RESUMO

The investigation of cultivation and management techniques that take into account behaviour and animal welfare becomes important due to their direct relationship with animal performance. This study aimed to classify juvenile Oreochromis niloticus by personality and compare their performance at exploration and competition tasks. The new object test was used to select bold and shy animals, which were then tested to see which first approached an object. The food competition test involved placing three feed pellets inside a cylinder and measuring the time taken until the pellet was eaten. The new object test found bold animals with a shorter mean time to approach the object than shy animals approaching 65% of the first object when compared to shy. The food competition test found that the bold animals ingested the first pellet more times and with a shorter average time of ingestion than shy animals, no difference in the second intake, whereas the results for the third were same as those for the first. Shy animals had higher intake of the second pellet. Bold animals explored their environment faster in both challenges, while shy individuals exhibited ability to learn from the challenges imposed in the study.

Aiming to test the capacity of retention of carotenoids in tissues, Lophiosilurus alexandri juveniles were fed diets containing 0, 25, 50, 100, 200, and 400 mg/kg of synthetic astaxanthin for 62 days. The inclusion of astaxanthin did not result in significant differences in growth, weight gain, apparent feed conversion, and feed efficiency of the fish. Blood biochemistry and liver histology did not change with the different treatments. At the level of 100 mg/kg of inclusion, there were the highest levels of carotenoids in the blood, and muscle and the smallest difference between the muscle x integument ratio.

Hypoxia is among the most critical environmental stressors for fish in aquatic environments, and several energetic alterations have been associated with it. The aim of the present study was to evaluate the involvement of the phosphoryl transfer network and its effects on adenosine triphosphate (ATP)-dependent enzymes during hypoxia, as well as the role of oxidative stress in the activity of the phosphoryl transfer network in pacamã (Lophiosilurus alexandri) subjected to severe hypoxia. Branchial creatine kinase (CK; cytosolic and mitochondrial fractions), adenylate kinase (AK), and pyruvate kinase (PK) activities were inhibited after 72 h of exposure to hypoxia compared to their respective normoxia groups, and remained low (except for AK) after 24 and 72 h of re-oxygenation. Activities of the branchial sodium-potassium pump (Na+, K+-ATPase) and proton pump (H+-ATPase) were inhibited in fish exposed to 72 h of hypoxia compared to the normoxia group, remained inhibited after 24 h of re-oxygenation, and were restored to physiological levels after 72 h of re-oxygenation. Levels of branchial reactive oxygen species (ROS) were higher in fish exposed to hypoxia for 72 h compared to the normoxia group, and increased during re-oxygenation. Lipid peroxidation (LOOH) levels were higher in fish subjected to 72 h of hypoxia compared to the normoxia group, and remained higher during re-oxygenation. On the other hand, protein sulfhydryl (PSH) levels were lower in fish exposed to hypoxia for 72 h compared to the normoxia group, and remained low during re-oxygenation. Based on this evidence, inhibition of the activities of enzymes belonging to phosphoryl transfer network contributed to impairing energetic homeostasis linked to ATP production and ATP utilization in gills of pacamã subjected to hypoxia, and remained inhibited during re-oxygenation (except AK activity). Moreover, inhibition of the phosphoryl transfer network impaired activity of ATP-dependent enzymes, which can be mediated by ROS overproduction, lipid peroxidation, and oxidation of SH groups.

Sexual differentiation and early gonadal development are critical events in vertebrate reproduction. In this study, the initial testis development and expression of the Vasa, Nanos2 and Sox9 proteins were examined in Nile tilapia Oreochromis niloticus submitted to induced sex reversal. To that end, 150O. niloticus larvae at 5 days post-hatching (dph) were kept in nurseries with no hormonal addition (control group) and 150 larvae were kept with feed containing 17α-methyltestosterone to induce male sex reversal (treated group). Morphological sexual differentiation of Nile tilapia occurred between 21 and 25 dph and sex reversal resulted in 94% males, whereas the control group presented 53% males. During sexual differentiation, gonocytes (Gon) were the predominant germ cells, which decreased and disappeared after that stage in both groups. Undifferentiated spermatogonia (Aund) were identified at 21 dph in the control group and at 23 dph in the treated group. Differentiated spermatogonia (Adiff) were found at 23 dph in both groups. Vasa and Nanos2 occurred in Gon, Aund and Adiff and there were no significant differences between groups. Vasa-labelled Adiff increased at 50 dph in both groups and Nanos2 presented a high proportion of labelled germ cells during sampling. Sertoli cells expressed Sox9 throughout the experiment and its expression was significantly greater during sexual differentiation in the control group. The results indicate that hormonal treatment did not alter initial testis development and expression of Vasa and Nanos2 in Nile tilapia, although lower expression of Sox9 and a delay in sexual differentiation was detected in the treated group.

The aim of this work was to investigate the effects of daily prey concentration during the first 15 days of active feeding of Hoplias lacerdae larvae, and the juvenile size on the feed training. In the first phase, the larvae received five Artemia nauplii concentrations (P). In the second phase, the juveniles from each treatment were trained to accept formulated diet. Superior growth was related to higher initial daily prey concentrations (900 and 1100 nauplii larvae-1). During feed training, the growth tendency was similar to that verified in the first phase. The lowest values of specific growth rate (SGR) were registered after the introduction of the semi-moist diet used in the feed training. However, the values of SGR recovered along the experiment and similar rates were found among the treatments. Survival, mortality and cannibalism were similar in the different treatments at the end of both phases. It can be concluded that: the prey concentration affects growth of H. lacerdae during the first 15 days of active feeding, and feed training can be initialized with juveniles of about 16 mm of total length.

In aquaculture, activities with anesthetic compounds are usually used in order to ensure the welfare of farmed fish, allowing handling out of water with decreased trauma by stress. Presently, there is no information about anesthetic action of eugenol in early life stages of Nile tilapia (Oreochromis niloticus). The objective of this study was to evaluate different concentrations of eugenol for larvae and juveniles of Nile tilapia. Sixty animals were used for each group of weight, group I = 0.02 g; group II = 0.08 g; group III = 0.22 g; group IV = 2.62 g; and group V = 11.64 g. The eugenol concentrations tested were 50, 75, 100, 125, 150 and 175 mg L-1. No mortality was reported during the tests with eugenol. Tilapia larvae with 0.02 g and juveniles around 11.64 g can be anesthetized with eugenol concentrations between 150 and 175 mg L-1, since they determine the shortest sedation time (23 and 72 seconds, for the group of lowest and highest weights, respectively).

The aim of this study was to evaluate commercial gelatin in the total replacement of bovine heart in feed training of "pacamã" Lophiosilurus alexandri in different water salinities. A completely randomized experimental design, in a 2 × 3 factorial arrangement, was performed using two types of moist ingredients (bovine heart and gelatin) and three water salinities (0.0; 2.0 and 4.0 g of salt L(-1)) with three replications. Juveniles (2.39 ± 0.08 cm standard length and 0.20 ± 0.03 g of weight) were conditioned to accept commercial diets by the technique of the gradual transition of ingredients. At the end of 36 days no differences were observed to weight gain, length gain and specific growth rate. The feed training efficiency was better (P < 0.05) with the gelatin use, 100.0%. There was a negative effect of salinity on the survival rate and management efficiency in the concentration of 4 g of salt L(-1), with values of 58.6 ± 12.0 % and 58.0 ± 12.0 %, respectively. Lophiosilurus alexandri juveniles could be feed-trained to accept commercial diets with gelatin in the total replacement of bovine heart in freshwater or salinity of 2 g of salt L(-1).

Temperature is a critical factor for fish management, especially during breeding season. This study evaluated gametogenesis and gonadal maturation in the catfish Lophiosilurus alexandri under farming conditions in two temperatures. The fish were divided in the following experimental groups: (S0) control group, before starting the experiment, (S1) 30 days at 23 °C, (S2) 70 days at 23 °C, (S3) 30 days at 27 °C, and (S4) 70 days at 27 °C. Gonad, liver, and blood samples were obtained to analyze reproductive and health parameters. In females, increased gonadosomatic index (GSI) was associated with a decreased proportion of perinucleolar follicles and increased diameter of vitellogenic follicles in S4. Postovulatory follicles were found only in fish kept at 27 °C, whereas atretic follicles were more frequent at 23 °C. In S4, females had higher hepatic levels of vitellogenin and lower levels of zona radiata proteins. In males, a lower proportion of spermatogonia was found at 27 °C (S4), whereas GSI and proportions of spermatocytes, spermatids, and spermatozoa were similar among the groups. Levels of cortisol were higher in females of group S3. Plasma 17ß-estradiol, 11-ketotestosterone and 17α-hydroxyprogesterone were significantly higher in S4. Hematocrit, erythrocytes, and glucose were higher at 27 °C. Together, the present study indicate that exposure of L. alexandri to 27 °C provides suitable conditions for germ cell development and gonadal maturation in captivity, promotes increase of sex steroids, follicular growth, and oocyte maturation, with onset of ovulation occurring at 30 days. At 23 °C, spermatogonia proliferation is favored in males, but it cannot be recommended for stimulating female maturation of L. alexandri in fish farming.

Considering that each personality type in animals presents distinct physiological and behavioural responses, this study evaluated the efficiency of the Novel Environment test to classify larvae of Lophiosilurus alexandri into bold and shy individuals, which were then investigated for growth, cannibalism and mortality in larviculture of pure and mixed groups. Larvae with an average weight of 24.0±1.7mg and length of 14.1±0.4mm, were subjected to a Novel Environment test to classify their personality type (bold and shy larvae). After the larvae were classified according to personality type, they were subjected to larviculture for 15days. Three treatments were tested: only bold larvae, only shy larvae, and a mixed treatment (bold larvae+shy larvae) at a density for 16 larvae/L, which were fed 3 times a day with Artemia nauplii. After larviculture, there were no differences in the final lengths of larvae of the bold, shy, and mixed treatments (26.9±0.76mm, 26.7±1.00mm, and 26.8±1.24mm, respectively); however, shy larvae possessed weighed less (0.22±0.01g) than the bold and mixed treatments, which did not differ significantly (0.25±0.02g and 0.27±0.02g, respectively). The bold and mixed treatments had the highest cannibalism rate (11.2±5.1% and 23.1±12.3%, respectively). Overall survival was lowest in the mixed treatment (62.5±13.0%), while that of the bold and shy treatments were similar (82.5±9.2% and 86.2±9.2%, respectively). The separation of L. alexandri larvae by traits can ensure a decrease in cannibalism and hence, more productive larviculture.

Understanding the influence of different temperature conditions on the spermatogenesis is important for improvement of the fish aquaculture. This study evaluated the influence of low temperature on structural and quantitative dynamics of the spermatogenesis in Oreochromis niloticus. Adult males were cultivated with room temperature water (20.28-22.46°C) and testes were collected for histological, ultrastructural and morphometric analyses. This species has unrestricted lobular testis with cystic spermatogenesis and type I spermiogenesis that results in a single anacrosomal aquasperm. Seminiferous lobules and spermatogenic cells had a radial arrangement toward the spermatic duct. Superior and central portions of testes had a greater lobular area than the inferior portion in all samplings. Spermatogonia (9.3%) were distributed in the inferior portion of testes, spermatocytes (25.3%) and spermatids (34.4%) in the central portion, while spermatozoids (39.4%) and secretory cells (4.6%) in the superior portion. Throughout the study, correlation between water temperature and lobular area characteristics was significant only in the inferior portion of testes (r(2)=0.95), although the lobular area in the other testicular portions increased when the water temperature increased by 2°C. Correlation between the water temperature and spermatogenic cells was significant for undifferentiated spermatogonia (r(2)=0.54) and number of spermatids (r(2)=0.67). It is concluded that low cultivation temperatures may positively influence the generation of primary spermatogonia in the inferior periphery of O. niloticus testes. In addition, males maintain reservoirs of germ cells at low temperatures and the radial zonation of spermatogenesis has an important role in the renewal and production of germ cells.

The balance between cell proliferation and apoptosis is crucial for tissue development and homeostasis. The present study investigated the contribution of proliferating cell nuclear antigen (PCNA) and apoptosis during ovarian remodeling after spawning in the Nile tilapia Oreochromis niloticus. Breeding females were kept in controlled conditions and ovary samples were collected weekly for TUNEL assay, immunohistochemistry for PCNA and caspase-3 and morphometric analysis. During the follicular growth, PCNA labeled mainly the nuclei of oocytes and follicular cells in a high proportion of follicles especially in primary growth, while a low occurrence of apoptosis in follicular and theca cells was detected. At 0-3 days post-spawning, post-ovulatory follicles showed no proliferative activity, however the follicular cells exhibited high rates of apoptosis. At 7-10 days, PCNA labeled the thecal cells in a low proportion of post-ovulatory follicles, which showed follicular cells with lower rates of apoptosis. PCNA labeled mainly the theca in the advanced and late stages of atretic follicles, while the follicular cells exhibited a significant increase of apoptosis along follicular atresia. We concluded that PCNA and apoptosis work cooperatively to ensuring the success of follicle development and maintaining of tissue homeostasis during follicular growth. PCNA and apoptosis are also essential mechanisms in the follicular regression during post-spawning ovarian recovery in the Nile tilapia.

Due to the ecological importance of Lophiosilurus alexandri, the present work evaluated its genetic representativeness by comparing wild stocks to broodstocks that were kept at three restocking hatcheries along the São Francisco River. A total of 97 samples were genotyped for newly developed microsatellite markers. Low levels of genetic diversity (average alleles number of 4.2 alleles) were detected in all cases, being more severe in captive groups. Significant pairwise FST and DEST values, Structure, and DAPC analyses showed that wild animals were structured in two groups, and a third group was formed by captive animals, evidencing the need to adopt genetic criteria to retain genetic diversity in the hatcheries. For this reason, three full-sib families were constructed to select the best relatedness estimator for L. alexandri and establish a cut-off value aimed to avoid full-sibling matings in the hatcheries. Two estimators, Wang (RW) and Lynch & Li (RLL), were accurate in reflecting the relatedness level for full-sibs in this species. According to them, less than 50% of the potential breeding matings in the three hatcheries are advisable. The innate low diversity of L. alexandri highlights the importance of minimizing inbreeding and retaining genetic diversity towards the species recovery.(AU)


Devido à importância ecológica de Lophiosilurus alexandri, o presente trabalho avaliou sua representatividade genética, comparando estoques selvagens com plantéis de reprodutores de três larviculturas ao longo do Rio São Francisco. Noventa e sete amostras foram genotipadas com marcadores microssatélites recém-desenvolvidos. Baixos níveis de diversidade genética (número médio de alelos de 4,2) foram detectados em todos os casos, sendo mais severo no cativeiro. Os valores de FST e DEST par a par, as análises do Structure e DAPC mostraram a estruturação dos animais selvagens em dois grupos, e um terceiro formado pelas larviculturas, evidenciando a necessidade de adoção de critérios genéticos para retenção da diversidade genética no cativeiro. Por essa razão, três famílias de irmãos completos foram construídas para selecionar o melhor estimador de parentesco para a espécie e estabelecer os valores mínimos de corte para evitar cruzamentos indesejados. Dois estimadores, Wang (RW) e Lynch & Li (RLL), foram eficientes em refletir as relações de parentesco para irmãos completos nessa espécie. Segundo eles, menos de 50% dos potenciais cruzamentos são recomendáveis nas três larviculturas. A baixa diversidade genética inerente ao L. alexandri destaca a importância de minimizar a consanguinidade e evitar perda de diversidade genética, visando a recuperação da espécie.(AU)

Lophiosilurus alexandri is a carnivorous freshwater fish endemic of São Francisco basin and an endangered species. In this study, we analysed the chemical composition (moisture, protein, ash and lipid), fatty acid profile, and nutritional quality (atherogenic index, thrombogenecity index, ratio between hypocholesterolemic and hypercholesterolemic fatty acids and ω6/ω3 = ratio) of L. alexandri in lotic (river), lentic (hydroelectric dams) environments, under natural fed, and in laboratory controlled conditions fed with commercial diets. Cultured and lentic fish had significantly higher lipid levels (1.5 and 1.9- fold, respectively) than lotic fish. Lentic L. alexandri had significantly higher eicosapentaenoic acid (EPA) levels (4×) than cultured or lotic L. alexandri. Docosahexaenoic acid (DHA) levels were the highest in lentic fish, followed by lotic fish. Lentic fish had greater proportion of fatty acid ω6/ω3 than lotic or cultured fish. The results of this study showed that L. alexandri is a lean fish (1-2% of total lipids) and that the environment has a great influence on the fatty acid profile. These results may be a reference for further studies, primarily as a source of information for conservation L. alexandri through restocking and the development of commercial projects of aquaculture.(AU)


Lophiosilurus alexandri é um peixe de água doce, carnívoro, endêmico da bacia do rio São Francisco e encontra-se vulnerável a extinção. Neste estudo, analisamos a composição química (umidade, cinzas, lipídeos e proteína), o perfil de ácidos graxos e qualidade nutricional (índices de aterogênicidade e trombogenicidade, razão entre ácidos graxos hipocolesterolêmicos e hipercolesterolêmicos e ω6/ω3) de L. alexandri em ambientes lóticos (rio), lênticos (hidrelétricas), sob alimentação natural, e em laboratório, com condições controladas e alimentados com dietas comerciais. Os peixes cultivados e capturados em ambientes lênticos apresentaram níveis lipídicos significativamente mais altos (1,5 e 1,9 vezes, respectivamente) do que os peixes cultivados em ambientes lóticos. Os animais cultivados em ambiente lênticos, apresentaram níveis significativamente mais altos de ácido eicosapentaenoico (EPA) (4×) que os animais cultivados em laboratório e ambiente lóticos. Os níveis de ácido docosaexaenoico (DHA) foram os mais altos em peixes lênticos, seguidos pelos peixes lóticos. Os peixes lênticos apresentaram melhor proporção de ácidos graxos ω6/ω3 que peixes lóticos ou de cativeiro. Os resultados deste estudo mostraram que L. alexandri é um peixe magro (1-2% do total de lipídios) e que o ambiente tem uma grande influência no perfil de ácidos graxos. Esses resultados podem ser uma referência para novos estudos, principalmente como fonte de informações para sua conservação por meio do repovoamento e desenvolvimento de projetos para sua criação comercial.(AU)

This study tested an external tag for juveniles of Lophiosilurus alexandri and measured the effects of tagging in different size classes of fishes. Experiment 1 evaluated the retention rate and influence of the tag on survival and growth of three hundred fishes divided Small, Medium and Large size classes. After 90 days of experiment the fishes had 100% survival and the tag had a higher retention rate for animals of the Large size class. Experiment 2 evaluated the tag's influence on hematology parameters of forty-two fishes at 5, 10 and 30 days post-tagging. In this experiment both tagged and untagged animals experienced 9.4% mortality. The hematocrit was higher on the 30th day for tagged animals than for untagged animals. No difference was observed for leukocytes, plasma protein, erythrocytes and mean corpuscular volume. It was concluded from Experiment 1 that the tag reduced growth performance when applied to small sized juvenile L. alexandri, and that the tag retention rate increased with increasing animal size. Among the hematological parameters studied for juveniles on the Experiment 2 the tag only influenced the hematocrit parameters, which shows that the tag can be used without considerable influence on the hematological parameters of juvenile L. alexandri.(AU)


Esse estudo testou a utilização de marcadores externos em juvenis de Lophiosilurus alexandri, assim como o tamanho mínimo ideal do peixe para marcação. No experimento 1 foi avaliado a taxa de retenção e a influência do marcador sobre a sobrevivência e crescimento dos peixes divididos por tamanho nas classes: Pequeno, Médio e Grande. Após 90 dias, os peixes apresentaram 100% de sobrevivência e uma taxa de retenção mais elevada para os animais classificados como grandes. O segundo experimento avaliou a influência do marcador sobre os parâmetros hematológicos dos animais após 5, 10 e 30 dias da marcação. Neste experimento, foram observados valores mais elevados para o hematócrito no 30º dia para os animais marcados, enquanto nenhuma diferença foi observada para leucócitos, proteína plasmática, eritrócitos e volume corpuscular médio. Concluiu-se o que o marcador reduziu a taxa de crescimento dos animais menores, e que a taxa de retenção aumentou com o aumento do tamanho do animal. Lophiosilurus alexandri is a carnivorous freshwater fish endemic of São Francisco basin and an endangered species. In this study, we analysed the chemical composition (moisture, protein, ash and lipid), fatty acid profile, and nutritional quality (atherogenic index, thrombogenecity index, ratio between hypocholesterolemic and hypercholesterolemic fatty acids and ω6/ω3 = ratio) of L. alexandri in lotic (river), lentic (hydroelectric dams) environments, under natural fed, and in laboratory controlled conditions fed with commercial diets. Cultured and lentic fish had significantly higher lipid levels (1.5 and 1.9- fold, respectively) than lotic fish. Lentic L. alexandri had significantly higher eicosapentaenoic acid (EPA) levels (4×) than cultured or lotic L. alexandri. Docosahexaenoic acid (DHA) levels were the highest in lentic fish, followed by lotic fish. Lentic fish had greater proportion of fatty acid ω6/ω3 than lotic or cultured fish. The results of this study showed that L. alexandri is a lean fish (1-2% of total lipids) and that the environment has a great influence on the fatty acid profile. These results may be a reference for further studies, primarily as a source of information for conservation L. alexandri through restocking and the development of commercial projects of aquaculture.(AU)
