To evaluate nasal cavity (NC) dimensions of individuals with cleft lip and/or palate (CL/P), obstructive sleep apnea (OSA), and primary snoring, by tomographic image analysis, compared to individuals with OSA without CL/P (N-CL/P).Cross-sectional and retrospective.Tertiary referral center.Patients were divided into 2 groups: (G1) CL/P + OSA or primary snoring, n = 11; (G2) N-CL/P + OSA, n = 13.NC tomographic images were reconstructed using ITK-SNAP software, and measurements were obtained from these three-dimensional models using SpaceClaim software.Total NC volumes, right and left NC volumes, and volumes of the nostril to the nasal valve (V1) and from the nasal valve to the superior limit of the nasopharynx (V2), cross-sectional areas, and perimeters.NC volumes (total, right, and left sides), V1, and V2, though smaller in the CL/P + OSA, did not differ significantly from the N-CL/P + OSA. Cross-sectional areas and perimeters of the superior limit of the nasopharynx, in the CL/P + OSA, presented significantly higher values compared to the N-CL/P + OSA (P ≤ .05).The internal nasal dimensions of patients with CL/P do not seem to be part of the etiopathogenesis, nor constitute a risk factor for OSA with greater severity, in this special group of patients.

OBJECTIVE: Pathological exercise is a dangerous behavior often observed in eating disorders. Data investigating associated characteristics of pathological exercise in men are lacking, despite college men and women being at equally elevated risk for developing eating disorders. PARTICIPANTS: Two hundred and twenty-four college men who exercise regularly completed a series of self-report questionnaires. METHODS: Latent profile analysis was used to identify empirically-derived homogenous subgroups of regular exercisers based on severity of other eating disorder attitudes and behaviors. Profiles were also compared on differences in exercise motivation and general psychopathology (i.e., depression, anxiety). RESULTS: Fit indices indicated a three-profile solution. Profiles described an eating psychopathology group, a low psychopathology group, and a high exercise frequency group without eating disorder features. CONCLUSIONS: Pathological exercise cannot be identified using exercise frequency alone. Other features like body dissatisfaction and exercise motivation style are relevant in identifying pathological exercise behavior in college men.

Proteases have received enormous interest from the research and medical communities because of their significant roles in several human diseases. Some examples include the involvement of thrombin in thrombosis, HIV-1 protease in Acquired Immune Deficiency Syndrome, cruzain in Trypanosoma cruzi infection, and membrane-type 1 matrix metalloproteinase in tumor invasion and metastasis. Many efforts has been undertaken to design effective inhibitors featuring potent inhibitory activity, specificity, and metabolic stability to those proteases involved in such pathologies. Protease inhibitors usually target the active site, but some of them act by other inhibitory mechanisms. The understanding of the structure-function relationships of proteases and inhibitors has an impact on new inhibitor drugs designing. In this paper, the structures of four proteases (thrombin, HIV-protease, cruzain, and a matrix metalloproteinase) are briefly reviewed, and used as examples of the importance of proteases for the development of new treatment strategies, leading to a longer and healthier life.

Herpes Simplex Virus (HSV) infections are among the most common human diseases. In this work, we assess the structural features and electronic properties of a series of ten 1-hydroxyacridone derivatives (1a-j) recently described as a new class of non-nucleoside inhibitors of Herpes Simplex Virus-1 (HSV-1). Based on these molecules, we applied rigid analogue and isosteric replacement approaches to design and synthesize nine new 3H-benzo[b]pyrazolo[3,4-h]-1,6-naphthyridine derivatives (2a-i). The biological and computational results of these new molecules were compared with 1-hydroxyacridones. An inhibitory profile was observed in 10-Cl substituted 3H-benzo[b]pyrazolo[3,4-h]-1,6-naphthyridine derivative (2f), which presents the same substituent at the analogous position of 1-hydroxyacridone derivative (1b). The structure-activity relationship (SAR) studies pointed out the 10-position next to nitrogen atom as important for the anti-HSV-1 profile in the pyrazolo-naphthyridine derivatives tested, which reinforced the promising profile for further experimental investigation. The most potent acridone and pyrazolo-naphthridine derivatives were also submitted to an in silico ADMET screening in order to determine their overall drug-score, which confirmed their potential antiviral profile.

Ecotin is a Escherichia coli-derived protein that has been characterized as a potent inhibitor of serine-proteases. This protein is highly effective against several mammalian enzymes, which includes pancreatic and neutrophil-derived elastases, chymotrypsin, trypsin, factor Xa, and kallikrein. In this work we showed that ecotin binds to human alpha-thrombin via its secondary binding site, and modulates thrombin catalytic activity. Formation of wild type ecotin-alpha-thrombin complex was observed by native PAGE and remarkably, gel filtration chromatography showed an unusual 2:1 ecotin:enzyme stoichiometry. Analysis of the protease inhibitor effects on thrombin biological activities showed that (i) it decreases the inhibition of thrombin by heparin/antithrombin complex (IC50=3.2 microM); (ii) it produces a two-fold increase in the thrombin-induced fibrinogen clotting; and (iii) it inhibits thrombin-induced platelet aggregation (IC50=4.5 microM). Allosteric changes on thrombin structure were then evaluated. Complex formation with ecotin caused a three-fold increase in the rate of thrombin inhibition by BPTI, suggesting a displacement of the enzyme's 60-loop. In addition, ecotin modulated the enzyme's catalytic site, as demonstrated by changes in the fluorescence emission of fluorescein-FPRCK-alpha-thrombin (EC50=3.5 microM). Finally, solid phase competition assays demonstrated that heparin and prothrombin fragment 2 prevents thrombin interaction with ecotin. Altogether, these observations strongly support an ecotin interaction with thrombin anion-binding exosite-2, resulting in modulation of its biological activities. At this point, ecotin might be useful as a new tool for studying thrombin allosteric modulation.

Changes are occurring within Brazilian institutes of higher education; currently several universities are reviewing their course offerings and teaching approaches to determine if they meet the needs of today's undergraduate students. When changes are made to the curriculum of experimental courses, there should be an understood guarantee that all efforts to avoid ethical and biosafety issues have been diligently considered. Ethical considerations lead us to create an alternative experimental session to be conducted that eliminated the use of rats, the conventional in vivo model employed for learning metabolism of glycogen in our university. To avoid possible biosafety issues, we prepared an alternative sample to simulate human urine, which we called guarurine. Using our new method, it is possible to verify positive results imitating a diabetic and starving people samples for detection of glucose and ketone. The alternative tool described herein is not only particularly suited to bypass the ethics of using animals for teaching, but also permits the discussion of significant aspects of pathological and physiological situations such as diabetics and starvation in a simple, safe, and interesting way.

In this work, we performed the design, synthesis, and the structure-activity relationship studies of 13 new derivatives of thieno[2,3-b]pyridine. These derivatives were prepared in high yields (96-70%) and their structures were elucidated by IR, (1)H, (13)C NMR, and MS. The biological results showed some derivatives as antiparasitic agents against Giardia lamblia. Computational analysis of HOMO and LUMO energy, HOMO orbital coefficient distribution, electrostatic potential map, dipole moment, and density HOMO was performed to gain insight into the SAR aspects. This study pointed the p-methoxy substituted derivative as a leading compound for the development of new microbicidal medicines based on thieno[2,3-b]pyridine analogs.

OBJETIVO: Revisar a estrutura e o funcionamento do sistema colinérgico central ressaltando seu papel na fisiologia e na fisiopatologia das doenças de Alzheimer e Parkinson, esquizofrenia, epilepsia e tabagismo. MÉTODO: Foi realizada uma pesquisa bibliográfica no MedLine, LILACS, PubMed e ISI, e na Biblioteca da Fundação Oswaldo Cruz, RJ, selecionando-se o período de 1914 a 2009, utilizando os descritores: "receptors", "cholinergic", "Alzheimer disease", "schizophrenia", "epilepsy" e "smoking", além de referências cruzadas dos artigos selecionados e análise adicional de referências na literatura específica do tema. RESULTADOS: Efeitos importantes da ativação de receptores colinérgicos nicotínicos e muscarínicos sobre o desenvolvimento do sistema nervoso central (SNC) têm sido descritos. A dessensibilização e a internalização dos receptores acoplados à proteína G mediadas pela ativação de proteínas cinases têm sido descritas em proliferação, diferenciação e morte celular, além de síndromes neuropsiquiátricas. CONCLUSÃO: As informações produzidas a partir de estudos do sistema de neurotransmissão colinérgica podem auxiliar no desenvolvimento de medicamentos mais específicos para o tratamento da doença de Alzheimer, esquizofrenia, epilepsia e tabagismo.


OBJECTIVES: To review articles regarding important topics about cholinergic system and its ionotropic and G-protein coupled receptors as well as their regulation, also enlightening its importance in central nervous system (CNS) development and in several neuropsychiatric conditions such as Alzheimer disease, schizophrenia, epilepsy and smoking. METHOD: Bibliographical research was completed through MedLine, LILACS, PubMed, ISI and the Fundação Oswaldo Cruz Library, RJ, specifically for 1914 to 2009, using the descritors: "receptors", "cholinergic", Alzheimer "disease", "schizophrenia", "epilepsy" and "smoking", in addition to the cross-reference of the articles selected and further analyses of bibliographical references on the theme. RESULTS: Currently literature describes important effects of nicotinic and muscarinic receptors activation on development of central nervous system (CNS). The protein G coupled receptors dessensibilization and internalization mediated by kinases have been described in proliferation, differentiation and cell death, and also in neurologic disorders. DISCUSSION: The importance of the cholinergic system and its relationship with pathologies such as Alzheimer disease, schizophrenia, epilepsy is evident. The data produced so far may help on planning medicaments more specific for these pathologies treatment.

RESUMO

A síndrome de Tourette (ST) é uma patologia de comprometimento psicossocial que acarreta alterações significativas na vida dos seus portadores e respectivos familiares. Este artigo aborda diversos aspectos relacionados a esta doença, incluindo etiologia, epidemiologia, aspectos neurobiológicos, quadro clínico, diagnóstico, patologias associadas e tratamento (clássico e alternativo). Neste trabalho, ainda comparamos a ST com outras doenças, envolvendo tiques e mencionamos as associações de apoio aos pacientes portadores de ST, que auxiliam no tratamento e na socialização do paciente afetado.
