RESUMO

Production of compost from cattle manure results in ammonia (NH3) and greenhouse gas emissions, causing the loss of valuable nitrogen (N) and having negative environmental impacts. Lignite addition to cattle pens has been reported to reduce NH3 emissions from manure by approximately 60%. However, the effect of lignite additions during the manure composting process, in terms of gaseous emissions of NH3, nitrous oxide (N2O), carbon dioxide (CO2), and methane (CH4) is not clear. This composting study was conducted at a commercial cattle feedlot in Victoria, Australia. Prior to cattle entering the feedlot, we applied 4.5 kg m-2 of dry lignite to a treatment pen, and no lignite to a control pen. After 90 days of occupancy, the cattle were removed and the accumulated manure from each pen was used to form two separate compost windrows (control and treatment). During composting we collected manure samples regularly and quantified gaseous emissions of NH3, N2O, CO2, and CH4 from both windrows with an inverse-dispersion technique using open-path Fourier transform infrared spectroscopy (OP-FTIR). Over the 87-day measurement period, the cumulative gas fluxes of NH3, N2O, CO2, and CH4 were 3.4 (± 0.6, standard error), 0.4 (± 0.1), 932 (± 99), and 1.2 (± 0.3) g kg-1 (initial dry matter (DM)), respectively for the lignite amended windrow, and 7.2 (± 1.3), 0.1 (± 0.03), 579 (± 50) and -0.5 (± 0.1) g kg-1 DM, respectively for the non-lignite windrow. The addition of lignite reduced NH3 emissions by 54% during composting, but increased total greenhouse gas (GHG) emissions by 2.6 times. Total N losses as NH3-N and N2O-N were approximately 11 and 25% of initial N for the lignite and non-lignite windrows, respectively. The effectiveness of retaining N was obvious in the first three weeks after windrow formation. A cost-benefit analysis indicated that the benefit of lignite addition to cattle pens by reduced NH3 emission could justify the trade-off of increased GHG emissions.

A continuous-wave, rapidly swept cavity-ringdown spectroscopic technique has been developed for localized atmospheric sensing of trace gases at remote sites. It uses one or more passive open-path optical sensor units, coupled by optical fiber over distances of >1 km to a single transmitter/receiver console incorporating a photodetector and a swept-frequency diode laser tuned to molecule-specific near-infrared wavelengths. Ways to avoid interference from stimulated Brillouin scattering in long optical fibers have been devised. This rugged open-path system, deployable in agricultural, industrial, and natural atmospheric environments, is used to monitor ammonia in air. A noise-limited minimum detectable mixing ratio of ~11 ppbv is attained for ammonia in nitrogen at atmospheric pressure.

Large ruminant production in developing countries is inefficient with low growth rates and likely high greenhouse gas emissions per unit of meat or milk produced. Trials conducted in Lao PDR from 2017 to 2020, studied ad libitum supplementation for 12 weeks with 20 kg high-quality molasses nutrient blocks (Four Seasons Pty Ltd., Brisbane, Australia), that were either non-medicated; fenbendazole-medicated (Panacur100®, Coopers Australia, 5 g/kg); triclabendazole-medicated (Fasinex®, Novartis Australia, 5 g/kg or 10 g/kg, respectively); or formulated with urea (8% or 10% urea, respectively). Average daily gains were determined for access to all molasses blocks and compared with access to control blocks, no supplementation, or previously determined free-grazing baseline average daily gains (55−84 g in cattle; 92−106 g in buffalo). Productivity was significantly improved following access to all molasses blocks. Average daily gains following access to 8% urea and control blocks were calculated for three age cohorts of cattle: young calves <8 m (238−298 g), growing cattle (143−214 g) and lactating cows (179−191 g). Modelling using IPCC Inventory software model V 2.69 of published data demonstrated a conservative net abatement of 350 kg CO2e was achievable over a 200-day feeding period. An additional trial of Emissions control blocks (n = 200) distributed to farmers (n = 60) and two educational institutions were conducted. Consumption rates (156 g/day) and farmer and institutional acceptance of these blocks were similar to our published findings with other molasses blocks, confirming all formulations of blocks improved animal productivity and body condition score, with healthier animals that were easier to manage. Modelling of changes in greenhouse gas emissions intensity identified an abatement of 470 kg CO2e per Emissions control blocks consumed, delivering a total project emissions abatement of 94 t CO2e. Provision of high-quality molasses blocks significantly improved smallholder large ruminant productivity and addition of greenhouse gas reducing agents is likely to achieve impressive abatement of greenhouse gas emissions due to improved efficiency of rumen fermentation and productivity.

This paper reports a reconnaissance survey of the concentrations of sterol compounds (as indicators of fecal contamination) in a large water supply system in southeast Australia comprising a network of rivers, channels, and drains. Levels of coprostanol and cholestanol were determined in surface water and bottom sediment using gas chromatography-mass spectrometry analysis across 17 strategic sampling sites and over 12 months. Clear differences in the levels of fecal contamination were observed among sites. Four sites routinely contained high levels of the fecal indicator sterols indicated from surface water and sediment sample analysis. Coprostanol concentrations at each location varied from 0 ng/L at the reference site to 11,327 ng/L in a surface water sample of a drain directly downstream of a knackery. The majority of the sites contained coprostanol in the range of 500 to 800 ng/L. Since no fecal-associated sterol compounds were detected at the external reference sites, these were assumed to be free from fecal contamination. Sewage water discharge and/or substantial water runoff maybe the principal factors contributing to fecal contamination of the supply drains and channels.

Manure composting is a common management practice for cattle feedlots, but gaseous emissions from composting are poorly understood. The objective of this study was to quantify ammonia (NH3 ), nitrous oxide (N2 O), carbon dioxide (CO2 ), and methane (CH4 ) emissions from windrow composting (turning) and static stockpiling (nonturning) of manure at a commercial feedlot in Australia. An inverse-dispersion technique using an open-path Fourier transform infrared (OP-FTIR) spectrometer gas sensor was deployed to measure emissions of NH3 , N2 O, CO2 , and CH4 over a 165-d study period, and 29 and 15% of the total data intervals were actually used to calculate the fluxes for the windrow and stockpile, respectively. The nitrogen (N) lost as NH3 and N2 O emissions represented 26.4 and 3.8% of the initial N in windrow, and 5.3 and 0.8% of that in the stockpile, respectively. The carbon (C) lost as CO2 and CH4 emissions represented 44 and 0.3% of the initial C in windrow, and 54.8 and 0.7% of that in the stockpile, respectively. Total greenhouse gas (GHG) emissions from the manure windrow were 2.7 times higher than those of the stockpiled manure. This work highlights the value that could be accrued if one could reduce emissions of NH3 -N and N2 O-N from composting, which would retain manure N content while reducing GHG emissions.

Chronic obstructive pulmonary disease (COPD) is characterized by abnormal inflammatory responses and impaired airway immunity, which provides an opportunistic platform for nontypeable Haemophilus influenzae (NTHi) infection. Clinical evidence supports that the COPD airways present increased concentrations of glucose, which may facilitate proliferation of pathogenic bacteria able to use glucose as a carbon source. NTHi metabolizes glucose through respiration-assisted fermentation, leading to the excretion of acetate, formate, and succinate. We hypothesized that such specialized glucose catabolism may be a pathoadaptive trait playing a pivotal role in the NTHi airway infection. To find out whether this is true, we engineered and characterized bacterial mutant strains impaired to produce acetate, formate, or succinate by inactivating the ackA, pflA, and frdA genes, respectively. While the inactivation of the pflA and frdA genes only had minimal physiological effects, the inactivation of the ackA gene affected acetate production and led to reduced bacterial growth, production of lactate under low oxygen tension, and bacterial attenuation in vivo. Moreover, bacterially produced acetate was able to stimulate the expression of inflammatory genes by cultured airway epithelial cells. These results back the notion that the COPD lung supports NTHi growth on glucose, enabling production of fermentative end products acting as immunometabolites at the site of infection. Thus, glucose catabolism may contribute not only to NTHi growth but also to bacterially driven airway inflammation. This information has important implications for developing nonantibiotic antimicrobials, given that airway glucose homeostasis modifying drugs could help prevent microbial infections associated with chronic lung disease.

Non-typeable Haemophilus influenzae (NTHi) is a major pathogen in upper and lower respiratory tract infections in humans, and is increasingly also associated with invasive disease. We have examined two unrelated NTHi invasive disease isolates, R2866 and C188, in order to identify metabolic and physiological properties that distinguish them from respiratory tract disease isolates such as Hi2019. While the general use of the Hi metabolic network was similar across all three strains, the two invasive isolates secreted increased amounts of succinate, which can have anti-inflammatory properties. In addition, they showed a common shift in their carbon source utilization patterns, with strongly enhanced metabolism of nucleoside substrates, glucose and sialic acid. The latter two are major compounds present in blood and cerebrospinal fluid (CSF). Interestingly, C188 and R2866 also shared a reduced ability to invade or survive intracellularly in 16HBE14 bronchial epithelial cells relative to Hi2019 (4-fold (4 h), 25-fold (24 h) reduction). Altered metabolic properties, such as the ones observed here, could arise from genomic adaptations that NTHi undergo during infection. Together these data indicate that shifts in substrate preferences in otherwise conserved metabolic pathways may underlie strain niche specificity and thus have the potential to alter the outcomes of host-NTHi interactions.

RESUMO

Radiative forcing of methane (CH4) is significantly higher than carbon dioxide (CO2) and its enteric production by ruminant livestock is one of the major sources of greenhouse gas emissions. CH4 is also an important marker of farming productivity, because it is associated with the conversion of feed to product in livestock. Consequently, measurement of enteric CH4 is emerging as an important research topic. In this review, we briefly describe the conversion of carbohydrate to CH4 by the bacterial community within gut, and highlight some of the key host-microbiome interactions. We then provide a picture of current progress in techniques for measuring enteric CH4, the context in which these technologies are used, and the challenges faced. We also discuss solutions to existing problems and new approaches currently in development.

Beef cattle feedlots are a major source of ammonia (NH3) emissions from livestock industries. We investigated the effects of lignite surface applications on NH3 and nitrous oxide (N2O) emissions from beef cattle feedlot pens. Two rates of lignite, 3 and 6kgm(-2), were tested in the treatment pen. No lignite was applied in the control pen. Twenty-four Black Angus steers were fed identical commercial rations in each pen. We measured NH3 and N2O concentrations continuously from 4th Sep to 13th Nov 2014 using Quantum Cascade Laser (QCL) NH3 analysers and a closed-path Fourier Transform Infrared Spectroscopy analyser (CP-FTIR) in conjunction with the integrated horizontal flux method to calculate NH3 and N2O fluxes. During the feeding period, 16 and 26% of the excreted nitrogen (N) (240gNhead(-1)day(-1)) was lost via NH3 volatilization from the control pen, while lignite application decreased NH3 volatilization to 12 and 18% of the excreted N, for Phase 1 and Phase 2, respectively. Compared to the control pen, lignite application decreased NH3 emissions by approximately 30%. Nitrous oxide emissions from the cattle pens were small, 0.10 and 0.14gN2O-Nhead(-1)day(-1) (<0.1% of excreted N) for the control pen, for Phase 1 and Phase 2, respectively. Lignite application increased direct N2O emissions by 40 and 57%, to 0.14 and 0.22gN2O-Nhead(-1)day(-1), for Phase 1 and Phase 2, respectively. The increase in N2O emissions resulting from lignite application was counteracted by the lower indirect N2O emission due to decreased NH3 volatilization. Using 1% as a default emission factor of deposited NH3 for indirect N2O emissions, the application of lignite decreased total N2O emissions.

In open beef feedlot systems, more than 50% of dietary nitrogen (N) is lost as ammonia (NH3). Here we report an effective and economically-viable method to mitigate NH3 emissions by the application of lignite. We constructed two cattle pens (20 × 20 m) to determine the effectiveness of lignite in reducing NH3 emissions. Twenty-four steers were fed identical commercial rations in each pen. The treatment pen surface was dressed with 4.5 kg m(-2) lignite dry mass while no lignite was applied in the control pen. We measured volatilised NH3 concentrations using Ecotech EC9842 NH3 analysers in conjunction with a mass balance method to calculate NH3 fluxes. Application of lignite decreased NH3 loss from the pen by approximately 66%. The cumulative NH3 losses were 6.26 and 2.13 kg N head(-1) in the control and lignite treatment, respectively. In addition to the environmental benefits of reduced NH3 losses, the value of retained N nutrient in the lignite treated manure is more than $37 AUD head(-1) yr(-1), based on the current fertiliser cost and estimated cost of lignite application. We show that lignite application is a cost-effective method to reduce NH3 loss from cattle feedlots.

OBJECTIVE: The objective of this study was to evaluate a model for a safe road transport system, based on some safety performance indicators regarding the road user, the vehicle, and the road, by using crashes with fatally and seriously injured car occupants. The study also aimed to evaluate whether the model could be used to identify system weaknesses and components (road user, vehicles, and road) where improvements would yield the highest potential for further reductions in serious injuries. METHODS: Real-life car crashes with serious injury outcomes (Maximum Abbreviated Injury Scale 2+) were classified according to the vehicle's safety rating by Euro NCAP (European New Car Assessment Programme) and whether the vehicle was fitted with ESC (Electronic Stability Control). For each crash, the road was also classified according to EuroRAP (European Road Assessment Programme) criteria, and human behavior in terms of speeding, seat belt use, and driving under the influence of alcohol/drugs. Each crash was compared and classified according to the model criteria. Crashes where the safety criteria were not met in more than one of the 3 components were reclassified to identify whether all the components were correlated to the injury outcome. In-depth crash injury data collected by the UK On The Spot (OTS) accident investigation project was used in this study. All crashes in the OTS database occurring between 2000 and 2005 with a car occupant with injury rated MAIS2+ were included, for a total of 101 crashes with 120 occupants. RESULTS: It was possible to classify 90 percent of the crashes according to the model. Eighty-six percent of the occupants were injured when more than one of the 3 components were noncompliant with the safety criteria. These cases were reclassified to identify whether all of the components were correlated to the injury outcome. In 39 of the total 108 cases, at least two components were still seen to interact. The remaining cases were only related to one of the safety criteria, namely, the road user (26), the vehicle (19), and the road (24). The criteria for the road and the vehicle did not address multiple event crashes, rear-end crashes, hitting stationary/parked vehicles, or trailers. CONCLUSIONS: The model for a safe road transport system was found useful to classify fatal and serious road vehicle crashes. It was possible to classify 90 percent of the crashes according to the safety road transport model. For all these cases it was possible to identify weaknesses and parts of the road transport system with the highest potential to prevent fatal and serious injuries. Injury outcomes were mostly related to an interaction between the 3 components: the road, the vehicle, and the road user.
