RESUMO

West Indian cherry cultivation has proved to be an important economic activity in northeastern Brazil. However, irrigation with brackish waters limits cultivation, requiring new strategies to minimize the effect of salt stress. In this context, the present study aimed to evaluate the effect of nitrogen (N), phosphorus (P), and potassium (K) combinations on the photosynthetic pigments and quantum yield of West Indian cherry cultivated under salt stress, in the second year of production. The assay was conducted in a protected environment by adopting an experimental design in randomized blocks, with treatments distributed in a 2×10 factorial arrangement referring to two electrical conductivity levels of irrigation water - ECw (0.6 and 4.0 dS m-1) and 10 NPK fertilization combinations - FC (80-100-100; 100-100-100; 120-100-100; 140-100-100; 100-80-100; 100-120-100; 100-140-100; 100-100-80; 100-100-120, and 100-100-140% of the recommendation, in the second year of production), with three replications, each consisting of one plant. Irrigation with the electrical conductivity of 4.0 dS m-1 negatively affected the synthesis of photosynthetic pigments and the photochemical efficiency of the West Indian cherry cv. Flor Branca. The NPK combinations did not attenuate the effects of salt stress on the analyzed variables. However, the combinations referring to 120-100-100%, 140-100-100%, and 100-120-100% of NPK recommendation improved the quantum yield of photosystem II by reducing the initial fluorescence and increasing the maximum fluorescence of the West Indian cherry cv. Flor Branca.

RESUMO

Improving plant germination is essential to guarantee better quality seedlings. Thus, this research aimed to evaluate whether the seed priming with light quality (LIQ) and the aqueous extract of Cyperus rotundus (AEC) tuber could modulate the germination and initial growth of Moringa oleifera L. seedlings. The experimental design was a completely randomized in the 4x4 factorial scheme, composed of four LIQ conditions (white, blue, red, and distant red light) and four AEC concentrations (0, 25, 50 and 100%). Seed priming with red light reduced the average emergence time, while blue, red, and extreme red lights associated with 50% of aqueous extract of C. rotundus increased shoot initial length and photosynthetic pigment accumulation. Seed priming with blue light resulted in seedlings with a shorter final shoot length. However, application of 100% of aqueous extract of C. rotundus reversed this. The white light in combination with concentrations of 50 and 100% of AEC promoted a higher relative shoot growth rate of seedlings. The research revealed that seed priming with light quality and aqueous extracts of C. rotundus tubers modulates the germination and initial growth of M. oleifera seedlings. More work needs to be done to determine the responsible compounds in AEC that is responsible for priming growth as phytohormones.

RESUMO

This research was carried out aiming at evaluating the effects of nitrate and ammonium ions on nutrient accumulation, biochemical components and yield of Italian zucchini (cv. Caserta) grown in a hydroponic system under salt stress conditions. The experiment was carried out in a greenhouse utilizing an experimental design in randomized blocks, arranged in a 2 x 5 factorial scheme, with 4 replications. The treatments consisted of two forms of nitrogen (nitrate - NO3- and ammonium - NH4+) and 5 electrical conductivity levels of irrigation water (ECw) (0.5, 2.0, 3.5, 5.0 and 6.5 dS m-1). The analysis of the results indicated that supply of N exclusively in NH4+ form promotes greater damage to the leaf membrane and reduction in accumulation of macronutrients and higher Na+/K+, Na+/Ca++ and Na+/Mg++ ratios in the shoots of zucchini plants. Electrical conductivity of irrigation water above 2.0 dS m-1 reduces the accumulation of nutrients in shoot and yield of Italian zucchini plant. The toxicity of NH4+ under Italian zucchini plants overlap the toxicity of the salinity, since its fertilization exclusively with this form of nitrogen inhibits its production, being the NO3- form the most suitable for the cultivation of the species.

RESUMO

BACKGROUND: There is evidence that intrauterine growth restriction, resulting in newborn girls that are small for gestational age (SGA), may be related to the onset of polycystic ovary syndrome (PCOS). Thus, we studied whether women born SGA have a higher prevalence of PCOS than women born appropriate for gestational age (AGA). METHODS: This was a prospective birth cohort study of 384 women born at term between June 1, 1978, and May 31, 1979, in Ribeirão Preto, Brazil. After exclusion, 165 women effectively participated in this study, of whom 43 were SGA and 122 were AGA. The prevalence of PCOS was analysed. At a mean age of 29 years, the women agreed to follow the study protocol, which included: anamnesis, physical examination, serum tests [follicle stimulating hormone, luteinizing hormone, total and free testosterone, dehydroepiandrostenedione sulphate, 17-OH-progesterone, fasting insulin, sex steroid-binding globulin (SHBG) and fasting glucose] and pelvic ultrasound. Data regarding gestational age, birthweight, age at menarche and maternal data were obtained from the files of the cohort. The adjusted relative risk (RR) values of the SGA, insulin resistance, body mass index, maternal smoking and parity variables were analysed using Poisson regression with robust adjustment of variance for the prediction of PCOS. RESULTS: The prevalence of PCOS was higher in the SGA group than in the AGA group [adjusted RR = 2.44, 95% CI (1.39-4.28)]. Hyperandrogenism was more prevalent in the SGA women than in the AGA women (P = 0.011). Circulating SHBG was lower in the SGA women than in the AGA women (P = 0.041), but fasting insulinemia was similar in both groups. CONCLUSIONS: The prevalence of PCOS in SGA women was twice as high as in AGA women in our study population.

RESUMO

The clinical manifestations of human Chagas disease are associated with distinct and complex host-parasite interactions that directly involve the host's immune system. In this study, we analysed the relationship between the production of intracytoplasmic cytokines after in vitro stimulation with the recombinant antigens CRA (cytoplasmatic repetitive antigen) or FRA (flagellar repetitive antigen) from Trypanosoma cruzi and the chronic cardiac or indeterminate clinical forms of Chagas disease. The chagasic patient groups consisted of 39 individuals, selected at the Chagas Disease Unit of the Oswaldo Cruz University Hospital, whom presented either a cardiac form without cardiac dilatation (CARD 1), cardiac form with cardiac dilatation (CARD 2) or indeterminate form (IND). Blood samples were obtained from these patients and cultured in the presence of CRA or FRA. The cytokines produced by lymphocytes and monocytes after antigen stimulation were analysed by flow cytometry. Our results showed that the IFN-γ and TNF-α, produced by CD8+ T lymphocytes after in vitro stimulation with CRA, differed among chagasic patients with CARD 1, CARD 2 or IND. We propose that these cytokines could be utilized as immunological markers for clinical cardiac forms of Chagas disease. In a prospective study of patients presenting IND and CARD 1, the assay performed in this paper could serve as a tool to monitor therapeutic interventions, thus improving the patient's quality of life.

Malassezia spp in skin microbiome scalp has been implicated in seborrheic dermatitis pathogenesis. Thus, treatment based in antifungal combined to topical keratolitic agents have been indicated as well as oral isotretinoin as it reduces the sebum production, glandular's size and possesses anti-inflammatory properties. This randomized, comparative and therapeutic trial aimed toper form the genotypic identification of Malassezia species before and after low-dose oral isotretinoin or topical antifungal treatments for moderate to severe seborrhea and/or seborrheic dermatitis on scalp. Scales and sebum of the scalp were seeded in the middle of modified Dixon and incubated at 32°C. For genotypic identification polymerase chain reaction primers for the ITS and D1/D2 ribossomal DNA were used and followed by samples sequencing. The procedure was conducted before and after therapeutic and randomized intervention for moderate to severe seborrhea/seborrheic dermatitis on the scalp, including oral isotretinoin, 10 mg, every other day and anti-seborrheic shampoo (piroctone olamine), over six months. The M. globosa and M. restricta were the most frequent species isolated on the scalp before and after both treatments. Other non-Malassezia species were also identified. The Malassezia spp. were maintained in the scalp after both treatments that were equally effective for the control of seborrhea/seborrheic dermatitis clinical signs.

RESUMO

Proper morphology is essential for the ability of Candida albicans to switch between yeast and hyphae and thereby sustain its virulence. Here we identified, by differential screening, a novel C. albicans AAA ATPase encoding gene, CaYLL34 (RIX7), with enhanced expression in hyphae. Phylogenetic analysis suggests that CaYLL34 belongs to a "VCP-like" subgroup of AAA ATPases essential for yeast viability and contains a bipartite nuclear localization signal. Inactivation of one copy of CaYLL34, by the URA-Blaster method, generated the heterozygous mutant strain M61. This strain has severe phenotypic alterations, such as a highly increased vacuole, abnormal cell shape and reduced growth in different conditions. Also, major pathogenicity factors are affected in M61, for instance, a significant decrease of hypha formation (>90%), surface biofilm adhesion (86%) and secreted aspartyl proteinase activity (76.5%). Our results show that the partial impairment of CaYll34p cellular levels is sufficient to affect the proper cellular morphology and pathogenicity factors and suggest that this protein is required for biogenesis of ribosomal subunits. Accordingly, we propose that the product of CaYLL34 could be tested as a novel target for antifungal drugs.

RESUMO

The incidence of C. dubliniensis in South America has not yet been determined. In the present study, oral swab samples were taken from 108 HIV-infected/AIDS individuals attending 6 separate Brazilian HIV-treatment centers to determine the incidence of C. dubliniensis in this population. Swabs were plated onto CHROMagar Candida medium and 155 isolates, presumptively identified as C. albicans or C. dubliniensis were further investigated. In a preliminary screen for C. dubliniensis, 13 of the 155 isolates showed no or poor growth at 42 degrees C, and all them were subjected to randomly amplified polymorphic DNA (RAPD) and polymerase chain reaction (PCR) analysis using C. dubliniensis-specific primers. We confirmed that 4 out of 13 isolates were C. dubliniensis, representing an incidence rate of 2.8% for the Brazilian HIV-infected population infected with yeasts exhibiting green colonies on CHROMagar Candida. This value is significantly lower than those reported in Ireland and the United States.

RESUMO

We describe the case of a newborn child with D-transposition of the great arteries who was found to have an uncommon single-origin coronary artery system. Surgical correction by arterial switch was performed but resulted in tension of the left coronary branch, which prevented discontinuation of cardiopulmonary bypass. The problem was eventually resolved by conversion into a dual coronary artery system through direct implantation of the left coronary branch into the aorta. The patient remains well, free from ischemia and with a patent left coronary artery, 2 years after operation.

RESUMO

The integrity of the pulmonary annulus was preserved in 21 of 22 children consecutively subjected to total correction of tetralogy of Fallot. There was one early death (4.8%). The 20 survivors were studied prospectively and constitute the basis of this report. Mean age was 59.3 +/- 35.3 months (range 6 months-12 years). Eight patients (40%) had previous Blalock-Taussig shunts and 3 (15%) had associated cardiac anomalies. Two-dimensional Doppler echocardiography had demonstrated right ventricle to main pulmonary artery peak systolic gradients of 75.4 +/- 12.7 mmHg (range 50-104 mmHg). Ventricular infundibulectomy was carried out in all patients but one, who had abnormal coronary anatomy. Extensive incision or resection of hypertrophied or abnormal muscle bands was performed. Pulmonary valve commissurotomy was performed in 14 patients (70%), 10 (50%) had autologous pericardium supravalvular enlargement of the pulmonary trunk and one (5%) had an infundibular bovine pericardial patch. Intraoperative, postrepair right/left ventricular pressure ratios were 0.67 +/- 0.18 (range 0.41-1.0). All ratios were accepted, because all patients were haemodynamically well and only two required inotropic (dopamine) support. There was no late mortality. During the early follow-up period (mean 13.0 +/- 5.4 months, range 7-25 months), all patients had two-dimensional Doppler echocardiography measured gradients at discharge (mean 44.2 +/- 22.6 mmHg, range 10-86 mmHg; p less than 0.001 vs. preoperative values), at 3 months (mean 31.1 +/- 14.5 mmHg, range 8-64 mmHg; p less than 0.011 vs. values at discharge) and at 6 months (mean 28.5 +/- 14.3 mmHg, range 12-57 mmHg; p = 0.009 vs. values at discharge). A similar favourable evolution of the diameters of the pulmonary annulus and of the right pulmonary artery was observed.(ABSTRACT TRUNCATED AT 250 WORDS)

RESUMO

Two experiments were performed on the predation of the planarian Dugesia tigrina (Girard) upon 2 mosquito prey species, Aedes albopictus and Culex quinquefasciatus. Bioassays were carried out in sectioned tires with 2 liters of water. In the 1st experiment, predation was evaluated using 4, 8, and 12 mature planarians against 40 2nd-stage larvae of each mosquito species alone. In the 2nd experiment, the same 3 predator densities were used with a pool composed of 20 2nd-stage larvae of each mosquito species. In the 1st experiment, final corrected mortality of Ae. albopictus reached 89.1, 98.8, and 99.6% and final corrected mortality of Cx. quinquefasciatus reached 29.4, 48.0, and 53.0%, respectively, with 4, 8, and 12 planarians. In the 2nd experiment and when subjected to the density of 4 planarians, Ae. albopictus was more susceptible to predation, with a selectivity index of 0.87, whereas this index was 0.13 for Cx. quinquefasciatus. Predation was more intensive during the 1st 4 days of the experiments, when most larvae were in the 2nd and 3rd stages. We observed that Cx. quinquefasciatus larvae were faster than Ae. albopictus in reacting to planarian contacts, resulting in more success in escaping from the predator attacks.

In order to evaluate the productive response and economic viability of diets that included hays as alternative forage for birds in a semi-intensive system, a performance test was held, in which 600 Label Rouge birds were distributed in a completely randomized design with five treatments, six replications, and twenty birds per plot. The control diet (RCO) was based on corn grain and soybean meal and used as a reference for the inclusion of forage hay, 20% by hays Silk Flower (Calotropis procera), Maniva Cassava (Manihot esculenta) , Mata Pasto (Senna obtusifolia) , or Leucaena (Leucaena leucocephala). The average for RCO feed intake, RMM, RFS, RMP, and RLE were 0.1050, 0.1470, 0.0888, 0.1411, and 0.1441 kg/bird/day, respectively, and feed prices were 1,489, 1,209, 1,189, 1,179, and 1,249 (R$/kg), respectively. Therefore, the total costs of food in the 84-day period of semi-confinement of the birds per ration were 1950,67, 1790,70, 1061,90, 1676,04, and 1813,60 (actual R$), respectively. These results showed a relative decline in power with regard to cost via including hay as an alternative forage in diets, but after evaluating the performance in weight gain and carcass yield of birds, the total revenue was lower in each of the feeds with hay forage and thus the economic efficiency and average cost rates were better with the RCO feed; however, the RMM had superior profitability compared to other diets with hay.(AU)
