OBJECTIVES: To achieve the therapeutic effects, antibiotics must penetrate rapidly into infection sites and bind to targets. This study reviewed updated knowledge on the ability of antibiotics to penetrate into the lung, their physicochemical properties influencing the pulmonary penetration and their ability to bind to targets on pneumococci. METHODS: A search strategy was developed using PubMED, Web of Science, and ChEMBL. Data on serum protein binding, drug concentration, target binding ability, drug transporters, lung penetration, physicochemical properties of antibiotics in low respiratory tract infection (LRTI) were collected. RESULTS: It was seen that infection site-to-serum concentration ratios of most antibiotics are >1 at different time points except for ceftriaxone, clindamycin and vancomycin. Most agents have proper physicochemical properties that facilitate antibiotic penetration. In antimicrobial-resistant Streptococcus pneumoniae, the binding affinity of antibiotics to targets mostly decreases compared to that in susceptible strains. The data on binding affinity of linezolid, clindamycin and vancomycin were insufficient. The higher drug concentration at the infection sites compared to that in the blood can be associated with inflammation conditions. Little evidence showed the effect of drug transporters on the clinical efficacy of antibiotics against LRTI. CONCLUSIONS: Data on antibiotic penetration into the lung in LRTI patients and binding affinity of antibiotics for pneumococcal targets are still limited. Further studies are required to clarify the associations of the lung penetration and target binding ability of antibitotics with therapeutic efficacy to help propose the right antibiotics for LRTI.

BACKGROUND: Most spinal meningiomas primarily grow in the intradural extramedullary location. Epidural meningiomas are uncommon; if detected, they usually coexist with intradural lesions. They inhere more aggressive and invasive characteristics compared with their counterparts inside the dura. CASE DESCRIPTION: We report a 22-year-old female who was admitted to the hospital with weakness and numbness in both lower limbs. Her cervical magnetic resonance imaging revealed an en-plaque and dumbbell-shaped lesion located from C5 to C8. After gadolinium injection, the whole mass was enhanced and unveiled two portions: intradural and extradural. The bone window of the computed tomography scan revealed calcification inside the lesion. The patient underwent tumor removal surgery. The pathology findings showed a psammomatous meningioma. After 6 months of surgery, the patient has been able to walk with walkers. CONCLUSION: We should consider spinal meningioma as a differential diagnosis when encountering an extradural lesion in the cervical region. The optimal surgical treatment for young patient with epidural meningiomas is radical surgery with dura attachment removal.

BACKGROUND: Carbon nanotube (CNT) characteristics, besides the processing conditions, can change significantly the microwave absorption behavior of CNT/polymer composites. In this study, we investigated the influence of three commercial multi-walled CNT materials with various diameters and length-to-diameter aspect ratios on the X-band microwave absorption of epoxy nanocomposites with CNT contents from 0.125 to 2 wt%, prepared by two dispersion methods, i.e. in solution with surfactant-aiding and via ball-milling. RESULTS: The laser diffraction particle size and TEM analysis showed that both methods produced good dispersions at the microscopic level of CNTs. Both a high aspect ratio resulting in nanotube alignment trend and good infiltration of the matrix in the individual nanotubes, which was indicated by high Brookfield viscosities at low CNT contents of CNT/epoxy dispersions, are important factors to achieve composites with high microwave absorption characteristics. The multi-walled carbon nanotube (MWCNT) with the largest aspect ratio resulted in composites with the best X-band microwave absorption performance, which is considerably better than that of reported pristine CNT/polymer composites with similar or lower thicknesses and CNT loadings below 4 wt%. CONCLUSIONS: A high aspect ratio of CNTs resulting in microscopic alignment trend of nanotubes as well as a good level of micro-scale CNT dispersion resulting from good CNT-matrix interactions are crucial to obtain effective microwave absorption performance. This study demonstrated that effective radar absorbing MWCNT/epoxy nanocomposites having small matching thicknesses of 2-3 mm and very low filler contents of 0.25-0.5 wt%, with microwave energy absorption in the X-band region above 90% and maximum absorption peak values above 97%, could be obtained via simple processing methods, which is promising for mass production in industrial applications. We have developed a pHANNIBAL-like silencing vector, pSilent-1, for ascomycete fungi, which carries a hygromycin resistance cassette and a transcriptional unit for hairpin RNA expression with a spacer of a cutinase gene intron from the rice blast fungus Magnaporthe oryzae. In M. oryzae, a silencing vector with the cutinase intron spacer (147 bp) showed a higher efficiency in silencing of the eGFP gene than did those with a spacer of a GUS gene fragment or a longer intron (850 bp) of a chitin binding protein gene. Application of pSilent-1 to two M. oryzae endogenous genes, MPG1 and polyketide synthase-like gene, resulted in various degrees of silencing of the genes in 70-90% of the resulting transformants. RNA silencing was also induced by a pSilent-1-based vector in Colletotrichum lagenarium at a slightly lower efficiency than in M. oryzae, indicating that this silencing vector should provide a useful reverse genetic tool in ascomycete fungi.
