RESUMO

We present an autopsy case of a 19-year-old man with a history of epilepsy whose unwitnessed sudden death occurred unexpectedly in the night. About 4 years before death, he was diagnosed with unilateral optic neuritis (ON). Demyelinating disease was suspected, but he was lost to follow up after the recovery. Six months before death, he received a second dose of mRNA coronavirus disease 2019 (COVID-19) vaccine. Three months before death, he experienced epileptic seizures for the first time. Seventeen days before death, he was infected with COVID-19, which showed self-limited course under home isolation. Several days before death, he complained of seizures again at night. Autopsy revealed multifocal gray-tan discoloration in the cerebrum. Histologically, the lesions consisted of active and inactive demyelinated plaques in the perivenous area of the white matter. Perivascular lymphocytic infiltration and microglial cell proliferation were observed in both white matter and cortex. The other major organs including heart and lung were unremarkable. Based on the antemortem history and postmortem findings, the cause of death was determined to be multiple sclerosis with suspected exacerbation. The direct or indirect involvement of cortex and deep gray matter by exacerbated multiple sclerosis may explain the occurrence of seizures. Considering the absence of other structural abnormalities except the inflammatory demyelination of the cerebrum, fatal arrhythmia or laryngospasm in the terminal epileptic seizure may explain his sudden unexpected death in the benign circumstances. In this case, the onset of seizure was preceded by COVID-19 vaccination, and the exacerbation of seizure was preceded by COVID-19 infection, respectively. Literature reporting first manifestation or relapse of multiple sclerosis temporally associated with COVID-19 vaccination or infection are reviewed.

In this study, the electrical resistance of the whole body and histological changes of skeletal muscle were investigated in rats according to the increase in radiation dose. A total of 15 male Sprague-Dawley rats (5-weeks-old) were randomly divided into 5 groups (each, n = 3). Each group received 1 Gy, 5 Gy, 10 Gy and 20 Gy systemic exposure, and the non-irradiated group was used as a control for morphological comparison. After attaching an electrode clip to the forelimb of the rat, an AC frequency was applied before and 4 days after irradiation using an impedance/gain-phase analyzer, and the measurement system was automatically controlled with LabVIEW. Comparing to before irradiation after 4 days, the difference in the average impedance values at 1 Gy, 5 Gy, 10 Gy, and 20 Gy was 1188±989 ohm, 3076±2251 ohm, 7650±6836 ohm, and 10478±6250 ohm, respectively. By comparing the normal group and the experimental group, muscle fiber atrophy and collagen fibers around blood vessels were observed (p < 0.05, control group vs 5 Gy or more high-dose group). These results confirmed the previously reported morphological changes of skeletal muscle and our hypothesis that whole-body impedance measurement enables to reflect tissue changes after irradiation.

We present autopsy findings of a 22-year-old man who developed chest pain 5 days after the first dose of the BNT162b2 mRNA vaccine and died 7 hours later. Histological examination of the heart revealed isolated atrial myocarditis, with neutrophil and histiocyte predominance. Immunohistochemical C4d staining revealed scattered single-cell necrosis of myocytes which was not accompanied by inflammatory infiltrates. Extensive contraction band necrosis was observed in the atria and ventricles. There was no evidence of microthrombosis or infection in the heart and other organs. The primary cause of death was determined to be myocarditis, causally-associated with the BNT162b2 vaccine.

This study analyzes the response of increasing radiation dose to the pork tenderloin tissue. Considering its significant cell structure, pork tenderloin tissue samples are selected for the experimental objects to measure their electrical impedance characteristics. This study proposes and investigates an effective approach to characterize the variation of the internal change of the components of pork tenderloin tissues caused by radiation. Changes in the pork tenderloin tissues are that the gap of the myotome is more far apart with increase of radiation dose because of the destroyed Myofibrils under the damage. With the increase of radiation dose, the impedance value of the pork tenderloin tissue decreases. Each of mean differences in the impedance values before and after irradiation dose under 1 Gy, 2 Gy and 4 Gy show 0.55±0.03, 1.09±0.14 and 1.97±0.14, respectively. However, the mean difference substantially increases to 13.08±0.16 at irradiation dose of 10 Gy. Thus, the cell membrane shows the most severe rupture at a radiation dose of 10 Gy. Changes in the microstructure of the irradiated pork tenderloin tissue samples are also checked and validated by a transmission electron microscope.

Sphingomonas is a Gram-negative, yellow-pigmented, chemoheterotrophic, strictly aerobic bacterium. The bacterium is known to be metabolically versatile and can utilize a wide range of natural compounds as well as some types of environmental contaminants, such as creosote, polychlorinated biphenyls, etc. Here, we report the draft genome sequence of Sphingomonas echinoides ATCC 14820, which will provide additional information to enhance our understanding of metabolic versatility of Sphingomonas.

RESUMO

The endosymbiotic bacterium Sphingomonas sp. strain PAMC 26605 was isolated from Arctic lichens (Ochrolechia sp.) on the Svalbard Islands. Here we report the draft genome sequence of this strain, which could provide further insights into the symbiotic mechanism of lichens in extreme environments.

RESUMO

The draft genome of Salinibacterium sp. PAMC 21357, isolated from permafrost soil of Antarctica, was determined. Here we present a 3.1-Mb draft genome sequence of Salinibacterium sp. that could provide further insight into the genetic determination of its cold-adaptive properties.

Fungal endophytes have been recorded in various plant species with a richness of diversity, and their presence plays an essential role in host plant protection against biotic and abiotic stresses. This study applied the Illumina MiSeq sequencing platform based on the amplification of fungal ribosomal ITS2 region to analyze fungal endophytic communities of two oak species (Quercus mongolica and Q. serrata) with different oak wilt disease susceptibilities in Korea. The results showed a total of 230,768 sequencing reads were obtained and clustered at a 97% similarity threshold into 709 operational taxonomic units (OTUs). The OTUs of Q. serrata were higher than that of Q. mongolica with the number of 617 OTUs and 512 OTUs, respectively. Shannon index also showed that Q. serrata had a significantly higher level of fungal diversity than Q. mongolica. Total of OTUs were assigned into 5 fungal phyla, 17 classes, 60 orders, 133 families, 195 genera, and 280 species. Ascomycota was the dominant phylum with 75.11% relative abundance, followed by Basidiomycota with 5.28%. Leptosillia, Aureobasidium and Acanthostigma were the most abundant genera detected in Q. serrata with the average relative abundance of 2.85, 2.76, and 2.19%, respectively. On the other hand, Peltaster, Cladosporium and Monochaetia were the most common genera detected in Q. mongolica with the average relative abundance of 4.83, 3.03, and 2.87%, respectively. Our results indicated that fungal endophytic communities were significantly different between two oak species and these differences could influence responses of host trees to oak wilt disease caused by Raffaelea quercus-mongolicae.

A psychrophilic yeast was isolated from an Arctic pond and its culture supernatant showed ice-binding activity. This isolate, identified as Leucosporidium sp. based on an analysis of the D1/D2 and ITS regions of its ribosomal DNA, produced a secretory ice-binding protein (IBP). Yeast IBP was purified from the culture medium to near homogeneity by the ice affinity method and appeared to be glycosylated with a molecular mass of approximately 26 kDa. In addition, the yeast IBP was shown to have thermal hysteresis (TH) and recrystallization inhibition (RI) activities. The full-length cDNA for yeast IBP was determined and was found to encode a 261 amino acid protein with molecular weight of 26.8 kDa that includes an N-terminal signal peptide and one potential N-glycosylation site. The deduced protein showed high sequence identity with other IBPs and hypothetical IBPs from fungi, diatoms, and bacteria, clustering with a class of ice-active proteins.

Oak wilt disease caused by Raffaelea quercus-mongolicae has emerged obviously in Korea. We selected antifungal isolates against R. quercus-mongolicae among 368 endophytic fungal isolates from different parts of oak and pine trees. The experiment was conducted in the primary and secondary screenings by dual culture test. The antifungal activity of the selected isolates was assessed in culture filtrate test based on the inhibition rates in mycelial growth, sporulation, and spore germination of oak wilt fungus. Five isolates, E089, E199, E282, E409 and E415, showed strong antifungal activity in culture filtrate test, and their antifungal activity decreased on the culture media supplemented with heated culture filtrate. Higher mycelial growth inhibitions on the unheated media were recorded in E409 (Colletotrichum acutatum), E089 (Daldinia childiae), E415 (Alternaria alternata) and E199 (Daldinia childiae) with the inhibition rates of 79.0%, 70.1%, 68.9% and 64.5%, respectively. These isolates also had the higher sporulation inhibitions on unheated media with the rates of 96.8%, 84.2%, 82.8% and 80.5%, respectively. The spore germination of the oak wilt fungus was completely inhibited by E282 (Nectria balsamea) on both unheated and heated media. These results showed that a higher number of potent antifungal isolates against oak wilt fungus was isolated from the petiole compared to the other parts. This study could contribute to the development of biological control approaches for the management of oak wilt disease caused by R. quercus-mongolicae.

BACKGROUND/AIM: Uniform treatment of hepatocellular carcinoma (HCC) with molecular targeted drugs (e.g., sorafenib) results in a poor overall tumor response when tumor subtyping is absent. Patient stratification based on actionable gene expression is a method that can potentially improve the effectiveness of these drugs. Here we aimed to identify the clinical application of actionable genes in predicting response to sorafenib. METHODS: Through quantitative real-time reverse transcription PCR, we analyzed the expression levels of seven actionable genes (VEGFR2, PDGFRB, c-KIT, c-RAF, EGFR, mTOR, and FGFR1) in tumors versus noncancerous tissues from 220 HCC patients treated with sorafenib. Our analysis found that 9 responders did not have unique clinical features compared to nonresponders. A receiver operating characteristic curve evaluated the predictive performance of the treatment benefit score (TBS) calculated from the actionable genes. RESULTS: The responders had significantly higher TBS values than the nonresponders. With an area under the curve of 0.779, a TBS combining mTOR with VEGFR2, c-KIT, and c-RAF was the most significant predictor of response to sorafenib. When used alone, sorafenib had a 0.7-3% response rate among HCC patients, but when stratifying the patients with actionable genes, the tumor response rate rose to 15.6%. Furthermore, actionable gene expression is significantly correlated with tumor response. CONCLUSIONS: Our findings on patient stratification based on actionable molecular subtyping potentially provide a therapeutic strategy for improving sorafenib's effectiveness in treating HCC.

Manganese superoxide dismutase (leMnSOD) cDNA was cloned from the Antarctic bivalve Laternula elliptica. The full-length cDNA of leMnSOD is 1238 bp in length and contains an open reading frame of 681 bp encoding 226 amino acid residues including a putative mitochondrial targeting peptide of 26 amino acids in the N-terminal region. The calculated molecular mass is 24.8 kDa with an estimated isoelectric point of 6.75. leMnSOD signatures from 185 to 192 (DVWEHAYY) and four conserved amino acids (H52, H11, D185, and H192) responsible for binding manganese were observed. Sequence comparison showed that leMnSOD had high levels of identity with MnSOD from Haliotis discus discus, Mizuhopecten yessoensis, and Crassostrea gigas (68%, 66%, and 59%, respectively). RT-PCR analysis revealed the presence of leMnSOD transcripts in all tissues examined. Quantitative real-time RT-PCR assay indicated that treatment with polychlorinated biphenyls (PCBs) significantly increased leMnSOD mRNA expression in an organ-, time-, and dose-dependent manner. The mRNA expression with exposure to PCBs at 0.1 and 10 ppb reached the highest level at 6 h and then recovered slightly from 6 to 48 h in the gill. In contrast, the expression of leMnSOD mRNA showed a different expression pattern related to PCB concentration in the digestive gland. The mRNA expression at 0.1 ppb PCBs increased up to 12 h and then decreased by 48 h, but increased immediately at 10 ppb PCBs. The leMnSOD was overproduced in Escherichia coli and purified. The recombinant leMnSOD showed maximum activity at pH 9.0, and it retained more than 50% of its original activity after incubation for 30 min at 40 degrees C.

Pinus densiflora Siebold et Zucc. is the major green canopy species in the mountainous area of Korea. To assess the response of resin acid biosynthetic genes to mechanical and chemical stimuli, we cloned cDNAs of genes encoding enzymes involved in the 2-C-methyl-d-erythritol 4-phosphate (MEP) pathway (1-deoxy-d-xylulose 5-phosphate synthase (PdDXS), 1-deoxy-d-xylulose 5-phosphate reductoisomerase (PdDXR) and 1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate reductase (PdHDR)) by the rapid amplification of cDNA ends (RACE) technique. In addition, we cloned the gene encoding abietadiene synthase (PdABS) as a marker for the site of pine resin biosynthesis. PdHDR and PdDXS occurred as two gene families. In the phylogenetic trees, PdDXSs, PdDXR and PdHDRs each formed a separate clade from their respective angiosperm homologs. PdDXS2, PdHDR2 and PdDXR were most actively transcribed in stem wood, whereas PdABS was specifically transcribed. The abundance of PdDXS2 transcripts in wood in the resting state was generally 50-fold higher than the abundance of PdDXS1 transcripts, and PdHDR2 transcripts were more abundant by an order of magnitude in wood than in other tissues, with the ratio of PdHDR2 to PdHDR1 transcripts in wood being about 1. Application of 1 mM methyl jasmonate (MeJA) selectively enhanced the transcript levels of PdDXS2 and PdHDR2 in wood. The ratios of PdDXS2 to PdDXS1 and PdHDR2 to PdHDR1 reached 900 and 20, respectively, on the second day after MeJA treatment, whereas the transcript level of PdABS increased twofold by 3 days after MeJA treatment. Wounding of the stem differentially enhanced the transcript ratios of PdDXS2 to PdDXS1 and PdHDR2 to PdHDR1 to 300 and 70, respectively. The increase in the transcript levels of the MEP pathway genes in response to wounding was accompanied by two orders of magnitude increase in PdABS transcripts. These observations indicated that resin acid biosynthesis activity, represented by PdABS transcription, was correlated with the selective transcriptions of PdDXS2 and PdHDR2. Introduction of PdDXS2, PdHDR1 and PdHDR2 rescued their respective knockout Escherichia coli mutants, confirming that at least these three genes were functionally active. Intracellular targeting of the green fluorescent protein fused to the N-terminal 100 amino acid residues of these genes in the Arabidopsis transient expression system showed that the proteins were all targeted to the chloroplasts. Our results suggest that the MEP pathway regulates resin biosynthesis in the wood of P. densiflora by differential transcription of the multiple PdDXS and PdHDR genes.

We investigated the ectomycorrhizal (ECM) fungal colonization status of Pinus thunbergii mature trees and regenerating seedlings varying in age in coastal pine forests on the east coast of Korea. We established one 20 x 20-m plot at each of two study sites at P. thunbergii coastal forests in Samcheok. Fifty soil blocks (5 x 5 x 15 cm) were sampled at regular intervals, and ten P. thunbergii seedlings of age 0, 1-3, 3-5, and 5-10 years were sampled in each study plot. In total of 27 ECM fungal taxa, Cenococcum geophilum was dominant, followed by Russula sp., Sebacina sp., and unidentified Cortinuris sp. in mature trees. In 0-year-old seedlings, some fungal species such as Sebacina sp., C. geophilum, and unidentified Cortinarius sp. were dominant whereas only C. geophilum was dominant after 1 year, and there were no apparent succession patterns in ECM fungal compositions beyond a host age of 1 year. Most ECM fungal taxa that had colonized seedlings of each age class were also observed in roots of mature trees in each site. These taxa accounted for 86.7-100% and 96.4-98.4% of ECM abundance in seedlings and mature trees, respectively. The results indicate that the species composition of ECM fungal taxa colonizing seedlings of different age in forests is similar to that of surrounding mature trees. Our results also showed that C. geophilum is a common and dominant ECM fungus in P. thunbergii coastal forests and might play a significant role in their regeneration.

It is apparently a common practice to fracture the inferior turbinate medially and superiorly to facilitate the fenestration of the inferior meatus, and to fracture medially and lateralize the inferior turbinate during turbinoplasty. However, it is also known that medial fracturing of the inferior turbinate may induce lateral displacement of the uncinate process. We investigated medial fracturing of the inferior turbinate to see whether it had any effect on changing the position of the uncinate process. A retrospective study was done on 23 patients who underwent medial fracturing of the inferior turbinate during submucosal turbinoplasty or turbinectomy from January 2004 through September 2006. By analyzing ostiomeatal-unit computed tomography, we measured the angle of the uncinate process, the minimal width of the ethmoid infundibulum, and the size of the maxillary sinus ostium. The angle of the uncinate process was 14.19-32.45 degrees , the minimal width of the ethmoid infundibulum was 0.61-2.45 mm, and the size of the maxillary sinus ostium was 2.14-6.77 mm. After the surgery, the angle of the uncinate process was 15.56-32.51 degrees , the minimal width of the ethmoid infundibulum was 0.53-2.52 mm, and the size of the maxillary sinus ostium was 2.18-7.01 mm. Pathologic change in the ostiomeatal-unit was not observed for an average period of 14.9 months (range 10.3-21.8 months) follow-up period. Our report suggests that the medial fracturing of the inferior turbinate does not alter the position of the uncinate process and the size of the maxillary sinus ostium. Clinically, it does not seem to affect normal physiology of the sinus function nor does it cause sinusitis.

The seroprevalence of cryptosporidiosis was examined using patients' sera collected from hospitals located in 4 different areas of the Republic of Korea. ELISA was used to measure antibody titers against Cryptosporidium parvum antigens from a total of 2,394 serum samples, which were collected randomly from patients in local hospitals; 1) Chungbuk National University Hospital, 2) Konkuk University Hospital, 3) local hospitals in Chuncheon, Gangwon-do (province), 4) Jeonnam National University Hospital, from 2002 through 2003. Of the 2,394 samples assayed, 34%, 26%, and 56% were positive for C. parvum-specific IgG, IgM, and IgA antibodies, respectively. Positive IgG titers were most common in sera from Jeonnam National University Hospital, Gwangju, Jeollanam-do, and positive IgM titers were most common in sera from Chungbuk National University Hospital, Cheongju, Chuncheongbuk-do. The seropositivity was positively correlated with age for both the IgG and IgA antibodies but was negatively correlated with age for the IgM antibodies. Western blotting revealed that 92%, 83%, and 77% of sera positive for IgG, IgM, and IgA ELISA reacted with 27-kDa antigens, respectively. These results suggested that infection with Cryptosporidium in hospital patients occurs more commonly than previously reported in the Republic of Korea.

In this study, we identified four canonical calmodulin genes in the Pacific abalone, Haliotis discus hannai. Their full-length cDNAs were variable in the 5' and 3' untranslated regions, but highly similar (91-97%) in the coding region. Each of the genes encoded 149 amino acids, with 93-97% similarity among themselves and 94-98% similarity with human CAM I. There were 54 substitutions distributed unevenly throughout the coding regions, found mostly in the third codon position. Gene structure analysis revealed that each of the calmodulin genes comprised five exons and four introns. The intron positions and phases were identical and there were no introns in the fourth exon. The corresponding introns differed in their sequences and sizes. Expression profiles of nine tissues from abalone revealed that the calmodulin genes were transcribed in common in gill and mantle tissue, but differentially in the other tissues. A phylogenetic analysis based on the amino acid sequences revealed that calmodulin C was the most common isoform in Gastropoda and calmodulin was the most diverged isoform. An in silico analysis of the calmodulin genes identified paralogous genes in other Haliotis species, indicating that gene duplication might have occurred in the last common ancestor of Haliotis. Two-hundred and fifty-five strains of actinomycetes isolated from soil samples were screened for their antagonistic activities against four well-known wood decay fungi (WDF), including a brown rot fungus, Gloeophyllum trabeum and three white rot fungi Donkioporia expansa, Trametes versicolor, and Schizophyllum commune. A dual culture assay using culture media supplemented with heated or unheated culture filtrates of selected bacterial strains was used for the detection of their antimicrobial activity against four WDF. It was shown that Streptomyces atratus, S. tsukiyonensis, and Streptomyces sp. greatly inhibited the mycelial growth of the WDF tested compared with the control. To evaluate the biocontrol efficacy of S. atratus, S. tsukiyonensis, and Streptomyces sp., wood blocks of Pinus densiflora inoculated with three selected Streptomyces isolates were tested for weight loss, compression strength (perpendicular or parallel to the grain), bending strength, and chemical component changes. Of these three isolates used, Streptomyces sp. exhibited higher inhibitory activity against WDF, especially G. trabeum, as observed in mechanical and chemical change analyses. Scanning electron microscopy showed that cell walls of the wood block treated with Streptomyces strains were thicker and collapsed to a lesser extent than those of the non-treated control. Taken together, our findings indicate that Streptomyces sp. exhibits the potential to be used as a biocontrol agent for wood decay brown rot fungus that causes severe damage to coniferous woods.

Lactarius and Lactifluus are milkcaps that are characterized by the secretion of latex. These two genera are part of a globally distributed cosmopolitan group of ectomycorrhizal fungi that is an important food resource in various ecosystems. Recently, the taxonomy of Lactarius and Lactifluus has been revised based on molecular phylogenetics. Despite the importance of these taxa, Korean species of both genera are poorly understood. In an effort to describe milkcap species that are indigenous to Korea, a long-term study has been initiated. During a recent survey, eight species of milkcaps that were previously unrecorded in Korea were detected based on morphological observation and sequence analysis of the internal transcribed spacer region: five Lactarius species (Lactarius atromarginatus, L. austrotorminosus, L. kesiyae, L. tabidus, and L. vietus) and three Lactifluus species (Lactifluus acicularis, Lf. pilosus, and Lf. pinguis). Detailed morphological descriptions and phylogenetic relationships of these species are provided in this article.

This prospective study attempts to explore the effect of glycerol on vestibular evoked myogenic potential (VEMP) in patients with definite unilateral Meniere's disease (MD) and to investigate whether the test reflects different pathologic states of endolymphatic hydrops (EH) in the inner ear by comparing the results of glycerol pure tone audiometry (PTA). Twenty-eight patients with definite unilateral MD were studied. As a control group, 11 patients with vestibular neuritis were selected. Sixteen ears of normal volunteers were enrolled to acquire normal range of the test. Each MD patient underwent glycerol testing measure with both conventional PTA and VEMP testing. Eleven vestibular neuritis patients also underwent glycerol-VEMP testing. The difference ratio was adopted to compare the changes in p1-n1 amplitude and latency of VEMP after the administration of glycerol (1.3 g/kg). Significant changes in the VEMP wave after the glycerol loading were seen in amplitude, but not in latency with MD patients. On the affected side, the amplitude of the p1-n1 biphasic wave significantly increased in 11 of 28 (39.3%) ears and decreased in 2 (7.1%) ears. On the unaffected side, 5 (17.9%) ears showed significant increase in amplitude. Significant changes in PTA were noted after administration of glycerol in 14 of 28 (50%) patients with MD. However, there was no statistically significant correlation between the test results. Glycerol administration had no significant effect on VEMP in patients with vestibular neuritis. The amplitude of VEMP in some patients with MD was increased, but the latency was not influenced by oral administration of glycerol. The severity of EH appears to vary in different parts of the inner ear. Glycerol-VEMP test results in patients with vestibular neuritis suggest that the VEMP reflects potential abnormality in the sacculocollic pathway, while glycerol-VEMP identifies the existence of EH in the saccule. VEMP and PTA after glycerol administration provide potential status of MD.
