RESUMO

This study reveals that it is possible to secrete truncated versions of outer membrane cytochromes into the culture supernatant and that these proteins can provide a basis for the export of heterologously produced proteins. Different soluble and truncated versions of the outer membrane cytochrome MtrF were analyzed for their suitability to be secreted. A protein version with a very short truncation of the N-terminus to remove the recognition sequence for the addition of a lipid anchor is secreted efficiently to the culture supernatant, and moreover this protein could be further truncated by a deletion of 160 amino acid and still is detectable in the supernatant. By coupling a cellulase to this soluble outer membrane cytochrome, the export efficiency was measured by means of relative cellulase activity. We conclude that outer membrane cytochromes of S. oneidensis can be applied as transporters for the export of target proteins into the medium using the type II secretion pathway.

OBJECTIVE: To investigate the influences of abutment water sorption and various aging parameters on the fracture resistance of zirconia copings. METHODS: Using a master die, identical replicas were made from three resin materials. The first was a melamine resin with very high water sorption (n = 48), the second an experimental resin core build-up composite with moderate water sorption (n = 40) and the third a commercially available core build-up composite with low water sorption (n = 40). On the abutment replicas, zirconia copings (n = 128) were made using a computer-aided design-computer-aided manufacturing system. The copings were luted onto the abutments using zinc oxide phosphate cement. In the melamine group, a subgroup of samples (n = 8) was cemented with a composite cement as controls. The forty specimens in every abutment material group were randomly divided into one of five subgroups, as follows: (i) not aged; (ii) mechanically (dry) loaded only (50 N; 1.2 × 10(6) cycles); (iii) stored for 10 days in water; (iv) thermally cycled (TC; 6000 × 5/55°C); and (v) TC and mechanically loaded (TCML; 50 N, 1.2 × 10(6); 6000 × 5/55°C). After aging, all copings were loaded to fracture. RESULTS: A statistically significant difference was found between the three abutment-die groups if the samples were aged by TCML. The zirconia copings cemented on abutments with high water sorption fractured during TCML, and the subgroup with moderate water sorption had significantly lower fracture resistance. A change of luting material had no impact on this behavior. CONCLUSION: Only the simultaneous combination of all chosen aging factors (TCML) was able to detect a difference in fracture behavior of a zirconia coping luted on abutments with varying water sorption.

An obscure sesquiterpene, rotundone, has been identified as a hitherto unrecognized important aroma impact compound with a strong spicy, peppercorn aroma. Excellent correlations were observed between the concentration of rotundone and the mean 'black pepper' aroma intensity rated by sensory panels for both grape and wine samples, indicating that rotundone is a major contributor to peppery characters in Shiraz grapes and wine (and to a lesser extent in wine of other varieties). Approximately 80% of a sensory panel were very sensitive to the aroma of rotundone (aroma detection threshold levels of 16 ng/L in red wine and 8 ng/L in water). Above these concentrations, these panelists described the spiked samples as more 'peppery' and 'spicy'. However, approximately 20% of panelists could not detect this compound at the highest concentration tested (4000 ng/L), even in water. Thus, the sensory experiences of two consumers enjoying the same glass of Shiraz wine might be very different. Rotundone was found in much higher amounts in other common herbs and spices, especially black and white peppercorns, where it was present at approximately 10000 times the level found in very 'peppery' wine. Rotundone is the first compound found in black or white peppercorns that has a distinctive peppery aroma. Rotundone has an odor activity value in pepper on the order of 50000-250000 and is, on this criterion, by far the most powerful aroma compound yet found in that most important spice.

RESUMO

Jasminum flexile flower absolute from the south of India and the corresponding vacuum headspace (VHS) sample of the absolute were analyzed using GC and GC-MS. Three other commercially available Indian jasmine absolutes from the species: J. sambac, J. officinale subsp. grandiflorum, and J. auriculatum and the respective VHS samples were used for comparison purposes. One hundred and twenty-one compounds were characterized in J. flexile flower absolute, with methyl linolate, benzyl salicylate, benzyl benzoate, (2E,6E)-farnesol, and benzyl acetate as the main constituents. A detailed olfactory evaluation was also performed.
