RESUMO

The ovicidal and larvicidal effect of a full aqueous extract (FE) and two phases: an aqueous (Aq-Ph) and an ethyl acetate (EtOAc-Ph) from Ruta chalepensis (Rc) stems and leaves against Haemonchus contortus (Hc) were assessed. The egg hatching inhibition (EHI) assay and larval mortality (LM) test were performed by triplicate in 96-well micro-titration plates (n=4 wells). The FE against Hc eggs and larvae was assessed at 1.25, 2.5, 5, 10 and 20 mg/mL; and 30, 60, 90, 120,150 and 200 mg/ mL, respectively. The ovicidal effect of Aq-F and EtOAc-F was assessed at 1.25, 2.5, 5, 10 and 20 mg/mL. Plates were incubated at 28 °C for 48 (ovicidal assay) and 72 h (larvicidal assay). The EHI results were considered based on the mean number of eggs hatching failure after 48 h exposure. The LM was recorded after 72 h exposure to the Rc phases and expressed as mortality percentage. The Rc FE caused 96 and 100% EHI at 10 and 20 mg/mL, respectively; meanwhile, 74% LM was recorded at 200 mg/mL (p<0.05). The Aq-Ph showed 78.5% EHI at 2.5 mg/mL. Likewise, the EtOAc-Ph caused 100% EHI in almost all concentrations. Eighteen compounds including alkaloids, cumarins, triterpens, flavonoids, tannins, saponins and sterols were identified by GC-MS analysis. The results indicate that the aqueous extract from Rc possesses bioactive compounds with in vitro nematocidal activity against Hc; mainly in the EtOAc-Ph. Further studies should be performed to elucidate those compounds searching for alternative methods of control of the sheep haemonchosis.

Ante-mortem studies of stress in rabbits are few and can potentially impact the haematologic response and physicochemical parameters of muscle meat. The aim of this study was to investigate the induction of stress by transport and simulation at two different times. Two hundred rabbits divided in a similar proportion of male and female were stressed by vehicular transportation and with motion simulation, using a vibration platform. They were divided into five groups: Control (without stress), Stress with Short Transportation (30 min), Stress with Long Transportation (60 min), Stress with Short Vibration Platform (30 min) and Stress with Long Vibration Platform (60 min). Transportation caused a decrease in the haematic potassium in both sexes (<5.2 mmol/L). The glucose and lactate concentrations increased in the transported rabbits (>219 and >61 mg/dl respectively). The differences between sexes showed that the P CO2 in males decreased <8.5 mmHg in stress groups compared to without stress group. Glucose concentration was 61.5 mg/dl for females and 38.0 mg/dl for males (p < .01). We concluded that in rabbits induced stress by transport increased the energetic metabolites in the blood tissue. This increase did not affect the production parameters and the quality of muscle meat.

The effects of exogenous fibrolytic enzymes (EFE; a mixture of two preparations from Trichoderma spp., with predominant xylanase and ß-glucanase activities, respectively) on colonization and digestion of ground barley straw and alfalfa hay by Fibrobacter succinogenes S85 and Ruminococcus flavefaciens FD1 were studied in vitro. The two levels (28 and 280 µg/ml) of EFE tested and both bacteria were effective at digesting NDF of hay and straw. With both substrates, more NDF hydrolysis (p<0.01) was achieved with EFE alone at 280 than at 28 µg/ml. A synergistic effect (p<0.01) of F. succinogenes S85 and EFE on straw digestion was observed at 28 but not 280 µg/ml of EFE. Strain R. flavefaciens FD1 digested more (p<0.01) hay and straw with higher EFE than with lower or no EFE, but the effect was additive rather than synergistic. Included in the incubation medium, EFE showed potential to improve fibre digestion by cellulolytic ruminal bacteria. In a second batch culture experiment using mixed rumen microbes, DM disappearance (DMD), gas production and incorporation of (15)N into particle-associated microbial N ((15)N-PAMN) were higher (p<0.001) with ammoniated (5% w/w; AS) than with native (S) ground barley straw. Application of EFE to the straws increased (p<0.001) DMD and gas production at 4 and 12 h, but not at 48 h of the incubation. EFE applied onto S increased (p<0.01) (15)N-PAMN at 4 h only, but EFE on AS increased (p<0.001) (15)N-PAMN at all time points. Prehydrolysis increased (p<0.01) DMD from both S and AS at 4 and 12 h, but reduced (p<0.01) (15)N-PAMN in the early stage (4 h) of the incubation, as compared to non-prehydrolyzed samples. Application of EFE to barley straw increased rumen bacterial colonization of the substrate, but excessive hydrolytic action of EFE prior to incubation decreased it.

This study was conducted to develop predictive equations for carcass characteristics and primal cut weights of native Mexican guajolotes using body measurements (BM). For this study, a total of 36 male guajolotes (Meleagris gallopavogallopavo), aged 6 to 10 months, and mean slaughter body weight (SBW) of 4543.14 ± 656.60 g, were used. The birds were kept under traditional extensive conditions. ThefollowingBMswererecorded24 h before slaughter: thoracicperimeter (TP), body circumference (BC), body length (BL), wing length (WL), keel length (KL), shank length (SL) and shank diameter (SD). After slaughter, hot carcass weight (HCW), cold carcass weight (CCW), hot dressing percentage (HDP), cold dressing percentage (CDP), organs and viscera weight (VIS) and abdominal fat weight (AFW) were recorded. The carcasses were dissected in to five primal cut (breast, thigh, drumstick, back and wing). The SBW and BMs showed moderate to high positive correlations (p<0.01; 0.34≤r<0.97) with carcass characteristics and primal cut weights. In the equations generated to predict HCW, CCW, HDP, CDP, VIS and AFW, the R2 ranged from 0.40 to 0.96, and the predictor variables were SBW, KL, BC, WL and SL. Regarding the equations developed to predict the primal cut weights, R2 ranged from 0.58 to 0.91. In these models, SBW, BC, SD, WL and KL explained most of the observed variation. The prediction equations obtained in the study had moderate to high accuracy; therefore, they can be used by researchers, technicians and poultry producers to obtain information on the carcass composition of native Mexican guajolotes.(AU)

The causes of mortality in kids under extensive conditions in the Mexico plateau were studied. Perinatal mortality of kids (0-7 days) was lower (P<0.05) than mortality in the other age groups (8-90 days). The weights of the animals at death were extremely low, group (i) 0-7 days, 2.3kg; (ii) 7-30 days, 2.8kg and (iii) 30-90 days, 4kg. The principal finding at necropsy was white muscle disease (WMD). The WMD lesions, suggested selenium (Se) deficiency was present in 49 out of the 74 kids evaluated in this study (P<0.05). In many cases, muscular dystrophy was present with other pathological conditions. Enteric and pneumonic conditions were also important causes. In the small intestine villus atrophy was observed in 14 of the 25 kids that died with enteric problems between 8 and 30 days of age. Pneumonic lesions were present in 12 out of 74 kids. Miscellaneous conditions as starvation (five cases), hepatic conditions (three cases), omphalophlebitic infection (two cases) were also present in lower proportions. In conclusion, WMD was considered the main cause of deaths in goat kids between 8 and 90 days of age. Evaluation of Se status of soil, forages and goat tissues is recommended.

The objective of this study was to determine the concentration of Selenium (Se) in soils, forages and the tissues of kids and does under extensive grazing conditions in two regions of the Tlaxcala state, Mexico (Carrillo Puerto and Ixtenco) in the dry and rainy season. There were no differences in the concentration of Se in soil (0.051 and 0.047ppm), pH of the soil (6.1 and 5.9), concentration of Se in the forage (0.052 and 0.075ppm) and blood serum of goats (0.02 and 0.021ppm) during the dry and rainy seasons, respectively. The Carrillo Puerto region had a significantly higher content of Se in the soil (31%), soil pH (7%), content of Se in forage (25.9%), and content of Se in blood serum of goats (16%) compared to the corresponding values obtained in Ixtenco. The mean concentration of Se in the serum of kids with clinical signs of white muscle disease, was significantly lower (36.3%) compared to the kids from the same farm that were apparently healthy. The results of this study suggest that soil, forages, and goats have a marginal Se status in both regions during the dry and rainy seasons. Further research is required to find the most appropriate method to correct the Se-deficiency in goats from these regions.

Twenty hair (Pelibuey x Katadin x BlackBelly) and 20 wool (Rambouillet x Criollo) lambs were fattened in an intensive feeding system to reach 40kg body weight at ten months of age. The yield of lamb sides and back fat (51.9 percent, 2.5mm vs. 47.0 percent, 2.3mm) were greater in wool lambs. Meat pH was not different between phenotypes but meat temperature was higher during the first hours postmortem in wool lamb sides (27, 23, and 20, vs. 24, 20, and 17ºC). Protein content in meat from the neck (braquicephalic and esternocephalic muscles) and loin (Longissimus lumborum muscle) were higher in hair lambs (21.1 and 20.4 vs. 19.7 and 19.3 percent), while moisture of the same muscles was higher in wool lambs (75.6 and 75.9 vs. 73.6 and 74.3 percent). Ash content was similar in both phenotypes, but palmitoleic acid was greater in hair lambs (2.9 and 2.7 vs. 2.7 and 2.5 percent). In fresh meat, pH, meat toughness, and water activity (Wa) of loin did not differ; but water holding capacity (WHC) was higher in hair lambs, and the same response was observed for cooked meat toughness from the neck, and in raw and cooked meat from loin. There were differences in the colour of meat from the neck, having values for L*, b* and tonalities greater in wool lambs, and a* greater for hair lambs. Sensorial parameters were similar in both phenotypes.(AU)

O rendimento de carcaça e a espessura da gordura foram maiores nos cordeiros lanados (51,9 por cento, 2,5mm vs. 47,0 por cento, 2,3mm). Não houve diferenças entre os dois grupos de cordeiros para pH, mas a temperatura da carne foi mais alta durante as primeiras horas pós-morte, nas meias-carcaças dos cordeiros lanados (27, 23, 20 vs. 24, 20, 17ºC). O conteúdo proteico da carne do pescoço (músculos braquicefálico e esternocefálico) e do lombo (músculo Longissimus lumborum) foi maior nos cordeiros deslanados (21,1 e 20,4 vs. 19,7 e 19,3 por cento) enquanto o teor de umidade foi maior nas lanados (75,6 e 75,9 vs 73,6 e 74,3 por cento). O conteúdo de cinzas não diferiu entre os dois grupos de cordeiros, mas o ácido palmitoleico foi maior nos cordeiros deslanados (2,9 e 2,7 vs 2,7 e 2,5 por cento). Three experiments were carried out to determine the optimum selenium-vitamin E injection level to maintain acceptable blood selenium (Se) status of does and kids, as well as to determine the relation of that status to mortality rates in kids. In experiment 1, 238 goats were assigned to one of three groups during the mating period: A1-control, A2- 0.06mgSe+0.8IU vitE/kgBW and A3-0.125mgSe+1.7IU vitE/kgBW. No differences (P>.05) for fertility and prolificacy were observed among the groups. Blood Se concentration did not differ among Se-vit E groups and control group before injection, and both groups showed Se deficient condition. There was a trend (P<.05, 32%) to increase Se blood level 60 days post-treatment, but difference was not observed between A2 and control groups, while difference (P<.05; 103%) was observed between A1 and A2 groups vs high Se injection (A3 group). In experiment 2, 48 goats were divided into four groups: B1- control, B2- 0.125mgSe+1.7IU vitE/kgBW, B3- 0.25mgSe+3.4IU vitE/kgBW and B4- 0.31mgSe+4.2IU vitE/kgBW. The B4 group reached the highest concentration at the third month after injection (0.11 ppm), then started to decline after 100 days, reaching a value slightly higher than B2 and B3 on the 135th day of pregnancy. Results of B2 and B3 groups were slightly higher that those of B1 (P<0.05). In experiment 3, 194 kids (3 to 7 days postpartum) born from Se-deficient goats were used to compare the effectiveness of Se injection. They were divided into three groups: C1- control, C2- 0.3mgSe+4.2IU vit E/kg BW and C3- 0.6mgSe+8.4IU vit E/kg BW. C1 showed the highest percentage of mortality (60%) as compared to treated Se groups, that scored equal percentage of deaths (22% averaged). The concentration of Se in blood, on day 20th after the onset of the treatment rapidly increased, according with level of Se injection.(AU)

O grupo B4 alcançou a concentração mais alta no terceiro mês depois da injeção (0,11ppm) e diminuiu depois de 100 dias, quando o valor foi apenas ligeiramente maior que os valores dos grupos B2 e B3, aos 135 dias da gestação. Os resultados dos grupos B2 e B3 foram ligeiramente mais altos que os do grupo B1 (P<0,05). No terceiro experimento, 194 cabritos (3 a 7 dias de idade), nascidos de cabras deficientes em Se, foram usados para comparar a efetividade da injeção de Se. Foram divididos em três grupos: C1- controle, C2- 0,3mgSe+4,2UI vit E/kg de peso-vivo e C3- 0,6mgSe+8,4UI vit E/kg de peso-vivo.A porcentagem de mortalidade foi mais alta em C1 (60%) e mais baixa nos grupos tratados com Se (22%).A concentração de Se no sangue no 20º após o início do tratamento aumentou rapidamente, de acordo com o nível de injeção de Se.(AU)
