RESUMO

The column chromatography with silica gel, reversed-phase C_(18), and Sephadex LH-20 was employed to separate the methanol extract of the aerial parts of Hypericum elatoides. The compounds were identified by the comprehensive analysis of IR, NMR, and MS data as methyl 8-O-ß-D-glucopyranosyl-(Z)-5-octenoate(1), methyl 3-O-ß-D-glucopyranosyl-4-methylhexanoate(2), byzantionoside B(3), 9-epi-blumenol C glucoside(4), corchoionoside C(5),(6S,9R)-roseoside(6), cis-p-coumaric acid 4-O-ß-D-glucopyranoside(7), trans-p-coumaric acid 4-O-ß-D-glucopyranoside(8), methyl 3-(4-hydroxyphenyl)propanoate(9),(E)-chlorogenic acid methyl ester(10), quercetin-3-O-ß-D-glucopyranoside(11), ß-sitosterol(12), stigmasterol(13), stigmast-4-en-3-one(14), ß-amyrin(15), daucosterol(16), sitoindoside Ⅰ(17), oleic acid(18), methyl α-linolenate(19), trilinolein(20), and cassipourol(21). Among them, compounds 1 and 2 were identified as new glycosides and named hyperelatosides G and H. Compounds 3-5, 7-9, 17, and 20-21 were isolated from the genus Hypericum for the first time. The remaining compounds were isolated from H. elatoides for the first time. The results of biological assays revealed that compound 11 exhibited significant anti-neuroinflammatory activity, and compounds 1, 3, and 19 displayed certain neuroprotective effects.

Four new δ- and γ-lactone derivatives, hyperelatolides A-D (1-4, respectively), were discovered from the aerial portions of Hypericum elatoides R. Keller. Their structures were elucidated by analysis of NMR spectra, HRESIMS, quantum chemical calculations of NMR and ECD spectra, and X-ray crystallographic data. Hyperelatolides A (1) and B (2) represent the first examples of δ-lactone derivatives characterized by a (Z)-(5,5-dimethyl-2-(2-oxopropyl)cyclohexylidene)methyl moiety and a benzoyloxy group attached to the ß- and γ-positions of the δ-lactone core, respectively, while hyperelatolides C (3) and D (4) are unprecedented γ-lactone derivatives featuring substituents similar to those of 1 and 2. All compounds were tested for their inhibitory effects on NO production in LPS-activated BV-2 cells. Lactones 1 and 2 exhibited considerable antineuroinflammatory activity, with IC50 values of 5.74 ± 0.27 and 7.35 ± 0.26 µM, respectively. Moreover, the mechanistic study revealed that lactone 1 significantly suppressed nuclear factor kappa B signaling and downregulated the expression of inducible nitric oxide synthase and cyclooxygenase-2 in LPS-induced cells, which may contribute to its antineuroinflammatory activity.

The technical problems of high in situ stress, high gas pressure, high gas content, and low coal seam permeability are widespread in deep soft coal seam excavation, which leads to frequent occurrences of dynamic phenomena, such as coal cannons and blowout holes. Based on the high-pressure hydraulic fracturing technology and process, this study puts forward a new technology of gas drainage in deep and soft coal seams by fracturing the overlying key strata to cut off the stress transmission path among coal and rock strata. According to the theories of key layer and masonry beams, the distribution locations of the main and subkey strata are determined, and based on uniaxial compression and Brazilian splitting experiments, the mechanical parameters of key stratum were tested. Combined with the results of numerical simulation and field test, initial pressure and fracturing radius of hydraulic fracturing technology for overlying key strata were determined, the stress relief effect and permeability variation law of coal seam after hydraulic fracturing in the main and subkey strata were analyzed, and then technical schemes for simultaneous fracturing of the key layer were designed. Field application results showed that the stress concentration phenomenon in soft coal seam excavation had been alleviated, and the stress relief effect of coal seam and the permeability were increased obviously. The volume and concentration of gas drainage were increased by 10 and 11%, respectively, the gas amount by 1.22 times; the frequency of dynamic phenomena such as coal cannons decreased by 95%, the gas concentration in return air flow by 20% during mining processes. This paper provided an innovative technical idea and process for gas control in deep and soft coal seam excavation, which could effectively solve the common and difficult problems about frequent occurrences of excessive gas concentration and dynamic phenomena.

Six undescribed C27-phytoecdysteroid derivatives, named superecdysones A-F, and ten known analogs were extracted from the whole plant of Dianthus superbus L. Their structures were identified by extensive spectroscopy, mass spectrometric methods, chemical transformations, chiral HPLC analysis, and the single-crystal X-ray diffraction analysis. Superecdysones A and B possess a tetrahydrofuran ring in the side chain and superecdysones C-E are rare phytoecdysones containing a (R)-lactic acid moiety, whereas superecdysone F is an uncommon B-ring-modified ecdysone. Notably, based on the variable temperature (from 333 K to 253 K) NMR experiments of superecdysone C, the missing carbon signals were visible at 253 K and assigned. The neuroinflammatory bioassay of all compounds were evaluated, and 22-acetyl-2-deoxyecdysone, 2-deoxy-20-hydroxyecdysone, 20-hydroxyecdysone, ecdysterone-22-O-benzoate, 20-hydroxyecdysone-20,22-O-R-ethylidene, and acetonide derivative 20-hydroxyecdysterone-20, 22-acetonide significantly suppressed the LPS-induced nitric oxide generation in microglia cells (BV-2), with IC50 values ranging from 6.9 to 23.0 µM. Structure-activity relationships were also discussed. Molecular docking simulations of the active compounds confirmed the possible mechanism of action against neuroinflammations. Furthermore, none compounds showed cytotoxicity against HepG2 and MCF-7. It is the first report about the occurrence and anti-neuroinflammatory activity of the phytoecdysteroids in the genus Dianthus. Our findings demonstrated that ecdysteroids may be used as potential anti-inflammatory drugs.

As an important ecosystem, alpine meadow in China has been degraded severely over the past few decades. In order to restore degraded alpine meadows efficiently, the underlying causes of alpine meadow degradation should be identified and the efficiency of restoration strategies should be evaluated. For this purpose, a mathematical modeling exercise is carried out in this paper. Our mathematical analysis shows that the increasing of raptor mortality and the decreasing of livestock mortality (or the increasing of the rate at which livestock increases by consuming forage grass) are the major causes of alpine meadow degradation. We find that controlling the amount of livestock according to the grass yield or ecological migration, together with protecting raptor, is an effective strategy to restore degraded alpine meadows; while meliorating vegetation and controlling rodent population with rodenticide are conducive to restoring degraded alpine meadows. Our analysis also suggests that providing supplementary food to livestock and building greenhouse shelters to protect livestock in winter may contribute to alpine meadow degradation.

Rodent pests bring great damage to human beings, while rodenticide and sterilant can be used to control the pests. After ingesting sterilant, rodent pests lose their fertility, but in some cases, the sterile individuals may gain their fertility again, produce offspring, and enlarge population size. In this paper, the dynamic models of rodent pest population under lethal control and shortacting contraception control were formulated, and, with the prerequisite of the seasonal breeding of rodent pest population, the models were used to regularly analyze their behaviors and the effects of the contraception rate, lethal rate, control interval, and sterilant valid period on the dynamics of the pest population. The results showed that larger contraception rate and lethal rate and shorter control interval could have better control effect, making the controlled population become smaller and even died out. Short-acting sterilant limited the control effect. At the later period of breeding season, the rodent pest population controlled with short-acting sterilant would have a weak recovery.
