RESUMO

The leaching of additives from plastics and elastomers (rubbers) has raised concerns due to their potential negative impacts on the environment and the development of antibiotic resistance. In this study, we investigated the effects of chemicals extracted from two types of rubber on microbiomes derived from a benthic sea urchin and two pelagic fish species. Additionally, we examined whether bacterial communities preconditioned with rubber-associated chemicals displayed adaptations to antibiotics. At the highest tested concentrations of chemicals, we observed reduced maximum growth rates and yields, prolonged lag phases, and increased alpha diversity. While the effects on alpha and beta diversity were not always conclusive, several bacterial genera were significantly influenced by chemicals from the two rubber sources. Subsequent exposure of sea urchin microbiomes preconditioned with rubber chemicals to the antibiotic ciprofloxacin resulted in decreased maximum growth rates. This indicates a more sensitive microbiome to ciprofloxacin when preconditioned with rubber chemicals. Although no significant interaction effects between rubber chemicals and ciprofloxacin exposure were observed in bacterial alpha and beta diversity, we observed log-fold changes in two bacterial genera in response to ciprofloxacin exposure. These findings highlight the structural and functional alterations in microbiomes originating from various marine species when exposed to rubber-associated chemicals and underscore the potential risks posed to marine life.

Arctic-breeding seabirds contain high levels of many anthropogenic contaminants, which they deposit through guano to the tundra near their colonies. Nutrient-rich soil in vicinity to seabird colonies are favorable habitats for soil invertebrates, such as springtails (Collembola), which may result in exposure to seabird-derived contaminants. We quantified a wide range of lipid-soluble and protein-associated environmental contaminants in two springtail species (Megaphorura arctica and Hypogastrura viatica) and their respective habitats (soil/moss) collected underneath seabird cliffs. Although springtails are commonly used in laboratory toxicity tests, this is the first study to measure concentrations of persistent organic pollutants (POPs) and mercury (Hg) in springtails from the field, and to study biotransportation of contaminants by seabirds to soil fauna. We categorized the sites a priori as of low, medium, or high seabird influence, based on the seabird abundance and species composition. This ranking was reflected in increasing δ15N values in soil/moss and springtails with increasing seabird influence. We found clear indications of seabirds impacting the terrestrial soil environments with organic contaminants, and that concentrations were higher in soil and moss close to the bird cliff, compared to farther away. However, we did not find a relationship between contaminant concentration in springtails and the concentrations in soil/moss, or with level of seabird influence. Our study indicates a low uptake of contaminants in the soil fauna, despite seabird-derived contamination of their habitat.

To examine whether natural stressors like predation risk affect responses to anthropogenic contaminants, we exposed nauplii of the copepod Tigriopus brevicornis to chemical cues from fish (kairomones) and copper (Cu). We tested effects of these treatments, singly and combined, on copepod age and size at maturity, and development stage sensitivity, while controlling for effects of genetic heterogeneity (clutch identity). Predation risk, Cu and clutch identity interacted in their effect on development time. Predation risk alone had minor effects, but potentiated Cu toxicity in the combined treatment by doubling the delay in age at maturity, as compared to Cu exposure alone. This potentiating effect on developmental delay appeared already at the first copepodite stage. The specific strength of response varied among nauplii from different females' clutches. There were no differences in copepod size at maturity among treatments. We did, however, find an interaction between the effect of Cu and clutch identity on copepod growth. Our results demonstrate the importance of ecological interactions for potentiating the toxicity of environmental contaminants. We also demonstrate the need to consider genetic heterogeneity in ecotoxicology. Natural variation in stressor responses has implications for the interpretation of results from toxicological studies using single-clone or inbred culture populations.

Anthropogenic pollutants produce oxidative stress in marine organisms, directly or following generation of reactive oxygen species (ROS), potentially resulting in increased accumulation of DNA strand breaks quantified. The aim of this study is to quantify baseline levels of DNA strand breaks in marine species from four phyla and to assess relative sensitivity to oxidative stress as well as ability to recover. DNA strand breaks were determined using a formamidopyrimidine DNA glycosylase (Fpg)-amended comet assay in circulating cells from blue mussel (Mytilus edulis), shore crab (Carcinus maenas), sea star (Asterias rubens), and vase tunicate (Ciona intestinalis). Lymphocytes from Atlantic cod (Gadus morhua) were used as a reference. In addition to immediate analysis, cells from all species were exposed ex vivo to two concentrations of hydrogen peroxide (H2O2) at 25 or 250 µM prior to assay. Mean baseline DNA strand breaks were highest for cells from sea star (34%) followed by crab (25%), mussel (22%), tunicate (17%), and cod (14%). Circulating cells from invertebrates were markedly more sensitive to oxidative stress compared to cod lymphocytes. DNA strand breaks exceeded 80% for sea star, crab, and mussel cells following exposure to the lowest H2O2 concentration. There was no recovery for cells from any species following 1 hr in buffer. This study provides an in-depth analysis of DNA integrity for ecologically important species representing 4 phyla. Data indicate that circulating cells from invertebrates are more sensitive to oxidative stress than cells from fish as evidenced by DNA strand breaks. Future studies need to address the extent to which DNA strand breaks may exert consequences for body maintenance costs in marine invertebrates.

With increasing oil and gas activities and transport in the Arctic, there is a need to understand how operational or accidental releases of substances affect marine organisms from a pristine environment. The aim of the current study was to describe and compare the responses of two marine fish species, Atlantic cod (Gadus morhua) and turbot (Scophthalmus maximus), following exposure to three levels (low, medium, high) of the water-soluble fraction of a North Sea crude oil for 16 days. The exposure system simulated environmental exposure by allowing clean seawater to percolate through gravel covered in weathered oil before being introduced to aquaria. Both polycyclic aromatic hydrocarbon (PAH) metabolite bile concentrations and cytochrome P4501A (CYP1A) levels and activity increased markedly in comparison with controls in both species, but there were no significant differences between the three exposures. Turbot possessed 4-5-fold higher concentrations of two PAH bile metabolites compared to Atlantic cod by day 8. In contrast, hepatic CYP1A activity in cod was consistently 2-6-fold higher than in turbot with increasing differences over the experimental period. Baseline DNA strand breaks in lymphocytes and kidney cells were low in both species, but was elevated for all treatments by day two. There were no marked indications of the treatments affecting immune functions in either species. This investigation demonstrated that there may be significant differences in responses between species receiving identical exposures and that DNA strand breaks in lymphocytes and kidney cells are sensitive to confinement stress. Data also indicate that some species, such as turbot, may adapt to treatments within days and weeks.

This study evaluated the effects of exposure medium and culture age on intracellular reactive oxygen species (ROS) development and cytotoxicity in fish hepatocytes following exposure to copper (Cu). ROS was quantified using the fluorescent probes DHR 123 and CM-H2DCFDA following exposure to Cu in Leibovitz' medium (L-15) or Tris-buffered saline (TBS). Similarly, culture age effects were investigated using 1-, 2- and 4-day-old cultured hepatocytes by exposing them to Cu in TBS. The exposure in L-15 resulted in significantly higher ROS compared to TBS using CM-H2DCFDA, but not DHR 123. The age of the primary cultures significantly affected the development of ROS for both probes. None of the exposures caused cytotoxicity in the hepatocytes. The results showed that both factors may affect responses to stressors, and suggested that the use of a simple medium such as TBS may be preferable for some applications. It is also preferable to use 1-day-old primary hepatocyte cultures.

Aryl hydrocarbon receptor (AhR) agonistic contaminants were identified in roadside snow samples. Snow was collected in Oslo, Norway, and compared to a background sample collected from a mountain area. The water and particulate fractions were analysed for AhR agonists using a dioxin-responsive, chemically activated luciferase expression (CALUX) cell assay and by gas chromatography coupled to high-resolution time-of-flight mass spectrometry with targeted analysis for polycyclic aromatic hydrocarbons (PAHs) and broad-spectrum non-target analysis. The AhR agonist levels in the dissolved fractions in the roadside samples were between 15 and 387 pg/L CALUX toxic equivalents (TEQ(CALUX)). An elevated AhR activity of 221 pg TEQ(CALUX) per litre was detected in the mountain sample. In the particle-bound fractions, the TEQ(CALUX) was between 1,350 and 7,390 pg/L. One possible explanation for the elevated levels in the dissolved fraction of the mountain sample could be the presence of black carbon in the roadside samples, potentially adsorbing dioxin-like compounds and rendering them unavailable for AhR interaction. No polychlorinated dibenzodioxins and dibenzofurans or polychlorinated biphenyls were detected in the samples; the occurrence of PAHs, however, explained up to 9 % of the AhR agonist activity in the samples, whilst comprehensive two-dimensional gas chromatography coupled to mass spectrometry GCxGC-ToF-Ms identified PAH derivatives such as polycyclic aromatic ketones and alkylated, nitrogen sulphur and oxygen PAHs in the particle fractions. The (large) discrepancy between the total and explained activity highlights the fact that there are other as yet unidentified AhR agonists present in the environment.

There are large and increasing discharges of statins into the aquatic environment. Statins are cholesterol-lowering pharmaceuticals, inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A reductase, an enzyme in the cholesterol synthesis pathway. Earlier studies have shown that statins will affect the expression of a range of genes in mammalian tissues and this group of pharmaceuticals has also been shown to affect membrane transporters. Changes in gene expression and ion transport in aquatic organisms may have dramatic consequences for the individual. The aim of the present study was to clarify whether waterborne exposure to a selected statin, atorvastatin, would affect gene expression in rainbow trout (Oncorhynchus mykiss) gill or liver or ion regulation in gills. Juvenile rainbow trout were exposed to two atorvastatin acid and atorvastatin lactone concentrations for 7 days (nominal concentrations 200 ng L(-1) and 10 µg L(-1)). The exposures caused up-regulated gene expression in gill, not liver, and only at the lowest concentration. Genes involved in membrane transport (pgp, mrp1), oxidative stress response (sod, mt), apoptosis (bax) and biotransformation (sult2b) were differentially expressed whereas the expression of genes involved in cholesterol biosynthesis (hmgr, fdps) or peroxisomal proliferation (ppar) were not affected. There were no significant changes in gill Na(+)/K(+) ATPase activity following exposure to atorvastatin. The pattern of differentially expressed genes in rainbow trout gills differ from responses previously observed in mammalian tissues following statin exposure.

Herring gulls (Larus argentatus) are opportunistic feeders, resulting in contaminant exposure depending on area and habitat. We compared contaminant concentrations and dietary markers between two herring gull breeding colonies with different distances to extensive human activity and presumed contaminant exposure from the local marine diet. Furthermore, we investigated the integrity of DNA in white blood cells and sensitivity to oxidative stress. We analyzed blood from 15 herring gulls from each colony-the urban Oslofjord near the Norwegian capital Oslo in the temperate region and the remote Hornøya island in northern Norway, on the Barents Sea coast. Based on d13 C and d34 S, the dietary sources of urban gulls differed, with some individuals having a marine and others a more terrestrial dietary signal. All remote gulls had a marine dietary signal and higher relative trophic level than the urban marine feeding gulls. Concentrations (mean ± standard deviation [SD]) of most persistent organic pollutants, such as polychlorinated biphenyl ethers (PCBs) and perfluorooctane sulfonic acid (PFOS), were higher in urban marine (PCB153 17 ± 17 ng/g wet weight, PFOS 25 ± 21 ng/g wet wt) than urban terrestrial feeders (PCB153 3.7 ± 2.4 ng/g wet wt, PFOS 6.7 ± 10 ng/g wet wt). Despite feeding at a higher trophic level (d15 N), the remote gulls (PCB153 17 ± 1221 ng/g wet wt, PFOS 19 ± 1421 ng/g wet wt) were similar to the urban marine feeders. Cyclic volatile methyl siloxanes were detected in only a few gulls, except for decamethylcyclopentasiloxane in the urban colony, which was found in 12 of 13 gulls. Only hexachlorobenzene was present in higher concentrations in the remote (2.6 ± 0.42 ng/g wet wt) compared with the urban colony (0.34 ± 0.33 ng/g wet wt). Arctic and sub-arctic pelagic organisms can be exposed to effluents and spills from offshore petroleum-related activities and thus it is important to understand how they respond to crude oil related contaminants such as polycyclic aromatic hydrocarbons (PAHs). The copepod species Calanus finmarchicus, Calanus glacialis and Calanus hyperboreus represent key links in the arctic marine food web. We performed a transcriptome analysis of the three species exposed to phenanthrene (Phe) and benzo[a]pyrene (BaP) representing low and high molecular weight PAHs, respectively. Differential expression of several genes involved in many cellular pathways was observed after 72 h exposure to Phe (0.1 µM) and BaP (0.1 µM). In C. finmarchicus and C. glacialis, the exposure resulted in up-regulation of genes encoding enzymes in xenobiotic biotransformation, particularly the phase II cytosolic sulfonation system that include 3'-phosphoadenosine 5'-phosphosulfate synthase (PAPSS) and sulfotransferases (SULTs). The sulfonation pathway genes were more strongly induced by BaP than Phe in C. finmarchicus and C. glacialis but were not affected in C. hyperboreus. However, a larger number of genes and pathways were modulated in C. hyperboreus by the PAHs including genes encoding xenobiotic biotransformation and lipid metabolism enzymes, suggesting stronger responses in this species. The results suggest that the cytosolic sulfonation is a major phase II conjugation pathway for PAHs in C. finmarchicus and C. glacialis. Some of the biotransformation systems affected are known to be involved in metabolism of endogenous compounds such as ecdysteroids, which may suggest potential interference with physiological and developmental processes of the copepod species.

Polychlorinated dibenzo-p-dioxins (PCDD) and polychlorinated dibenzofurans (PCDF) have been discharged to the Frierfjord (Eastern Norway) for 50 yr. The discharges, dominated by PCDF, were highest in the early 1970s and subsequently reduced 3-4 orders ofmagnitude to 1-2 g toxic equivalents (TEQ)/yr in 1992-2002. Atlantic cod (Gadus morhua) and semipermeable membrane devices (SPMD) were placed in cages for 6 wk in the Frierfjord and at two locations in the adjacent Langesundsfjord. The objective was to clarify the importance of PCDD/PCDF accumulation from the water directly compared to important food items. Little accumulation of PCDD and a significant accumulation of PCDF were observed in the SPMD. Accumulation of PCDF was observed in fish at Kastebukta, closest to the original source, but not at the other two stations. The PCDF concentrations detected in caged fish at Kastebukta were low compared to concentrations found in locally caught fish. Data indicate that it would take 4-55 yr to reach the PCDF-levels observed in locally caught cod if they were exposed only through water. Our results indicate that accumulation of PCDF/PCDD from prey is more important than accumulation from water.

The European Community Regulation on the registration, evaluation, authorisation and restriction of chemicals (REACH) introduced exposure scenarios describing safe use quantitatively, and enhancing the importance of scientific based exposure assessments. This paper presents methods to determine exposure from the airless spray application of anti-corrosive paint and leaching of painted articles submerged in seawater, to establish whether it is possible to test these exposures in a reproducible and feasible way. The paper also presents results from using the methods in order to assess how well the default values recommended under REACH coincide with the tested values and corresponding values available in literature. The methods used were feasible under laboratory conditions. The reproducibility of the application study was shown to be good and all analyses of the leaching showed concentrations below detection limit. More replicates will be required to validate the reproducibility of the growth inhibition tests. Measured values for the present overspray scenario were between, and the leaching values below, values from REACH guidelines and emission scenario documents. Further development of the methods is recommended.

Traditional ecotoxicology methods involving copepods have focused on exposure of pooled individuals and averaged responses, but there is increasing awareness of the importance of individual variation. Many biological traits are density dependent, and decisions to use single-individual or pooled exposure may affect responses to anthropogenic stressors. We investigated how conspecific density as a biotic stressor affects behavioral and respiratory responses to copper (Cu) exposure in the coastal copepod Tigriopus brevicornis. Adults were incubated at densities of 1, 2, or 4 individuals per replicate in 3.2 mL of exposure medium (23 µg Cu L-1 or control). Our results show an interaction of Cu exposure and density on respiration. The Cu exposure increased respiration, but this effect diminished with increasing density. We also found reduced swimming activity with increasing density. We propose 2 nonexclusive alternative explanations for the density-dependent respiratory increase of Cu exposure: 1) a behavioral stress response to low conspecific density, or 2) increased Cu exposure due to increased swimming activity. The present study deals with genotoxicity assessment of freshwaters using caged carp (Cyprinus carpio). Carps were transplanted from a fish-farm to three differently polluted sites in eastern Croatia. Two polluted sites were situated in the river Drava, downstream from the cities of Belisce and Osijek, while the reference site was in the Nature Park Kopacki rit, a preserved wetland area with limited anthropogenic influence. Exposure lasted for 3 weeks and was repeated for 3 years (2002-2004). DNA damage was assessed in erythrocytes of the exposed animals by the Comet assay and micronucleus test (MNT). In order to evaluate possible differences in stress responses to polluted water in situ and in aquaria a laboratory exposure was performed with water from the studied location in the second year of the study. Carp from the sites with high anthropogenic influence (Belisce and Osijek) had higher average DNA damage as expressed in both the MNT and Comet assay. Of the two, the Comet assay appeared to be more sensitive following both caging and aquaria exposures. The results from this study suggest that 3 weeks caging exposure of C. carpio may be a useful strategy to monitor for genotoxic agents in freshwater ecosystems.

Natural biotic and anthropogenic stressors can interact to alter contaminant toxicity. Energetic restrictions are potential mechanisms causing this pattern. To identify processes underlying observed effects of predation risk and copper (Cu) on delayed copepod age at maturity, we examined how these 2 stressors affect respiration rates. We tested 2 very different copepod species: the large, pelagic calanoid Calanus finmarchicus and the small, semibenthic harpacticoid Tigriopus brevicornis. Adult individuals were exposed for 12 h to the treatments: predation risk, Cu (23 µg L-1 ), combined predation risk and Cu (23 µg L-1 ), or control. Oxygen concentrations were monitored continuously. The 2 species differed in their responses. We found no clear effects of either stressor in C. finmarchicus. In T. brevicornis, predation risk increased respiration rates, whereas Cu alone had little impact. In contrast, combined exposure to predation risk and Cu interacted to reduce respiration rates to less than expected. We further observed an effect of sex because female-biased T. brevicornis replicates were more sensitive to both predation risk (increased respiration rates) and Cu exposure (reduced respiration rates). The present study provides further evidence that predation risk can interact with copepod responses toward Cu exposure. It is generally assumed that there should be some relationship between tissue residues of contaminants and related biomarker responses in the same individual, with a possible additional modulation through physiological processes. The main basis for such assumptions originated from experimental studies, but it has been less easy to detect and quantify any such relationships in field-collected material. The aim of the current study was to clarify whether the responses of selected biomarkers (cytochrome P-4501A [CYP1A], delta-aminolevulinic acid dehydratase [ALA-D], metallothionein [MT]) could be correlated with physiological parameters and/or contaminant levels. Atlantic cod (Gadus morhua L.) were sampled over a decade at a number of locations along the Norwegian coast. The locations included both polluted and reference areas. In addition to contaminant-related responses (biomarkers), physiological characteristics were registered for each individual and the contaminant load determined. There were clear relationships between responses in all three biomarkers and tissue contaminants, but additional factors such as age or size and liver lipid or liver somatic index also contributed in multiple-regression models. The results indicated that responses in all three biomarkers reflected relevant, contaminant-related processes in the cod even though none of them provided a simple separation between polluted and pristine locations.

Bile metabolites are widely accepted as measures of exposure to polycyclic aromatic hydrocarbons (PAH) and have also been used to assess exposure to alkyl phenols (AP). The aim of this study was to clarify relationships between exposure (through water or diet) and subsequent accumulation of specific PAH and AP metabolites in Atlantic cod (Gadus morhua). Atlantic cod were exposed through water or diet to a synthetic mixture of PAH, alkylated PAH, and AP, simulating the composition of North Sea produced water (formation water; separated from oil or gas on the platform). Fish were exposed through water for 11 mo and the results reported here are from 2 and 8 mo. Fish were subjected to one of four exposures: "control," "low," "high," or "pulsed high." Bile samples were analyzed by gas chromatography/time-of-flight mass spectroscopy (GC/MS-ToF) for identification of PAH and AP metabolites. Cod exposed through diet were divided into six groups receiving different groups of compounds (AP, PAH, or alkylated PAH), a mixture of all compounds, a low-concentration mixture, and controls. A dose-dependent relationship was found for metabolites for most of the PAH and AP, although results were less clear for the more volatile substances such as phenol and naphthalene. The concentration of bile metabolites from fish exposed through water rose with increasing lipophilicity, but this relationship was less clear for fish exposed through diet. Overall, data indicated that bile metabolites of AP and PAH in fish are reliable markers of both water exposure and dietary exposure to such substances, although with the possible exception of the more volatile species such as phenol and naphthalene.

The extent to which accumulations of polycyclic aromatic hydrocarbons (PAH) by semipermeable membrane devices (SPMD) may be correlated to the concentration of PAH metabolites in fish was examined. Atlantic cod (Gadus morhua) and SPMD were exposed over a 4-wk period to a mixture that was designed to simulate the composition of PAH in produced water discharges in the North Sea. Fish bile and whole SPMD were analyzed after exposure in a high, low, and a control treatment tank. PAH metabolites were detected and quantified by means of high-performance liquid chromatography (HPLC) or gas chromatography-time-of-flight mass spectroscopy (GC-ToF-MS) and PAH in SPMD by gas chromatography-mass spectroscopy (GC-MS). The level and duration of exposure were generally reflected in both matrices. Exposure water concentrations, calculated from SPMD accumulations, showed some considerable differences from nominal concentrations for volatile compounds. Concentration factors (CF) for fish based on SPMD-derived water concentrations ranged from 2.9 to 354.3 L/g. CF increased with hydrophobicity with more variation at higher log K(ow). Regression analysis of accumulations of individual compounds in fish and SPMDs showed reasonable but compound-specific correlation, r2 = .54-.85 (C1-phenanthrenes/anthracenes and pyrene, respectively). Comparison of the two exposures suggests that fish CF may not be entirely independent of water concentrations. Overall the results show that accumulation of up to four-ring PAH in SPMD may be related to internal exposure of fish to these substances when exposed through water, as evidenced by metabolite concentrations. The usefulness of using SPMD to measure exposure concentrations in a large-scale flow-through study is also examined.

Environmental contaminants released into water tend to accumulate in sediment, leading to exposure for sediment-dwelling organisms. This study aimed to clarify whether chronic (56 d) exposure to a mixture of environmentally relevant concentrations of contaminants (tributyltin [TBT] and perfluorononanoic acid [PFNA]) spiked in clean sediment would affect the sediment-dwelling polychaete Hediste diversicolor. Endpoints were burial time, biomass change, and cellular energy allocation (CEA). Sediments were spiked with these two contaminants to produce different levels of contamination. Chemical analyses of the sediment following the exposure indicated that the spiking procedure was only partially successful and treatments were grouped into four categories according to contaminant concentrations: "background," "low," "moderate," and "high." No significant differences were found between treatments with respect to burial time, biomass change, or energy storage fractions (proteins, lipids, carbohydrates) in the polychaetes. A significant increase in cellular respiration was found for polychaetes in "moderate" and "high" treatment groups compared to the "background" treatment group. This resulted in significantly lower cellular energy allocation values for "moderate" and "high" treatment groups compared to "background." Although the effects were not marked, the results show that sediment-dwelling organisms may be affected following chronic exposure to environmental contaminants. There is a need to clarify whether chronic exposures exert a negative effect on sediment-dwelling organisms.

Marine tailings disposal from mineral production is expected to have an environmental impact. In this case study we use a discharge of limestone processing tailings to a Norwegian fjord to describe an adaptive management process. The aim of the paper is to describe the development of an environmental adaptive management system (EAMS), contrasted with management simply based on the quantity of the discharge. The main driver for developing a new management system for the submarine tailings deposits was a desire to establish a system based on what was perceived as important to all stakeholders, that is, environmental impact. Involvement of stakeholders is essential, and a resource group with members from fisheries, local interest organizations, scientists, independent experts, and managers from the mining company jointly defined common sets of acceptance criteria to evaluate impact. Introduction of an EAMS has resulted in a change in the company's view of the impact their activity has on the environment and in an increased willingness to initiate monitoring and research to reduce knowledge gaps and uncertainty and impact on the marine environment. Environmental adaptive management has facilitated the development of a more ecologically relevant, integrated, and focused submarine tailings deposits management.
