The aim of this study was to determine the prevalence of polymorphisms in the glutathione S-transferase genes GSTM1 and GSTT1 in patients with lens opacity (cataract). Peripheral blood samples were obtained from male and female patients (N = 23) with cataract. The GSTM1 and GSTT1 polymorphic regions were amplified by polymerase chain reaction, and the amplification products were electrophoresed on a 2% agarose gel. The obtained bands were by staining with ethidium bromide. The results were compared by a chi-square test using the BioEstat software (v.5.0). The frequencies of the GSTM1- and GSTT1-null genotypes were higher than those of the GSTM1- and GSTT1-present genotypes. The frequency of GSTT1-null genotypes was approximately 1.7 times higher than that of GSTM1, which was a statistically significant difference (P = 0.0019). Although a consensus remains to be reached on the correlation between genetic polymorphisms in GSTs and cataract susceptibility, the observations from most scientific studies are similar to those reported in this study. Thus, we conclude that the absence of these genes, particularly GSTT1, is correlated with the development of lens opacity.

RESUMO

Leishmaniasis is a vectorborne disease caused by Leishmania protozoa, which is a major health problem and a neglected disease common in many regions of the world. Leishmania is an intracellular parasite transmitted by sand flies that causes clinical manifestations ranging from a severe and potentially fatal disease named visceral leishmaniasis to less severe but in many cases disfiguring diseases that mainly affect the skin or mucosal tissues, known as cutaneous leishmaniasis. Despite the detection of Leishmania parasites in the brain and cerebrospinal fluid of human patients and dogs, epidemiological data, as well as information about the mechanisms of central and peripheral nervous system alterations, are poorly described. This review is focused on the current knowledge about the neurological manifestations and immunopathogenic mechanisms in human patients and animals infected with Leishmania.

Polycystic Ovary Syndrome (PCOS) is the most common cause of subfertility associated to metabolic disorders. The aim of this study was to correlate metabolic and proinflammatory factors in women with PCOS. The frequency of Plasminogen Activator Inhibitor-1 (PAI-1) promoter 4 G/5 G polymorphism was also compared to healthy controls. We evaluated 79 PCOS and 79 healthy women. PAI-1 levels are positively correlated with proinflammatory factors in PCOS group. 4 G allele in PAI-1 gene was more frequent in PCOS and the 4G/4 G genotype was associated with increased PAI-1 levels. A correlation between insulin resistance and proinflammatory and overweight was also observed. C-reactive protein, serum levels of vascular cell adhesion molecule-1 (sVCAM-1), Lipid Accumulation Product (LAP) and vitamin D are good tools to evaluated factors associated to cardiovascular risk in women with PCOS.

Many plants may accumulate rainwater, forming phytotelmata, aquatic microhabitats inhabited by various organisms. The aim of this study was to conduct an inventory of heterotrophic flagellates associated with phytotelmata of the bromeliad Aechmea distichantha Lem., found in rocky cliffs on the Upper Paraná River. The bromeliads were removed manually from the rocky wall, the water was removed and cultures of organisms of each plant were mounted in Petri dishes. Sixteen species of heterotrophic flagellate were recorded, drawn and described, among them one species belonging to the Amorpha Domain and 15 species to the Diaphoretiches Domain. The groups with most species were Euglenida and Kinetoplastea. The low diversity of heterotrophic flagellates recorded in this study, compared to the plankton of lakes and reservoirs, is probably related to the fact that phytotelmata are habitats with extreme environmental conditions, thus selecting organisms tolerant to these environments.

BACKGROUND: Photodynamic therapy (PDT) is an anticancer therapy that associates the photosensitizer (PS), oxygen and light to destroy cancer cells. Methylene blue (MB) is considered a second generation phenothiazine dye with excellent photochemical properties. AIM: To evaluate whether MB-mediated PDT can induce oxidative stress and inflammation, therefore, interfering tumor growth. MATERIALS AND METHODS: The study was conducted on Wistar rats transplanted with Walker 256 carcinosarcoma (W256). The proinflammatory interleukins levels (IL-1ß, IL-6, IL-10, TNF-α) were determined by ELISA, mRNA expression of COX-1, COX-2, iNOS and eNOS by RT-PCR, lipid peroxidation was measured by the TBARS method. Moreover, myeloperoxidase (MPO) activity in neutrophils was determined by MPO activity assay. All indices mentioned above were determined in tumor tissue. Kaplan - Meier and Gehan - Breslow - Wilcoxon tests were used for survival analysis. RESULTS: We found that the treatment of W256 with 0.1% MB + 1 J/cm2 provoked a significant increase in the interleukins levels (IL-1ß, IL-6, IL-10, TNF-α), prostaglandin E2, the mRNA expression of COX-2, iNOS, lipid peroxidation and MPO activity in tumor tissue, which were statistically different (p < 0.05) compared to other experimental and control groups. The results of the estimation of survival curves show a greater probability of survival in 0.1% MB + 1 J/cm2 (total energy dose =142.8 J/cm2) treated group. CONCLUSION: Our results suggest that treatment of W256 with 0.1% MB + 1 J/cm2 was able to promote cytotoxic effects in tumor tissue by the generation of reactive oxygen species causing inflammation and thus interfering in the tumor growth.

Carbohydrate antigen 125 (CA125) has long been used as an ovarian cancer biomarker. However, because it is not specific for ovarian cells, CA125 could also be used to monitor congestion and inflammation in heart disease. Acute heart failure (HF) is used to identify patients with a worse prognosis in ST-segment elevation myocardial infarction (STEMI). We aimed to determine the association of CA125 with acute HF in STEMI and to compare CA125 with N-terminal pro brain natriuretic peptide (NTproBNP) with a cross-sectional study. At admission, patients were examined to define Killip class and then underwent coronary angioplasty. Blood samples, preferably taken in the hemodynamic ward, were centrifuged (1500 g for 15 min at ambient temperature) and stored at -80°C until biomarker assays were performed. Carassius auratus is a teleost fish that has been largely used in behavioral studies. However, little is known about potential environmental influences on its performance of learning and memory tasks. Here, we investigated this question in C. auratus, and searched for potential correlation between exercise and visuospatial enrichment with the total number of telencephalic glia and neurons. To that end, males and females were housed for 183 days in either an enriched (EE) or impoverished environment (IE) aquarium. EE contained toys, natural plants, and a 12-hour/day water stream for voluntary exercise, whereas the IE had none of the above. A third plus-maze aquarium was used for spatial and object recognition tests. Different visual clues in 2 of its 4 arms were used to guide fish to reach the criteria to complete the task. The test consisted of 30 sessions and was concluded when each animal performed three consecutive correct choices or seven alternated, each ten trials. Learning rates revealed significant differences between EE and IE fish. The optical fractionator was used to estimate the total number of telencephalic cells that were stained with cresyl violet. On average, the total number of cells in the subjects from EE was higher than those from subjects maintained in IE (P=0.0202). We suggest that environmental enrichment significantly influenced goldfish spatial learning and memory abilities, and this may be associated with an increase in the total number of telencephalic cells.

The objective of this study was to verify the agreement between pre-programmed and executed pacing during race walking and whether level of the athletes experience and performance influenced this relationship. Twenty-nine national and international race walkers participated in this study (14 males, 24.0±7.1 years old, and 15 females, 23.3±7.3 years old). Pre-programmed pacing for 10- and 20-km official walking races was self-selected via demonstrative pacing charts prior to races, while executed pacing was analyzed by a specialist investigator via an individual plot of current velocity versus distance. There was no agreement between pre-programmed and executed pacing (P=0.674). There was no association between the ability to match the pre-programmed pace with the executed pace and race walking experience or level of performance. Low- and high-performance athletes pre-programmed a similar pacing profile (P=0.635); however, high-performance athletes generally executed an even pacing strategy, while low-performance athletes generally adopted a positive pacing strategy (P=0.013). Race walkers did not faithfully match their pre-programmed with their executed pacing, and this seemed to be independent of previous experience and level of performance. High-performance athletes, however, tended to execute an even pacing strategy, even though this had not been pre-programmed.

We investigated the effect of caffeine ingestion combined with a 2-wk sprint interval training (SIT) on training-induced reductions in body adiposity. Twenty physically-active men ingested either 5 mg/kg of cellulose as a placebo (PLA, n=10) or 5 mg/kg of caffeine (CAF, n=10) 60 min before each SIT session (13×30 s sprint/15 s of rest). Body mass and skinfold thickness were measured pre- and post-training. Energy expenditure was measured at rest, during exercise, and 45 min after exercise in the first SIT session. Body fat was similar between PLA and CAF groups at pre-training (P>0.05). However, there was a significant decrease in body fat after training in the CAF group (-5.9±4.2%, P<0.05) but not in PLA (1.5±8.0%, P>0.05). There was no difference in energy expenditure at rest and during exercise between PLA and CAF groups (P>0.05), but the post-exercise energy expenditure was 18.3±21.4% greater in the CAF than in the PLA group (P<0.05). In conclusion, caffeine ingestion before SIT sessions induced a body fat loss that may be associated with higher post-exercise energy expenditure.

Visceral leishmaniasis (VL) is one of the most important reemerging parasitic disease in the world. The domestic dog is the main reservoir in urban environments. The aim of this work was to extend the knowledge on canine Leishmania infection in the city of Fortaleza in northeastern Brazil, identifying the risk factors inherent in dog susceptibility to the infection. Two populations were analyzed, domestic dogs from clinics and the Veterinary Hospital Unit of Ceará State University and stray dogs captured by the Center for Zoonosis Control in Fortaleza. Blood samples were collected and centrifuged and the sera were stored at -20 degrees C. ELISA, with soluble crude Leishmania chagasi antigens (LTCC - WDCM731) was used for diagnosis. A total of 1,381 samples were tested, 750 from domestic and 631 from stray dogs. The seroprevalence of canine VL was 21.4% (135/631) in stray dogs and 26.2% (197/750) in domestic dogs. The seroprevalence of Leishmania infection in the six administrative regions of the city (Secretarias Executivas Regionais, or SER) among stray dogs was highest in SER V, representing 31.4% of the cases, with large dogs more infected (27.7%). Among domestic dogs Leishmania infection was most prevalent in SER V (38.5%) and VI (37.6%). The dogs' age (1-6 years), large size, environment with dense vegetation and presence of clinical signs compatible with Leishmania infection were associated with the illness in domestic dogs. The frequency of the infection varied seasonally. The seroprevalence was greatest in July and December. These results confirm Fortaleza is an endemic area for canine VL and suggest some variables associated with increasing infection risk in dog populations.

The aim of this study was to test the hypothesis that reduced pre-exercise carbohydrate (CHO) availability potentiates fat oxidation after an exhaustive high-intensity exercise bout. Eight physically active men underwent a high-intensity exercise (∼95% V̇O2max) until exhaustion under low or high pre-exercise CHO availability. The protocol to manipulate pre-exercise CHO availability consisted of a 90-min cycling bout at ∼70% V̇O2max + 6 × 1-min at 125% V̇O2max with 1-min rest, followed by 48 h under a low- (10% CHO, low-CHO availability) or high-CHO diet (80% CHO, high-CHO availability). Time to exhaustion was shorter and energy expenditure (EE) lower during the high-intensity exercise in low- compared to high-CHO availability (8.6±0.8 and 11.4±1.6 min, and 499±209 and 677±343 kJ, respectively, P<0.05). Post-exercise EE was similar between low- and high-CHO availability (425±147 and 348±54 kJ, respectively, P>0.05), but post-exercise fat oxidation was significantly higher (P<0.05) in low- (7,830±1,864 mg) than in high-CHO availability (6,264±1,763 mg). The total EE (i.e., exercise EE plus post-exercise EE) was similar between low- and high-CHO availability (924±264 and 1,026±340 kJ, respectively, P>0.05). These results suggest that a single bout of high-intensity exercise performed under low-CHO availability increased post-exercise fat oxidation, and even with shorter exercise duration, both post-exercise EE and total EE were not impaired.

Ethanol is an important risk factor for the occurrence of several brain disorders that depend on the amount, period and frequency of its consumption. Chronic use of ethanol often leads to the development of neurodegenerative syndromes, which cause morphological and functional impairments such as foetal alcohol syndrome in newborns exposed to ethanol during pregnancy, Wernicke-Korsakoff Syndrome and, more rarely, Marchiafava-Bignami disease (MBD). MBD is characterized by primary degeneration of the corpus callosum, without inflammation and is associated with oxidative stress and hypovitaminosis, as well as altered mental status, to mention dementia, seizures, depression and so on. This review discusses MBD and poor nutrition as a risk factor for the development of such alcoholic syndrome, with focus on diagnosis, pathogenic aspects, signs and symptoms, as well as therapeutic perspectives. On the basis of the inclusion/exclusion criteria adopted, the performed search in scientific databases (Pubmed, Scielo and Google Scholar) resulted in 100 studies that are being presented and discussed in the present work. Review, case-control and cohort studies on alcoholism-associated hypovitaminosis, oxidative stress, MBD and ethanol metabolism pathways were admitted as relevant. We highlight that MBD is a poorly described, diagnosed, insidious and progressive condition, for which evidence suggests a synergism between ethanol-induced neurotoxic effects and hypovitaminosis B. Present treatment consists of vitamin B1(thiamine) supplementation. Nonetheless, other strategies such as the inclusion of antidepressants or steroidal anti-inflammatories as add-on therapies have been employed as an attempt to improve the damage. Indeed, both the diagnosis and treatment are difficult, and death occurs within few years.

We investigated if carbohydrate (CHO) availability could affect the excess post-exercise oxygen consumption (EPOC) after a single supramaximal exercise bout. Five physically active men cycled at 115% of peak oxygen uptake (V̇O2 peak) until exhaustion with low or high pre-exercise CHO availability. The endogenous CHO stores were manipulated by performing a glycogen-depletion exercise protocol 48 h before the trial, followed by 48 h consuming either a low- (10% CHO) or a high-CHO (80% CHO) diet regime. Compared to the low-CHO diet, the high-CHO diet increased time to exhaustion (3.0±0.6 min vs 4.4±0.6, respectively, P=0.01) and the total O2 consumption during the exercise (6.9±0.9 L and 11.3±2.1, respectively, P=0.01). This was accompanied by a higher EPOC magnitude (4.6±1.8 L vs 6.2±2.8, respectively, P=0.03) and a greater total O2 consumption throughout the session (exercise+recovery: 11.5±2.5 L vs 17.5±4.2, respectively, P=0.01). These results suggest that a single bout of supramaximal exercise performed with high CHO availability increases both exercise and post-exercise energy expenditure.

Resumo Em sua superfície externa, muitas plantas podem acumular água da chuva formando fitotelmata, ou seja, micro-habitat aquáticos colonizados por diversos organismos, entre eles, os flagelados protistas. Fez-se um inventário de flagelados heterotróficos associados ao fitotelmata da bromélia Aechmea distichantha Lem., encontrada em paredões rochosos em um trecho da bacia do alto rio Paraná. As bromélias foram retiradas manualmente de paredão rochoso e, em laboratório, a água foi retirada dos tanques de cada planta, e foram montadas culturas dos organismos em placas de Petri. Os flagelados heterotróficos foram, então, observados in vivo sob microscópio óptico Olympus BX51, com sistema de contraste de interferência diferencial (DIC) acoplado. Dezesseis espécies flagelados heterotróficos foram registradas, desenhadas e descritas, sendo uma espécie pertencente ao Domínio Amorpha e 15 espécies ao Domínio Diaphoretiches. Os grupos mais especiosos foram Euglenida e Kinetoplastea. A diversidade relativamente reduzida de flagelados heterotróficos, quando comparada ao plâncton de lagos e reservatórios, pode estar relacionada ao fato de os fitotelmata serem habitats limitados, com condições ambientais extremas, selecionando, assim, organismos tolerantes a esses ambientes.

Many plants may accumulate rainwater, forming phytotelmata, aquatic microhabitats inhabited by various organisms. The aim of this study was to conduct an inventory of heterotrophic flagellates associated with phytotelmata of the bromeliad Aechmea distichantha Lem., found in rocky cliffs on the Upper Paraná River. The bromeliads were removed manually from the rocky wall, the water was removed and cultures of organisms of each plant were mounted in Petri dishes. Sixteen species of heterotrophic flagellate were recorded, drawn and described, among them one species belonging to the Amorpha Domain and 15 species to the Diaphoretiches Domain. The groups with most species were Euglenida and Kinetoplastea. Creatinine plays a key role in the function and maturation of fetal kidneys throughout pregnancy. It is important to identify other markers that may help in the diagnosis of renal dysfunction. Our aim was to determine the profile of and the correlation between biochemical markers to be used to assess renal function and maturation of the fetus in the amniotic fluid during pregnancy and to determine the distribution of normal values for creatinine, N-acetyl-beta-D-glucosaminidase (NAG), beta2-microglobulin, glucose, urea, sodium, potassium, phosphorus, calcium, uric acid, albumin, and osmolality in three gestational age groups. This was a cross-section study that assessed 115 samples of amniotic fluid during three different periods of pregnancy, i.e., 13 to 20, 27 to 34, and 36 to 42 weeks. Concentrations of creatinine, NAG, urea, potassium and uric acid increased during pregnancy (P<0.05). Beta2-microglobulin, glucose, sodium, phosphorus, calcium, and albumin concentration and osmolality decreased (P<0.05), whereas beta2-microglobulin, glucose and uric acid presented significant correlations with gestational age and creatinine, respectively (r>0.6, P<0.05). Urea, potassium and phosphorus showed mild correlations with both (r>0.5, P<0.05). NAG, sodium, albumin and osmolality did not show significant correlations (r<0.5, P<0.05). These tests confirmed the important role of creatinine in terms of correlation with gestational age. beta2-Microglobulin, glucose and uric acid were significant as markers of function and maturation of fetal kidneys, whereas NAG did not demonstrate a useful role for the assessment of renal maturation.

This study was conducted on 32 mares during the first 30 d of the postpartum period to characterize the first estrous cycle, assessing ovarian cyclicity by determining plasma progesterone concentration and by transrectal palpation. The total pregnancy rate of the breeding season was 81.25%. The present results show that the incidence of estrus occurring at the beginning of the breeding season were early, long and anovulatory. The mares that did not become pregnant ovulated on average 14.5 d post partum, and those that became pregnant ovulated at 19.6 d post partum (P<0.05). On the basis of clinical and hormonal data, we divided the animals into 4 groups, all presenting signs of estrus: Group 1, animals that did not ovulate (n=7) and that presented basal P(4) levels (0.01-2.34 ng/ml) during the first 30 postpartum days; Group 2, animals that ovulated and did not become pregnant (n=13); Group 3, animals that ovulated and became pregnant (n=8). Maximal P(4) levels ranged from 4.40 to 13.50 ng/ml (Group 2) and from 3.70 to 20.50 ng/ml (Group 3). Group 4 were animals that presented high plasma P(4) levels before any clinical sign of ovulation (n=3). The absence of pregnancy could not be attributed to a failure of the corpus luteum, since the groups of mares that became pregnant exhibited similar plasma P(4) levels as the group of nonpregnant mares. Our findings demonstrated that mares exhibited differences in the timing of the first postpartum estrus, the duration of the first postpartum estrus and the timing of the first postpartum ovulation according to the month of the breeding season in which foaling occurs under tropical conditions. Furthermore, our results indicate that the foal heat may be used since its utilization did not affect the total pregnancy rate of the breeding season.

RESUMO

Two HIV-seronegative patients with cryptococcal disease refractory to conventional antifungal therapy were submitted to an evaluation of the immune system. Hypogammaglobulinemia was found in both and associated with abnormal function of cell-mediated immunity. Hypogammaglobulinemia is considered as a possible predisposing factor for cryptococcal infection. The importance of the antibodies on the control of Cryptococcus neoformans infection is discussed.

In view of the scarcity of data about the topography and syntopy of abdominopelvic viscera of the giant anteater (Myrmecophage tridactyla - Linnaeus, 1758), the present study aimed to elucidate these characteristics and to compare them with the other animal species, especially the domestic ones. Three specimens, two males and one female, were donated by the Environmental Military Police of Franca to the Anatomy Veterinary Laboratory of the University of Franca, after death by road killings. The animals were fixed and maintained in aqueous 10% formaldehyde solution, followed by conventional dissection of the abdominopelvic cavities for subsequent direct inspection and topographic description of the viscera, aiming at comparative analyzes with other species, whose positioning and particularities are already established in the literature. It was observed that most of the viscera of these cavities have similar location and syntopy to domestic animals, except for the kidneys and testicles. In view of the established methodology and the results obtained, it is accepted that more specimens of anteater, both genera, should be evaluated and registered scientifically to confirm the data of the current research and anatomical preconization of the abdominopelvic cavity, inasmuch anatomical individual variation are possible between animals of the same species.(AU)


Diante da escassez de dados sobre a topografia e a sintopia das vísceras abdominopélvicas do tamanduá-bandeira (Myrmecophage tridactyla - Linnaeus, 1758), o presente estudo teve como objetivo elucidar essas características e compará-las com as demais espécies animais, mormente as domésticas. Utilizaram-se três espécimes, dois machos e uma fêmea, provenientes de doação da Polícia Militar Ambiental de Franca ao Laboratório de Anatomia Veterinária da Universidade de Franca, após óbitos por atropelamentos. Os animais foram fixados e mantidos em solução aquosa de formaldeído a 10%, seguidos de dissecação convencional das cavidades abdominopélvicas para posterior inspeção direta e descrição topográfica das vísceras, visando a análises comparativas com outras espécies, cujo posicionamento e cujas particularidades já são bem estabelecidos na literatura. Observou-se que a maioria das vísceras dessas cavidades possuem localização e sintopia similares aos animais domésticos, exceto os rins e os testículos. Diante da metodologia estabelecida e dos resultados obtidos, admite-se que mais espécimes de tamanduás-bandeiras, de ambos os gêneros, devam ser avaliados e registrados cientificamente, visando à confirmação dos dados da atual pesquisa e à preconização anatômica da cavidade abdominopélvica, visto que variações anatômicas individuais são passíveis entre animais da mesma espécie.(AU)
