RESUMO

A series of polyelectrolyte multilayers (PEMs) has been fabricated using polyanions and polycations that have repeat units (i) similar in structure and composition (matched), (ii) partially similar in structure and composition (semimatched), and (iii) very different in structure and composition (mismatched). The primary aim of this investigation was to determine whether the matching of the polyelectrolytes can significantly influence the permeability properties of hyperthin PEMs. While matching, per se, was not found to be a key factor in defining membrane permeability, large differences in permeability were observed (the permeances of N2 varied by a factor of 20), which were correlated with the concentration of pendant aryl groups present, i.e., the greater the concentration of these groups, the higher the permeability. Analysis by AFM indentation measurements further revealed that high-permeability PEMs tend to be more compliant than low-permeability PEMs. These findings underscore the need for considering a broad range of polyelectrolyte combinations when optimizing a particular functional property of PEMs.

Bacterial peptidoglycan is a mesh-like network comprised of sugars and oligopeptides. Transpeptidases cross-link peptidoglycan oligopeptides to provide vital cell wall rigidity and structural support. It was recently discovered that the same transpeptidases catalyze the metabolic incorporation of exogenous D-amino acids onto bacterial cell surfaces with vast promiscuity for the side-chain identity. It is now shown that this enzymatic promiscuity is not exclusive to side chains, but that C-terminus variations can also be accommodated across a diverse range of bacteria. Atomic force microscopy analysis revealed that the incorporation of C-terminus amidated D-amino acids onto bacterial surfaces substantially reduced the cell wall stiffness. We exploited the promiscuity of bacterial transpeptidases to develop a novel assay for profiling different bacterial species.

Emergency remote education (ERD) adopted by universities amidst the COVID-19 pandemic has pursued to maintain students' satisfaction. The current research inquiries into perception of the satisfaction towards the quality of classes during ERD. The research is carried out through a mixed sequential approach. Six focus groups in its qualitative phase, and 2074 students from the business undergraduate program in Colombia, Peru, and Mexico in its quantitative phase. Measure of satisfaction towards ERD establishes three dimensions: Concerns about academic quality, teaching strategies used by professors, and perceptions of access limitations. This study identifies a moderating effect of the perceptions of access limitations on the relationship of the teaching strategies and concerns about academic quality. In high constraints, =.-016, p < 0.0001, the relationship is weaker than medium constraints, = -0.22, p < 0.0001. In turn, the medium access limitation condition shows a weaker effect than the high limitation condition, = -0.28, p < 0.0001.

RESUMO

Stereotactic body radiation therapy is a novel treatment option for refractory ventricular tachycardia. We present a case of ventricular tachycardia, with epicardial origin located in large inferior infarct scar, that recurred despite treatment with multiple antiarrhythmic drugs, catheter ablation, and cardiac sympathetic denervation. Stereotactic body radiation therapy safely and effectively terminated the arrhythmia. (Level of Difficulty: Advanced.).

BACKGROUND: Progressive fibrosing interstitial lung disease (ILD) is characterised by parenchymal scar formation, leading to high morbidity and mortality. The ability to predict this phenotype remains elusive. We conducted a proteomic analysis to identify novel plasma biomarkers of progressive fibrosing ILD and developed a proteomic signature to predict this phenotype. METHODS: Relative plasma concentrations for 368 biomarkers were determined with use of a semi-quantitative, targeted proteomic platform in patients with connective tissue disease-associated ILD, chronic hypersensitivity pneumonitis, or unclassifiable ILD who provided research blood draws at the University of California (discovery cohort) and the University of Texas (validation cohort). Univariable logistic regression was used to identify individual biomarkers associated with 1-year ILD progression, defined as death, lung transplant, or 10% or greater relative forced vital capacity (FVC) decline. A proteomic signature of progressive fibrosing ILD was then derived with use of machine learning in the University of California cohort and validated in the University of Texas cohort. FINDINGS: The discovery cohort comprised 385 patients (mean age 63·6 years, 59% female) and the validation cohort comprised 204 patients (mean age 60·7 years, 61% female). 31 biomarkers were associated with progressive fibrosing ILD in the discovery cohort, with 17 maintaining an association in the validation cohort. Validated biomarkers showed a consistent association with progressive fibrosing ILD irrespective of ILD clinical diagnosis. A proteomic signature comprising 12 biomarkers was derived by machine learning and validated in the University of Texas cohort, in which it had a sensitivity of 0·90 and corresponding negative predictive value of 0·91, suggesting that approximately 10% of patients with a low-risk proteomic signature would experience ILD progression in the year after blood draw. Those with a low-risk proteomic signature experienced an FVC change of +85·7 mL (95% CI 6·9 to 164·4) and those with a high-risk signature experienced an FVC change of -227·1 mL (-286·7 to -167·5). A theoretical clinical trial restricted to patients with a high-risk proteomic signature would require 80% fewer patients than one designed without regard to proteomic signature. INTERPRETATION: 17 plasma biomarkers of progressive fibrosing ILD were identified and showed consistent associations across ILD subtypes. A proteomic signature of progressive fibrosing ILD could enrich clinical trial cohorts and avoid the need for antecedent progression when defining progressive fibrosing ILD for clinical trial enrolment. A series of polycations bearing one, two, and three pendant quaternary ammonium groups per repeat unit has been synthesized and combined with poly(sodium 4-styrenesulfonate) to produce hyperthin polyelectrolyte multilayers. Incremental addition of quaternary ammonium groups per repeat unit leads to decreased permeances for H2, CO2, and N2 and increased H2/CO2 and CO2/N2 permeation selectivities. These results, together with analysis of the composition from X-ray photoelectron spectra and values of Young's modulus from nanoindentation analysis, show that oligo pendant ions provide a means for increasing volume charge density and tightening polyelectrolyte multilayers.
