RESUMO

Etlingera pavieana (Pierre ex Gagnep.) R.M.S. is a rhizomatous plant in the Zingiberaceae family which could be freshly eaten, used as a condiment or as a traditional remedy. Our work investigated the chemical composition and antimicrobial activity of the E. pavieana essential oils extracted from the rhizome (EOEP). We extracted the EOEP from the rhizome by hydrodistillation and analyzed the chemical composition by headspace solid-phase microextraction coupled with gas chromatography/mass spectrometry (HS-SPME-GC/MS). A total of 22 volatile compounds were identified where trans-anethole (78.54%) and estragole (19.36%) were the major components in the EOEP. The antimicrobial activity of EOEP was evaluated based on the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) values using the broth dilution method and enumerating cell death overtime. Our work shows that the EOEP exhibits potent antibacterial activity against foodborne pathogenic Gram-positive bacteria, namely Bacillus cereus, Staphylococcus aureus and Listeria monocytogenes in the range of 0.1-0.3% (v/v). We further investigated the mechanism of EOEP inhibition using Synchrotron Fourier transformation infrared (FTIR) microspectroscopy. Here, we show significant differences in DNA/nucleic acid, proteins and cell membrane composition in the bacterial cell. To conclude, EOEP exhibited antimicrobial activity against foodborne pathogens, especially the Gram-positive bacteria associated with ready-to-eat (RTE) food and, thus, has the potential to serve as a natural preservative agent in RTE products.

RESUMO

The utilization of biopolymers incorporated with antimicrobial agents is extremely interesting in the development of environmentally friendly functional materials for food packaging and other applications. In this study, the effect of calcium oxide (CaO) on the morphological, mechanical, thermal, and hydrophilic properties as well as the antimicrobial activity of carboxymethyl chitosan (CMCH) bio-composite films was investigated. The CMCH was synthesized from shrimp chitosan through carboxymethylation, whereas the CaO was synthesized via a co-precipitation method with polyethylene glycol as a stabilizer. The CMCH-CaO bio-composite films were prepared by the addition of synthesized CaO into the synthesized CMCH using a facile solution casting method. As confirmed by XRD and SEM, the synthesized CaO has a cubic shape, with an average crystalline size of 25.84 nm. The synthesized CaO exhibited excellent antimicrobial activity against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) (>99.9% R). The addition of CaO into CMCH improved the mechanical and hydrophobic properties of the CMCH-CaO films. However, it resulted in a slight decrease in thermal stability. Notably, the CMCH-CaO10% films exhibited exceptional antimicrobial activity against E. coli (98.8% R) and S. aureus (91.8% R). As a result, such bio-composite films can be applied as an active packaging material for fruit, vegetable, or meat products.

RESUMO

Several outbreaks of Listeria monocytogenes originated mainly from contaminated contact surfaces. Thus, this study investigated the antilisterial effect of natural surfactants in terms of their use as a 2-in-1 sanitizing washer on a food contact surface and evaluated their modes of action. The antilisterial activity of alkyl polyglycosides (APGs), namely capryl glucoside (CA), coco glucoside (CG), and decyl glucoside (DG), was evaluated based on the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) using broth dilution assay. The results showed that CG had the strongest antilisterial activity. Therefore, CG was selected for further investigation. The time-kill assay showed a lethal effect of 0.5 % (w/w) CG by inactivating 4 Log reduction (99.99 %) of L. monocytogenes within 3 s. Furthermore, 1 % (w/w) CG with slight mechanical force in washing (by shaking) was efficient for sanitizing a stainless-steel coupon surface based on its ability to cause a total reduction of deposited L. monocytogenes (99.9 %) within 10 min. Scanning electron microscopy and applying Fourier-transform infrared spectroscopy revealed that CG chemically disrupted the cell wall and plasma membrane of L. monocytogenes within 5 min after a gentle wash. The results showed it had potent antimicrobial activity and was bactericidal against L. monocytogenes. Overall, our results supported the use of CG as a natural antibacterial surfactant to alter the chemical sanitizer and the possibility of its practical use in the food industry and for household use.

RESUMO

Wolffia globosa is an interesting alternative plant-based protein source containing up to 40% protein dry weight. Dried duckweed protein extract (PE) was obtained using ultrasound-assisted extraction (UAE) before isoelectric precipitation (pH 3.5) to yield protein concentrate (PC) and protein solution (PS). The PC was hydrolyzed using Alcalase enzyme to obtain protein concentrate hydrolysate (PCH). Among all fractions, PCH exhibited antimicrobial properties by decreasing populations of Vibrio parahaemolyticus and Candida albicans at 0.43 ± 1.31 log reduction (66.21%) and 3.70 ± 0.11 log reduction (99.98%), respectively. The PE and PS also showed high solubilities at pH 8 of 90.49% and 86.84%, respectively. The PE demonstrated the highest emulsifying capacity (EC) (71.29%) at pH 4, while the highest emulsifying stability (ES) (~98%) was obtained from the PE and PS at pH 6 and pH 2, respectively. The major molecular weights (Mw) of the PE, PC, PCH and PS were observed at 25, 45, 63 and 100 kDa, with a decrease in the Mw of the PCH (<5 kDa). The PCH contained the highest total amino acids, with aspartic acid and glutamic acid being the major components. The results revealed the antimicrobial and functional properties of duckweed protein and hydrolysate for the first time and showed their potential for further development as functional food ingredients.

RESUMO

Seafood is a highly economical product worldwide. Primary modes of deterioration include autolysis, oxidation of protein and lipids, formation of biogenic amines and melanosis, and microbial deterioration. These post-harvest losses can be properly handled if the appropriate packaging technology has been applied. Therefore, it is necessary for packaging deterioration relevance to be clearly understood. This review demonstrates recent polymeric packaging technology for seafood products. Relationship between packaging and quality deterioration, including microbial growth and chemical and biochemical reactions, are discussed. Recent technology and trends in the development of seafood packaging are demonstrated by recent research articles and patents. Development of functional polymers for active packaging is the largest area for seafood applications. Intelligent packaging, modified atmosphere packaging, thermal insulator cartons, as well as the method of removing a fishy aroma have been widely developed and patented to solve the specific and comprehensive quality issues in seafood products. Many active antioxidant and antimicrobial compounds have been found and successfully incorporated with polymers to preserve the quality and monitor the fish freshness. A thermal insulator has also been developed for seafood packaging to preserve its freshness and avoid deterioration by microbial growth and enzymatic activity. Moreover, the enhanced biodegradable tray is also innovative as a single or bulk fish container for marketing and distribution. Accordingly, this review shows emerging polymeric packaging technology for seafood products and the relevance between packaging and seafood qualities.

RESUMO

Foodborne pathogens mostly contaminate ready-to-eat (RTE) meat products by post-process contamination and cause foodborne disease outbreaks. Preventing post-process contamination and controlling microbial growth during storage by packing the RTE meats with active antimicrobial film from chitosan combined with the synergism of Thai household essential oils was investigated. Here, we analyzed antimicrobial activity and mechanical properties of chitosan films incorporated with essential oil of fingerroot (EOF) and holy basil (EOH) based on their fractional inhibitory concentration and isobolograms. We showed that antimicrobial activities of chitosan film and chitosan films formulated with EOF:EOH displayed a dramatical reduction of Listeria monocytogenes Scott A concentration by 7 Log in 12 h. Chitosan film incorporated with EOF:EOH at ratio 0.04:0.04% v/v/w strongly retarded growth of total viable count of L. monocytogenes on vacuum-packed bologna slices during seven days of storage at 4 and 10 °C. Combined EOF and EOH added to chitosan films did not alter thickness, elongation (%) and colors (L*, a* and b*) of the chitosan film, but it increased water vapor transmission rate and decreased film tensile strength. Results suggested that chitosan film had strong antibacterial properties. Its effectiveness in inhibiting foodborne pathogenic bacteria in ready-to-eat meat products was enhanced by adding a combination of EOF:EOH.

RESUMO

The antibacterial mode of action of chitosan using real-time, horizontal attenuated total reflectance, Fourier-transform infrared (HATR-FTIR) spectroscopy and transmission electron microscopy (TEM) was investigated. Listeria innocua was treated with chitosan solution. HATR-FTIR revealed an increased lethality and substantially metabolomics response on cell components. The main changes in FTIR and their 2nd derivative spectra were at 1045 cm-1 (carbohydrates in cell wall). Principal component analysis clearly segregated untreated and treated cells. Loadings plot revealed the functional groups in cell wall, cell membrane, phospholipid and protein regions of spectrum that are responsible for the classification of treated and control spectra. Kinetic traces of the metabolomics change suggested that cell wall and cell membrane seemed to be the initial target of the antimicrobial mechanism of chitosan. In agreement with the TEM images, which showed breakage of cell wall integrity. The cell wall, cell membrane, phospholipids, proteins and nucleic acids of FTIR spectral data recorded during the cell inactivation were shown to be linked to the metabolomics cell response in the lethality rate and structure of the cells. This work clearly showed, using HATR-FTIR spectroscopy, how bacteria can change their metabolomics response substantially during the first 45 min of contact time.

RESUMO

Active films from rice starch/carboxymethyl chitosan (RS/CMCh) incorporated with propolis extract (ppl) were developed and characterized. The effect of the ppl content (0⁻10% w/w based on RS/CMCh) on the developed films' properties were determined by measuring the optical, mechanical, thermal, swelling, barrier, antimicrobial, and antioxidant attributes. The thermal stability and biodegradability of the films were also investigated. As the ppl content increased, free radical scavenging and a* and b* color values increased, whereas luminosity (L*) and swellability of the films decreased. The active films with 5⁻10% ppl possessed antimicrobial ability against Gram-positive bacteria (Staphylococcus aureus and Bacillus cereus). The active film with 10% ppl displayed increased flexibility and thermal stability, without a change in oxygen permeability. The results indicated that incorporation of ppl into RS/CMCh film could enhance the films' antioxidant and antimicrobial properties.
