RESUMO

Response and growth kinetics of microbes in contaminated medium are useful indices for the screening and selection of tolerant species for eco-friendly bio-augmentative remediation of polluted environments. In this study, the heavy metal (HM) tolerance, bioaccumulation and growth kinetics of seven bacterial strains isolated from mining sites to 10 HMs (Cd, Hg, Ni, Al, Cr, Pb, Cu, Fe, Mn and Zn) at varied concentrations (25-600 mgL-1) were investigated. The isolates were phylogenetically (16S rRNA gene) related to Lysinibacillus macroides, Achromobacter spanius, Bacillus kochii, B. cereus, Klebsiella pneumoniae, Pseudomonas mosselii and P. nitroreducens. Metal tolerance, effects on lag phase duration and growth rates were assessed using the 96-well micro-titre method. Furthermore, metal bioaccumulation and quantities within cells were determined by transmission electron microscopy and electron dispersive x-ray analyses. Tolerance to Ni, Pb, Fe and Mn occurred at highest concentrations tested. Growth rates increased with increasing Fe concentrations, but reduced significantly (p < .05) with increasing Zn, Cu, Hg, Cd and Al. Significantly higher (p < .05) growth rates (compared to controls) was found with some isolates in Hg (25 mgL-1), Ni (100 mgL-1), Cr (150 mgL-1), Mn (600 mgL-1), Pb (100 mgL-1), Fe (600 mgL-1) and Al (50 mgL-1). Lag phase urations were isolate- and heavy metal-specific, in direct proportion to concentrations. A. spanius accumulated the most Mn and Zn, while B. cereus accumulated the most Cu. Metals accumulated intra-cellularly without cell morphology distortions. The isolates' multi-metal tolerance, intra-cellular metal bioaccumulation and growth kinetics suggest potentials for application in the synergetic biodegradation and bioremediation of polluted environments, especially HM-rich sites.

To date, only two haemogregarine parasite species have been described from South African anurans: Hepatozoon ixoxo, infecting toads of the genus Sclerophrys (syn. Amietophrynus); and Hepatozoon theileri, parasitising the common river frog, Amietia quecketti. Both species have been characterised using limited morphology, and molecular data from PCR amplified fragments of the 18S rRNA gene. However, no ultrastructural work has been performed thus far. The aim of this study was to add descriptive information on the two species by studying their ultrastructural morphology. Mature gamont stages, common in the peripheral blood of infected frogs, were examined by transmission electron microscopy. Results indicate that H. ixoxo and H. theileri share typical apicomplexan characteristics, but differ markedly in their external cellular structure. Hepatozoon ixoxo is an encapsulated parasite presenting a prominent cap at the truncate pole, and shows no visible modifications to the host cell membrane. In comparison, H. theileri does not present a capsule or cap, and produces marked morphological changes to its host cell. Scanning electron microscopy was performed to further examine the cytopathological effects of H. theileri, and results revealed small, knob-like protrusions on the erythrocyte surface, as well as notable distortion of the overall shape of the host cell.

Copepods of the genus Achtheinus Wilson, 1908 (Pandaridae) are parasites of elasmobranchs that attach to their fins, gill slits and around the nostrils. Specimens of Achtheinus pinguis Wilson, 1912 were collected and examined using histology and scanning electron microscopy to determine their way of attachment to the host and the possible effect on the host. They insert their antennae deep into the dermis of the shark's skin, which causes the most damage due to possible tissue compression and/or fibrosis as well as rupture of the connective tissue. Additionally, the presence of the copepod on the skin causes cell erosion of the epidermal cells and thus reduces the number of epidermal layers. The maxillipeds are used to attach to the placoid scales that cover the shark's skin and probably serve to keep the copepod and inserted antennae in position. This is accomplished by the insertion of the placoid scales into the flaccid corpus of the maxillipeds. Observed damage seems to be negligible to the shark apart from the possibility of secondary infection.

Gold nanoparticles (AuNPs) have shown great potential for use in nanomedicine and nanotechnologies due to their ease of synthesis and functionalization. However, their apparent biocompatibility and biodistribution is still a matter of intense debate due to the lack of clear safety data. To investigate the biodistribution of AuNPs, monodisperse 14-nm dual-radiolabeled [14C]citrate-coated [198Au]AuNPs were synthesized and their physico-chemical characteristics compared to those of non-radiolabeled AuNPs synthesized by the same method. The dual-radiolabeled AuNPs were administered to rats by oral or intravenous routes. After 24 h, the amounts of Au core and citrate surface coating were quantified using gamma spectroscopy for 198Au and liquid scintillation for the 14C. The Au core and citrate surface coating had different biodistribution profiles in the organs/tissues analyzed, and no oral absorption was observed. We conclude that the different components of the AuNPs system, in this case the Au core and citrate surface coating, did not remain intact, resulting in the different distribution profiles observed. A better understanding of the biodistribution profiles of other surface attachments or cargo of AuNPs in relation to the Au core is required to successfully use AuNPs as drug delivery vehicles.

Oil-in-water (o/w) emulsions are commonly converted into solid-like powders in order to improve their physical and chemical stabilities. The aim of this study was to investigate whether whey protein/polysaccharide-stabilized o/w emulsions could be converted into stable oil powders by means of freeze-drying. Moreover, during this study, the effects of pH and polymer type on release and trans(dermal) delivery of salicylic acid, a model drug, from these oil powders were investigated and compared to those of the respective template emulsions and redispersed oil powders. Physical characterization of the various formulations was performed, such as droplet size analysis and oil leakage, and relationships drawn with regards to release and trans(dermal) delivery. The experimental outcomes revealed that the oil powders could be redispersed in water without changing the release characteristics of salicylic acid. pH and polymer type affected the release of salicylic acid from the oil powders, template emulsions, and redispersed powders similarly. Contrary, the transdermal delivery from the oil powders and from their respective redispersed oil powders was differently affected by pH and polymer type. It was hypothesized that the release had been influenced by the electrostatic interactions between salicylic acid and emulsifiers, whereas the transdermal performance could have been determined by the particle or aggregate sizes of the formulations.

BACKGROUND: Human immunodeficiency virus (HIV) and antiretroviral treatment (ART) are both associated with hypercoagulability. Altered clot properties could be a potential mechanism thereof. We aimed to investigate the association of HIV and ART, with fibrinogen and plasma clot properties in a group of Black South Africans. METHODS: At baseline, 151 newly diagnosed people living with HIV (PLWH) and 176 controls were recruited. Some PLWH subsequently commenced with ARTs (n = 70) while others remained ART-naïve (n = 81). Fibrinogen and clot properties (turbidity assay) were investigated from baseline to 5-year follow-up. A sub-group of 21 women (n = 10 ART-treated; n = 11 ART-naïve) with HIV was systematically selected and matched with 12 controls, and additional clot properties (rheometry, permeability and fibre diameter) were investigated. RESULTS: Fibrinogen was lower in the HIV groups compared to the controls, while % γ' fibrinogen was higher. PLWH had shorter lag times and lower maximum absorbance than the controls (p<0.05). Their CLTs on the other hand were longer. Most variables increased over time in all groups, but differences in the degree of change over time was observed for lag time (p = 0.024) and permeability (p = 0.03). Participants who commenced with ART had a tendency of delayed clot formation (p = 0.08) and increased clot permeability (p = 0.005). CONCLUSION: PLWH had lower total fibrinogen concentration and formed less dense clots. They also formed clots that were more difficult to lyse, which likely not resulted from altered clot properties. ART use (NNRTI's) had a moderately protective effect, delaying clot formation, and increasing clot permeability.

Dust pollution can be severe in urban centers near mines and smelters. Identification of dust sources and assessing dust capturing plant morphological traits may help address the problem. A chromium (Cr) mining and ferrochrome smelting region in Sekhukhuneland, South Africa, was investigated to identify the sources of Cr in soil and plant leaf surfaces and to evaluate the association between Cr sources and plant morphology. Combinations of bi- and multivariate statistical analysis techniques were applied. Non-significant relation between Cr quantities in surface soil and on leaf surfaces suggested negligible Cr dust contribution from soil to leaves. Association among Cr, Fe, Mg, Al, and Si levels on leaf surfaces confirmed their shared origin, possibly from chromite containing dust dispersed by mines, smelters, roads, and tailings. Both plant morphology and Cr sources (number and proximity to mines and roads) conjointly determined Cr dust deposition on leaf surfaces. Air mass movement patterns further identified local polluters, i.e., mines, ferrochrome smelters, and roads, as dominant dust sources in the region. Common plant species showed Cr dust adhesion favouring traits (plant tallness, larger leaf area, dense epicuticular wax structures, and larger stomata) and projected dust mitigation prospects for Sekhukhuneland.

Diplobodes africanus Mahunka, 1987, is redescribed by using Scanning Electron Microscopy (SEM) in order to complement optical microscopy. Diplobodes thailande sp. nov. from Thailand is described. For both species we include inclined views, to permit detailed analysis of ventral regions and to aid understanding of the complex relationships between elevated and depressed zones. Diplobodes thailande sp. nov. presents previously undescribed structures on legs III, which are involved in the leg folding process. Rwandabodes kayoveae gen. nov., sp. nov. presents a series of particular characteristics, such as: elevated interlamellar process on prodorsum, composed of a triangular structure, externally delimited by a low lamellar furrow; in setae inserted on elevated interlamellar process; large free lamellar tip; notogaster lacking ridges; anterior genital furrow extends into an oblique lateral depression; depressed area anterior to anal zone; both zones clearly delimited. These characteristics permit easy differentiation from related genera.

Gold nanostars are being used more regularly in the biosensing field. Despite their useful attributes, there is still a need to optimize aspects of the synthesis and stability. The seedless, synthetic method comprising 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES) is a facile, rapid method; however, it produces heteromorphic nanostars. The modification of a HEPES method resulted in a silver-assisted, seedless gold nanostar synthesis method. The nanostars resulting from this method were monodispersed, multi-branched and approximately 37 ± 2 nm in diameter. It proved to be a repeatable method that produced homogeneous and robust nanostars. Once functionalized with polyvinylpyrrolidone 10 000, the new nanostars were observed to be stable in various environments such as salt, ionic strength and cell culture medium. In conclusion, the addition of the silver nitrate improved the morphology of the reported HEPES nanostars for the purpose of nanobiosensor development.

Synkrotima tsalakpmenoi sp. nov. is described from material collected in Zimbabwe and Kenya. This species presents the following characteristics: notogastral integument formed by longitudinally aligned cuticular cords; thick, lanceolate notogastral setae; and genital neotrichy. Synkrotima tsalakpmenoi sp. nov. is compared to S. zimbabwae. Congocepheus thailandae sp. nov. is the first species of this genus described from Thailand, with the following present: large finger-like notogastral projection; notogastral setae c1, c2, da, dm, dp, la, lm, lp, h1, h2 situated on promontories; setae c1, c2 located adjacent and parallel.        A complementary study of Cavaecarabodes hauseri, making use of Scanning Electron Microscopy (SEM), allows for clarification of particularities regarding complex setae as well as some aspects of the integument, and provides a more detailed diagnosis.

Two very particular new species of the family Lohmanniidae were studied and described using optical and Scanning Electron Microscopy (SEM). Meristacarus perikopesissp. n. displays complex cuticular microsculpture with cross-shaped grooves and pusticulate porose areas; ten transversal bands, with reticulate-foveate microsculpture; S4, S5, S7, S10 not crossing medial notogastral plane, amongst other characters. Torpacarus eidikoteraisp. n. with: prodorsum - rostrum weakly bilobate with small central structure and CSO present. Six transversal depressions present, transversal bands absent; but six transversal depressions present, none of the depressions crossing medial notogastral plane.

The nematode Caenorhabditis elegans is used as an ecotoxicological model species in both aqueous medium and solid substrates. It is easy and of low cost to maintain in the laboratory and it produces hundreds of offspring within a short period of time. It also has a small body size (1 mm), making it possible for in vivo assays to be conducted in 12-well plates. Engineered nanomaterials (ENMs) are a class of emerging pollutants. Nanogold (nAu) is used in many consumer products and in vivo drug delivery. These materials can be released into the aquatic environment during production or discarding of consumer products. As nAu is insoluble in water, the sediment would become the final depository for the materials. It has become increasingly important to use sediment dwelling organisms to screen for possible toxicity of these ENMs. In this study C. elegans was exposed to a range of concentrations of nAu and ionic gold in M9-media, acting as a substitute for pore water. After 96-hour growth, fertility and reproduction were determined. Internal structure damage and internalisation of particles in C. elegans were determined by using SEM and CytoViva® Darkfield Imaging. From these images the nanomaterials are distributed around the oocytes in the reproductive organs, as well as the pharynx. Results obtained indicate that nAu affects reproduction more than growth due to internal gonad damage, albeit at very high exposure concentrations, indicating no toxicity at environmentally relevant concentrations. Ionic Au is more toxic than nAu and effects fertility and reproduction due to ion release. These results give more information regarding the toxicity and in vivo uptake of nAu and form part of an environmental risk assessment of ENMs.

In this study, the conversion of silver ions into ∼30.74 nm sized silver nanoparticles (AgNPs) was achieved in 30 min at a reaction temperature of 80-90°C in aqueous leaf extract of Artemisia afra. The synthesised AgNPs showed surface plasmon resonance in the range of 423-438 nm. Spherical and face-centred cubic nanoparticles were confirmed by transmission electron microscope (TEM) and X-ray diffraction (XRD) analysis, respectively. Fourier transform infra-red (FTIR) results indicated that the obtained nanoparticles were stabilised and capped through the carbonyl and carboxylate ion groups possibly from flavonoids, terpenoids, phenolics and esters content of the extracts. In addition, the AgNPs were assessed for their biological potentials against some microbes and, also, their free radical scavenging ability was established. The AgNPs exhibited interesting antimicrobial and antioxidant properties better than the aqueous extract of A. afra.

Antimicrobial resistance is an imminent threat to the effective prevention and treatment of bacterial infections and alternative antimicrobial strategies are desperately needed. Antimicrobial peptides (AMPs) may be promising alternatives to current antibiotics or act as adjuvants to enhance antibiotic potency. Additionally, the use of biodegradable lipid nanoparticles can enhance the antibacterial activity of antibiotics and antimicrobial peptides. In this study, the interaction of the AMPs, nisin Z and melittin, with conventional antibiotics was investigated on Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli. The effectiveness of nanostructured lipid carriers (NLCs) for the entrapment of nisin Z was also evaluated. Findings revealed that nisin Z exhibited additive interactions with numerous conventional antibiotics. Notable synergism was observed for novobiocin-nisin Z combinations. The addition of the non-antibiotic adjuvant EDTA significantly improved the antimicrobial activity of free nisin Z towards E.coli. NLCs containing nisin Z were effective against Gram-positive species at physiological pH, with an increase in effectiveness in the presence of EDTA. Results indicate that nisin Z may be advantageous as an adjuvant in antimicrobial chemotherapy, while contributing in the battle against antibiotic resistance. NLCs have the potential to enhance the antibacterial activity of nisin Z towards Gram-positive bacterial species associated with skin infections.

The newer generation of environmental scanning electron microscopes (ESEMs) allows samples to be viewed under a range of different vacuum conditions. No specific sample preparation protocols are required with the ESEMs, as fresh, unfixed samples are used and discarded later. We have worked out a method that preserves aerial hyphae on biltong that closely resemble fresh specimens and may be stored for viewing at a later date. Another advantage is that fixed samples are more resilient to the variable vacuum encountered in the ESEM. When biltong samples with fungal growth were first studied, we observed that vacuum-related artifacts were induced unless vacuum conditions and changes in pressure were carefully controlled. Damage readily occured in conidia and its delicate hyphae. Fresh, unfixed samples are very vulnerable to these artifacts. In addition, biltong proved to be a problematic study sample because of its high salt content, its hygroscopic nature as well as being laden with spices. To eliminate these artifacts, the preservation of specimens by OsO4 vapor fixation combined with a special sputter-coating technique is described. Previous studies confirmed that OsO4 vapor fixation is superior to traditional immersion fixation methods for the examination of hyphae and conidia of various fungi in a conventional SEM. However, both preparation methods induce sample shrinkage. We observed that OsO4 vapor fixation followed by Au coating under strictly controlled vacuum conditions induced fewer artifacts and gave the best images with minimum distortion in low pressure (LP) mode. The proposed method allows samples to be viewed both in ESEM and LP mode. There are however some disadvantages inherent to ESEM mode. Even when viewing fixed, coated samples, care should be taken to maintain a pressure of not lower than 0.2 Torr in the specimen chamber. It is critical that different samples have the same vacuum exposure history, as shrinkage and collapse were found to be directly related to the lowest pressure the samples were subjected to, both in the sputter coater chamber and in the electron microscope specimen chamber.

The synthesis, spectroscopic characterisation, and thermal studies of pyridyl adducts of Zn(II) and Cd(II) complexes of N-ethyl-N-phenyl dithiocarbamate, represented as [ZnL2py] and [CdL2py2], are reported. Single-crystal X-ray structural analysis of the Zn compound showed that it is five-coordinate with four sulphurs from dithiocarbamate and one nitrogen from pyridine in a distorted square pyramidal geometry. The thermogravimetric studies indicate that the zinc and cadmium compounds undergo fast weight loss, and the temperature at maximum rate of decomposition is at 277 °C and 265 °C respectively, to give the metal (Zn or Cd) sulphide residues. These compounds were used as single molecule precursors to produce nanocrystalline MS (M = Zn, Cd) after thermolysis in hexadecylamine. The morphological and optical properties of the resulting MS nanocrystallites were investigated using transmission electron microscopy (TEM), scanning electron microscopy (SEM), UV-Vis absorption and photoluminescence (PL) spectroscopy, and powdered X-ray diffraction (XRD). By varying the growth time, the temporal evolution of the optical properties and morphology of the nanocrystals were investigated.

Symbiotic nitrogen fixation is sensitive to dark chilling (7 degrees C-15 degrees C)-induced inhibition in soybean (Glycine max). To characterize the mechanisms that cause the stress-induced loss of nodule function, we examined nodule structure, carbon-nitrogen interactions, and respiration in two soybean genotypes that differ in chilling sensitivity: PAN809 (PAN), which is chilling sensitive, and Highveld Top (HT), which is more chilling resistant. Nodule numbers were unaffected by dark chilling, as was the abundance of the nitrogenase and leghemoglobin proteins. However, dark chilling decreased nodule respiration rates, nitrogenase activities, and NifH and NifK mRNAs and increased nodule starch, sucrose, and glucose in both genotypes. Ureide and fructose contents decreased only in PAN nodules. While the chilling-induced decreases in nodule respiration persisted in PAN even after return to optimal temperatures, respiration started to recover in HT by the end of the chilling period. The area of the intercellular spaces in the nodule cortex and infected zone was greatly decreased in HT after three nights of chilling, an acclimatory response that was absent from PAN. These data show that HT nodules are able to regulate both respiration and the area of the intercellular spaces during chilling and in this way control the oxygen diffusion barrier, which is a key component of the nodule stress response. We conclude that chilling-induced loss of symbiotic nitrogen fixation in PAN is caused by the inhibition of respiration coupled to the failure to regulate the oxygen diffusion barrier effectively. The resultant limitations on nitrogen availability contribute to the greater chilling-induced inhibition of photosynthesis in PAN than in HT.
