RESUMO

Legionella is a bacterium that is ubiquitous in natural and artificial aquatic environments. Some species of Legionella are recognized as opportunistic potential human pathogens. We investigated the distribution of Legionella at seventeen WWTPs throughout Taiwan. Legionella were detected in 10 of the 17 WWTPs (58.8%) and 25 of 41 samples (61.0%). In the integrated, hospital, industrial and domestic wastewater systems were 13/18 (72.2%), 7/12 (58.3%), 2/7 (28.6%) and 3/4 (75.0%) of the samples were positive for Legionella, respectively. The most frequently encountered species were L. donaldsonii and uncultured L. spp., which were both found in 10 samples (24.4% of all samples), then followed by L. lytica (4.9%) and L. pneumophila (4.9%). L. anisa was detected in one sample (2.4%). The results of this survey confirm that Legionella are ubiquitous in WWTPs in Taiwan. Therefore, long-term investigations should be conducted to evaluate the overall occurrence of Legionella in WWTPs in Taiwan.

Icariin is a prenylated flavonol glycoside contained in the herb Epimedium, which has long been used to improve bone fracture healing or prevent osteoporosis because of the belief that the herb has bone-strengthening action. We have previously demonstrated that icariin enhances the osteogenic differentiation of rat bone marrow stromal cells, and partially explained the bone-strengthening mechanism of the herb. In the present study, the effect of icariin on osteoclastogenesis and bone resorption activity was investigated in mouse bone marrow culture. It was found that icariin dose-dependently inhibited the growth and differentiation of hemopoietic cells from which osteoclasts were formed. Far less TRAP+ multinuclear cells appeared in the 10 microM icariin group than in the control. The bone resorption pits formed in the 10 microM icariin group was also significantly less than that of the control. RT-PCR analysis showed that the gene expression of TRAP, RANK and CTR was obviously lower than that of the control. It can be concluded that icariin has the ability to inhibit the formation and bone resorption activity of osteoclasts, which suggests that icariin should be the effective component for the bone-strengthening action of herb Epimedium.

RESUMO

We present and derive a formula that is useful for the design and evaluation of gene cloning experiments in which a complete gene library of the entire genome of an organism is desired. The formula n = ln(1-phi f)/ln(1-f) (in which n is the number of recombinant clones required to ensure a probability, phi, of obtaining at least one of each of all possible gene sequences, and f is the fraction of the genome contained in an average-sized DNA fragment) applies to construction of libraries, in which at least one copy of all the genetic information of a genome is required. The use of this formula for quantitative evaluation of genomic libraries should give greater assurance that a given library would be complete.

RESUMO

A case of malignant melanoma arising from an ovarian cystic teratoma is reported. The tumor was resected, but the patient died of metastases in the abdominal wall, peritoneal cavity, liver, and lungs. The possible histogenesis of the tumor is briefly discussed.

Previous studies have reported that glass-ionomer and adhesive resin cements can bond to various alloys, while zinc phosphate cements lack this adhesive property. This study evaluated the bonding properties of three luting cements during the first seven days after cementation. Thirty cylinders were cast with a high-noble porcelain-fused-to-metal (PFM) alloy and luted in pairs with one of the cements. The joints were stored in water at 37 degrees C for one, two, or seven days before being fractured in shear. The cylinders were re-used to provide 40 joints within each test group. The data were subjected to a Weibull analysis, a curve-fitting method shown to be appropriate for comparing the bond strengths of dental materials. The results showed that the zinc phosphate cement was the weakest material, whereas the adhesive resin produced the strongest joints. Microscopic observations of the fractured samples did not reveal any specific differences between the samples in terms of their mechanism of fracture. The glass-ionomer cement reached its maximum bond strength after two days, whereas storage time had no influence on the zinc phosphate cement. The adhesive resin cement was slightly, but not significantly, weaker after one week in water. We suggest that excessive loading of restorations cemented with glass ionomer should be avoided for the first two days after the placement. The use of an adhesive resin cement can be recommended on endodontically treated teeth, but further studies are needed to evaluate its biocompatibility and adhesion to dentin.

RESUMO

Human choriogonadotropin (HCG) of high immunoactivity was isolated from hydatidiform mole by acid and salt precipitation, immunoaffinity and DEAE-Sephadex chromatography. Polyacrylamide gel electrophoresis, immunodiffusion, immunoelectrophoresis and N-terminal amino acid analyses showed that the product obtained is essentially homogeneous. Molar HCG was found to resemble HCG from urine of normal pregnant women in amino acid composition bu to differ from it in having a lower content of mannose and N-acetylglucosamine.

The specific activity and isoenzyme pattern of beta-glucuronidase (beta-D-glucuronide glucurononosohydrolase, EC 3.2.1.31) from first-trimester placenta, term placenta and hydatidiform mole were compared. Enzyme activity is highest in term placenta and lowest in hydatidiform mole, reflecting the steroid conjugation requirements of the tissues. The difference in enzyme activity is due to variation in the level of one isoenzyme form (isoenzyme B) of beta-glucuronidase, which is absent in molar tissue. The possible cause for the changing isoenzyme pattern is discussed.

In response to the challenge posed by falls in the elderly, the Jewish Institute for Geriatric Care established a Falls Clinic. The coordinated expertise of a geriatrician, neurologist, cardiologist, and physiatrist were combined with resources in audiology, ophthalmology, and podiatry. Thirty-six patients enrolled in the study had sustained a total of 36 falls, which resulted in 13 fractures and seven soft-tissue injuries. Falls were a daily occurrence for three patients, weekly for five patients, monthly for ten, semiannually for 14, and yearly for four patients. After a one-year follow-up, 77% of the patients experienced no further falls. We suggest that falls are a multidisciplinary issue and recommend a team approach for successful management.
