of habitats. Unknown nesting ecology, species identification characters supported with micrographs has been given which will help researchers for further ecological studies.

RESUMO

Objective: The purpose of this study was to use ultrasonographic data to rule out and distinguish diseases that cause acute pelvic pain. Material and method: The literature was reviewed using a systematic search of the databases Google Scholars and PubMed, as well as through hand searching. We looked through a total of 35 articles, but only 26 were selected after preliminary screening. Furthermore, 14 articles were left out because they required a membership, copyright clearance, or featured non-English references. There were a total of 12 articles included in the final revuew. Among all the study-related articles, only original research studies and one systematic review that sonographically explored the gynecological etiology of acute pelvic pain were selected. Results: Acute pelvic pain in women might be difficult to identify between gynecologic and non-gynecologic causes based solely on patient history and examination. Advanced imaging, like ultrasound, aids in determining the reason. Pelvic inflammatory disease can be difficult to diagnose, and clinicians should use a low threshold for starting presumptive treatment in order to avoid significant long-term effects such as infertility. Conclusions: Pelvic pain can be acute, chronic or functional. Imaging investigations such as CT, ultrasonography, and MRI can assist in establishing a diagnosis. Particularly ultrasound scanning makes it possible to arrive at a diagnosis with a high degree of precision.

RESUMO

Soil receiving discharges from Pb-acid batteries dismantling and restoring units (PBS) can have a high concentration of phytoavailable Pb. Reducing Pb phytoavailability in PBS can decline Pb uptake in food crops and minimize the risks to humans and the environment. This pot study aimed to reduce the concentration of phytoavailable Pb in PBS through Aspergillus niger (A. niger)-mediated release of PO43- from fish bone [Apatite II (APII)] products. The PBS (Pb = 639 mg kg-1 soil) was amended with APII powder (APII-P), APII char (APII-C), and A. niger inoculum as separate doses, and combining A. niger with APII-P (APII-P + A. niger) and APII-C (APII-C + A. niger). The effects of these treatments on reducing the phytoavailability of Pb in PBS and its uptake in fenugreek were examined. Additionally, enzymatic activities and microbial biomass carbon (MBC) in the PBS and the indices of plant physiology, nutrition, and antioxidant defense machinery were scoped. Results revealed that the APII-C + A. niger treatment was the most efficient one. Compared to the control, it significantly reduced the Pb phytoavailability (DTPA-extractable Pb fraction) in soil and its uptake in plant shoots, roots, and grain, up to 61%, 83%, 74%, and 92%. The grain produced under APII-C + A. niger were safe for human consumption as Pb concentration in grain was 4.01 mg kg-1 DW, remaining within the permissible limit set by WHO/FAO (2007). The APII-C + A. niger treatment also improved soil pH, EC, CEC, MBC, available P content and enzymatic activities, and the fenugreek quality parameters. A. niger played a significant role in solubilizing PO43- from APII-C, which reacted with Pb and formed insoluble Pb-phosphates, thereby reducing Pb phytoavailability in PBS and its uptake in plants. This study suggests APII-C + A. niger can remediate Pb-polluted soils via reducing Pb phytoavailability in them.

RESUMO

Several strains of beta-hemolytic Streptococci produce streptokinase enzyme that can bind and activate human plasminogen to plasmin. Streptokinase degrades the fibrin lump by its explicit lysine joining site and so it is applied as a remedy in thrombolytic therapy. The purpose of the study was to subject wild strain of Streptococcus equisimilis to strain development technique, using random mutagenesis by UV irradiation for enhanced production of streptokinase. To evaluate the hyper production of streptokinase after mutagenesis of wild Streptococcus equisimilis by means of UV irradiation. Randomized study. 2012-2014. Enzyme Biotechnology Laboratory, Department of Biochemistry, University of Agriculture, Faisalabad-Pakistan. UV lamp [TUP 40w lamp which has about 90% of its radiation at 2540-2550 A[0]] was used for the mutation of Streptococcus equisimilis cells [1x 10[7] cells mL[-1]] for enhanced production of streptokinase. 10 mL fresh inoculum was transferred to sterile petri plates, which were exposed to UV light for 30, 60, 90, 120, 150, 180, 210, 240 and 270 minutes. The exposure was carried out at distance of 20cm from the centre of lamp. A dose producing 87% killing was selected as optimum dose, after preparing kill curve. The kill/ survival curve was prepared and time of exposure giving [210 minutes] 3 log kill was selected for mutation of the Streptococcus equisimilis for hyper production of streptokinase enzyme. Enzyme assay was performed for both wild and mutant strains. Dose of 210 minutes was selected as best dose which was followed by the selection using triton X-100. Finally the selected strain S. equisimilis EBL-UV-210 showed 480 U mL[-1] of streptokinase activity in quantitative blood clot liquefaction test, which is quite higher than wild strain [370 U mL[-1]]. This maximum yield of streptokinase was obtained after 24h, at CSL 4%, pH 7.5, 37[degree sign] C, KH[2]PO[4] 0.04%, K[2]HPO[4] 0.05%, MgSO[4]. 7H[2]O 0.04%, NaHCO[3] 0.15%, CaCO[3] 0.004%, CH[3]COONa. CH[3]CO 0.10%, FeSO[4]. 7H[2]O 0.04%, MnCl[2]. 4H[2]O 0.02%, glucose 2%, yeast extract 3% and 5% inoculum size in liquid state fermentation. Results showed that mutated strain gave enhanced streptokinase activity in comparison to the wild strain. Our current study focused on streptokinase production from this UV mutated streptococcus equisimilis species and purification of this enzyme by ammonium sulfate precipitation, Ion exchange and gel filtration chromatography. The activity of streptokinase was determined by using quantitative blood clot liquefaction method

RESUMO

To investigate the inhibitory effect of Captopril on level of glycation [in vivo]. [2] To study glycation inhibition in vivo. Study Case study. Period: Sep. 2006 to March. 2008. One year seven months. Department of Biochemistry University of Agriculture, Faisalabad. Different parameters like fluorescence, total proteins, TBA [thiobarbituric acid] method, periodate borohydride assay were used to check the effect of inhibitor on glycation. Thirty two combinations were made and all these combinations were placed at 37°C, at same time for five weeks. 3mL of blood sample was drawn after 1st, 3rd and 5th week of incubation to perform the experiments for glycation and glycation inhibition. Along with the same temperature [37°C], different combinations of glucose and inhibitor were used. Effective concentration of inhibitor helped to decrease the level of glycation. All concentrations of glucose [G, G and G] showed 1 2 3 glycation with protein. The inhibitor Captopril [all concentrations] showed variations in inhibition of glycation at one temperature [37°C with different parameters [Fluorescence, TBA and Periodate] but the most effective concentration of inhibitors at each condition is I [1mM] but I [10 3 1 mM] and I [5 mM] were also equally effective after I. Periodate borohydride Assay is more effective for glycation determination than 2 3 thiobarbituric acid assay. Captopril can be used as glycation inhibitor in future. As it enhances the activity of transketolase, it can produce 3DG compound which can block the AGEs. However, more experimentations should be done on animal or on large scale before its application in diabetic patients

RESUMO

Dengue is an important vector borne viral disease of public health significance. Dengue fever is on the rise in Pakistan with considerable morbidity and mortality in recent outbreaks. We undertook the study to analyze the prevalence of Dengue fever in Rawalpindi and Islamabad region. The object of our study was to estimate the burden fever in Rawalpindi and Islamabad region. Another aim of our study was to evaluate and if possible alleviate the anxiety associated with dengue fever. We wanted to find out the factual prevalence of the disease. We aimed to determine the most common age group and gender of the illness as well. Cross Sectional Study. This study was conducted in Benzair Bhutto Hospital [BBH] and Pakistan Institute of Medical Sciences [PIMS] in Rawalpindi and Islamabad from 1[st] September to 15[th] November 2011. People of all ages and both sexes with suspicion of Dengue were included in the study. A total of 16616 people turning up at these hospitals were tested for dengue virus. Only 2269 were positive for disease [13.6%]. Male to female ratio came out to be 2.1:1 and maximum number of cases belonged to age group 21-30 years. The number of cases turning up with Dengue suspicion is out of proportion to actual disease burden. Thus we need to alleviate the anxiety associated with the disease. There is a genuine need for strict adherence to preventive control measures and research needs to be undertaken in order to reduce the mortality and morbidity associated with the Dengue

RESUMO

Gastric cancer is among the most common malignancies in Pakistan. The aim of our study are i] to document different histological types of gastric malignancies as per age and sex ii] discuss its possible association in patients who underwent endoscopy and iii] compare our data with local and abroad studies. Retrospective study. This study was conducted in the Department of Pathology, Basic Medical Science Institute, Jinnah Postgraduate Medical Centre, Karachi, Pakistan, from 1[st] January 2003 to 31 December 2007. A total of 740 cases of gastric endoscopic biopsies were received for histopathological evaluation at the Department of Pathology, Basic Medical Science Institute, JPMC, Karachi. Out of these cases, a total of 70 gastric neoplasms were separated, analyzed and classified according to the WHO classification system and their relative frequencies were recorded. Gastric adenocarcinoma was the commonest neoplasm with male predominance in 5th to 7[th] decade of life, account for 88.57% [62/70] of all gastric neoplasms. Gastric lymphoma was the second common with 8.57%[06/70] and gastric carcinoid was least common with 1.43%[1/70] of all gastric neoplasms. This study highlights that adenocarcinoma is the most common gastric malignancy seen in most high risk group includes elderly males followed by lymphoma with no gender difference. So we advise regular endoscopic biopsies surveillance at least in high risk age group for the early detection of cancer

RESUMO

The keratinizing tumors are less radiosensitive and non-keratinizing are more radio responsive. This study was carried out to detect the presence of keratin in apparently non- keratinizing poorly differentiated squamous cell carcinomas by immunoperoxidase staining. It is a cross sectional and descriptive study conducted at King Edward Medical College, Post Graduate Medical Institute, Lahore. A total of 100 patients suffering from squamous cell carcinomas were included in the study. Both H and E and immunoperoxidase staining were performed. The results of both types of staining were compared for each case. Four groups were identified. Forty nine cases showed obvious keratinization on both H and E and immunoperoxidase staining. Twenty cases had doubtful keratinization on H and E but showed more obvious keratinizaton on immunoperoxidase staining. Ten cases were non-keratinizing on H and E as well as on immunoperoxidase analysis. Cytokeratin can be used as a tumour marker for squamous cell carcinoma. The immunohistochemical technique can help us in revising and modifying our H and E impression of a squamous cell carcinoma
