RESUMO

Groups of approximately 20 one-day-old chickens were inoculated with G-4260, the reference strain of avian nephritis virus (ANV), or saline. Based on mortality rates from severe nephritis in comparable experiments, light Sussex chickens generally were more susceptible than Rhode Island red (RIR) chickens. Mortality was greater in those given broiler starter than those given other feeds, and was greater when light Sussex chickens were given broiler starter feed and cold-stressed at 15 +/- 1 C for 2 hr daily during the first week rather than brooded normally. Inoculation with G-4260 either orally or by intraperitoneal injection produced similar results in RIR chickens. Thirty-three inoculated chickens died of severe nephritis between 4 and 12 days postinoculation, and 24 (73%) of them had visceral urate deposits. Inoculated inbred white leghorn Line 15 chickens with maternal antibody to ANV were brooded normally and given broiler feed: they were susceptible to infection as evidenced by subsequent histological lesions in the kidneys and serology, but mortality was not a feature. There were no deaths from nephritis in inoculated non-inbred white leghorn chickens free of maternal antibody to ANV that were given broiler feed and brooded normally. These results have implications in standardizing experimental conditions for the study of mortality induced by G-4260 and similar viruses.

RESUMO

This report describes inclusion body hepatitis in a 12-week-old leghorn-type cockerel. The cockerel had come from a specific-pathogen-free flock and was reared in a positive-pressure isolator until 6 weeks old. Grossly, the cockerel was jaundiced, and the liver was swollen and had small white foci throughout. Histologically, there were foci of hepatic necrosis and basophilic inclusion bodies in the nuclei of numerous hepatocytes. A fowl adenovirus, isolated from the liver, was found to belong to serotype 2/12. The bursa of Fabricius had some degenerative changes, but these were not consistent with infectious bursal disease, nor was there evidence of seroconversion to infectious bursal disease virus in the remaining chickens.

This series of case reports details observations on toxicity episodes in poultry due to a variety of chemotherapeutic agents. These problems arose owing to overdosage, variation in species susceptibility, potentiation of the toxic effects of one substance by the presence of another substance, and particular disease or other on-farm factors. Ignorance and accident were responsible for some of these situations. The episodes involved monensin, salinomycin, nicarbazin, sulphaquinoxaline, dinitolmide, dimetridazole, nitrofurans, streptomycin, and 3-nitro-4-hydroxyphenylarsonic acid.

The role of selenium deficiency in the etiology of the runting-stunting syndrome (RSS) of broiler chickens in Australia was investigated. Commercial broiler chickens maintained on selenium-deficient developed signs consistent with selenium deficiency of exudative diathesis and markedly reduced plasma glutathione peroxidase activity, but they did not develop pancreatic atrophy and fibrosis or elevated plasma amylase activity, which are the other lesions associated with RSS. Supplementation of the diets of birds from a RSS-susceptible flock with a mixture of selenium, vitamin E, cysteine, and sulfate had no effect on the incidence of runting in the treated birds. In field outbreaks of RSS there were no observable differences between affected and unaffected birds in the concentration of selenium in tissue samples. Furthermore, evidence is presented which suggests that in cases of RSS, pancreatic atrophy and elevations in plasma amylase precede reductions in plasma glutathione peroxidase activity.

In November of 1997 an outbreak of highly pathogenic avian influenza occurred near the town of Tamworth, in northern New South Wales, Australia. The viruses isolated from chickens on two commercial chicken farms were identified as H7N4 viruses, with hemagglutinin cleavage site amino acid sequences of RKRKRG and intravenous pathogenicity indices of 2.52 and 2.90, respectively. A virus with an identical nucleotide sequence, but with an intravenous pathogenicity index of 1.30, was also isolated from cloacal swabs collected from asymptomatic emus kept on a third property.

An ascites syndrome was induced in 17 to 28 per cent of specific pathogen-free (SPF) Light Sussex (LSX) chickens given a low protein (16 per cent crude protein) high calcium (3.5 per cent calcium) layer crumble feed on two separate occasions 6 months apart. Affected chickens had increased right ventricular weight as a proportion of either total heart weight or live-weight at 3 weeks of age, compared with non-affected LSX chickens on the same feed, thus indicating right ventricular hypertrophy. The incidence of ascites was not increased by infection with avian nephritis virus, nor by limited cold-stress during brooding. It was not produced in LSX chickens given other feeds, nor in SPF Rhode Island Red chickens.

Mortality rates in 11 inbred and partially inbred chicken lines inoculated with a very virulent strain (CS89) of infectious bursal disease virus (IBDV) varied considerably, being highest (almost 80%) in a Brown Leghorn line (BrL). Bursa of Fabricius to body weight ratios were depressed in the survivors in each line, but no differences were observed between lines. However, histological examination of bursae from survivors showed that, although bursal damage occurred in every line, it was most severe in the two lines (BrL and White Leghorn W1) in which the highest mortality was recorded. Experiments with F1 matings between highly susceptible and highly resistant lines showed that resistance was partially dominant and that there were no maternal effects. Experiments using F2 and backcross chicks suggested the involvement of a single gene and indicated no involvement of the MHC. There was considerable variation between lines in IBDV-specific antibody, measured by ELISA, both in the vaccinated parent hens and in the amounts of inherited maternal antibody and its rate of decay in the progeny.

Details of cases involving the inadvertent exposure of birds to eight toxic substances are recorded. The organophosphate insecticides dichlorvos, diazinon and malathion produced respiratory symptoms which in the former and latter cases were initially thought to be caused by infectious disease. Birds which consumed feed containing fenitrothion showed nervous signs before death. On three separate occasions feral starlings (Sternus vulgaris) were found dead and their gizzard contents contained mevinphos. The rodenticide warfarin was associated with petechial haemorrhages in the skeletal muscles and on the serosal surfaces of one hen. Cyanogenic glycosides from Eucalyptus cladocalyx were responsible for the sudden deaths of ducks and guinea fowl. 'Ornamental dough' containing sodium chloride was fed to birds which were deprived of water and they showed diarrhoea and nervous disorders before death.

A mortality survey was carried out on three flocks of broiler chickens on a Victorian grower farm. A total of 61,400 chicks were placed and 2883 (4.7 per cent) died or were culled during rearing. Only 1028 (36 per cent) dead and culled birds were available for autopsy and interpretation was based on figures extrapolated from the findings. The most common condition encountered was death in good condition associated with congestion of the lungs (39 per cent of chickens autopsied). Inclusion body hepatitis was diagnosed in 30 per cent of chickens over 21 days of age which died. Mortality during the first 10 days was 1.1 per cent of chicks placed. The incidence of various other conditions is presented with relevant observations and comments.

The following unusual diseases were diagnosed in birds submitted to the Veterinary Research Institute, Victoria, between 1978 and 1987: the viral diseases beak and feather disease of psittacines, infectious laryngotracheitis in peafowls, a papovavirus-like inclusion body disease in psittacines, and pox; chlamydiosis; the bacterial diseases actinomycosis, listeriosis and mycobacteriosis; the fungal diseases favus, yeast infections and systemic zygomycosis; the protozoan diseases cryptosporidiosis, hexamitiasis, suspected leucocytozoonosis, sarcosporidiosis, toxoplasmosis, trichomoniasis and an unidentified protozoan-like organism which caused pneumonia in ducks; a variety of parasites; the metabolic disorders curled-toe paralysis in pheasant poults, encephalomalacia and parenchymatous goitre; toxicity due to dimetridazole and the ingestion of the leaves of the tobacco tree; and other non-infectious conditions including asphyxiation, burns, cataracts, cerebellar degeneration and atrophy, cystic right oviducts and exertional rhabdomyolysis.

RESUMO

Observations on a runting and stunting syndrome in broiler chickens in Victoria, Australia, based on general observations from 1980 to 1983 on 2244 chickens from 109 affected broiler chicken flocks, are summarised. The details on 156 of these birds from five affected flocks with varying runting percentages are presented. Typically affected birds were presented with atrophy of the pancreas, the thymus and the bursa of Fabricius.

A number of plasma biochemical parameters were examined in five outbreaks of runting in broiler chickens. In four of the five outbreaks, runts showed consistent elevations in plasma amylase activity and reductions in glutathione peroxidase activity. In two of the five outbreaks the plasma vitamin E concentration was reduced, as was the activity of plasma alkaline phosphatase. A highly significant number of runted chickens were found to have pancreatic degeneration, elevated plasma amylase activity and reduced plasma glutathione peroxidase activity, compared with non-runted chickens. The implications of these changes are discussed in relation to the aetiology of runting and stunting syndrome and, in particular, the possible involvement of selenium.

Observations were made on the long bones of 16 young ratite birds (10 rheas, 3 emus and 3 ostriches), from 1-day-old to 12 weeks of age. At hatching all long bones contained large cartilaginous cones which were continuous with the growth plates, and an osseous cortex. At one week of age ossification had commenced on the periphery of these embryonic cones and in some bones the cones had become separated from the growth plates. At 3 weeks of age the embryonic cones of cartilage were still present in the proximal and distal tibio-tarsi and narrow cartilaginous bridges connected the cones to the growth plates. Embryonic cones were not present in other long bones of this 3-week-old rhea nor in the long bones of ratites 6, 8 and 12 weeks of age. Other praecocial birds such as turkey poults and chickens have cones of embryonic cartilage in their long bones at hatching and these persist in the tibio-tarsi till one to 2 weeks of age. The presence of large cones of embryonic cartilage in the tibio-tarsal bones of 3-week-old ratite birds is probably a normal phenomenon. Awareness of this feature is necessary for the correct differential diagnosis of the prevalent musculoskeletal disorders of ratite birds.

Utility pigeons on 3 farms were affected by a severe neurological disturbance manifested by fine tremors, rolling gait and incoordination. Affected birds had necrosis of the Purkinje cells of the cerebellar cortex. The birds from the 3 farms had been fed a pelleted diet obtained from a single mix of feed prepared by a stockfeed manufacturer. When feed obtained from the farms was given to utility pigeons under controlled conditions at the laboratory, they suffered the same clinical signs and lesions as seen in birds from the farms. Cross-bred cockerels were not affected when given this feed from day-old to 6 weeks of age. Samples of the feed contained 185 to 226 ppm of the coccidiostat dinitolmide. Utility pigeons given a prepared diet containing a similar level (234 ppm) of dinitolmide developed an identical condition after 3 to 4 weeks feeding. Laying hens were not affected by this feed. Dimetridazole, which was also present in some of the original feed samples, did not exacerbate the condition when included in the diet at a rate of 233 ppm. Production of squabs from the affected farms was dramatically reduced, and many of the breeder had to be culled because of the long-standing neurological disease.

The resistance to anti-microbial agents of bacteria isolated from pathological conditions of birds in Victoria, 1978 to 1983, was determined for isolates of Escherichia coli, Salmonella species, Staphylococcus aureus, Pasteurella multocida, P. anatipestifer, Yersinia pseudotuberculosis and Haemophilus paragallinarum. The isolates of E. coli had a high prevalence of resistance to tetracycline and sulphonamides, and a lower prevalence of resistance to furazolidone and sulphamethoxazole-trimethoprim. The isolates of Salmonella spp commonly had resistance to tetracycline, sulphonamides, furazolidone and sulphamethoxazole-trimethoprim. Almost half the isolates of S. aureus showed resistance to lincomysin and many showed resistance to penicillin. Resistance to tetracycline was found in isolates of P. multocida, P. anatepestifer and Y. pseudotuberculosis. Some isolates of H. paragallinarum showed resistance to sulphonamides, streptomycin and sulphamethoxazole-trimethoprim.

Between 1967 and 1977 a study was made of diseases of cattle in the Solomon Islands. Tuberculosis was found in only 3 herds and was eradicated by 1975. Brucellosis serology revealed very few reactors and by 1977 the herds involved were considered free of the disease. Significant serological reactions were found to Leptospira interrogans serovars pomona, hardjo, autumnalis and jez-bratislava. There was evidence that infectious bovine rhinotracheitis and mucosal disease were present. Seventeen parasites were identified of which Haematobia irritans exigua, Haemonchus placei, Oesophagostomum radiatum and Ceylonocotyle streptocoelium were widely distributed. Nutritional stress occurred under some forms of husbandry but environmental stress was minimal. The Solomon Islands are therefore in a most favourable situation with regard to diseases of cattle.

RESUMO

An infectious bursal disease vaccine, registered for use in breeder flocks, was studied for efficacy on the day-old offspring of vaccinated hens and for virulence in susceptible day-old and 6-week-old chickens. When given to susceptible day-old chicks and 6-week-old cockerels, the vaccine was found to induce atrophy and pathology of the bursa of Fabricius similar to that observed in field infections. Chicks vaccinated at day-old had markedly lowered titres in the haemagglutination inhibition test to Newcastle disease virus, when this was given 2 weeks later, but the response of the 6-week-old cockerel was similar to that of control birds. Maternal antibody induced by the vaccine protected chicks against infection at day-old.
