Background: The current work involved monitoring two biomarkers in the plasma of children with ASD: the cofactor thiamine that is involved in neurotransmitters modulation for acetylcholine, and the compound histamine, which acts as a neuromodulator by regulating the release of other neurotransmitters. This is the first report to highlight the potential utilization of plasma levels of the selected two brain-related biomarkers in children with ASD.Methods: A total of 43 children with ASD of both genders (age 4-12 years) were involved in this study and compared to age and gender-matched control children (n = 42). The diagnosis of ASD was based on the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM5), followed by an additional assessment using the Childhood Autism Rating Scale (CARS). All participants were Jordanian children on Mediterranean diet, and had no history of chronic illness or medications. Measurement of thiamine and histamine in plasma was performed using enzyme-linked immunosorbent assay (ELISA).Results: The outcomes revealed that average histamine levels (31.7 ± 18.5 ng/ml) of ASD group were 5.3× higher (p < .001) compared to their control (0.013 ± 0.011 ng/ml; 6.03 ± 4.25 ng/ml), while thiamine (10.78 ± 7.49 ng/ml) levels of ASD group were significantly lower (p < .001) than the control (37.92 ± 26.87 ng/ml; 0.209 ± 0.054 ng/ml).Conclusions: The study is proposing that monitoring of the plasma levels of thiamine and histamine as biomarkers for future evaluation and development of ASD therapies and nutritious diets.

BACKGROUND: Elemental analysis has been increasingly used for biomonitoring heavy metals and trace elements. METHODS: This study monitored the levels of two heavy metals (Al and Pb), and seven trace elements (Macroelements Mg, K, P and Ca; Microelements Zn, Cu, Fe) in scalp hair of 57 children with severe autism spectrum disorder (ASD) and 50 age-matched controls, using Inductively Coupled Plasma Atomic Emission Spectrophotometry (ICP-AES). RESULTS: Compared to controls, significantly higher levels of Al (p =  0.001), Pb (p =  0.001) and K (p =  0.021), with lower levels of Mg and Zn (p = 0.038) were observed for the ASD group. ASD boys had higher levels of Al (p =  0.001), Pb (p =  0.001) and K (p =  0.017) than control boys, while ASD girls had higher Pb levels (p =  0.005) than control girls. The ASD subgroup exposed to passive smokers had higher levels of Al (p =  0.033) and Pb (p =  0.001, and the ASD subgroup not exposed to passive smoke had higher levels of Al (p =  0.011), Pb (p =  0.001), K (p =  0.003); and lower levels of Mg (p =  0.011) than their controls. Other confounding factors and the correlation between these elements were also investigated. CONCLUSION: This data suggests that exposure to Al and Pb, increase intake of K, and decreased intake of magnesium and zinc, may contribute to ASD etiology.

Vipera palaestinae is responsible for many venomous incidents in the Middle East. However, this species is not included in the antigenic pool of venoms for the production of the regionally available polyvalent antivenoms. In an attempt to develop a potential complementary alternative therapy for snakebite patients, this study is investigating the antagonistic effect of Eryngium creticum against V. palaestinae venom. In this context, the concentration of the venom as well as the electrophoretic profile, and the venom LD50 were determined by intraperitoneal injection (ip). The methanolic leaf extract was prepared, and its safety on rats was examined. Adult male Sprague-Dawley rats were divided into 8 groups (n = 6); G1-G3 were injected subplantar in the right hind paws with 2.5, 3.125, and 3.75 mg kg-1 then 200 mg kg-1 extract ip. G4-G6 were given the same venom dose with no extract, respectively. Controls were G7 that only had the extract ip, and G8 that was injected subplantar with PBS. The swollen paws were measured at Hour 0 (before injection), Hour 1, Hour 6, and Hour 24. IL-6 and TNF-α were measured in serum using ELISA. Histopathological changes were examined in paw sections. The pooled venom concentration was 176.93 ±â€¯35.81 mg ml-1, revealed 10 protein bands (5-80 kDa), and the LD50 via ip rout was 6.56 mg kg-1. Paw edema peaked at Hour 1. At Hour 6, edema in G1 was significantly reduced (p < 0.05) compared to G6, while at Hour 24 there was no significant difference between all groups including the controls. Treated animals in G1-G3 expressed IL-6 significantly lower (p < 0.001) than untreated G4-G6, respectively. Levels of TNF-α in G1 and G2 were significantly (p < 0.001) lower than G3-G6, while G5 and G6 were significantly (p < 0.001) higher than G1-G4. Histopathological changes showed intensifying edema, hemorrhage, and inflammation with incrementing venom doses. Sections from treated animals expressed less adverse changes compared to untreated animals. Together, the outcomes are encouraging future utilization of E. creticum as a supportive remedy for snakebite cases.

Bromide (Br-) is a bromine atom with a negative charge which is released mainly in the production of pesticides and flame retardants. It is also found naturally in seawater. Br¯ has been associated with many detrimental effects such as respiratory problems, gastric hemorrhages, and dermal burns. The aim of the study was to monitor serum bromide in humans and to correlate its level with genotoxicity and apoptosis in human. The study utilized comet assay, to measure DNA damage in peripheral leukocytes (i.e. T%DNA), enzyme-linked immunosorbent assay were used to determine fortilin level as an apoptosis marker, and spectrophotometry to measure serum Br¯ in two populations at the Dead Sea area, which are located close to and far from a local bromine factory: Ghor As-safi and Deir Alla, respectively. The biomarkers were compared with the correlating serum Br¯. A total of 397 individuals were involved in the study. The serum Br- and the genotoxicity biomarker were significantly higher (p < 0.001) in Ghor As-safi than in Deir Alla. In contrast, serum fortilin did not differ significantly between the two regions (p > 0.05). T%DNA was significantly correlated (r = 0.867, p < 0.01) to serum Br¯. In conclusion, residing near a bromide source site is increasing the bromide body burden, and enhancing genotoxicity with no detectible apoptosis. Furthermore, the selected biomarkers could serve as tools to assess the toxicity of bromide as a consequence of environmental exposure.

The acute toxicity of polychlorinated dibenzo-p-dioxins/dibenzofurans (PCDD/Fs) was investigated in the earthworm Eisenia andrei using filter paper toxicity test. Protein content, catalase (CAT) activity, and histology of intestinal wall (chloragogen cells and intestinal epithelium) were investigated in earthworms exposed for 48 h to 0 (control), 0.5, 1.0, and 1.5 ng/cm2 PCDD/Fs. The results showed an increase in the total protein content 1.56- (p = 0.104), 1.66- (p = 0.042), and 2.26-fold (p < 0.001), respectively, compared to control. The average ± standard deviation of tissular CAT activity showed no significant differences; it was 36.01 ± 7.65, 36.17 ± 9.45, 36.08 ± 9.80, and 40.01 ± 6.98 U/g tissue, respectively. However, the average specific activity of CAT ± standard deviation was significantly decreased (p < 0.001) at all doses compared to control; it was 2.93 ± 0.42, 1.93 ± 0.53, 1.80 ± 0.38, and 1.53 ± 0.44 U/mg protein, respectively. There was a progressive damage in both of the intestinal villi and the chloragogenous tissue associated with the incrementing doses. Since the toxic mixture altered the investigated biomarkers of E. andrei within 48 h, the cellular and molecular alterations resulted from the filter paper contact test could be utilized as a rapid toxicity assessment tool of environmental contamination with dioxins/furans and to assess consequent potential adverse effects on soil biota and other organisms in the ecosystem.
