BACKGROUND: Cocaine- and amphetamine-regulated transcript (CART) peptide is the substance distributed in various parts of the nervous system. The majority of previous studies described this substance in the brain, where it takes part in regulatory processes connected with the food intake. CART is also present in the peripheral nervous system, especially in the enteric neurons and nerves located in the wall of the stomach and intestine, but many aspects of distribution and functions of this peptide in the digestive organs remain unknown. The aim of the present study was to investigate the changes of CART-positive nerves in the porcine gallbladder after administration of low-dose Salmonella Enteritidis lipopolysaccharide (LPS) using the single immunofluorescence technique. MATERIALS AND METHODS: Seven days after the injection of 5 µg/kg b.w. LPS S. Enteritidis the gallbladders were collected. CART-positive nerves were studied with standard single immunofluorescence method and counted per observation field (0.1 m2). RESULTS: In control animals the average number of CART-positive nerves per observation field (0.1 mm2) amounted to 5.38 ± 0.32, 11.11 ± 1.56 and 2.97 ± ± 0.24 in gallbladder neck, body and fundus, respectively. LPS administration caused the increase in the number of CART-positive fibres in all parts of gallbladder, and these values amounted to 12.74 ± 0.51, 19.75 ± 0.19 and 5.1 ± 0.05 in the gallbladder neck, body and fundus, respectively. CONCLUSIONS: The obtained results suggest that CART is involved in the neuronal regulatory processes in the porcine gallbladder under physiological conditions, but also during pathological processes, but exact functions of this peptide in this organ remain unexplained and require the further investigation.

Mitochondria play central roles in diverse physiological and pathological conditions associated with cell survival and death. Delocalized lipophilic cations, such as dequalinium (DQA), are accumulated in cancer cells attracted by the highly negative mitochondrial transmembrane potential of these cells. DQA showed a potent anticancer activity in cells from different malignancies. Here, we report the effect of DQA on PC-3 prostate cancer cells. Incubation with DQA at concentrations between 1.5 and 100 microM from 24 to 48 h decreases cell viability. The decrease in cell viability together with a loss of mitochondrial transmembrane potential induced an increase in reactive oxygen species production and cell death via caspase-3 dependent apoptotic pathway. QA was shown to cause moderate to strong cell death in a time and concentration dependent manner, causing a most advantageous effect at a concentration of 10 microM applied for a long 48 h time period, which might be a consequence of the kinetics of intracellular DQA accumulation in mitochondria, but also of the mechanisms of DQA-induced cell death. This data shows DQA as a promising agent against the human prostate cancer PC-3 cell line, activating the caspase-3 dependent apoptotic pathway. This fact might be beneficial for possible future applications in cancer therapy.

European beaver (Castor fiber), the largest rodent species inhabiting a wide area of Eurasia, feeds mainly on dry parts of plants, bark or wood. Such kind of nourishment needs to be properly digested in each part of the gastrointestinal tract. The time of stomach digestion, which directly influences all the following steps of the digestion process, is precisely controlled by the pylorus and its innervation. However, virtually no data is available on the organization of the enteric nervous system in most of the wild animal species, including beavers. On the other hand, a pecu- liar diet consumed by beavers, suggests that the arrangement of their stomach intramural nerve elements can be atypical. Therefore, the present study investigated the distribution and chemical coding of neurons and nerve fibers in the pylorus of the European beaver. The experiment was performed on stomachs obtained from a group of 6 beavers caught in Northeastern region of Poland (due to beaver overpopulation). Pyloric wall tissue cryosections were double immunostained with a mixture of antibodies against pan-neuronal marker PGP 9.5 (to visualize enteric neurons) and ChAT (cholinergic marker), nNOS (nitrergic marker), SP, CGRP, Gal (peptidergic markers). Confocal microscopy analysis revealed that the majority of enteric nerve cells were clustered forming submucosal and myenteric ganglia and all the studied substances were expressed (in various amounts) in these neurons. We conclude, that the anatomical arrangement and chemical coding of intramural nerve elements in the beaver pylorus resemble those found in other mammalian species.

L-asparaginase is widely used in the treatment of acute lymphoblastic leukemia in children and adults. Use of L-aspa E. Coli as well as Erwinase is not possible in all cases because of the side effects, mainly allergic reactions and disfunction of pancreas. Recently, the new form of the enzyme PEG-L-asparaginase was introduced. Binding L-asparaginase E. coli to polyethylene glycol a decreased its toxicity, extended its plasma half-live, not significantly affecting the efficacy. The aim of the study was to examine the results of PEG-L-asparaginase administration in five children with acute lymphoblastic leukemia, and the symptoms of intolerance to L-aspa E. Coli or Erwinase. There were three children with newly diagnosed ALL and two children with first relapse of ALL, treated according to New York Protocol and BFM-90 Protocol for ALL relapses respectively. PEG-L-asparaginase (Oncaspar) was administered in the dose of 2500 IU/m2. According to the protocol four children received 11 courses of treatment with the full dose of the drug. The number of doses for individual patient varied from one to six. The short-lived nettlerash was observed in one patient during two subsequent infusions of the drug. Hydrocortisone and antihistamine drugs were administered. Treatment with PEG-asparaginase was discontinued in one child, who developed dyspnea, nausea, vomiting and face rash during the third dose of the drug. Oncaspar is the valuable drug, which enabled continuation of treatment according to protocol in four out of five children with bad tolerance to routinely used L-asparaginase preparations.

In the time period from February to October 1988 the indicators of hepatitis virus B infection, transaminase activity, and bilirubin level were studied in 89 children with acute leukaemias and malignant lymphomas. The age of the children was from 7 months to 16 years. Fifteen children were examined before starting the treatment with cytostatics, 48 during intensive chemotherapy, and 26 during maintenance treatment. HBV markers were found in 53 children during intensive therapy and maintenance treatment. Out of 36 children with negative HBV infection markers 31 were examined before starting the treatment with cytostatics or during the first 6 months of treatment. Nine out of 53 children in whom HBV infection markers were found had aminotransferase activity raised over 100 u/l and/or bilirubin level over 5 mg/dl and in nine cases transaminase activity and bilirubin level were raised only slightly (below 100 u/l and below 5 wg/dl). In 53 children the results were normal. The high incidence of HBV infection in children with acute leukaemias and lymphomas indicates the necessity of routine prophylactic treatment in this group of patients.

In 11 patients with compensated and 10 with decompensated liver cirrhosis the following determinations were carried out: total thyroxine serum concentration (T4), triiodothyronine-binding coefficient (T3BC), free thyroxine by the dialysis method (DFT4), free thyroxine index (FT4I) and absolute value of free thyroxine (ADFT4). A statistically significant rise of T3BC, DFT4, ADFT4 and FT4I was observed only in the group of decompensated cirrhosis. It is supposed that these changes develop not as a result of excessive production of thyroid hormones but due to disturbances in the binding capacity of the proteins which bind thyroid hormones and also due to an impaired degradation of these hormones in the liver.
