BACKGROUND: Although cis-diamminedichloro-platinum (CDDP) exhibits strong therapeutic effects in cancer chemotherapy, its adverse effects such as peripheral neuropathy, nephropathy, and vomiting are dose-limiting factors. Previous studies reported that chronotherapy decreased CDDP-induced nephropathy and vomiting. In the present study, we investigated the influence of dosing times on CDDP-induced peripheral neuropathy in rats. METHODS: CDDP (4 mg/kg) was administered intravenously at 5:00 or 17:00 every 7 days for 4 weeks to male Sprague-Dawley rats, and saline was given to the control group. To assess the dosing time dependency of peripheral neuropathy, von-Frey test and hot-plate test were performed. RESULTS: In order to estimate hypoalgesia, the hot-plate test was performed in rats administered CDDP weekly for 4 weeks. On day 28, the withdrawal latency to thermal stimulation was significantly prolonged in the 17:00-treated group than in the control and 5:00-treated groups. When the von-Frey test was performed to assess mechanical allodynia, the withdrawal threshold was significantly lower in the 5:00 and 17:00-treated groups than in the control group on day 6 after the first CDDP dose. The 5:00-treated group maintained allodynia throughout the experiment with the repeated administration of CDDP, whereas the 17:00-treated group deteriorated from allodynia to hypoalgesia. CONCLUSIONS: It was revealed that the severe of CDDP-induced peripheral neuropathy was inhibited in the 5:00-treated group, whereas CDDP-treated groups exhibited mechanical allodynia. These results suggested that the selection of an optimal dosing time ameliorated CDDP-induced peripheral neuropathy.

RESUMO

A novel exopolysaccharide-producing bacterium, designated strain SE3(T), was isolated from Pacific Ocean sediment. The strain was Gram-stain-negative, motile, strictly aerobic, oxidase-positive and catalase-positive, and required Na(+) for growth. Its major isoprenoid quinone was ubiquinone-8 (Q-8), and its cellular fatty acid profile mainly consisted of C16 : 1ω7c, C16 : 0 and C18 : 1ω7c. The DNA G+C content was 46.9 mol%. 16S rRNA gene sequence analysis suggested that strain SE3(T) is a member of the genus Pseudoalteromonas. Strain SE3(T) exhibited close phylogenetic affinity to Pseudoalteromonas arabiensis JCM 17292(T) (99.0 % 16S rRNA gene sequence similarity), Pseudoalteromonas lipolytica LMEB 39(T) (98.39 %) and Pseudoalteromonas donghaensis HJ51(T) (97.65 %). The DNA-DNA reassociation values between strain SE3(T) and P. arabiensis JCM 17292(T), P. lipolytica JCM 15903(T) and P. donghaensis LMG 24469(T) were 31, 26 and 44 %, respectively. Owing to the significant differences in phenotypic and chemotaxonomic characteristics, phylogenetic analysis based on 16S rRNA gene sequences and DNA-DNA relatedness data, the new isolate merits classification as a representative of novel species, for which the name Pseudoalteromonas shioyasakiensis is proposed. The type strain is SE3(T) ( = JCM 18891(T) = NCIMB 14852(T)).

The Last Interglacial (LIG: 130,000-115,000 years ago) was a period of warmer global mean temperatures and higher and more variable sea levels than the Holocene (11,700-0 years ago). Therefore, a better understanding of Antarctic ice-sheet dynamics during this interval would provide valuable insights for projecting sea-level change in future warming scenarios. Here we present a high-resolution record constraining ice-sheet changes in the Wilkes Subglacial Basin (WSB) of East Antarctica during the LIG, based on analysis of sediment provenance and an ice melt proxy in a marine sediment core retrieved from the Wilkes Land margin. Our sedimentary records, together with existing ice-core records, reveal dynamic fluctuations of the ice sheet in the WSB, with thinning, melting, and potentially retreat leading to ice loss during both early and late stages of the LIG. We suggest that such changes along the East Antarctic Ice Sheet margin may have contributed to fluctuating global sea levels during the LIG.

Magnetic biomonitoring techniques and in situ topsoil magnetic susceptibility measurements have been shown to be rapid, cost-effective, and useful methods for investigating roadside pollution. However, combustible vegetation in samples makes it very difficult to use them in high-temperature magnetic experiments although the thermal alteration of spontaneous magnetization is a fundamental magnetic property and can be used to identify reliably the magnetic minerals. Here, we report the first magnetic biomonitoring results of dust deposited on plant leaves along the Tateyama-Kurobe Alpine route at the highly protected Midagahara wetland areas of Mt. Tateyama in Toyama, Japan. In-field magnetic susceptibility from 15 sites (161 points) shows higher susceptibilities near the roadside. Dust deposited on the leaves of Sasa kurilensis, or dwarf bamboo, has been wiped off at 12 sites (64 samples) with a commercial ethanol wipe sheet or silica wool damped in ethanol and subjected to rock magnetic analyses. Thermomagnetic remanence curves and low-temperature behaviors for samples collected near the roadside using silica wool show clearly that the main magnetic mineral in the dust is partially oxidized magnetite. Further, detailed rock magnetic analyses and elemental analyses of leaves in the study area indicate that (a) the magnetic mineralogy on leaves' surface is consistent throughout the study area and (b) higher saturation isothermal remanent magnetization intensities as well as higher concentrations of Pb, Fe, Cr, and Y are observed near the roadside, i.e., the closer to the roadside, the more anthropogenic materials, including partially oxidized magnetite, are present. Also, microscopic observations show the lack of spherical grains, indicating that dust on the roadside leaves is derived from passing vehicle rather than industrial process. Both rock magnetic and geochemical results show that S. kurilensis would be an excellent candidate for investigating air pollution. Also, the proposed wiping-off method of collecting dust from plant surfaces is an effective non-destructive method that can be applied even in highly restricted collection areas, because detailed magnetic mineralogy, including superparamagnetic minerals, could be determined with the use of silica wool for use in both high- and low-temperature experiments.

Warming of high northern latitudes in the Pliocene (5.33-2.58 Myr ago) has been linked to the closure of the Central American Seaway and intensification of North Atlantic Deep Water. Subsequent cooling in the late Pliocene may be related to the effects of freshwater input from the Arctic Ocean via the Bering Strait, disrupting North Atlantic Deep Water formation and enhancing sea ice formation. However, the timing of Arctic freshening has not been defined. Here we present neodymium and lead isotope records of detrital sediment from the Bering Sea for the past 4.3 million years. Isotopic data suggest the presence of Alaskan glaciers as far back as 4.2 Myr ago, while diatom and C37:4 alkenone records show a long-term trend towards colder and fresher water in the Bering Sea beginning with the M2 glaciation (3.3 Myr ago). We argue that the introduction of low-salinity Bering Sea water to the Arctic Ocean by 3.3 Myr ago preconditioned the climate system for global cooling.
