RESUMO

We report on a new approach for creating water-soluble functionalized vesicles employing N-alkyl-3-boronopyridinium triflates (alkyl = Me, C12H25, C16H33) as sensors for monosaccharides. The nanoaggregate properties were studied by means of DLS, TEM, high-resolution (1)H NMR, and the solvatochromic dyes Reichardt's betaine and Methyl Orange. The vesicles were shown to have 30-200 nm diameters depending on the amphiphile chain length. Diol binding to the vesicles was studied by steady-state fluorescence and UV-vis using Alizarin Red S as a probe in the solution at pH 7.4 in the presence and in the absence of D-glucose and D-fructose. Strong sensing ability of boronic acid functional moieties in the order D-fructose > D-glucose was demonstrated, and apparent binding constants were estimated.

Chloroaluminum phthalocyanine (ClAlPc) is a promising sensitizer molecule for photodynamic therapy, but its hydrophobicity makes it difficult to formulate. In this study, we have efficiently encapsulated ClAlPc into gelled soybean oil particles dispersed in water. 12-Hydroxystearic acid (HSA) and polyethyleneimine (PEI) were the gelling and stabilizing agents, respectively. The preparation process involved hot emulsification above the gelation temperature (Tgel), followed by cooling to room temperature, which gave a colloidal dispersion of gelled particles of oil in aqueous medium. The gelled particles containing ClAlPc had a medium diameter of 280 nm, homogeneous size distribution (polydispersity index ≈0.3) and large positive zeta potential (about +50 mV) and showed a spherical morphology. The gelled oil particle formulations exhibited good physical stability over a 6-month period. ClAlPc interfered with the HSA self-assembly only slightly, and decreased the gelation temperature to a small extent; however it did not affect gelation process of the oil droplets. The amounts of PEI and HSA employed during the preparation allowed us to control particle size and the dispersion stability, a phenomenon that results from complex electrostatic interactions between the positively charged PEI and the negatively charged HSA fibers present on the gelled particles surface. In summary, by using the right ClAlPc, HSA, and PEI proportions, we prepared very stable dispersions of gelled soybean oil particles with excellent ClAlPc encapsulation efficiency. The obtained colloidal formulation of gelled oil particles loaded with ClAlPc shall be very useful for photodynamic therapy protocols.

In a first step towards chemical sensors using molecular imprinted materials, the complexing characteristics of diethyl 4-nitrobenzylphosphonate, an organophosphate pesticide analogue, have been studied. Two molecules have been assessed as potential interacting moieties, specifically a fluoroalcohol and an aromatic acid. The interactions have been first characterized by regular methods, such as 1H, 31P NMR and IR spectroscopy. These showed a stoichiometry 1/1 for both complexes and association constants, respectively, close to 40+/-10 and 12+/-2 M(-1). In a second step, isothermal titration calorimetry was used and a method was developed to obtain low-association constants. The association constant could be obtained for the fluoroalcohol ligand and was found equal to 63+/-0.7 M(-1). For the acidic molecule, an appropriate model could not be found, preventing the evaluation of this constant.

Several liquid crystalline imprinted materials have been synthesized from polysiloxanes or polyacrylates bearing mesogenic side-chains and low contents of cross-linkers. They were imprinted by various achiral or chiral templates, then tested for molecular recognition or assessed as specific catalysts. All mesogenic imprinted networks exhibit a much higher affinity towards the template than non-imprinted networks. On the other hand, the molecular trapping capacity was shown to be much greater than that of most of the previously studied non-mesomorphous systems. Moreover, it was shown that mesomorphic order provides significant enhancement to the bonding between the template and the liquid crystalline network and reinforces the shape memory of the imprinted cavities. Some of these materials were used to catalyze the isomerization of benzisoxazole. They exhibited an acceleration effect close to 100 between imprinted sites and non-imprinted ones. Lastly, cholesteric networks, that were imprinted by a chiral template, showed good properties for the enantiomer separation leading to an enantiomeric excess of 35% and a capacity of around 1 mmol g(-1).

RESUMO

A hydrogen-bonded complex between an aromatic acid and an enantiopure chiral amine has been dissolved in a nematic solvent, giving rise to a cholesteric medium. Fourier transform infrared (FT-IR) experiments have been performed at various temperatures on both sides of the cholesteric-isotropic transition. Liquid crystalline order provides significant enhancement to the strength of interaction, inducing a discontinuous jump in concentration of the complex at the cholesteric-isotropic transition.

RESUMO

Liquid crystalline thin films elastomers that are able to bind pesticides have been developed. The synthesis involves grafting mesogen and crosslinkable groups on a polysiloxane chain in the presence of a template molecule. The molecular imprinted material is obtained after thin film deposition, UV crosslinking and washing. Experiments of readsorption of pesticide are presented. Development of a multisensor platform based on thermal and capacitive sensors is described and tests of deposition of the polymer film are presented.

Two series of water-soluble dendritic catanionic assemblies, acting as multisite analogues of galactosylceramide (Galbeta1cer), have been prepared with the goal of blocking HIV infection prior to the entry of the virus into human cells. Trifunctional and hexafunctional cinnamic acid-terminated dendrimers have been synthesized from phosphorus-containing dendrimers bearing aldehyde end groups. A classical acid-base reaction performed in water between acid-terminated dendrimers and stoichiometric amounts of N-hexadecylamino-1-deoxylactitol provided the expected catanionic assemblies. Antiviral assays on these supramolecular entities confirmed the crucial roles both of multivalency effects and of lipophilicity on the biological activity of Galbeta1cer analogues. Moreover, correlation between in vitro tests and molecular modeling highlights the specific influence of the assembly shape on the anti-HIV efficiency, with the tri- and hexafunctional cored dendrimers, both decorated with 12 sugar moieties, exhibiting IC50 values of 1.1 and 0.12 microM, respectively.
